Nephropathies Associated with Sickle Cell Trait and How to Study Them.

Sickle cell trait (SCT), which carries a single point mutation in the hemoglobin-β (HBB) gene, has long been considered a benign condition. However, epidemiological evidence challenges this assumption, revealing that individuals with SCT face an elevated risk of renal dysfunction. However, this field of study remains ill-defined as it has focused on sickle cell disease (SCD), where renal complications are severe. As SCT is more prevalent than SCD, consequences of nephropathies in this group translate into a substantial and largely unaddressed public health burden. Clinical data, primarily obse

Phenome- and laboratory-wide meta-analyses of sickle cell trait reveal multi-system disease associations.

Sickle cell trait (SCT) is increasingly recognized as a risk factor for adverse health outcomes. We utilized three large US electronic health record-based biobanks (Vanderbilt University Medical Center's BioVU, Penn Medicine Biobank, and All of Us) to conduct phenome-wide association (PheWAS) and clinical laboratory-wide association (LabWAS) meta-analyses of 4,813 individuals with SCT among 58,830 African genetic ancestry participants (∼60% female). Significant associations were replicated using a published PheWAS of SCT from the Million Veteran Program. Our PheWAS meta-analysis confirmed the

Uganda's Mandatory Newborn Sickle Cell Disease Screening Programme: Translating a Decade of Evidence into Differentiated Implementation.

Uganda's sickle cell disease (SCD) prevalence of 1.3% and sickle cell trait (SCT) prevalence of 13.3% nationally represent a substantial public health burden, with an estimated 20,000 affected infants born annually. On 9 February 2026, Uganda's Ministry of Health launched a nationwide mandatory newborn screening programme for SCD, placing the country among the first in sub-Saharan Africa to implement universal birth screening. This policy builds on over a decade of evidence, including the Uganda Sickle Surveillance Study, and cost-effectiveness analyses showing dried blood spot (DBS) screening

A machine-learning capillary electrophoresis-based pattern to identify suspected sickle cell trait and α-thalassemia co-inheritance.

OBJECTIVES: Sickle cell trait (HbAS) and α-thalassemia are common conditions. Although their hematological laboratory findings have been extensively characterized individually, there is limited evidence on the hematological pattern when they are co-inherited. Early and simple diagnosis of HbAS/α-thalassemia allows for optimization of the clinical approach and provision of appropriate genetic counselling. The objective is to identify, using machine learning algorithms, the most relevant hematological quantities and to establish a useful cut-off value for screening patients with suspected HbAS/α

Clinical Implications of Sickle Cell Thalassemia on Pregnancy Outcomes: A Systematic Review.

Background/Objectives: Sickle cell thalassemia (HbSβ-thal) and related sickle cell disease (SCD) genotypes are associated with substantial maternal, fetal, and neonatal morbidity during pregnancy. This systematic review aimed to summarize pregnancy outcomes and clinical management strategies in women with HbSβ-thal and related hemoglobinopathies, with an additional narrative comparison to sickle cell trait (SCT)-a distinct, heterozygous, and generally benign carrier state that is not a form of SCD. Methods: A systematic literature search was conducted in PubMed, MEDLINE, Web of Science, EMBASE

Exploring the anti-quorum sensing potential of synthetic lipstick dyes against Pseudomonas aeruginosa.

Lip care products such as balms, glosses, and lipsticks are widely used to enhance lip health, appearance, and protection. Lipsticks provide aesthetic appeal, social and psychological benefits, that act as a protective barrier against environmental stressors. However, growing concerns have emerged regarding chronic lip infections associated with multidrug-resistant bacteria, particularly Pseudomonas aeruginosa. This is especially concerning in individuals with conditions such as juvenile bullous pemphigoid, hypochromic microcytic anemia, and sickle cell trait. The persistence of P. aeruginosa

Classifying Sickle Cell Disease Subtypes from Clinical Reports: Algorithm Validation and ICD-10 Accuracy Assessment in Five French Hospitals.

INTRODUCTION: Research on rare diseases using large hospital and medico-administrative databases is expanding, yet precise patient characterization remains challenging. Sickle cell disease (SCD), given its different subtypes and the presence of sickle cell trait (SCT), illustrates these difficulties. METHODS: We developed and validated an algorithm using regular expressions to classify SCD subtypes and SCT from unstructured clinical reports. We included all documents issued between 2013 and 2025 of adult patients with at least one ICD-10 code for SCD in five French expert centers of the Great

Predicting locus of control for health in adults with sickle cell disease via mental health visits.

OBJECTIVE: This study examined the relationship between healthcare-seeking behavior and health locus of control orientations among individuals with sickle cell disease, specifically investigating whether mental health service utilization predicts internal versus external control beliefs while accounting for demographic factors. METHODS: Thirty-two African American adults with sickle cell disease were recruited from a Southeastern Comprehensive Sickle Cell center. Participants completed the Multidimensional Health Locus of Control scale (MHLC), which assesses internal control beliefs and exter

Genotype Differences and Hydroxyurea Utilization Among Adults With Moderate to Severe Sickle Cell Disease.

BACKGROUNDS: Hydroxyurea (HU) remains underutilized in adults with sickle cell disease (SCD) despite proven benefits. Current HU guidelines primarily target sickle cell anemia (SCA), overlooking other genotypes. OBJECTIVES: This study examined HU utilization patterns across genotypes among adults considered to have moderate to severe SCD manifestations by the 2014 National Heart, Lung, and Blood Institute (NHLBI) guideline criteria and identified factors associated with early HU use. METHODS: This retrospective cohort study analyzed electronic health records from the University of Pittsburgh

High concordance of physician-attestation with manual data abstraction for sickle cell type: an ASH RC Data Hub study.

Sickle cell disease (SCD) is the most common inherited blood disorder in the United States. The American Society of Hematology Research Collaborative (ASH RC) has built an SCD Data Hub (DH) to facilitate research and quality improvement using real-world data. To our knowledge, we report here the first analyses from the DH, focusing on cohort demographics and accurate identification of SCD diagnosis type. DH sites have data use agreements with ASH RC to extract and transfer electronic health record (EHR) data at least quarterly. Principal investigators (PI) provided additional attestation of SC

Evaluating alloimmunization risk in patients with sickle cell disease and in prenatal care patients with RHCE variants.

BACKGROUND AND OBJECTIVES: Clinically significant alloantibodies complicate transfusion and prenatal care, especially in individuals with genetic variants affecting high-frequency antigens. Many patients from African descent carry RHCE*ceVS alleles, which alter the expression of c (RH4), e (RH5) and hrB (RH31). However, the risk and clinical impact of alloimmunization remain uncertain. We evaluated the alloimmunization in a cohort of patients with a RHCE*ceVS genotype. MATERIALS AND METHODS: We conducted a retrospective study on 48 patients with a RHCE*ceVS genotype divided into three categor

Direct healthcare costs associated with sickle cell disease complications: a retrospective cohort study using routinely collected healthcare data in England.

OBJECTIVES: Due to the multisystemic nature of sickle cell disease (SCD), complications can occur together and thus discerning costs associated with individual complications requires a methodology that can estimate the costs of a given complication while accounting for the presence of other complications. In this study, we aimed to estimate period-based incremental costs associated with specific chronic complications in patients with SCD in England while accounting for multimorbidity. DESIGN/SETTING: All-cause primary and secondary care healthcare resource utilisation (HCRU) was obtained for

Epidemiological and comorbidity burden in transfusion-dependent patients with thalassemia and sickle cell disease in Greece.

TDT and SCD are hereditary hemoglobinopathies that require lifelong red blood cell transfusions. Modern treatment has increased survival, exposing patients to transfusion-related problems such iron excess. Despite this movement toward an elderly patient population, transfusion-dependent adult comorbidity data is scarce, especially in Greece. A retrospective, descriptive observational analysis of 105 adult transfusion-dependent patients (92 with TDT and 13 with SCD) followed at a Greek tertiary hemoglobinopathy center in 2024 was conducted. Organ system-specific chronic comorbidities were ident

Screening for sickle cell retinopathy: A comparison of systematic angiography and ultra-widefield imaging.

PURPOSE: Sickle cell disease is a prevalent genetic hemoglobinopathy affecting approximately 120 million people worldwide. Fluorescein angiography in the seven ETDRS fields is currently the gold standard for detecting proliferative forms of retinopathy, but ultra-widefield imaging (UWF) may represent a non-invasive alternative. We sought to compare the two screening strategies implemented in the Centre-Val de Loire region: systematic 7-field ETDRS fluorescein angiography at the University Hospital of Orléans versus non-angiographic ultra-widefield imaging (Optos©) at the University Hospital of

Priapism as an Initial Presentation of Sickle Cell Disease: A Case Report.

Priapism is a urological emergency defined as an erection lasting more than four hours unrelated to sexual stimulation. Most cases are ischemic and often related to hematologic disorders, especially sickle cell disease (SCD). About 30 to 40 percent of males with SCD experience priapism, usually starting in adolescence. The pathophysiology is linked to nitric oxide depletion, dysregulation of phosphodiesterase type 5, oxidative stress, and genetic factors. Although common, priapism is often overlooked and undertreated, which can lead to erectile dysfunction and psychological problems. A 21-year
