BACKGROUND/AIM: This retrospective study examined whether long-term SGLT-2i use is associated with differences in thyroid-stimulating hormone (TSH) and the systemic immune-inflammation index (SII) in patients with type 2 diabetes mellitus (T2DM) without known thyroid disease. The neutrophil-to-lymphocyte ratio (NLR), renal function (eGFR), diabetes duration, and metformin use were also evaluated as potential covariates.
METHODS: This was a single-center, retrospective observational study. Adults with T2DM who had used empagliflozin or dapagliflozin for at least six months were compared with a

BACKGROUND: Autosomal dominant polycystic kidney disease (ADPKD) is characterized by progressive cyst growth, kidney enlargement, and decline in renal function. Increasing evidence suggests that inflammatory processes may contribute to disease progression. This study aimed to evaluate the association between tolvaptan use, systemic inflammatory markers, and total kidney volume (TKV) progression in patients with ADPKD.
METHODS: This retrospective study included 67 patients with ADPKD, including 40 receiving tolvaptan and 27 untreated controls. Neutrophil-to-lymphocyte ratio (NLR), platelet-to-

Sickle cell disease (SCD) is characterized by hemolytic anaemia, painful vaso-occlusive crises, and chronic inflammation resulting partly from platelet activation. It has been shown that platelets express the nucleotide-binding domain leucine-rich repeat containing protein 3 (NLRP3) and the apoptosis-associated speck-like protein containing a CARD (ASC), forming the NLRP3 inflammasome, which controls activation of caspase-1 and cleavage of interleukin-1β (IL-1β). Bruton tyrosine kinase (BTK), known to activate the NLRP3 inflammasome, is also expressed in platelets. In SCD, platelets are activa

Neutrophil extracellular trap (NET) formation, termed NETosis, is a key driver of lung tissue injury in acute lung injury/acute respiratory distress syndrome (ALI/ARDS). However, the precise mechanisms that trigger neutrophil NETosis remain incompletely understood. Here, we observed that neutrophils from both ALI mice and ARDS patients exhibited NETosis alongside a pronounced selective metabolic shift, characterized by enhanced oxidative phosphorylation (OXPHOS) and attenuated glycolytic activity. Phosphoproteomic profiling revealed that this shift was driven by aberrant activation of pyruvate

BACKGROUND: Psoriasis is a chronic inflammatory skin disease characterized by aberrant keratinocyte proliferation and dysregulated immune responses. Increasing evidence suggests that reciprocal communication between tissue-resident keratinocytes and infiltrating immune cells shapes the inflammatory microenvironment and governs disease progression. However, the molecular mechanisms underlying this immune-parenchymal crosstalk remain incompletely understood.
METHODS: Here, we generate epidermal-specific Med1 conditional knockout (cKO) mice using the Cre-loxP system and induce psoriasis model wi

Lactate, traditionally regarded as a product of glycolytic metabolism, is increasingly recognized as a metabolite that can influence cellular signaling and protein post-translational modification, including lysine lactylation (Kla). Neutrophils rely on glycolytic metabolism to support rapid effector functions and encounter substantial metabolic changes during inflammation, infection, ischemic injury, and tumor progression. Emerging studies have implicated histone and non-histone lactylation in the regulation of neutrophil-associated processes, including migration, inflammatory activation, neut

Pneumonia in children is frequently accompanied by excessive inflammatory responses and immune dysregulation, which may lead to severe clinical outcomes. Catechin has shown anti-inflammatory activity in multiple disease settings, but its role and molecular mechanisms in pneumonia remain unclear. Public bulk RNA-sequencing data from pediatric pneumonia (GSE261482) and public mouse lung single-cell RNA-sequencing data from an experimental pneumonia model (GSE295484) were analyzed using weighted gene co-expression network analysis, differential expression analysis, cell-type annotation, and gene

Peri-implant diseases, including peri-implant mucositis and peri-implantitis, constitute significant human health challenges that threaten implant longevity despite advances in implant medicine. These inflammatory disease entities compromise peri-implant tissue integrity and oral wellbeing, with growing public health relevance due to the widespread use of dental implants. Increasing evidence implicates tribocorrosion-induced titanium particle release in provoking foreign body-mediated inflammation, facilitating disease progression from a reversible inflammatory illness to an irreversible destr

Inborn errors of immunity (IEI) are a diverse group of inherited disorders caused by detrimental variants in the genome that compromise the immune system and lead to an increased susceptibility to infections, autoimmunity, and/or malignancy. Although the diagnostic process traditionally begins with a clinical workup and laboratory screening, such as complete blood counts, serum immunoglobulin measurements, and lymphocyte subset enumeration, functional assays serve as a vital bridge to diagnosis by providing a real-time assessment of immune system performance. Such assays include lymphocyte pro

Oncolytic bacteria are microorganisms with cytotoxic activity capable of lysing cancer cells while simultaneously activating both innate and adaptive immune responses to exert anti-tumor effects. However, the innate immune activation targeting oncolytic bacteria within the tumor microenvironment (TME) is a limiting factor. Here we find that the CD11b-targeting agonist GB1275 can modulate bacteria-immune interaction and inhibit the infiltration of CD11b⁺ innate immune cells such as neutrophils and macrophages, thereby suppressing the antimicrobial immune defenses in the TME in mice. This then a

BACKGROUND: Crohn's disease (CD) is a chronic inflammatory bowel disease with rising global incidence. Its multifactorial etiology involves genetic, environmental, microbial, and immunological factors. Current diagnostic methods are invasive and costly, underscoring the need for improved biomarkers and non-invasive tools for early detection and monitoring.
METHODS: Using bioinformatics and machine learning, we analyzed datasets from the Gene Expression Omnibus (GEO) to identify diagnostic biomarkers and immune infiltration patterns. Multi-omics integration and immune profiling were employed.

Hidradenitis suppurativa (HS) is a chronic inflammatory skin disorder with a high unmet need, particularly for treatment-resistant tunnels. Integrated multiomics analysis revealed that HS tunnels are spatially organized, self-amplifying inflammatory circuits driven by synergistic IL-17/IL-36γ activation of a unique keratinocyte subset, which activates neutrophils and inflammatory fibroblasts. This mechanistic insight coupled with spatial coenrichment of IL-17A and IL-36R provided the rationale for HB0043, a bispecific antibody that leverages IL-36R binding to locally dual-block both cytokines.

Severe acute pancreatitis (SAP) has no targeted therapeutic options, largely owing to incomplete understanding of the mechanisms governing the transition from tissue injury to repair. Neutrophils are central effectors of sever acute pancreatitis, but their temporal contribution to injury and repair remains unresolved. Neutrophil depletion was therefore timed to defined disease stages using ROSA26iDTR/MRP8-Cre mice, in which diphtheria toxin administration was initiated on day 0, day 1, or day 2 after caerulein-induced pancreatitis; depletion efficiency was confirmed by flow cytometry. Depletio
