Hepatic focal nodular hyperplasia (FNH)-like lesions mimicking metastasis have been reported as rare complications of cytotoxic chemotherapy, but they have not previously been described in association with CDK4/6 inhibitors alone. We present the case of a 32-year-old chemotherapy-naïve woman with de novo hormone receptor (HR)-positive/HER2-negative metastatic breast cancer who was treated with first-line ribociclib and letrozole. After four cycles, imaging revealed a complete response of her bone metastases alongside a newly developed 5.5-cm hepatic lesion suspicious for visceral progression.

This Correspondence responds to the prospective study by Anuszkiewicz et al., which assessed dynamic indocyanine green (ICG) lymphography drainage time as a marker of axillary nodal burden. We raise three methodological points relevant to how the results should be read. First, the cohort included only patients selected for axillary lymph node dissection, so it represents a narrow, non-representative spectrum in which discrimination tends to be optimistic; the reported area under the curve of 0.995 was obtained without internal or external validation. Second, an ipsilateral-to-contralateral rat

BACKGROUND: For breast cancer patients with clinically node-positive (cN+) and residual disease after neoadjuvant chemotherapy (NAC), axillary lymph node dissection (ALND) remains the standard of care, though alternative strategies may be selected by multidisciplinary teams in certain cases. This study evaluated factors associated with omission of ALND for cN+ patients who were pathologically node-positive after NAC (ypN+).
METHODS: In this retrospective observational study, the National Cancer Database was queried for patients with clinical T1-T4, N1-N3 breast cancer from 2016 to 2021 who re

PURPOSE: To assess intra- and inter-reader agreement of the Node-RADS v1.0 score for axillary lymph node evaluation in breast cancer (BC) patients undergoing preoperative breast MRI (bMRI), and to explore its patient-level association with pathological nodal involvement. Contrast-enhanced CT was evaluated as a subgroup exploratory analysis.
METHODS: This retrospective study included 100 women with biopsy-proven BC who underwent preoperative bMRI; 25 also underwent contrast-enhanced CT. Four readers (two expert breast radiologists and two junior readers) independently assigned Node-RADS v1.0 s

BACKGROUND: This study aimed to develop a dynamic nomogram in which clinical indicators are integrated with magnetic resonance imaging (MRI) radiomics to predict axillary lymph node metastasis (ALNM) in patients with breast cancer.
METHODS: Our retrospective study included 221 pathologically confirmed patients with breast cancer. Radiomic features were extracted from dynamic contrast-enhanced MRI (DCE-MRI) and fat-suppressed T2-weighted imaging (FS-T2WI) datasets. After feature screening, a support vector machine (SVM) algorithm was employed to establish radiomic models and calculate radiomic

BACKGROUND: Accurate assessment of axillary lymph node status after neoadjuvant systemic therapy (NST) is essential for de-escalating surgery in node-positive breast cancer. Although various axillary surgeries based on sentinel lymph node biopsy (SLNB) are widely reported, they still have limitations in this setting. Since metastatic lymph nodes inherently encompass sentinel nodes, we hypothesized that directly marking and retrieving these nodes could enable reliable restaging without SLNB.
MATERIALS AND METHODS: In this single-arm, prospective cohort study, patients with pathologically confi

Background/Objectives: Tumor-infiltrating lymphocyte (TIL) therapy is an important option for patients with metastatic melanoma progressing after standard systemic therapy, but real-world data on treatment delivery, toxicity monitoring, and immune recovery remain limited. We evaluated clinical outcomes, treatment tolerance, immune reconstitution, and cardiac biomarker dynamics across three Mayo Clinic sites. Methods: We retrospectively analyzed adults with metastatic melanoma who received lymphodepleting chemotherapy followed by TIL infusion and high-dose interleukin-2 (IL-2) between April 202

Neutrophil extracellular traps (NETs) may facilitate metastasis of tumor cells through interaction with the CCDC25 located along cell membrane, resulting high mortality of patients. Although cationic materials can destruct NETs and inhibit tumor metastasis, they lack targeting ability in physiological environments. Herein, we designed and synthesized a peptide-dendrimer conjugates, AAPVRnS(5-n)-G2.5, that may target neutrophil elastase (NE), a component of NETs, expose their cationic cores by NE degradation, and finally neutralize DNAs of NETs via charge interaction. It combined a triple strat

BACKGROUND: To compare HER2-targeted with ¹⁸F-FDG PET/CT for detecting axillary lymph node metastasis in patients with newly diagnosed HER2-positive or HER2-low breast cancer.
METHODS: In this prospective study, HER2-expressed breast cancer participants underwent both 18 F-FDG and Al 18 F-NOTA-HER2-BCH PET/CT within 1 week. On the basis of IHC and/or FISH, participants were categorized as HER2-positive (n=25) or HER2-low (n=26). Histopathology or imaging follow-up served as reference standards. Statistical analyses utilized the paired McNemar test for diagnostic performance, and the Wilcoxon

BACKGROUND: Axillary management in early breast cancer (EBC) is increasingly moving toward surgical de-escalation. While sentinel lymph node biopsy (SLNB) has reduced morbidity compared with axillary lymph node dissection, it still carries risks and may be unnecessary in selected patients with very low nodal risk. An accurate estimate of the risk of sentinel lymph node (SLN) metastasis may support risk-adapted omission strategies. This study aimed to validate existing prediction models, develop a cohort-derived model, and evaluate whether ensemble modeling improves the prediction of SLN macrom

The central nervous system (CNS) is surrounded by specialized immune interfaces, including the meninges and the olfactory bulb, which can serve both as permissive routes for metastatic colonization and as entry points for anti-tumor immune responses. Understanding how immune cell dynamics evolve within these structures during metastatic progression is essential for identifying mechanisms that either support or restrict tumor growth in the brain. This protocol details the establishment of a breast cancer brain metastases (BCBM) mouse model through intracranial injection of E0771 breast cancer c

Triple-negative breast cancer (TNBC) is an aggressive subtype of breast cancer defined by a paucity of targeted therapies, high metastatic propensity, and an immunosuppressive tumor microenvironment rich in tumor-associated macrophages (TAMs). Here, we developed a liposomal immunoadjuvant system co-encapsulating Poly(I:C) and QS-21 aimed at reprogramming TAMs and bolstering antitumor immunity. In an orthotopic 4T1 TNBC mouse model, intratumoral injection of the combined formulation markedly suppressed primary tumor growth and pulmonary metastasis, with no observable systemic toxicity. BACKGROUND: Completion axillary lymph node dissection (ALND) is no longer routinely recommended for patients with breast cancer and minimal nodal involvement. In 2012, Danish national guidelines were revised to omit ALND for patients with sentinel node (SN) micrometastases (pN1mi) or isolated tumor cells (pN0[i+]). Long-term evidence on axillary recurrence (AR) and overall survival (OS), particularly after mastectomy, remain limited. This nationwide retrospective register-based study evaluated long-term outcomes after omission of ALND.
METHODS: Clinically node-negative women with pN1mi/pN0(i+

BACKGROUND: Survivorship considerations have gained increasing importance in patients with early breast cancer. Every surgical procedure poses a risk of complications and a potential negative impact on patient-reported outcomes (PROs), thereby driving growing interest in de-escalation strategies in breast cancer surgery. In this context, we aimed to assess complication rates, the potential role of physiatric rehabilitation in managing these complications, and the impact of axillary surgery on PROs.
METHODS: This retrospective single-center cohort study included breast cancer patients who unde

PURPOSE: Developing a deep learning model to simultaneously evaluate lymph node status and distinguish between benign and malignant breast masses has been a challenging clinical task. This study aimed to use radio frequency (RF) signal data to create a deep learning, multimodal multitasking model, incorporating Class Activation Mapping (CAM) heatmaps to assist ultrasonographers in assessing the overall patient state based on their expertise. Research has shown a correlation between lymph node metastases and the heterogeneity of neutrophil distribution in malignancies. Therefore, the study also

The necessity of sentinel lymph node biopsy (SLNB) in early-stage, node-negative (cN0) breast cancer is increasingly questioned in the era of surgical de-escalation and biologically driven treatment strategies. While SLNB has long replaced axillary lymph node dissection to reduce morbidity, it nevertheless carries a measurable risk of complications, including lymphedema, pain, and reduced arm mobility. Recent high-quality evidence suggests that SLNB may be safely omitted in selected patients without compromising oncologic outcomes. The SOUND randomized trial demonstrated that omission of SLNB

BACKGROUND: Triple-negative breast cancer (TNBC) is characterized by a high propensity for metastatic relapse and a robust infiltration of tumor-associated macrophages (TAMs), which orchestrate a pro-metastatic microenvironment. Hedyotis diffusa Willd-Scutellaria barbata (HS), a classical herbal pair, exhibits potent antitumor activities; however, its specific role in modulating TAM-dependent metastasis remains unclear.
PURPOSE: This study investigated whether HS suppresses TNBC metastasis by modulating the functional state of TAMs, and sought to define the bioactive constituents and their mo
