Phenotypic profiling supports that Listeria costaricensis lacks in vitro pathogenicity but exhibits persistence traits.

Listeria costaricensis was recently isolated from a food-processing facility. However, its pathogenic potential and persistence capacity remain poorly characterized at the phenotypic level, raising concerns about food safety. Therefore, we assessed its pathogenic potential through phenotypic invasion and adhesion assays in HeLa cells, showing that L. costaricensis cannot invade HeLa cells despite displaying a cell adhesion capacity comparable to that of Listeria monocytogenes and Listeria innocua reference strains. Given the industrial origin of the strain, we further evaluated its susceptibil

Large-scale phenotyping and comparative genomics reveal genetic features of Listeria persistence in epithelial cells.

During infection in epithelial cells, after invading the cytosol, multiplying, and spreading, Listeria monocytogenes (Lm) ceases to produce ActA and becomes trapped in Listeria-containing vacuoles (LisCVs). These persistence acidic vacuoles harbor bacterial subpopulations that resist to stress in a metabolically dormant state. Although LisCVs have been proposed as a hallmark of Lm persistence in epithelial cells, their prevalence across strains and the bacterial factors underlying their formation remain uncharacterized. Given the significant genetic diversity within the species, it is importan

The conserved Pss exopolysaccharide is a common component of Listeria biofilms.

Listeria monocytogenes (Lm) is a major foodborne pathogen capable of persisting on fresh produce and in food-processing environments. Although exopolysaccharides (EPSs) are recognized as central components of resilient bacterial biofilms, Lm biofilms have long been considered to be EPS-less or EPS-poor. We previously identified a listerial EPS, Pss, whose biosynthesis is encoded by the highly conserved pss operon and is stimulated by elevated c-di-GMP levels. However, the prevalence of Pss in wild-type strains remained unclear, in part, due to the lack of a specific detection probe. Here, we e

Genetic Diversity, Biofilm Formation, and Antibiotic Resistance in Listeria monocytogenes Isolated from Meat-Processing Plants.

Listeria species are ubiquitous microorganisms that can be present all over the food chain. They can survive under adverse conditions and are frequently found in food-processing plants. In this study, 19 Listeria innocua and 19 Listeria welshimeri strains were isolated from meat product manufacturing companies in Spain, and biofilm formation capabilities were analyzed. In addition, 37 Listeria monocytogenes strains were also isolated, and their genetic diversity, biofilm formation capabilities, and antibiotic resistance were analyzed too. The species distribution was similar between two food-p

Genomic characterization of Listeria monocytogenes isolated from normally sterile human body fluids in Lithuania from 2016 to 2021.

Listeria monocytogenes is a saprophytic gram-positive bacterium and opportunistic foodborne pathogen that can cause listeriosis in humans. The incidence of listeriosis has been rising globally and, despite antimicrobial treatment, the mortality rates associated with the most severe forms of listeriosis such as sepsis, meningitis and meningoencephalitis remain high. The notification of listeriosis in humans is mandatory in Lithuania, and up to 20 cases are reported annually. However, no studies have described the detailed virulence and antimicrobial susceptibility profiles of any clinical L. mo

Chicken Embryo as an In Vivo Model to Revive Viable but Non-Culturable Pathogens.

The chicken embryo has emerged as a popular in vivo model with increasing application in biomedical research due to its simplicity, affordability, and adaptability in the study of various biological phenomena. This model has been used to investigate microbial pathogenicity and is becoming a useful tool to study bacterial dormancy. The viable but non-culturable (VBNC) state is a dormant state in which bacteria become metabolically quiescent and resistant to cultivation to preserve their viability in harsh environments. Under favorable conditions, VBNC bacteria can wake up back into a metabolica

Survival of Listeria monocytogenes biofilms and planktonic cells on cooked ham in a contamination-storage-digestion process.

In 2023, listeriosis experienced a significant increase in Europe, with 2952 invasive cases and 19.70 % mortality rate, being the ready-to-eat foods category identified as the mostly involved. This study focused on the survival of L. monocytogenes through a process of food contamination-storage-digestion. For this, cooked ham was artificially contaminated with biofilm or planktonic cells, followed or not by vacuum-packed storage at 7 °C for 7 days, and submitted to in vitro digestion. Enumeration analysis was performed at different stages during the contamination-storage-digestion process. Con

Ampicillin-induced resistant variants of Listeria monocytogenes: effects on growth, survival, and virulence.

Listeria monocytogenes is a major foodborne pathogen whose rising antibiotic resistance poses increasing public health concerns. Understanding how resistance acquisition affects bacterial physiology is essential for accurate microbial risk assessment. In this study, five antibiotic-sensitive foodborne L. monocytogenes strains were subjected to ampicillin (AMP) exposure to obtain AMP-resistant variants. Paired susceptible and resistant strains were compared for growth kinetics at 25 °C, 30 °C, and 37 °C; tolerance to heat (60 °C) and acid stress (pH 2.5); biofilm formation; and in vivo virulenc

The chaperone PrsA2 regulates the secretion, stability, and folding of listeriolysin O during Listeria monocytogenes infection.

UNLABELLED: Pathogenic bacteria rely on secreted virulence factors to cause disease in susceptible hosts. However, in Gram-positive bacteria, the mechanisms underlying secreted protein activation and regulation post-membrane translocation remain largely unknown. Using proteomics, we identified several proteins that are dependent on the secreted chaperone PrsA2. We followed with phenotypic, biochemical, and biophysical assays and computational analyses to examine the regulation of a detected key secreted virulence factor, listeriolysin O (LLO), and its interaction with PrsA2 from the bacterial

Molecular definition of the endogenous Toll-like receptor signalling pathways.

Innate immune pattern recognition receptors, such as the Toll-like receptors (TLRs), are key mediators of the immune response to infection and central to our understanding of health and disease1. After microbial detection, these receptors activate inflammatory signal transduction pathways that involve IκB kinases, mitogen-activated protein kinases, ubiquitin ligases and other adaptor proteins. The mechanisms that connect the proteins in the TLR pathways are poorly defined. To delineate TLR pathway activities, we engineered macrophages to enable microscopy and proteomic analysis of the endogeno

Aquatic environment drives the emergence of cell wall-deficient dormant forms in Listeria.

Stressed bacteria can enter a dormant viable but non-culturable (VBNC) state. VBNC pathogens pose an increased health risk as they are undetectable by growth-based techniques and can wake up back into a virulent state. Although widespread in bacteria, the mechanisms governing this phenotypic switch remain elusive. Here, we investigate the VBNC state transition in the human pathogen Listeria monocytogenes. We show that bacteria starved in mineral water become VBNC by converting into osmotically stable cell wall-deficient coccoid forms, a phenomenon that occurs in other Listeria species. We reve

Current methodologies available to evaluate the virulence potential among Listeria monocytogenes clonal complexes.

Listeria monocytogenes is a foodborne pathogen that causes listeriosis in humans, the severity of which depends on multiple factors, including intrinsic characteristics of the affected individuals and the pathogen itself. Additionally, emerging evidence suggests that epigenetic modifications may also modulate host susceptibility to infection. Therefore, different clinical outcomes can be expected, ranging from self-limiting gastroenteritis to severe central nervous system and maternal-neonatal infections, and bacteremia. Furthermore, L. monocytogenes is a genetically and phenotypically diverse

Isolation, characterization and genomic analysis of the novel Listeria bacteriophage LMLPA3 as a potential antimicrobial in foods.

Listeria monocytogenes (LM) is an opportunistic foodborne pathogen responsible for listeriosis in both humans and animals. The disease manifests in a variety of ways, including febrile gastroenteritis, septicemia, meningitis, and in some cases, preterm birth and spontaneous abortion. It is therefore crucial to develop effective strategies to control this bacterium. In this study, we isolated and characterized a novel Listeria phage, named LMLPA3. Morphological and genomic analyses revealed that phage LMLPA3 belongs to the class Caudovirales, family Herelleviridae, Myovirus-like. Phage LMLPA3 d

Marked inter-strain heterogeneity in the differential expression of some key stress response and virulence-related genes between planktonic and biofilm cells in Listeria monocytogenes.

Listeria monocytogenes is a facultatively intracellular pathogenic bacterium that can provoke invasive listeriosis, a severe foodborne infection in humans. Outside the host, this is capable to survive for long periods in soil, and water, as well as on plants, while, like many other microorganisms, this can also attach to abiotic surfaces, such as food contact ones, forming biofilms on them. It has been suggested that inside those sessile communities, L. monocytogenes cells not only display an increased stress tolerance but may also boost their pathogenicity. In this work, the expression of ten

Large Multicountry Outbreak of Invasive Listeriosis by a Listeria monocytogenes ST394 Clone Linked to Smoked Rainbow Trout, 2020 to 2021.

Whole-genome sequencing (WGS) has revolutionized surveillance of infectious diseases. Disease outbreaks can now be detected with high precision, and correct attribution of infection sources has been improved. Listeriosis, caused by the bacterium Listeria monocytogenes, is a foodborne disease with a high case fatality rate and a large proportion of outbreak-related cases. Timely recognition of listeriosis outbreaks and precise allocation of food sources are important to prevent further infections and to promote public health. We report the WGS-based identification of a large multinational liste

Polysome-CAGE of TCL1-driven chronic lymphocytic leukemia revealed multiple N-terminally altered epigenetic regulators and a translation stress signature.

Some of the phylogenetic lineages of Listeria monocytogenes are more likely to cause invasive disease in humans than are strains from other phylogenetic lineages. This suggests that strains belonging to these lineages display different levels of pathogenicity. To investigate this, we carried out a plaque-forming assay with HT-29 cells to evaluate the virulence of six perinatal strains from the three lineages that compose the species. All of the strains were largely over the 3.34 cutoff (between 4.29 and 5.97 mean log) with the HT-29 model and can therefore be considered to be equally virulent.
