BACKGROUND: The value proposition for Shigella vaccines is strengthened by the potential for vaccines to prevent linear growth faltering. However, because expected effect sizes in Phase 3 vaccine trials are small due to limited Shigella incidence, a simple comparison of growth by randomized vaccine arm is likely underpowered and may yield null or even inverse results.
METHODS: We consider a new approach that estimates vaccine effects in the subgroup that would be infected in absence of vaccination, termed the naturally infected. In simulations parameterized by multi-site studies of diarrhea,

Shigella flexneri 2a is the most common cause of shigellosis, a major public health concern in developing countries. Rapid and reliable diagnostic tools are critical for timely outbreak detection and management. Leveraging clustered regularly interspaced short palindromic repeats (CRISPR) technology, we developed a CRISPR-Cas12a-based assay for the rapid and specific detection of S. flexneri 2a and validated its performance using stool specimens from patients. Two guide RNAs targeting the gtrII and gtrX genes, unique markers of the S. flexneri 2a serotype, were designed to ensure specificity.

Shigellosis is one of the leading causes of diarrhoeal disease worldwide. It mostly affects children under 5 years and elderly persons over 70 years in low- and middle-income countries (LMICs). The high disease incidence in LMICs and the rise of antimicrobial resistance have prompted efforts to develop novel vaccines against shigellosis. Several multicomponent vaccines are currently under development; to date, however, no correlate of protection against shigellosis has been defined. The Serum Bactericidal Activity assay (SBA) is regularly applied to measure the complement-mediated functional a

Background: As a major regulator, methyltransferase-like 3 (METTL3) catalyzes N6-methyladenosine (m6A) modification in mRNA. The m6A modifications mediated by METTL3 influence RNA splicing, nucleocytoplasmic distribution, stability, and other functions, thereby playing a vital and indispensable role in genetic regulatory network. Although several studies have shown its critical role in mRNA fate, the global pattern of mRNA methylation alteration driven by METTL3 remain unclear. Methods: Here, a HEK293T cell line with METTL3 depletion was constructed, and RNA sequencing (RNA-seq) and methylated

The review article is a critical examination of the growing global problem of multidrug-resistant (MDR) Salmonella and Shigella, focusing on their epidemiology, molecular resistance mechanisms, and clinical effects. It compiles current information on increasing resistance to major antibiotics, such as fluoroquinolones, third-generation cephalosporins, and sulfonamides, that is caused by a variety of factors, including ESBL production and plasmid-mediated gene transfer, especially in Asia, Africa, and the Middle East. The minireview discusses the new therapeutic approaches such as bacteriophage

INTRODUCTION: Salmonella and Shigella infections are two of the leading causes of global diarrheal deaths in children younger than 5 years. Licensed vaccines against Salmonella Typhi have been shown to be effective, and other Salmonella and Shigella vaccine candidates are being evaluated in clinical trials.
METHODS: We have previously described two 4-valent vaccines against Salmonella and Shigella strains separately. Here, we tested the combination of Salmonella and Shigella vaccines in animal models. ELISA was used to determine antibody titers against antigens, and bactericidal and opsonopha

ETHNOPHARMACOLOGICAL RELEVANCE: Baiyaozi, the dried tuberous root of Stephania cepharantha Hayata, has long been used in traditional Chinese medicine for acute hepatitis, bacillary dysentery, acute gastroenteritis, abdominal pain, and bleeding disorders, indications that collectively correspond to hepatic inflammation, intestinal mucosal injury, and the bleeding tendency characteristic of cirrhosis-associated intestinal barrier injury. This ethnopharmacological correspondence provides a rationale for evaluating Baiyaozi (Stephania cepharantha Hayata) as a candidate therapy for the cirrhotic in

Background. Recent outbreaks of gastrointestinal symptoms among gay, bisexual and other men who have sex with men (GBMSM) have been caused by multidrug-resistant (MDR) Shigella sonnei carrying bla CTX-M-27 or bla CTX-M-15. To date, the bla CTX-M-3 variant has not been associated with sexually transmitted shigellosis in the UK.Methods. Routine surveillance identified an outbreak S. sonnei among men in England in June 2025. Short-read sequencing data were analysed to identify antimicrobial resistance (AMR) determinants and to investigate the phylogenetic context of the outbreak. Long-read Oxford

BACKGROUND: Shigella causes severe diarrheal disease, and S. flexneri and S. sonnei are the targets for multivalent vaccine development. Culture-based agglutination has been the gold standard for serotyping, but it is limited by logistics, subjectivity, and the availability of antisera for emerging serotypes. Newer methods, including a real-time PCR-based approach and whole-genome sequencing offer alternatives, but their performance in Shigella endemic populations are not well documented.
METHODS: Shigella isolates obtained from the Enterics for Global Health (EFGH) study in Iquitos, Peru wer

BACKGROUND: Shigella causes severe bacterial diarrhea, especially in children in low- and middle-income countries. Rising antimicrobial resistance (AMR), including multidrug-resistant (MDR) and ESBL-producing strains, threatens empirical treatment, particularly cephalosporins.
METHODS: We conducted a structured narrative review of PubMed, Scopus, Web of Science, ScienceDirect, and WHO GLASS. Included studies comprised randomized trials, cohorts, case series, and relevant in vitro studies. Quality was appraised using Joanna Briggs Institute checklists; findings were synthesized narratively.

BACKGROUND: This study aimed to investigate the azithromycin resistance rate and the related genes in clinical Shigella species obtained from children with diarrhea in Ahvaz, southwest Iran. METHODS AND RESULTS: Shigella isolates were collected from children with diarrhea and identified using biochemical tests and polyvalent antisera. Antibiotic susceptibility testing was performed using disk diffusion. Minimum inhibitory concentrations (MICs) of azithromycin were determined by the agar dilution method in the presence and absence of efflux inhibitor, phenylalanine-arginine β-naphthylamide (PAβ

Background/Objectives: Shigella is the leading bacterial cause of diarrheal disease worldwide. Although multiple vaccine candidates are under development and in clinical trials, no Shigella vaccine is currently available on the market. Shigella comprises four species: S. dysenteriae, S. flexneri, S. boydii, and S. sonnei. S. flexneri has been recognized as the most prevalent species, particularly in low- and middle-income countries (LMICs), and the top serotypes are S. flexneri 2a, 3a and 6. Conversely, S. sonnei has a single serotype and predominates in high-income countries (HICs). Invasion

INTRODUCTION: The emergence of shigellosis caused by Shigella spp. poses a significant threat to global health, and the prevalence of multidrug-resistant (MDR) strains necessitates the development of effective antimicrobial therapies. Bacteriophages present a promising approach for both prevention and treatment of MDR infections.
METHODS: Ten lytic bacteriophages targeting Shigella flexneri 2457T were isolated from wastewater using the double-layer agar method. Phage morphology, lytic activity spectrum, latent period, burst size, bacterial killing efficiency, and efficacy under ex vivo microb

Shigella species frequently cause bacillary dysentery and remain a major public health concern in low- and middle-income countries. We report draft genomes of 22 Senegalese Shigella isolates including S. sonnei, S. flexneri, and S. boydii. Genome sizes range from 4.34 to 5.54 Mb, with GC contents ranging from 50.25% to 50.73%. These data provide a resource for genomic epidemiology and antimicrobial resistance of Shigella in West Africa.

BACKGROUND: Shigella is a leading cause of childhood diarrheal disease and a priority target for vaccine development, particularly in low-resource settings where its true burden is often underestimated. A clear understanding of local Shigella epidemiology and risk factors is essential for guiding control strategies and vaccine introduction.
METHODOLOGY/PRINCIPAL FINDINGS: We conducted a 12-month longitudinal cohort study enrolling 750 children under five years of age in a peri-urban area of Ouagadougou, Burkina Faso, from December 2020 to March 2021, with follow-up through March 2022. Active

Antimicrobial resistance (AMR) in Shigella is rising globally, complicating shigellosis management. Whole-genome sequence analysis (WGSA) has advanced our understanding of AMR and transmission dynamics, yet contemporary whole-genome sequencing data from Shigella in sub-Saharan Africa remain scarce. In this study, we applied WGSA to 27 Shigella isolates collected from children presenting with diarrhoea at Ndirande Health Centre in Malawi (2022-2023), as part of the Enterics for Global Health Shigella surveillance study. Serotyping, AMR profiling and phylogenetic analysis revealed Shigella sonne

The genus Shigella comprises four species: Shigella dysenteriae (serogroup A), Shigella flexneri (serogroup B), Shigella boydii (serogroup C), and Shigella sonnei (serogroup D), which cause shigellosis. The emergence and global spread of multidrug resistance make these species a severe threat to public health, prompting the World Health Organization (WHO) to designate fluoroquinolone-resistant Shigella as a high-priority pathogen. Shigella employs a repertoire of virulence factors to evade the host immune response, among which the lipopolysaccharide (LPS), a primary endotoxin, plays a key role

BACKGROUND: There are no licensed vaccines to protect against Shigella or enterotoxigenic Escherichia coli (ETEC), yet they are among the most common causes of bacterial diarrhea. We engineered an attenuated Shigella flexneri 2a expressing colonization factor antigen 1 (CFA/I) and heat-labile enterotoxin B subunit (LTB) from ETEC and conducted a first-in-human randomized-controlled trial of ascending doses of the vaccine (CVD 1208S-122).
METHODS: Dose-escalating cohorts received a single oral dose of 108-1010 colony-forming units (CFU) of CVD 1208S-122 or placebo. A fourth cohort received 1 o

BACKGROUND: Flooding from natural disasters has become increasingly common worldwide, increasing the risk of infectious disease transmission, particularly waterborne and vector-borne infections. Although many studies report outbreaks following floods, the evidence remains fragmented across regions and pathogens. A comprehensive review is needed to clarify these associations and inform effective prevention and response strategies.
METHODS: A systematic search of PubMed, Embase, and the Cochrane Library was conducted between November 2024 and January 2025, identifying studies published from Jan

The month of March is dedicated to Colon Cancer Awareness. Worldwide, colorectal cancer (CRC) incidence has been on the rise. It is currently the third most common cancer in men (746,000 cases, 10.0% of the total) and the second in women (614,000 cases, 9.2% of the total).1 Arecent meta-analysis reported a 61% risk reduction in CRC incidence with colonoscopy.2 Unlike screening programs for breast and prostate cancers, not only has CRC screening reduced mortality from colon cancer and detected early CRC, it has also decreased the incidence of CRC through detection and removal of pre-cancerous l
