Pulmonary fibrosis (PF) is a major cause of morbidity and mortality. Although increased oxidative stress and altered metabolism are implicated in PF pathobiology, our knowledge regarding the contribution of the glucose metabolism to the synthesis of extracellular matrix (ECM) is still incomplete. Therefore, our objective was to determine altered metabolic pathways that contribute to bleomycin (BLM; 5 mg/kg) sulfate-induced PF in rats. We determined the effects of nebulized BLM on PF in CRISPR-edited rats expressing glucose-6-phosphate dehydrogenase (G6PD) variant (S188F; G6PDS188F) and their w

BACKGROUND: Glucose-6-phosphate dehydrogenase (G6PD) deficiency (G6PDd) is an common enzyme deficiency disease, inducing hemolysis, hyperbilirubinemia, and jaundice, as well as liver dysfunction, in approximately 400 million patients. Patients with G6PDd are more susceptible to viruses infection than those without. However, the incidence and severity of hepatitis E virus (HEV) infection in patients with G6PDd are largely unknown.
METHODS: The prevalence of HEV in patients with G6PDd was investigated. Susceptibility to HEV was evaluated in a hepatoma cell line with G6PD knockdown, and the inte

Hepatolenticular degeneration (HLD, MIM:277900) is an autosomal recessive disorder characterized by excessive copper accumulation in hepatocytes, leading to hepatic and neurological abnormalities, hepatocellular injury, neurodegeneration, and copper deposition in the cornea. Glucose-6-phosphate dehydrogenase (G6PD) deficiency is the most common genetic cause of hemolytic anemia, which can be triggered by chronic conditions, drugs, food, or infections, and exhibits highly variable severity. In some cases, multiple genetic factors may collectively influence disease presentation and progression.

BACKGROUND: Glucose-6-phosphate dehydrogenase deficiency (G6PDd) is a common genetic disorder that impairs the cellular antioxidant response and has been hypothesized as a potential risk factor for severe outcomes in viral infections, including COVID-19. However, clinical evidence remains limited, especially in regions with high G6PDd prevalence.
METHODS: We conducted a retrospective cohort study using secondary data from four health information systems from health facilities in the Brazilian Amazon (E-SUS Notifica, SIVEP-Gripe, SIM, and a G6PD enzyme activity database). The study population

Glucose-6-phosphate dehydrogenase (G6PD) deficiency is the most common human enzyme defect, predisposing individuals to acute hemolytic anemia upon oxidative stress. Hereditary elliptocytosis (HE) is a distinct, typically autosomal dominant disorder caused by red blood cell (RBC) membrane skeleton defects, leading to chronic hemolysis. The co-occurrence of G6PD deficiency and HE is exceedingly rare, often resulting in more severe and complicated clinical presentations. Here we report the first case of a five-year-old Saudi male from a consanguineous marriage who presented in the neonatal perio

BACKGROUND: Outside of sub-Saharan Africa, Plasmodium vivax has become the dominant species of malaria. Focal mass drug administration (fMDA) is a potential strategy to support elimination efforts, but controlled studies are lacking.
METHODS: The FocaL mass drug Administration for Plasmodium vivax Malaria Elimination (FLAME) study is a 3-year cluster randomized controlled trial to determine the impact and safety of fMDA to reduce P. vivax transmission. The study will be conducted in Loreto, Peru, where standard interventions have reduced P. vivax cases, but transmission persists due to a high

Glucose-6-phosphate dehydrogenase (G6PD) deficiency is an X-linked recessive genetic disorder that can cause severe anemia in affected individuals exposed to oxidative stress. This risk is particularly relevant in patients treated with the antimalarial drug primaquine. In Myanmar, primaquine has been widely administered as a Plasmodium vivax malaria treatment; however, prevalence of G6PD deficiency among the population remains insufficiently characterized. This study investigated the prevalence of G6PD variants among various minority ethnic subgroups residing in Kachin State, Myanmar. Blood sa

BACKGROUND: Malaria, caused by Plasmodium falciparum and Plasmodium vivax, is commonly reported in Ethiopia. The World Health Organization (WHO) recommends treating P. vivax malaria with a schizonticidal drug and a 14-day course of primaquine (0.25-0.5 mg/kg body weight daily) to prevent relapses. In 2018, Ethiopia adopted a policy of primaquine radical cure (0.25 mg/kg for 14 days), with close monitoring for adverse events, but without prior glucose-6-phosphate dehydrogenase (G6PD) testing. This study aimed to assess the safety of this policy by monitoring the risk of haemolysis and other adv

Plasmodium vivax malaria remains a significant global health challenge, complicated by the parasite's ability to form dormant liver stages (hypnozoites) that cause relapses. Radical cure of P. vivax malaria requires administration of a hypnozoitocidal drug, such as primaquine or tafenoquine. However, these drugs can cause severe haemolysis in individuals with glucose-6-phosphate dehydrogenase (G6PD) deficiency. G6PD deficiency is caused by more than 230 different variants at the gene level that confer different degrees of deficiency phenotypically. Understanding the distribution of different G

BACKGROUND: Both Duffy blood antigen expression and G6PD deficiency are known to be associated with ethnic origin. Updates in epidemiological data on the prevalence of polymorphisms in these two human genes are key information for guiding national programs to eliminate Plasmodium vivax malaria.
METHODS: Duffy genotypes and their predicted phenotypes were determined in 943 blood samples from Mauritanian patients belonging to different ethnic groups (n = 432 White Moors and n = 511 individuals of black African ancestry) with a known G6PD genotype determined by PCR-restriction fragment length po

Dengue fever (DF) is a viral fever transmitted by the Aedes species of mosquitoes. Globalisation has led to worldwide spread of DF including Oman. High incidence of glucose-6-phosphate dehydrogenase (G6PD) deficiency is present in Oman's population but is often undiagnosed. We report 3 G6PD-deficient patients admitted in a tertiary care hospital in Muscat, Oman in 2023 with DF that later triggered haemolysis. It proved fatal in 1 case and caused irreversible renal damage requiring dialysis in the other case. Both DF and G6PD deficiency can cause bleeding. Both need supportive treatment. G6PD d

Glucose-6-phosphate dehydrogenase (G6PD) deficiency is the most common enzymopathy in humans that may exacerbate clinical outcomes during viral infections such as dengue, particularly in regions where both conditions are endemic. This study aimed to characterize the spectrum of G6PD mutations and explore trends in hemoglobin levels among adult dengue patients in Thailand. Samples from 231 adult patients diagnosed with dengue were analyzed. G6PD deficiency was identified in 24 individuals (10.4%), while G6PD mutations were detected in 111 patients (48.1%). The most frequently observed mutations

Glucose-6-phosphate dehydrogenase (G6PD) deficiency is a well-known red blood cell enzymopathy and a cause of intravascular hemolysis. This case report presents a child with underlying G6PD deficiency who experienced an acute episode of extensive intravascular hemolysis induced by a scrub typhus infection. The key takeaway from this report is that scrub typhus infection can trigger extensive hemolysis in patients with even "mild" G6PD deficiency, and normal G6PD levels found during the acute phase of hemolysis do not rule out the possibility of underlying G6PD deficiency.

Typhoid is an acute febrile illness primarily caused by Salmonella enterica serotype typhi (S. Typhi) which could be challenging to diagnose in children, owing to its non-specific clinical signs and symptoms which may resemble other febrile illnesses. Here, we present a case of typhoid which was atypically presented as fever of unknown origin (FUO) in a two-year-old boy with underlying glucose-6-phosphate dehydrogenase (G6PD) deficiency. This child was initially diagnosed and managed as acute tonsillopharyngitis, however remained febrile despite medications. A series of investigations were per

BACKGROUND: Glucose-6-phosphate dehydrogenase (G6PD) deficiency is the main risk factor for severe haemolysis following treatment with 8-aminoquinolines (8AQ). The World Health Organization recommends G6PD testing prior to 8AQ-based hypnozoitocidal treatment.
METHODS: We undertook an individual level meta-analysis of the performance of commercially available quantitative point-of-care diagnostics (PoCs) compared with reference spectrophotometry. A systematic literature search (PROSPERO: CRD42022330733) identified 595 articles of which 16 (2.7%) fulfilled pre-defined inclusion criteria and wer

BACKGROUND: Measurement of glucose-6-phosphate dehydrogenase (G6PD) activity guides hypnozoitocidal treatment of P vivax malaria. The G6PD Standard (SDBiosensor, Republic of Korea) here referred to as "Biosensor" is a quantitative point-of-care diagnostic that measures G6PD activity in U/gHb. The manufacturer recommends cutoffs to define G6PD deficient (≤ 4.0U/gHb), intermediate (4.1- ≤ 6.0U/gHb) and normal (> 6.0U/gHb) individuals. The aim of this individual patient data (IPD) meta-analysis was to evaluate these cutoffs (CRD42023406595).
METHODS: A systematic review identified studies report

OBJECTIVES: Glucose-6-Phosphate Dehydrogenase Deficiency (G6PD) is the most common enzymopathy globally. Early studies suggested an association with severe psychotic illness; however, changes to laboratory testing and diagnostic classification renders the association unclear. This study aims to explore the interaction between G6PD deficiency and psychotic symptoms, in particular to identify specific patterns of presentation or impact on outcomes.
METHODS: Pubmed, Embase, and PsycInfo databases were searched from inception to May 2023. Descriptive statistics and narrative review of were used t
