Molecular Disambiguation of Heart Rate Control by the Nucleus Ambiguus.

The nucleus ambiguus (nAmb) provides parasympathetic control of cardiorespiratory functions as well as motor control of the upper airways and esophagus. A subset of nAmb neurons innervates the heart through the vagus nerve to control cardiac function at rest and during key autonomic reflexes such as the mammalian diving reflex. Yet, how these cardiovagal nAmb neurons differ from other nAmb neurons in the adult brain remains unclear. We therefore classified adult male and female mouse nAmb neurons molecularly, anatomically, and functionally. First, our integrated analysis of single-nucleus RNA-

B-Cell Lymphoma Following an Indolent Course Over 30 Years with Immunophenotypic Changes at Each Recurrence.

INTRODUCTION: Diffuse large B-cell lymphoma (DLBCL) comprises biologically distinct subtypes, but whether cell-of-origin-related features remain stable over long disease courses is unclear. CASE DESCRIPTION: A woman was initially diagnosed with CD20-positive B-cell lymphoma in 1988 and treated with cyclophosphamide, vincristine and prednisolone (COP) chemotherapy. She experienced a first relapse in 1998 and received cyclophosphamide, doxorubicin, vincristine and prednisolone (CHOP) chemotherapy. The second relapse occurred in 2008 and was managed with rituximab. At the third relapse in 2011,

Multimodal Novel Recurrent Laryngeal Nerve Monitoring Techniques in Anterior Cervical Spine Surgery: Initial Experience and Proof of Concept.

Injury to the recurrent laryngeal nerve (RLN) during anterior cervical spine surgery is an uncommon yet impactful sequela. This study evaluates the feasibility and intraoperative behavior of laryngeal adductor reflex (LAR) and motor-evoked potentials with vocalis recordings (MEP-VC) during anterior cervical spine surgery. LAR and MEP-VC were employed for 13 patients undergoing anterior cervical spine surgery. LAR alerts were issued when clinically correlated reductions in amplitude, typically ≥20% in previously stable, non-habituating recordings, and/or temporal dispersion of the LAR waveform

Paraventricular nucleus of the hypothalamus activation and forebrain neuroinflammation in salt-loaded rats.

Salt loading (SL) in rats for 7 days raises mean arterial pressure (MAP) and induces astrogliosis in the paraventricular nucleus of the hypothalamus (PVN). However, the exact role of the PVN in elevating MAP and sympathetic nerve activity (SNA), as well as its effects on astrocytes in forebrain osmosensitive areas, remain unclear. Male Holtzman and Wistar rats (300-320 g) received only 2% NaCl solution (SL) to drink for 7 days, while controls drank water (WT). Daily fluid and food intake, body weight, urinary volume, and MAP were recorded. On day 7, blood and cerebrospinal fluid (CSF) were col

Dexmedetomidine induces bradycardia through vagal-nucleus tractus solitarius glutamatergic circuit.

BACKGROUND: Dexmedetomidine (DEX) often causes bradycardia. While sympathetic suppression is involved, its effects on vagal component remains elusive. METHODS: C57BL/6J male mice were treated with different doses of DEX (50, 100, or 200 μg/kg), heart rate was recorded with an electrophysiology recording system. Neuronal activity in the vagal nodose ganglion (NG) was assessed by in-vitro two-photon calcium imaging, patch-clamp recordings, phosphorylated ERK (p-Erk) staining, in-vivo electrophysiological recordings. Chemo-inactivation was targeted on vagal glutamatergic neurons and the vagal-nu

Pathological tau propagation from the brain to the colon via the vagal efferent pathway in Alzheimer's disease.

BACKGROUND: Alzheimer's disease (AD) is a neurodegenerative disorder marked by the accumulation of amyloid-β (Aβ) plaques and tau neurofibrillary tangles, leading to cognitive decline. Recent research has highlighted the involvement of the gut-brain axis (GBA) in AD progression, suggesting that the disease may also affect the gut. OBJECTIVE: To investigate the transmission of tau from the brain to the gut via the vagus nerve and its impact on gut function, we aimed to develop a novel in vitro system to simulate the GBA. DESIGN: We used an AD animal model to examine the spread of tau from the

Ultrasound Guidance for Botulinum Toxin Injection of Muscles Innervated by the Facial Nerve: A Systematic Review of Anatomical Precision, Safety, and Outcomes.

Botulinum toxin type A (BoNT-A) chemodenervation is commonly used for facial synkinesis and aesthetic indications, but landmark-based techniques are limited by anatomical variability and risk of off-target delivery. High-resolution ultrasound (US) can be used to enhance precision and safety. This systematic review explores the role of US-guided BoNT-A facial chemodenervation in evaluating anatomical accuracy, clinical efficacy, complications, and patient satisfaction. A comprehensive search of 6 databases through April 2025 identified studies assessing US-guided BoNT-A chemodenervation for fac

Immunohistochemical Localization and Anatomical Distribution of Enteric Glia Cells in Enteric Nervous System.

The enteric nervous system (ENS) comprises highly heterogeneous glia cell populations, which play critical roles in the regulation of intestinal neuromuscular function, providing physical and immune protection to neuronal cells and processes in physiological and pathological conditions. Due to the high muscle/mucosa content, thinness, and softness, freezing and sectioning of intestinal tissue are difficult, and distortion of tissue morphology might occur. However, whole-mount immunostaining for glia characterization in the 3D environment of the ENS is a better choice, as this powerful techniqu

Immunohistochemical Staining Protocol of Enteric Glial Cells in Myenteric Plexi.

Despite the development of several genetic approaches to visualize enteric glia within the enteric nervous system (ENS), multiple fluorescence labeling remains an essential tool for simultaneously identifying several target molecules. Fluorescence microscopy is widely used to differentiate between distinct glial subtypes based on their morphological appearance and chemical content, with a high degree of specificity. The dynamic nature of enteric glial cells drives extensive changes in morphology and/or the expression of markers, such as GFAP and S100β, in response to extracellular cues and und

Isolation of Distinct Populations of Enteric Glia from the Muscularis Externa and Mucosa of the Mouse Intestine.

Enteric glia, integral to the enteric nervous system (ENS), are crucial for intestinal motility, secretomotor function, and immunity. These glia occupy diverse niches from the serosa to the lumen, yet their transcriptional diversity across these compartments remains incompletely understood. Traditionally, studies on enteric glia have mostly focused on the myenteric plexus, omitting the large populations of glia located in the circular muscle and mucosa. Here, we present a method to rapidly isolate enteric glia from the muscularis externa and mucosal compartments of the mouse intestine in tande

Calcium Imaging Studies in Enteric Glia.

Calcium (Ca2+) imaging is a versatile technique to study cellular activity and is a particularly important tool to study electrically passive cells such as glia. Enteric glia play important roles in gut physiology, and their activity can be captured using Ca2+ imaging. Ca2+-encoded activities command glial processes such as gliotransmission and intercellular signaling. This chapter describes a protocol that will allow users to study glial Ca2+ activity in ex vivo whole-mounts of circular muscle-myenteric plexus. This chapter will include comprehensive steps to execute Ca2+ imaging experiments,

DPP-4 inhibitor alleviates gut-brain axis pathology in Parkinson's disease.

BACKGROUND: Dipeptidyl peptidase-4 inhibitors (DPP-4is) have been reported to exhibit therapeutic effects in Parkinson's disease (PD), increasing their potential for drug repurposing. One aspect of PD pathogenesis is thought to be associated with the gut-brain axis, where α-synuclein from the gut is transmitted to the brain via the vagus nerve (VN). OBJECTIVE: We explored whether sitagliptin, a DPP-4i, exhibits a protective effect in a low-dose rotenone-treated gut-brain axis-associated PD model. DESIGN: To explore the effect of sitagliptin, we used the oral rotenone-treated mouse model, whi

Electroacupuncture Alleviates Brain Injury Through Vagus Nerve Activation and Gut Microbiota in a Rat Model of Ischemic Stroke.

BACKGROUND: Emerging evidence implicates gut microbiota dysbiosis in exacerbating stroke pathogenesis via the gut-brain axis, suggesting novel therapeutic targets. While electroacupuncture (EA) demonstrates anti-inflammatory effects through vagus nerve activation, its neuroprotective mechanisms via vagus nerve-microbiota crosstalk remain unexplored. METHODS: Rats with middle cerebral artery occlusion received daily ST36 (Acupoint Zusanli) EA for 1 to 7 days postischemia. Subdiaphragmatic vagotomy and fecal microbiota transplant were implemented to validate pathway specificity. Multimodal asse

Cardiac vagal tone is associated with physical activity but not with depressive symptoms, work stress, and social support: A large-scale 10-year follow-up study.

Cardiac vagal tone has been associated with mental and physical health and presents opportunities for preventive and therapeutic applications in mental health and cardiovascular medicine. Vagally-mediated heart rate variability (vmHRV), a marker of cardiac vagal tone, has been associated with multiple factors related to well-being. Recent research further suggests that stress regulation, social connection, and lifestyle jointly contribute to improved well-being. In a large-scale study (n = 2252), we measured depressive symptoms, work stress, social support, and physical activity (T1) and then

Infectious Models in Mice for Study of Gut Inflammation: Primary Cultures of Myenteric Neurons.

The human gut has 200-600 million neurons coordinated by an integrative system called the enteric nervous system (ENS). The ENS forms a vast network that controls all the complex activities of the gastrointestinal tract (GIT). In some intestinal alterations, this complex control is impaired by the loss or reduction of enteric neurons/glial cells due to inflammation, which affects neurotransmitter and motility activities and leads to the development of disease. Elucidating the mechanism of intestinal inflammation-induced dysfunction of GIT is one of the major gaps in inflammatory disease resear
