Listeria monocytogenes poses a significant threat in food service environments due to its environmental robustness and pathogenicity. In this study, L. monocytogenes contamination across 24 catering facilities in Chengdu highlighted potential public health concerns, with a 6.43% (40/622) prevalence rate and meat processing areas as primary hotspots (13.48%, 19/141; cleaned meat: 25.0%, 6/24; cleaning pools: 20.83%, 5/24). Cross-contamination pathways from raw to cooking area were identified in 25% of facilities. Genomic characterization showed serotype 1/2a dominance (65.0%, 26/40), with 82.5%

Bacterial pathogens must possess finely tuned physiological adaptations to adapt to their infectious niche. One such niche inhabited by Listeria monocytogenes (L. monocytogenes) is the host cell cytosol, a compartment characterized by significant barriers to entry, metabolic limitation, and immune surveillance. Previously, we identified L. monocytogenes transposon mutants defective for intracellular survival due to disruptions in key metabolic pathways, including cell wall biosynthesis, menaquinone production, and pyruvate metabolism. Here, we demonstrate that mutations in the pyruvate dehydro

Chitin, the second most abundant polymer in nature, is recognized for biocompatibility and diverse effects on microbes and the host. However, the role of dietary chitin against foodborne pathogens such as Listeria monocytogenes remains underexplored. Here, we show that chitin functions as an environmental signal that attenuates L. monocytogenes virulence and mitigates infection outcomes. Consistent with previous studies, exposure of L. monocytogenes to chitin resulted in downregulation of key virulence genes and we demonstrate that this is accompanied by reduced pathogenesis in intestinal epit

BACKGROUND: Human listeriosis, caused by Listeria monocytogenes, is a serious food borne illness. Information from Pakistan on human listeriosis is sparse. This study reports the emergence and genomic characteristics of human L. monocytogenes cases from Pakistan between 2017 and 2023. L. monocytogenes cases were identified from our laboratory database in specimens submitted for culture and a commercially available multiplex PCR assay for cerebrospinal fluid from 2017 to 2023. Whole genome sequencing was performed on randomly selected isolates collected during 2022-2023. Frequency and geographi

Methylglyoxal is a reactive aldehyde produced by macrophages as part of their antimicrobial innate immune arsenal. Our prior work showed that Listeria monocytogenes relies on glyoxalase A (GloA) and bacterial glutathione to detoxify methylglyoxal and that loss of GloA severely impairs bacterial virulence in mice and results in a 100 to 1,000 increase in bacterial mutation frequency. Glutathione is required for both methylglyoxal detoxification and for allosteric activation of the master virulence regulator PrfA, underscoring its central, yet complicated role in pathogenesis. We previously demo

Interferon-induced transmembrane (IFITM) proteins underpin antiviral responses, yet their role in bacterial infections remains underexplored, particularly for parasites. We probed the role of IFITM1 in Mycobacterium tuberculosis (Mtb), Listeria monocytogenes (Lm), and Leishmania major infection using IFITM1 knockout mice. Notably, IFITM1 was upregulated in murine and human macrophages, as well as in PBMCs during active tuberculosis (TB), subsiding with therapy. IFITM1 also accumulated in the lungs of outbred mice and macaques that progressed to TB. IFITM1-deficient mice displayed no difference

Two cross-sectional studies determined the prevalence of antimicrobial resistance (AMR) in Listeria monocytogenes and Listeria spp. isolated from cattle farms in Mpumalanga and North West provinces in South Africa. A total of 100 isolates comprise 12 and 44 of L. monocytogenes and Listeria spp., respectively, from Mpumalanga province and 3 and 41 from North West province. The disc diffusion method was used to determine the isolates' resistance to 16 antimicrobial agents. The studies also investigated the factors (district, farm type, and sample type) associated with AMR. All 100 isolates teste

Mucosal-associated invariant T (MAIT) cells are among the most conserved and abundant innate-like T cells in humans that recognize microbial-derived riboflavin precursors and elicit potent antimicrobial responses. The foodborne pathogen Listeria monocytogenes is a broad host-range facultative intracellular pathogen that lacks the riboflavin biosynthetic pathway, leading us to hypothesize that this deficiency is pathoadaptive and allows the pathogen to evade MAIT cells. Here, we show that L. monocytogenes strains engineered to produce riboflavin (L. monocytogenes-ribDEAHT) are attenuated in wil

Listeria monocytogenes is a Gram-positive foodborne pathogen that causes listeriosis with a high mortality rate among high-risk populations. Although combination therapy with ampicillin and gentamicin is the standard treatment, the emergence of multidrug-resistant strains poses a growing public health threat. To explore the mechanism of multidrug resistance, the strain 466-3LM was analyzed using antimicrobial susceptibility testing, plasmid stability assessment, whole genome sequencing, and conjugation experiments in this study. Results showed that 466-3LM was resistant to trimethoprim/sulfame

Foodborne pathogen Listeria monocytogenes causes listeriosis, a rare but deadly condition. Internalin A (InlA) and Listeriolysin O (LLO), its main virulence factors, facilitate adhesion, invasion, intracellular survival, and intercellular spreading, making them interesting therapeutic targets. L. monocytogenes infections are becoming harder to treat because of antibiotic resistance; hence, flavonoids are being considered. An integrated in-silico technique was used to test plant-derived flavonoids' inhibitory efficacy against these proteins. For both targets, three modes of docking (HTVS, SP, a

Listeria monocytogenes (LM) is found in various environmental sources, including animal intestinal tracts, soil, and sewage. LM can contaminate various food products, including meat, seafood, cheese, fruits, and vegetables. Individuals with underlying medical conditions, the elderly, pregnant women, and infants, may develop meningitis and sepsis upon consuming LM-contaminated food. A rapid and standardized quantification method for evaluating initial contamination levels of LM in food remains to be established. This study aimed to develop a rapid quantification method to monitor LM contaminati

BACKGROUND: Listeria monocytogenes is a rare ocular pathogen, and posterior segment infection is particularly uncommon. When it involves the retina, its presentation may overlap with viral hypertensive uveitis or necrotizing viral retinitis, especially in eyes with unilateral ocular hypertension, iris atrophy, and chronic recurrent inflammation. This diagnostic overlap may delay recognition of an unusual bacterial cause.
FINDINGS: A 23-year-old male presented with progressive visual loss in the left eye after a five-year history of recurrent unilateral ocular redness and previously documented

Innate immune activation is tightly coupled to metabolic remodeling, yet how STING-associated signaling intersects with macrophage metabolism during bacterial infection remains incompletely understood. Here, using Listeria monocytogenes infection models, we show that infection is accompanied by increased glycolytic gene expression, glucose consumption, and lactate production in macrophages, and that STING deficiency attenuates these metabolic changes, whereas cGAS deficiency has a comparatively limited effect under our experimental conditions. Reduced glucose availability, 2-deoxy-d-glucose, a

Listeriosis, caused by Listeria monocytogenes (L. monocytogenes), is a severe foodborne infection linked to neonatal meningitis and high mortality. However, long-term epidemiological trends in China remain undercharacterized. We conducted a 60-year literature-based review of listeriosis cases in China (1961-2020) and integrated publicly available genomic data from Chinese clinical L. monocytogenes isolates to explore molecular epidemiology and evolutionary patterns. Our analysis identified three distinct epidemiological periods: Sentinel (-1999), Growth (2000-2009), and High-Incidence Plateau

Listeria monocytogenes is an important foodborne pathogen and is a common contaminant of raw milk and meat products. It is also found in many sources such as soil, water, and vegetables. Pathogenic L. monocytogenes is the etiological agent of listeriosis that mainly affects certain risk groups such as the elderly, pregnant women, neonates, and immunocompromised patients. Of note, L. monocytogenes is almost exclusively transmitted to humans through food. New biological and environmental tools are needed to inhibit the proliferation of foodborne pathogens and to prevent spoilage microorganisms t

Peeled hard-cooked eggs (HCEs) are ready-to-eat foods susceptible to Listeria monocytogenes contamination during postprocessing. A 2019 multistate listeriosis outbreak linked to HCEs treated with 0.3% citric acid highlighted the need for effective antimicrobial treatment. This study evaluated the efficacy of organic acid treatments (0.3% and 2% citric, and 2% acetic, lactic, and malic acids) at 5 °C and 25 °C against low-level contamination (1 log CFU/HCE) on HCEs during 28-day refrigerated storage (5 °C). Two contamination scenarios were assessed: inoculation prior to acid treatment (simulati

Listeriosis, caused by Listeria monocytogenes (L. monocytogenes), is an important foodborne disease characterized by low incidence but high case fatality rates. Its prevalence varies across regions and is influenced by hygiene practices, food handling, and environmental conditions. In developing countries like Ethiopia, data on L. monocytogenes is often limited due to a lack of awareness. This study aims to assess the prevalence of L. monocytogenes in retail meat and dairy products and evaluate their antimicrobial profiles. A total of 422 food samples were collected from May 2022 to March 2023

Zoonotic infections constitute a major cause of reproductive failure and perinatal mortality in humans and animals. Among the most relevant etiological agents are Listeria monocytogenes, Brucella spp., and Toxoplasma gondii, which, despite distinct biological properties, share the capacity to breach placental defenses, establish intracellular niches, and impair fetal development. This review aims to synthesize current knowledge on the pathological and pathogenic mechanisms underlying fetal and neonatal death associated with these pathogens, with a focus on placental infection, vertical transmi

Listeria monocytogenes is an important food-borne pathogen that is responsible for contamination of a variety of food products. It causes listeriosis which is one of the most severe and serious foodborne diseases, whose main severe clinical forms include septicaemia, meningitis, and mateoneonatal infections. Also because of its ability to adhere to industrials' surfaces, it is difficult for them to come up against this danger. Although there are researches in L. monocytogenes proteome, most of them are based on the study of a single strain, yet little is known about the proteome of more of the

In Canada in 2024, an outbreak of Listeria monocytogenes occurred, linked to pasteurised plant-based dairy alternative beverage products. The outbreak involved 20 confirmed cases (age range: 7-89 years; 13 females, seven males; 14 having underlying medical conditions) and resulted in three deaths. All cases were infected with the same genetic strain of L. monocytogenes. Nineteen of the 20 outbreak-confirmed cases reported consuming the same brand of plant-based beverage product prior to illness onset, and the outbreak strain was found in six open leftover plant-based beverage samples collected
