BACKGROUND Mast cell hyperplasia can present as systemic mastocytosis (SM) or reactive mast cell hyperplasia. Distinguishing between these 2 diseases is a critical challenge. Here, we report a rare case of concurrent mast cell hyperplasia, multiple myeloma (MM), and low-level myeloid blastocytosis. The potential mechanisms underlying the coexistence of the aforementioned 3 diseases were also analyzed. CASE REPORT A patient with muscle pain was initially diagnosed with monoclonal gammopathy of undetermined significance (MGUS). Three years later, she progressively developed pancytopenia and rena

Lipoid lesions of the thyroid are extremely rare. Adipose changes can be seen in both malignant (carcinomas) and benign conditions. The two most common benign entities are thyrolipoma and thyrolipomatosis, which have been reportedly associated with systemic amyloidosis. Radiologically, these lesions can mimic malignancy, especially when presenting as a biochemically active condition. We present a rare case of concurrent thyroid lipomatosis and serum amyloid A amyloidosis (AA) manifesting as goiter and subclinical hyperthyroidism. A 72-year-old male presented to Danbury Hospital with complaints

Multiple myeloma (MM) is the second most common hematological malignancy, characterized by a clonal expansion of malignant plasma cells in bone marrow. Monoclonal gammopathy of undetermined significance (MGUS) is the premalignant condition of MM. The tumor microenvironment is thought to influence the progression from premalignant conditions. Myeloid-derived suppressor cells (MDSCs) are a heterogenous group of different cellular subsets with myeloid origin, characterized by their ability to inhibit T-cell responses. MDSC are thought to play an important immunoregulatory role in different diseas

BACKGROUND: Monoclonal gammopathy of undetermined significance (MGUS) is a premalignant plasma cell disorder whose prevalence increases with age and may progress to multiple myeloma. The tumor microenvironment plays a crucial role in MGUS pathogenesis and progression. Systemic immune-inflammatory markers can reflect tumor microenvironment(TME) dynamics, but their association with MGUS odds remains understudied.
METHODS: This cross-sectional study analyzed data from 12,080 adults aged ≥ 50 years (including 350 MGUS cases) in the National Health and Nutrition Examination Survey (NHANES III and

Monoclonal gammopathy of undetermined significance (MGUS) is a necessary precursor of multiple myeloma (MM). Compared with non-Hispanic White (White) patients, African American individuals experience approximately twofold greater, and Asian American individuals 50% lower, occurrence of MGUS and MM. We examined determinants of MGUS among Japanese American, African American, Latino, Native Hawaiian, and White participants in a case-control study nested in the Multiethnic Cohort (MEC) study. Presumptive cases included MEC participants aged ≥65 years initially screened using the Medicare billing c

Multiple myeloma (MM) is a haematological malignancy characterised by clonal plasma cell proliferation principally in the bone marrow. Up to 80% of individuals with MM experience osteolytic bone disease, characterised by an increased risk of pathological fractures and significant bone pain impacting quality of life. MM is preceded by monoclonal gammopathy of undetermined significance (MGUS) and smouldering multiple myeloma (SMM), both of which are usually asymptomatic. Although patients with these precursor conditions do not present with detectable osteolytic bone disease and generally are not

Immunoglobulin G4-related disease (IgG4-RD) is an immune-mediated chronic fibrotic inflammatory disorder. The coexistence of IgG4-RD with monoclonal gammopathy of undetermined significance (MGUS) is relatively rare, with only a few cases reported in the literature. This article reports a case of a 79-year-old male patient with clinical manifestations of persistent upper abdominal pain of unknown cause and recurrent pleural effusion. Auxiliary examinations indicated positive serum antinuclear antibodies and persistently positive IgG4; three-dimensional imaging reconstruction showed mild pancrea

Monoclonal gammopathy of undetermined significance (MGUS) is a common plasma cell disorder with well described risks of progression to myeloma and lymphoplasmacytic lymphoma. Using data from an established UK population-based cohort of hematological malignancies and premalignancies, we investigated patient and disease characteristics, subsequent hematological malignancy, and survival in 4,651 people diagnosed with MGUS between 2005 and 2019. The 5-year net (relative) survival (disease-specific estimate of the probability of survival) for MGUS patients was 87.8% (95% confidence interval [CI]: 8

Monoclonal gammopathy of undetermined significance (MGUS) is a premalignant plasma cell disorder with a risk of progression to multiple myeloma. Metabolic syndrome (MetS) is implicated in cancer development, yet its association with MGUS remains unclear. We examined MetS as a risk factor for MGUS in a large Korean cohort. In a retrospective cohort study using the National Health Information Database, we analyzed 4,453,504 adults undergoing health checkups in 2012, followed through up to 2022. MetS was defined by NCEP-ATP III criteria. Over a median 9.3-year follow-up, 1,241 MGUS cases were ide

BACKGROUND: Monoclonal gammopathy of undetermined significance (MGUS) is a precursor to multiple myeloma (MM), but the mechanisms of progression remain unclear.
METHODS AND OBJECTIVES: Transcriptomic datasets procured from the Gene Expression Omnibus (GEO) underwent thorough analysis to ascertain disease-related modules using weighted gene co-expression network analysis. A prognostic model (MGUSscore) was constructed via least absolute shrinkage and selection operator (LASSO) regression within the GSE136337 cohort and validated across independent datasets (The Cancer Genome Atlas - multiple m

BACKGROUND: Multiple myeloma (MM) health disparities are well-documented, with non-Hispanic Black (NHB) and male individuals experiencing higher disease burdens than their non-Hispanic White (NHW) or female counterparts. However, no studies have shown that how these disparities translate into differences in life expectancy, particularly for monoclonal gammopathy of undetermined significance (MGUS), a precursor to MM. This study quantified the remaining life years and life years lost associated with MGUS and MM to inform MM prevention and control priorities.
METHODS: We developed a discrete-ev

Monoclonal gammopathy of undetermined significance (MGUS) represents a premalignant clinical state with an evolving footprint beyond its established role as a precursor to multiple myeloma. This review is intended for hematologists, oncologists, and translational investigators involved in MGUS risk stratification and prevention research and confronts four unresolved controversies shaping modern MGUS management: (1) The debate surrounding population screening-weighing potential benefits against economic/psychological burdens; (2) Limitations of current risk-stratification models and the promise

Genome-wide association studies have linked major histocompatibility complex (MHC) region loci to immune-related diseases, but their role in multiple myeloma (MM) progression from monoclonal gammopathy of undetermined significance (MGUS) remains unclear. This case-control study included 238 individuals with MGUS (> 5 years follow-up without MM progression) and 618 MM patients from MCW Froedtert Hospital (2011-2020). Using multivariable logistic regression, Omnibus test and conditional haplotype tests, we identified HLA alleles and amino acid positions associated with MM risk compared to MGUS.

OBJECTIVE: Current evidence regarding the association between obesity and monoclonal gammopathy of undetermined significance (MGUS) remains inconsistent. This study aims to evaluate the relationship between objectively measured obesity markers and prevalent MGUS using nationally representative data from the U.S.
METHOD: Data came from the third National Health and Nutrition Examination Survey III (1988-1994) and continuous NHANES (1999-2004). We estimated multivariable-adjusted odds ratios (aORs) and 95 % confidence intervals (CIs) for the association between MGUS and seven obesity markers (i

Eosinophilic fasciitis (EF) is a rare sclerosing disorder characterized by limb edema evolving to woody induration with peau d'orange texture and the groove sign. Diagnosis typically requires a full-thickness fascial biopsy, while magnetic resonance imaging (MRI) can noninvasively demonstrate fascial thickening and guide biopsy. EF has recognized associations with hematologic conditions, including monoclonal gammopathy. A previously healthy 64-year-old woman developed progressive lower extremity hyperpigmentation, edema, and painful induration after travel. Over months, she experienced forearm

Monoclonal gammopathy of undetermined significance (MGUS) is an asymptomatic pre-cancerous condition that precedes plasma cell dyscrasias, including multiple myeloma (MM). Current clinical guidelines report that MGUS's rate of malignant progression to haematological malignancy (HM) is ~1% per year; however, reported rates have varied widely. To address this discrepancy, a systematic review was conducted, investigating MGUS's rate of malignant progression to HM and MM alone. Four electronic databases were searched from inception to 28 March 2024 for articles reporting these outcomes; articles w

Monoclonal gammopathy of undetermined significance (MGUS) is a necessary precursor condition to multiple myeloma (MM). Given the role of autophagy in modulating MM risk, we investigated whether genetic variation in autophagy-related genes influences susceptibility to MGUS. We analyzed the association of 34,042 common autophagy-related single nucleotide polymorphisms (SNPs) with MGUS across six independent cohorts, five from Europe and one from North America, comprising 2317 MGUS cases and 282,358 controls. We also assessed their impact on immune parameters, including absolute counts of 91 bloo

Multiple myeloma (MM) is the most common plasma cell dyscrasia in the United States with tremendously high burden. MM is preceded by a premalignant condition, monoclonal gammopathy of undetermined significance (MGUS). Although several risk factors for progression have been identified (e.g., older age, male sex, black race, obesity, chemical exposure), their relative contributions remain unclear. Unlike other malignancies, such evidence is lacking for MM despite its high burden. To identify potential intervention strategies that can effectively prevent progression in patients with MGUS, we quan
