Low-selenium diet, bone, and articular cartilage in rats.

The pathophysiology of secondary osteoarthritis remains largely obscure. Our attention has been drawn to Kashin-Beck disease (KBD), which has been attributed to Se deficiency. To obtain information regarding the prevention, prediction of progression, and treatment of this condition, we performed histological and biochemical studies on bone and articular cartilage specimens obtained from rats fed a low-Se diet. A low-Se diet was prepared and fed to Wistar rats for 3-11 mo, after which the rats were killed under general anesthesia, and their articular cartilages were studied microscopically and

Age-related distribution of elastic fibers in the rabbit knee.

The distribution and histochemical properties of elastic fibers in the knee of newborn, young, adult, and old New Zealand White rabbits were studied. In the growing rabbit, elastic fibers, especially oxytalan fibers, are abundant in the perichondrium and the fibrous layer of the periosteum. They are homogeneously distributed in tendons, ligaments, and menisci, and are absent in the epiphyseal cartilage and growth plate and at the enthesis. In mature rabbits, the articular capsule, the synovial membrane, the peritenon, the periosteum, the perimysium, and the perivascular connective tissue are r

Preoperative staging of osteosarcoma: efficacy of MR imaging in detecting joint involvement.

OBJECTIVE: Selecting the appropriate surgical procedure for treatment of patients with osteosarcoma requires accurate preoperative evaluation of tumor extent. Establishing the presence or absence of joint involvement is particularly important. Accordingly, we studied the efficacy of MR imaging for detecting joint involvement in 46 patients with osteosarcoma around joint spaces. SUBJECTS AND METHODS: Preoperative MR examinations were performed in 46 consecutive patients with osteosarcoma whose tumors were located around the knee (n = 33), the hip (n = 8), or the shoulder (n = 5). T2-weighted a

The presence of lysylpyridinoline in the hypertrophic cartilage of newly hatched chicks.

The presence of lysylpyridinoline (LP) as a nonreducible cross-link in appreciable quantities has primarily been limited to the mineralized tissues, bone and dentin. However, the results reported here show that LP is not only present in the hypertrophic cartilage of the tibiotarsus isolated from newly hatched broiler chicks, but it is approx. 4-fold as concentrated as hydroxylysylpyridinoline (HP). Bone and articular cartilage surrounding the hypertrophic cartilage do not contain measurable quantities of LP. Purified LP has a fluorescent scan similar to purified HP and literature values, confi

[Assay of cross-linking molecules of collagen (pyridinolines) in the study of the degradation of bone tissue and articular cartilage].

Recent studies aimed at studying the degradation of the fibrillar collagens of bone and articular cartilage have lead to the discovery of naturally fluorescent crosslinking compounds derived from lysine and hydroxylysine. These small molecules characterized by an aromatic 3-hydroxypyridinium ring are present in significant amounts only in bone (pyridinoline and deoxy-pyridinoline) and cartilage (pyridinoline). Diseases characterized by an increased degradation of the fibrillar collagenous network are associated with an increased urinary excretion of both compounds: Pyridinoline and deoxypyridi

Effect of ibuprofen on the healing and remodeling of bone and articular cartilage in the rabbit temporomandibular joint.

The purpose of this study was to determine the effect of ibuprofen on the healing and remodeling of bone and cartilage in the temporomandibular joint of the rabbit. Forty-two rabbits were operated on to create a groove and a hole in the articular surface of both the right and left mandibular condyles. Following surgery, the animals were divided into three groups. Group A (12 rabbits) was used as a control and the animals did not receive any medication. Group B (15 rabbits) was given a daily dose of 17 mg/kg of ibuprofen. Group C (15 rabbits) was given a daily dose of 34 mg/kg of ibuprofen. All

Quantitative study of articular cartilage and subchondral bone remodeling in the knee joint of dogs after strenuous running training.

All tissues of the joint are affected in some way in osteoarthritis because the joint is an interactively functioning unit. Our goal was to investigate the combined responses of articular cartilage and subchondral bone to altered loading conditions to improve our understanding of the physiology of these two components and, ultimately, the pathophysiology of osteoarthritis. A group of 20 female beagle dogs were divided pairwise into runners (n = 10) and controls (n = 10). The running training on a treadmill started at the age of 15 weeks, and during the following 40 weeks the running distance w

Deformation of the distal femur: a contribution towards the pathogenesis of osteochondrosis dissecans in the knee joint.

Osteochondrosis dissecans (OD) is a process of subchondral bone necrosis occurring predominantly in young individuals at specific sites. The aetiology of this disease remains controversial with mechanical processes due to trauma and/or ischaemic factors being proposed. This study aims at explaining the aetiology of OD in the knee joint as a result of the particular deformation of the condyles. A finite element analysis of the distal third of the femur was performed. A three-dimensional model was developed based on computed tomography scans of a normal femur, consisting of cortical bone, cancel

Absorbed dose profiles for radionuclides of frequent use in radiation synovectomy.

Absorbed dose profiles are presented for radionuclides of frequent or potential use in radiation synovectomy, an intraarticular technique aimed at treating rheumatoid arthritis through direct and highly selective irradiation of inflamed synovium. The radionuclides investigated were 198Au, 165Dy, 32P, 186Re, 90Y, and 166Ho. These profiles reveal the absorbed dose imparted per unit activity of injected radionuclide (Gy/mCi) as a function of penetration distance (mm) to major components of the arthritic synovial joint. Their usefulness is twofold: they can be employed to select the radionuclide b

Magnetic resonance imaging of injuries to bone and articular cartilage. Emphasis on radiographically occult abnormalities.

Magnetic resonance imaging (MRI) has emerged as the premier noninvasive imaging method for evaluation of the musculoskeletal system. Among the most widely recognized and accepted applications of MRI is that of trauma-related imaging. While the initial emphasis on MRI was directed toward the assessment of soft-tissue structures previously difficult to image (eg, knee menisci and cruciate ligaments), it has become increasingly apparent that MRI is superbly suited to the depiction of a wide spectrum of injuries to bone and articular cartilage. MRI has proved capable of depicting abnormalities ran

Physical therapy management of hand fractures.

Hand fractures can be a complicated management dilemma for both the general clinician and the specialist. To better equip the therapist to treat fractures in the hand, a brief review of bone and articular cartilage healing and the effects of immobilization are reviewed. Active, passive, and resistive exercises for the patient with hand fracture are reviewed in addition to treatment of the associated problems of scar formation, edema, and pain. Static and dynamic splinting techniques are also discussed.

Effect of transforming growth factor beta on cell proliferation and glycosaminoglycan synthesis by rabbit growth-plate chondrocytes in culture.

The effects of the transforming growth factor beta (TGF-beta) on the growth and glycosaminoglycan synthesis of rabbit growth plate-chondrocytes in culture were studied. In serum-free medium, TGF-beta caused dose-dependent inhibition of DNA synthesis by chondrocytes, measured as [3H]thymidine incorporation (ED50 = 0.1-0.3 ng/ml). The inhibitory effect was maximal at a dose of 1 ng/ml, and extended for a duration of 16-42 h. In contrast, TGF-beta potentiated the synthesis of DNA stimulated by fetal calf serum (FCS). Addition of TGF-beta (1 ng/ml) to cultures containing 10% FCS increased [3H]thym

Collagen cross-linking in human bone and articular cartilage. Age-related changes in the content of mature hydroxypyridinium residues.

The concentration in collagen of hydroxypyridinium cross-linking amino acids was measured in samples of bone and cartilage from human subjects aged from 1 month to 80 years. Cortical and cancellous bone samples were dissected and analysed separately. In both bone and cartilage, the content of this mature form of cross-link reached a maximum by 10-15 years of age (the amount in cartilage being 5-10 times that in bone), then stayed essentially in the same range throughout adult life. In bone the ratio of the two chemical variants of the mature cross-link, hydroxylysylpyridinoline to lysylpyridin

Evidence that ecto-nucleoside-triphosphate pyrophosphatase serves in the generation of extracellular inorganic pyrophosphate in human bone and articular cartilage.

Extracellular inorganic pyrophosphate (PPi) is important in the regulation of mineralisation of bone, and in the pathogenesis of chondrocalcinosis, an arthritic disease in which calcium pyrophosphate dihydrate crystals form in articular cartilage. Nucleoside-triphosphate pyrophosphatase, which catalyses the formation of PPi, was previously observed at the surface of human articular chondrocytes in culture. A similar enzyme has been identified in osteoblast-like human bone cells in culture, and is active towards purine and pyrimidine nucleoside triphosphates. The enzyme has high affinity for AT

Effects of mechanical loading on the tissues of the rabbit knee.

We studied changes in subchondral bone and articular cartilage in an animal model of osteoarthrosis. In this model we applied repetitive impulsive loads to rabbits' knees. Their legs were held in short leg splints so the rabbits were unable to dampen the peak applied load with ankle flexion. After sacrifice, at 1 day to 6 weeks, we studied proximal tibial load-bearing cartilage histologically, biochemically, and with radioactive sulfate uptake. We also studied the subchondral bone under that cartilage histologically, histomorphometrically, with bone scan (99mTc pyrophosphate), and by tetracycl

Upper tibial osteotomy.

Arthritis of the knee usually produces a change in the coronal plane of the normal femorotibial axial alignment. This change occurs if the arthritis is mostly unicompartmental. Most unicompartmental degenerative arthritis involves the medial compartment, producing a varus deformity. In rheumatoid patients, however, a valgus deformity is commonly produced. For at least the last 25 years, upper tibial osteotomy has been used to correct this malalignment. The rationale is to unload the more involved compartment and transfer the load to the less involved side. While the vast majority of upper tibi

A bipedicled chest wall flap to cover an open elbow joint in a burned infant.

A 7-month-old girl sustained a deep and extensive burn of the posterior aspect of the left upper arm and forearm, complicated by an open wound of the elbow joint with exposed bone and articular cartilage. A bipedicled chest wall flap was used to cover the defect. The limb was preserved along with good function in the elbow joint. An above-elbow amputation was thus avoided.

Replacement of epiphyseal bone with methylmethacrylate: its effect on articular cartilage.

Methylmethacrylate cement was used as a substitute for the epiphyseal bone of the canine proximal femora, after femoral neck osteotomy. The effects of different volumes of acrylic cement on the integrity of trabecular bone and articular cartilage and the effects of different types of fixation of the osteotomy site were assessed clinically, radiologically and histologically. Degenerative changes occurring in the articular cartilage were greatest when a large area of epiphyseal bone was replaced with methylmethacrylate. These degenerative changes varied proportionately with the rigidity of fixat

Proteolytic enzymes in joint destruction.

An increase in both neutral and acid proteolytic activity and proteinase inhibitors is reported in synovial fluid of osteoarthrotic and rheumatoid joints, compared with controls, but in subchondral bone and articular cartilage only Cathepsin D and in synovial tissue only neutral caseinolysis were elevated. Especially high proteolytic activities were found in joint compartments of seronegative rheumatoid and in sera of osteoarthrotic patients. The source of the main part of neutral caseinolytic activity in joint fluid is inflammatory cells, in the case of Cathepsin D the tissues of origin seem

Survival of articular cartilage after controlled impact.

Survival characteristics of forty-three specimens of living human bone and articular cartilage from the knees of eight renal-transplant donors were studied, using a drop-tower device. Autoradiography and light and scanning electron microscopy revealed no evidence of chondrocyte death or structural damage until stress levels of twenty-five newtons per square millimeter were reached, corresponding to strains on the order of 20 to 30 per cent and involving energy absorption of one millijoule per cubic millimeter. The data for strain rates of 500 and 1000 s-1 suggest that impact loads sufficient t
