Serum erythropoietin and its determinants and associations in patients with haemoglobin E β-thalassaemia.

Erythropoietin is a hormone that stimulates erythropoiesis. The role of erythropoietin in the pathophysiology of HbE β-thalassaemia, a subtype of thalassaemia, is understudied. Here, we aim to evaluate the determinants and associations of erythropoietin in HbE β-thalassaemia patients. We conducted a cross-sectional study in Thalassaemia Centres of Colombo North Teaching Hospital, Sri Lanka, from September to December 2024. All patients with HbE β-thalassaemia were recruited. Clinical details were collected using an interviewer schedule. Haemoglobin, ferritin and erythropoietin were measured us

CRISPR/Cas12a-SERS biosensor based on sea urchin-like AuNPs for the detection of β-thalassemia mutant gene CD31.

Beta-thalassemia is a single-gene recessive disorder caused by mutations in the HBB gene, and approximately 1.5% of the global population are carriers of β-thalassemia. It is therefore vital to establish a rapid and sensitive method to detect the mutant genes of β-thalassemia. In this study, a CRISPR/Cas12a-mediated amplification-free surface-enhanced Raman spectroscopy (SERS) biosensor was developed. This biosensor uses sea urchin-shaped gold nanoparticles (SUGNPs) as the SERS enhancement substrate and 4-mercaptobenzoic acid (4-MBA) as the Raman reporter. It couples the SUGNPs/4-MBA with magn

Blood that was far from red: hypertriglyceridaema thalassaemia syndrome: case report.

Thalassaemia is the most common inherited haemoglobin disorder worldwide and a major health problem. Beta-thalassaemia major generally presents with a normal serum lipid profile, but there have been a few reports of its association with hypertriglyceridaemia, the exact pathogenesis of which remains unknown, and where early and regular blood transfusions ameliorate hypertriglyceridemia, thereby preventing its complications and the need for medical treatment of hypertriglyceridemia. This entity was seen in a 6-month-old infant girl who presented with severe anaemia, failure to thrive and organom

Burden of transfusion-transmitted infections among patients with thalassaemia and sickle cell anaemia in India: A systematic review and meta-analysis.

Lifelong blood transfusions, crucial for severe cases of thalassaemia and sickle cell disease (SCD), contribute to a high risk of transfusion-transmitted infections (TTIs). The current systematic review and meta-analysis determined the prevalence of transfusion-transmitted infections (TTIs) among patients with thalassaemia and sickle cell anaemia in India. A systematic search was conducted in PubMed, Scopus, ProQuest, EMBASE and Web of Science for articles published up to September 13, 2025. Title abstract screening followed by full-text review and data extraction was done by two independent r

Deferiprone therapy improves the oxidative status of LDL in patients with β-thalassaemia/HbE.

BACKGROUND: Oxidative modifications of low-density lipoproteins (LDL) have been reported in patients with β-thalassaemia/haemoglobin E (HbE) and are related to cardiovascular complications. Deferiprone (L1) is an iron chelator that decreases iron overload and, consequently, reduces oxidative stress. This study assesses the protective effect of L1 on the oxidative status of LDL in patients with β-thalassaemia/HbE. METHODS: Twenty-nine patients with β-thalassaemia/HbE treated with L1 were recruited. The study included a 4-week washout period followed by 4 and 12 weeks of L1 treatment. Non-trans

Comprehensive genetic screening of in vitro fertilized embryos using preimplantation genetic testing for monogenic gene disorders via the Sanger sequencing technique.

BACKGROUND: Preimplantation genetic testing for monogenic disorders (PGT-M) in conjunction with in vitro fertilization (IVF) has dramatically improved the ability to eliminate the development of genetic disorders among newborns. The incorporation of Sanger sequencing with PGT-M has enhanced accurate diagnosis and decreased the risk of developing genetic disorders. METHODS: A total of 295 embryos were collected from 47 IVF cases, and purification was performed after extraction; spectrophotometric analysis with a Nanodrop was conducted to measure the concentration and purity of total DNA. Genetic mutations of Transfusion Dependent Thalassaemia (TDT) in paediatric patients.

INTRODUCTION: Thalassaemia comprises inherited disorders characterised by impaired synthesis of globin chains, leading to varying phenotypes from mild to severe anaemia, transfusion-dependent. The Malaysian Thalassaemia Registry reports that 57.69% of patients are classified as Transfusion-Dependant Thalassaemia (TDT). Despite diagnostic advancements, local data linking genetic mutations with clinical and laboratory features of TDT remain limited. This study aims to address this gap by evaluating paediatric TDT patients at Hospital Tuanku Azizah (HTA), Kuala Lumpur, Malaysia. MATERIALS AND ME

Advances in Gene Therapy for Inherited Haemoglobinopathies.

Haemoglobinopathies, including β-thalassaemia and sickle cell disease (SCD), are among the most common monogenic disorders worldwide and remain major causes of morbidity and early mortality. Historically, management of these life-altering diseases has relied on supportive treatment and symptom management and, although these treatments reduce symptoms and ease disease burden, they do not correct the underlying genetic defect. Allogenic haematopoietic stem cell transplantation (HSCT) has been the only established curative option; however, it comes with substantial risks that significantly restri

In-silico identification and functional analysis of high-risk oncogenic nsSNPs in the human SDHB gene and their implications for cancer susceptibility.

BACKGROUND AND AIM OF THE STUDY: Although next-generation cardiac prostheses have shown favorable results in transcatheter aortic valve implantation (TAVI), these have mostly been documented in intermediate-risk patients. Whether this could be translated to high-risk patients is not known. Hence, the safety and clinical performance of the new, repositionable CoreValve Evolut R-System (ERS) was evaluated by comparison with a non-repositionable CoreValve-System (CVS), in 96 high-risk/inoperable (HRI) patients. METHODS: The primary safety end points were mortality and stroke, defined by VARC-2 c

Serum folate and dietary folate intake in beta thalassaemia trait: a case-control study from Sri Lanka.

OBJECTIVES: Ineffective erythropoiesis, although at a mild degree, could make individuals with beta thalassaemia trait (BTT) vulnerable to folate deficiency. This could be more pronounced in communities where dietary intake of folate is substantially lower. We aimed to assess serum folate levels and dietary folate consumption in individuals with BTT and compare them with healthy matched controls. DESIGN: A case-control study. SETTING: This study was conducted between June 2021 and June 2022 at a regional thalassaemia centre in Sri Lanka. PARTICIPANTS: The study includes 100 sets of samples,

Combined hereditary spherocytosis and β-thalassaemia trait: A rare co-existence.

Haemoglobinopathies, the most prevalent haemolytic disease in India, make up the majority of patients in most haematology outpatient clinics. The most prevalent hereditary haemolytic anaemia is beta-thalassaemia trait (βTT). It often remains undiagnosed as it has an asymptomatic clinical course. However, βTT needs be identified to provide genetic counselling to the affected families and to reduce the number of affected children born, which will lower their overall financial burden. Better screening methods for haemoglobinopathies have increased the detection of mixed haemolytic anaemia. We rep

Case-control study of maternal and fetal outcomes in beta thalassaemia trait during pregnancy.

Individuals with beta thalassaemia trait are not expected to have clinically significant morbidities besides mild anaemia. Pregnancy would exaggerate the anaemia in beta thalassaemia traits, but how this could affect maternal and fetal outcomes is unclear. Previous studies on maternal and fetal outcomes in beta thalassaemia trait have been inconsistent and even contradictory. Thus, we aimed to study the outcomes of pregnancy in pregnant women with beta-thalassemia trait. The prospective case-control study included 120 pregnant women with beta thalassaemia trait and 120 normal pregnant women. P
