Occurrence and antimicrobial resistance profile of Listeria monocytogenes isolated from chilled processed meat products in the Metropolitan Region of Recife, Brazil.

The presence of Listeria monocytogenes in food represents a major challenge to food safety, especially in ready-to-eat products stored under refrigeration. This risk is associated with the ability of the pathogen to survive and multiply during prolonged refrigerated storage, making these foods potential sources of infection and a relevant public health concern. In this context, the aim of this study was to investigate the presence of Listeria monocytogenes in chilled processed meat products sold in bulk in the Metropolitan Region of Recife, PE, Brazil, as well as to evaluate the antimicrobial

LIPI-4 and lmo0180 cooperatively regulate flagellar motility and virulence in Listeria monocytogenes.

INTRODUCTION: Deletion of Listeria pathogenicity island 4 (LIPI-4) in Listeria monocytogenes (L. monocytogenes) impairs motility, disrupts flagellar assembly, and markedly upregulates the lmo0180 gene. METHODS: In this study, the Δlmo0180 single-gene deletion mutant and ΔLIPI-4/lmo0180 double-gene deletion mutant were constructed by homologous recombination. Bacterial motility was assessed, and flagellar biogenesis was observed by transmission electron microscopy. The adhesion, invasion, intracellular proliferation, and cell-to-cell spread capacities were compared across strains. Virulence wa

Clinical Characteristics and Outcomes of Invasive Listeria Monocytogenes Infection: A 45-Year Retrospective Study at a US Tertiary Center.

BACKGROUND: Invasive listeriosis remains a leading cause of foodborne infection-related mortality in the United States and is associated with substantial case fatality despite appropriate antimicrobial therapy. Owing to the relative rarity of invasive disease, contemporary data evaluating clinical characteristics, optimal antimicrobial strategies, and predictors of mortality remain limited. METHODS: We conducted a retrospective cohort study of adults with laboratory-confirmed invasive Listeria monocytogenes infection at Mayo Clinic Health System sites from 1980 to 2024. Clinical syndromes wer

Aggressive brainstem listeria abscess in an immunocompetent individual: A case report.

Listeria monocyotogenes, a gram-positive bacterium is transmitted to humans through ingestion of contaminated foods, including deli meats, soft cheeses, and raw vegetables but can also thrive in soil, water, sewage, manure and animal feed. Its infection in immunocompetent patients is fairly limited to gastrointestinal distress but carries a high mortality rate in immunocompromised patients especially when affecting the central nervous system. We present a challenging case of an immunocompetent patient in their late 50s with neurolisteriosis presenting as multifocal brainstem abscesses with an

Phenotypic profiling supports that Listeria costaricensis lacks in vitro pathogenicity but exhibits persistence traits.

Listeria costaricensis was recently isolated from a food-processing facility. However, its pathogenic potential and persistence capacity remain poorly characterized at the phenotypic level, raising concerns about food safety. Therefore, we assessed its pathogenic potential through phenotypic invasion and adhesion assays in HeLa cells, showing that L. costaricensis cannot invade HeLa cells despite displaying a cell adhesion capacity comparable to that of Listeria monocytogenes and Listeria innocua reference strains. Given the industrial origin of the strain, we further evaluated its susceptibil

Large-scale phenotyping and comparative genomics reveal genetic features of Listeria persistence in epithelial cells.

During infection in epithelial cells, after invading the cytosol, multiplying, and spreading, Listeria monocytogenes (Lm) ceases to produce ActA and becomes trapped in Listeria-containing vacuoles (LisCVs). These persistence acidic vacuoles harbor bacterial subpopulations that resist to stress in a metabolically dormant state. Although LisCVs have been proposed as a hallmark of Lm persistence in epithelial cells, their prevalence across strains and the bacterial factors underlying their formation remain uncharacterized. Given the significant genetic diversity within the species, it is importan

The conserved Pss exopolysaccharide is a common component of Listeria biofilms.

Listeria monocytogenes (Lm) is a major foodborne pathogen capable of persisting on fresh produce and in food-processing environments. Although exopolysaccharides (EPSs) are recognized as central components of resilient bacterial biofilms, Lm biofilms have long been considered to be EPS-less or EPS-poor. We previously identified a listerial EPS, Pss, whose biosynthesis is encoded by the highly conserved pss operon and is stimulated by elevated c-di-GMP levels. However, the prevalence of Pss in wild-type strains remained unclear, in part, due to the lack of a specific detection probe. Here, we e

AddaVax-Adjuvanted Inactivated Vaccine Confers Protection Against Listeria monocytogenes Challenge in Mice.

Listeria monocytogenes (Lm) is a facultative intracellular pathogen responsible for life-threatening listeriosis, characterized by high mortality rates. Currently, there is no vaccine available for the prevention or treatment of listeriosis. Here, we developed two adjuvanted inactivated vaccines and evaluated their safety, immunogenicity, and protective efficacy. The hypervirulent Lm strain XYSN was inactivated with β-propiolactone (BPL) and formulated with AddaVax or Alum to produce inactivated vaccines (ADIV and ALIV) respectively. Local reactogenicity and histopathological observations reve

Genomic and pathogenic characterization of Listeria ivanovii subsp. londoniensis from ovine farms in China.

BACKGROUND: Listeria ivanovii (Liv) is an anaerobic intracellular pathogen primarily responsible for listeriosis in ruminants, presenting with clinical manifestations such as gastroenteritis, bacteremia and abortion. Liv contains two validated subspecies: Listeria ivanovii subsp. ivanovii (Liv-I) and Listeria ivanovii subsp. londoniensis (Liv-L). Hitherto, the genomics and pathogenic features of Liv-L remain poorly understood. RESULTS: Here, we characterized the genomic diversity and pathogenic traits of 10 Liv isolates from five ovine farms in eastern China with a high prevalence of listerios

Bacteriophages as Antibacterial Agents Against Bovine Pathobionts Associated with Foodborne Human Morbidity.

Rates of foodborne infectious disease are increasing globally. The One Health zoonoses report shows increasing cases of shigatoxigenic Escherichia coli, campylobacteriosis, salmonellosis and listeriosis in the last 5 years. The ESKAPE pathogens are the top priority due to their alarming rate of resistance to broad-spectrum beta-lactams, carbapenems, glycopeptides, fluoroquinolones, aminoglycosides and biocide solutions. Research assessing alternative biocontrol options highlight the advantages of bacteriophages in the control of resistant bacterial species. Phage formulations including ListShi

Listeria ivanovii-an underestimated pathogen in veterinary medicine.

Listeria (L.) ivanovii is a Gram-positive, facultatively intracellular rod-shaped bacterium. It is predominantly associated with animal infections, particularly in ruminants. Unlike L. monocytogenes, which is widely recognised as a major zoonotic and foodborne pathogen, L. ivanovii is often overlooked, despite documented cases of abortion, stillbirth, and neonatal septicemia in sheep, goats, and cattle. This review provides an overview of L. ivanovii taxonomy, microbiological characteristics, virulence determinants, host adaptation, epidemiology, transmission routes, diagnostic challenges, ant

The two-component system histidine kinase LiaS contributes to the stress resistance and virulence of zoonotic Listeria monocytogenes.

LiaS is a histidine kinase receptor in two-component systems that senses environmental stress signals. This study investigated the roles of LiaS in Listeria monocytogenes stress resistance and virulence. Phenotypic assays showed that deletion of liaS (ΔliaS) resulted in significantly impaired growth under acidic, alkaline, and osmotic stresses, as well as reduced survival under strong acidic conditions. These defects were partially restored in the complemented strain (CΔliaS). The growth defects were also observed in the ΔliaS under Cu2+ or H2O2 exposure. Furthermore, the ΔliaS strain exhibite
