A rising threat: Emergence of Listeria monocytogenes infection from hilly terrain of North India.

Listeria monocytogenes is a foodborne pathogen capable of causing severe invasive infections, particularly meningitis, in neonates, immunocompromised individuals, and pregnant women. This report represents a neonate with multiple complications including meningitis. This case underscores the diagnostic difficulties and therapeutic challenges posed by L. monocytogenes, especially in high-risk populations. Early recognition, timely antimicrobial therapy, and close long-term follow-up are critical for improving outcomes in affected patients and minimizing the risk of neurological sequelae or fatal

Automated high-throughput microscopy screening unveiled new Listeria monocytogenes genes involved in cell infection.

To uncover novel genetic factors required for Listeria monocytogenes cell infection, we developed an automated high-throughput microscopy screening pipeline that integrates GFP-expressing bacteria with machine learning-based image analysis. Using this approach, we screened a mariner transposon library comprising 4224 L. monocytogenes EGDe mutants and identified 58 with significantly reduced numbers of intracellular bacteria. Sequencing revealed 24 unique insertion sites corresponding to 14 genes, including previously known virulence factors and nine novel candidates not previously implicated i

Evolved populations of Listeria monocytogenes related to biofilm formation and biocide stress in the context of food production environment niches.

Cleaning and disinfection of food production environments (FPE) are fundamental components of food safety programmes designed to control microbial pathogens and prevent food contamination. Yet, FPE can still harbour foodborne pathogens, including Listeria monocytogenes, a significant concern to food manufacturers and health authorities due to the high mortality rate associated with invasive listeriosis. Mechanisms contributing to L. monocytogenes persistence in FPE include biofilm formation and reduced susceptibility to biocides, such as benzalkonium chloride (BC), for which several mechanisms

Protective role of the gut microbiota against Listeria monocytogenes: From colonization resistance to therapeutic approaches.

Listeria monocytogenes (LM) is a significant foodborne pathogen with considerable resilience in diverse environments. Following ingestion via contaminated food, LM can breach the intestinal barrier and infect target organs, causing systemic infection. This breach represents a critical step in its pathogenesis. The gut microbiota, a key component of intestinal defense, can restrict the colonization and invasion of the pathogen through mechanisms such as nutrient competition and bacteriocin production. In response, LM has evolved counterstrategies to enhance its survival and invasiveness in the

Dietary antigens drive the generation of functional cytotoxic intraepithelial lymphocytes for early defense against foodborne pathogens.

INTRODUCTION: Intraepithelial lymphocytes (IELs) constitute the largest lymphocyte population in the body and exhibit direct cytotoxic effector functions. Despite their abundance and importance in mucosal immunity, the mechanisms governing the generation and maintenance of functional IELs remain incompletely understood. Given the predominance of dietary components in the small intestine, dietary antigens may play a critical role in regulating the generation, maintenance, and functional maturation of IELs. METHODS: To assess the relative contributions of gut microbiota and dietary antigens to

Rapid Raman Fingerprinting Identification of Listeria monocytogenes via Immunomagnetic Enrichment Coupled with Droplet-Shrinkage-Assisted SERS.

Rapid identification of Listeria monocytogenes (L. monocytogenes) is critical to preventing outbreaks of foodborne listeriosis. Conventional methods required immunoassay or nucleic acid sequence recognition for indirect/labeled detection or identification of L. monocytogenes, separately. Herein, a facile label-free fingerprinting method was developed for direct identification of L. monocytogenes using immunomagnetic enrichment coupled with droplet-shrinkage-assisted surface-enhanced Raman spectroscopy (SERS). Immunomagnetic beads were used for specific enrichment of L. monocytogenes from the c

Evaluating the Performance of Loop-Mediated Isothermal Amplification for the Detection of Listeria monocytogenes Biofilms on Stainless Steel Surfaces.

L. monocytogenes is the causative agent of human listeriosis, a deadly disease with fatality rates up to 20%. L. monocytogenes has the ability to grow under harsh environmental conditions. It can form biofilms in food industries, making it capable of persisting in facilities. Given this scenario, it is of utmost importance to rapidly detect this bacterium not only in foods but also on food-contact surfaces. For the successful outcome of any given detection technology, it is imperative to properly process the samples. In the present work, PBS, LPT, and LPT-Pronase were compared to determine whi

Dryocrassin ABBA Inhibits the Function of LLO and SortaseA to Alleviate the Virulence of Listeria monocytogenes.

Listeria monocytogenes is a deadly foodborne pathogen, its virulence factors Listeriolysin O (LLO) and Sortase A (SrtA) play a critical role in its infection and have been identified as key targets for developing its inhibitor. This study reveals that the natural compound Dryocrassin ABBA (ABBA) alleviates the toxicity of L. monocytogenes by effectively inhibiting the functions of LLO and SrtA. ABBA bound to the binding pocket of LLO and inhibited the formation of its oligomers, which in turn reduced the ability of LLO to lyse mammalian erythrocytes. When co-cultured with L. monocytogenes, ABB

Tissue-specific microbiota dictates the competitive dynamics of listeria species colonization.

The genus Listeria is heterogeneous and contains pathogenic and nonpathogenic species. Pathogenic L. monocytogenes and L. ivanovii have different environmental distributions, infect different hosts, and cause distinct syndromes. Here, we evaluated whether responses of different Listeria species to diverse host niches contribute to virulence heterogeneity and influence their environmental distribution. We assessed resistance to gastric and intestinal fluids, gut and uterus microbiota, and semen. L. monocytogenes was more resistant than L. ivanovii in gastric fluid, whilst L. seeligeri and L. va

Taurine attenuates Listeria monocytogenes-induced inflammation and pyroptosis in mouse model by regulating MAPK and NLRP3/caspase-1/GSDMD pathways.

UNLABELLED: Listeria monocytogenes, as a significant foodborne pathogen, is not frequently encountered; however, when infections do occur, they can prove highly lethal to specific populations. Antibiotics are still regarded as the primary treatment option for Listeria infections. Nevertheless, under the global antibiotic crisis, there is an urgent demand for innovative and alternative strategies. In our study, we identified taurine, a sulfur-containing free amino acid that can be extracted from a wide variety of foods, as an effective inhibitor of Listeria growth. Furthermore, our findings rev
