Bridging the Literacy Gap in Breast Reconstruction Patient Education Using Artificial Intelligence.

BACKGROUND: Artificial intelligence is increasingly integrated into healthcare, demonstrating applications across surgical prediction, intraoperative precision, and patient communication. However, there is limited literature examining the role of artificial intelligence in improving patient comprehension of educational materials for breast reconstruction. METHODS: 30 texts from online education resources for breast reconstruction surgery were selected. Readability of the texts was then evaluated using the Flesch-Kincaid grade level, the Gunning Fog index, and the Coleman-Liau index. ChatGPT 4

Automated Aesthetic Evaluation After Breast Reconstruction Using Routine Postoperative Photographs: A No-Code AutoML Approach.

BACKGROUND: Aesthetic assessment after breast reconstruction remains subjective and is often affected by inter-rater variability. A practical and reproducible evaluation method is needed for routine clinical use. This study aimed to develop an automated algorithm for classifying postoperative breast aesthetics into three categories: excellent, fair, and bad using Google Cloud AutoML Vision, without requiring advanced programming expertise. METHODS: A total of 519 frontal two-dimensional photographs obtained 1 year after breast reconstruction were retrospectively collected. Images were annotat

Diagnostic Performance and Concordance of a BI-RADS-Output AI System in Mammography: A Retrospective Study of Radiologists-AI Collaboration.

The identification of breast cancer using mammography and ultrasonography is influenced by inter-observer variability. Artificial intelligence (AI) models for breast classification have been investigated, but limited reports exist on Breast Imaging Reporting and Data System (BI-RADS)-output AI systems. This study attempted to assess a BI-RADS-output AI system by comparing breast cancer detection rates among radiologist-only, AI-only, and combined interpretations, while also focusing on the AI's ability to identify cancers missed by radiologists and analyzing diagnostic concordance concerning i

Gendered ageism among women undergoing mastectomy across two life stages: early adulthood and later life in Pakistan.

This qualitative study examines how gender and age shape breast cancer and mastectomy experiences among women in Pakistan. Using critical phenomenology and drawing on 12 in-depth interviews with women across two distinct age groups (early adulthood and later life), the study identifies three key themes regarding age- and gender-based inequalities that shape women's access to care. These inequalities are produced not only within the healthcare system but also through social and familial structures: [1] From Surveillance to Neglect: Age-Stratified Gatekeeping and Delayed Diagnosis, [2] Sacrifici

Advancing women's health in Egypt: an economic evaluation of the presidential initiative for nationwide breast cancer early detection and treatment.

BACKGROUND: Breast cancer (BC) is the leading cause of cancer-related mortality among women worldwide and poses a major public health and economic challenge in Egypt, where late-stage diagnosis remains high. Egypt launched the Presidential Initiative for Women's Health, a nationwide, publicly funded program providing free early detection, diagnostic, and treatment services. This study presents a model-based economic evaluation of the initiative. METHODS: A hybrid decision tree and Markov model compared the Presidential Initiative pathway with opportunistic case detection, from a societal pers

Mitochondrial transfer between breast cancer cells promotes ROS-dependent proliferation.

Previously, we showed that macrophages transfer mitochondria to breast cancer cells, promoting proliferation in acceptor cancer cells. Transferred mitochondria were depolarized and accumulated reactive oxygen species (ROS), and the mitochondrial transfer-induced proliferation was dependent upon ROS signaling (Kidwell et al. 2023). Our unexpected findings supported a model in which transferred mitochondria act as a signal for proliferation in acceptor cancer cells rather than a direct source of increased bioenergetics. It remains unclear whether this unexpected signaling mechanism is unique to

Durable partial response with low-dose, intermittent nivolumab in combination with gemcitabine for high PD-L1 metastatic triple-negative breast cancer: a case report.

Immune checkpoint inhibitors (ICIs) have improved outcomes in selected patients with metastatic triple-negative breast cancer (mTNBC), particularly those with high programed death-ligand 1 (PD-L1) expression. However, optimal dosing strategies balancing efficacy, toxicity, and cost remain unclear. We report the case of a 34-year-old woman with recurrent mTNBC and high PD-L1 expression (combined positive score, CPS ≥ 60; immunohistochemistry 22C3 pharmDx assay). She achieved durable disease control over 22 months (September 2024 to June 2026) using a combination of gemcitabine maintenance chemo

Tumor-antigen-specific polyclonal antibodies suppress tumor growth and metastasis in heterogeneous solid tumors.

INTRODUCTION: Tumor heterogeneity and signaling redundancy limit the efficacy of monospecific antibody therapies in solid malignancies. Here we used our platform of glyco-humanized polyclonal antibodies (GH-pAb), to investigate whether targeting multiple tumor-associated antigens provides integrated anti-tumor activity. METHODS: GH-pAb (hereinafter XON7) were generated by immunization of double-knockout pigs with human tumor cells. Antigen specificity of XON7 was assessed using a human proteome array and validated by ELISA. Anti-tumor mechanisms were evaluated in vitro on 2D or 3D-cultures. E

Boundary-aware geometric Mamba network for breast tumor segmentation in 3D MRI.

INTRODUCTION: Accurate breast tumor segmentation in Dynamic Contrast-Enhanced Magnetic Resonance Imaging (DCE-MRI) is important for reliable diagnosis and treatment planning. However, complex tumor morphology, diverse spatial structures, and indistinct boundaries remain challenging for existing segmentation methods. Convolutional neural network-based methods have limited ability to model long-range dependencies, while Transformer-based methods often introduce substantial computational overhead. Although Mamba-based approaches provide an efficient alternative, conventional scanning strategies m

Peer-led, faith-based co-designed intervention to improve Muslim women's cancer screening: results from the IMCAN study.

INTRODUCTION: Cancer screening saves lives, but Muslim women are less likely to participate in screening than the general population. This makes them less likely to benefit from earlier diagnosis. Faith-based cancer communications provide an acceptable strategy for addressing screening barriers. Hence, we evaluated the impact of a faith-based intervention on knowledge, attitudes, and breast, cervical and colorectal (bowel) screening practices of Muslim women in the UK. Our intervention was a co-designed, peer-led, two-hour workshop informed by the Integrated Screening Action Model (I-SAM) and

Effects of the AI-HEALS intervention based on the Fogg behavior model on self-management in postoperative breast cancer patients: a randomized controlled trial study protocol.

BACKGROUND: Breast cancer is the most common malignancy among women, and postoperative self-management is critically associated with long-term quality of life. Traditional clinical nursing and mobile health interventions frequently lack behavioral theory support, thereby failing to meet individualized rehabilitation needs. The Fogg Behavior Model (FBM) provides a theoretical framework for precise intervention through the dimensions of motivation, ability, and prompts. Accordingly, this study developed an FBM-based AI-HEALS intervention and will validate its effectiveness via a Randomized Contr

Phytochemical modulation of DNMT1-mediated DNA methylation: computational insights into breast cancer.

CONTEXT: Aberrant DNA methylation, particularly DNMT1-mediated hypermethylation of tumor suppressor and DNA repair genes, contributes to breast cancer progression. The limitations associated with existing DNMT inhibitors highlight the need for alternative and potentially safer epigenetic modulators. Here, we developed a computational workflow to identify phytochemicals targeting the catalytic methyltransferase domain of DNMT1. Screening of the SelleckChem phytochemical library identified Echinacoside (-9.56 kcal/mol), 1,2,3,4,6-O-Pentagalloylglucose (-9.49 kcal/mol), and Complanatoside A (-9.2

Investigation of DNA methylation levels before and after treatment in breast cancer patients receiving neoadjuvant chemotherapy.

PURPOSE: Drug resistance is a major cause of treatment failure in breast cancer patients. Numerous genetic and epigenetic markers have been proposed to influence Neoadjuvant Chemotherapy (NACT) response, and genes that regulate the effectiveness of chemotherapeutic agents undergo expression regulation via methylation. In this study, we aimed to investigate the methylation levels of KDM5A, KDM5B, and KDM5C, as well as their associated genes, EZH2, TFAP2C, ZMYND8, and SIN3B (Exon 1-2 and 19-20), in blood samples collected from patients before and after NACT treatment. Besides, we also examined p

Paraptosis, a new type of cell death that can be applied to cancer treatment.

Paraptosis is a form of non-apoptotic cell death characterized by cytoplasmic vacuolization and swelling of the mitochondria and endoplasmic reticulum (ER). Recent studies have demonstrated that inducing paraptosis in cancer cells can circumvent apoptosis resistance and synergize with other forms of cell death and tumor immunity, providing a novel strategy for anti-tumor therapy. The underlying mechanism of paraptosis involves ER stress triggered by inducers, leading to the swelling of the ER and mitochondria. This subsequently alters the cytoskeleton and disrupts molecular processes, such as

From Intestinal Origins to Mammary Targets: Exploring the Potential Role of the Brain-Gut-Microbiota Axis in Pre-Metastatic Conditioning of Breast Tissue in Colorectal Cancer.

Historically viewed as a localized gastrointestinal malignancy, colorectal cancer (CRC) is increasingly recognized as a systemic pathology that actively programs distant microenvironments. This review delineates a novel mechanistic framework wherein CRC-driven dysregulation of the brain-gut-microbiota axis (BGMA) acts as a systemic architect of the breast pre-metastatic niche (PMN). We synthesize how localized intestinal oncogenic stress propagates through three convergent conduits: (1) Immune Reprogramming, where gut-derived inflammatory mediators and extracellular vesicles (EVs) recruit myel

Maternal Cannabis Use and Secretory IgA and IgM Concentrations in Breast Milk.

BACKGROUND: Breast milk is the gold standard for infant nutrition, providing essential nutrients and bioactive factors such as secretory immunoglobulins A (SIgA) and M (SIgM). The association between maternal cannabis consumption and SIgA and SIgM concentrations in breast milk remains poorly understood, despite the rising trends in cannabis consumption among women of reproductive age. OBJECTIVE: To examine the association between maternal characteristics and cannabis use patterns and SIgA and SIgM concentrations in breast milk. METHODS: This prospective cohort study enrolled 160 pregnant and

Association of Circulating Eosinophils With Systemic Immune Activation and Reduced Recurrence in Hormone Receptor-Positive/Human Epidermal Growth Factor Receptor 2-Negative Breast Cancer.

PURPOSE: Hormone receptor-positive/human epidermal growth factor receptor 2-negative (HER2-) breast cancer, the most common subtype, lacks simple, noninvasive biomarkers for recurrence. We evaluated circulating eosinophil count as a clinically applicable biomarker for recurrence in this population. METHODS: Associations between preoperative eosinophil count and patient outcomes were evaluated in a multimodal discovery cohort (N = 472) and validated via propensity score overlap weighting in an independent expanded cohort (N = 23,170). We assessed the additive value of eosinophils in patients w
