Myeloid cell reprogramming drives enhanced defense against Streptococcus pneumoniae lung infection following exposure to commensal Prevotella.

Clinical data link the prevalent respiratory tract anaerobe Prevotella with reduced pneumonia mortality, but the mechanisms directing Prevotella regulation of lung immune homeostasis are unclear. Here, single-cell RNA sequencing was employed to define the transcriptional immune signatures underlying improved clearance of Streptococcus pneumoniae following lung exposure to Prevotella melaninogenica. Overall, we observed a substantial shift in myeloid cell transcriptional programming from interferon-dominant to a more antibacterial profile in S. pneumoniae-infected mice after pre-exposure to P.

Functional characterization of neutrophils in patients with multiple myeloma.

Multiple myeloma is a hematologic cancer caused by abnormal plasma cell proliferation in the bone marrow and is associated with significant immune dysfunction. Neutrophils, essential components of the innate immune response, may function poorly in patients with multiple myeloma, increasing the risk of infection and affecting disease progression. This study aimed to systematically evaluate the extent of neutrophil dysfunction in patients with newly diagnosed and relapsed multiple myeloma. The study employed a cross-sectional analytical design and recruited 72 participants: 30 multiple myeloma p

IGF2BP2 remodels the lung metastatic immune microenvironment to promote bladder cancer metastasis.

Bladder cancer lung metastasis poses a major therapeutic challenge because of pronounced tumor heterogeneity. However, the mechanisms regulating bladder cancer metastasis remain elusive. Here, we established a lung-tropic bladder cancer subline through iterative in vivo selection, enabling stepwise enrichment of metastatic competence while minimizing intratumoral variability. Using integrated multiomics analyses combined with functional validation, we identified an IGF2BP2-m⁶A-complement axis that is progressively engaged during metastatic evolution and contributes to lung colonization by blad

The IL-17A/TNFSF10 axis promotes vulvovaginal candidiasis pathogenesis by inducing neutrophil necroptosis.

INTRODUCTION: Vulvovaginal candidiasis (VVC) is a common infection of the female reproductive tract and is closely associated with dysregulated mucosal immunity. This study investigated how IL-17A/TNFSF10 signaling regulates neutrophil death and VVC progression. METHODS: Clinical specimens, RNA sequencing, cell-based assays, and mouse models were used. RESULTS: IL-17A enhanced neutrophil extracellular trap formation and increased TNFSF10/TNFRSF10 expression, together with increased phosphorylation of RIP3 and MLKL. In vivo, IL-17A aggravated mucosal injury and neutrophil infiltration, wherea

Histone neutralization protects the ischemic brain against the consequences of stroke-associated pneumonia.

Bacterial pneumonia aggravates ischemic stroke via mechanisms that still remain to be determined. In ischemic stroke patients, we show that stroke-associated pneumonia is associated with poor clinical outcome and long-term neutrophil deregulation. In mice exposed to transient middle cerebral artery occlusion (MCAO), experimental S. pneumoniae pneumonia induced at 3 days after stroke impaired neurological recovery and increased blood-brain barrier breakdown, brain neutrophil infiltrates, cerebral microvascular thrombosis, and progressive brain atrophy. The antibiotic amoxicillin only partially

Functional divergence of regulatory and conventional bovine neutrophils following Mycobacterium bovis infection.

INTRODUCTION: Bovine tuberculosis (bTB), due to Mycobacterium bovis (Mb) infection, is a chronic cattle disease and neglected cause of zoonotic tuberculosis. The role of neutrophils in bTB is overlooked. We recently identified a new neutrophil subset in cattle and mice with a similar morphology to conventional inflammatory neutrophils (Nconv). However, unlike Nconv, these regulatory neutrophils (Nreg) express MHC-II at their surface and can suppress lymphocyte proliferation. METHODS: In this study, we compared the responses of bovine Nconv and Nreg to infection with a virulent Mb strain circu

Epigenetic reprogramming by BCG (Bacillus Calmette-Guérin) vaccination: A novel strategy to reverse immunodeficiency in chronic kidney disease.

Chronic kidney disease (CKD) is associated with reflecting innate immune impairment. Chronic inflammation, oxidative stress, metabolic dysregulation, and prolonged uraemia are the primary causes of this condition. Monocytes, macrophages, and their progenitors may undergo drastic epigenetic changes in response to these systemic stresses. This has lowered the activation of histone marks, such as H3K4me3, and destabilised repressive marks, like H3K9me3. Critical immune response genes are silenced by chromatin closure. This epigenetic change exhausts the immune system and impairs its ability to cl

Profiling peripheral immune cells in Parkinson's disease: A scoping review.

Parkinson's disease (PD) is increasingly recognized as a multi-system disorder with immune dysregulation extending beyond the central nervous system. Although numerous studies have examined peripheral immune alterations in people with PD, findings remain heterogeneous and difficult to reconcile. To clarify the current landscape, we conducted a comprehensive scoping review of human studies profiling peripheral blood immune cells in PD. Following PRISMA-ScR guidelines, we systematically screened the literature and curated studies reporting in vivo and ex vivo immune characterizations from PD pat

Age-dependent associations of peripheral immune-inflammatory indices with cerebrovascular remodeling after revascularization in ischemic moyamoya disease.

OBJECTIVE: Peripheral immune-inflammatory activity may influence postoperative vascular remodeling in Moyamoya disease (MMD), but the relevant pathways may differ by developmental stage. We investigated age-dependent associations of blood cell-derived inflammatory indices with two complementary imaging outcomes: collateralization angiogenesis ratio (CAR) and arterial residual volume (ARV). METHODS: This retrospective observational study included 108 patients with ischemic MMD who underwent revascularization (76 adults and 32 pediatric patients). Associations of neutrophil-to-lymphocyte ratio

Integrative Transcriptomic and Genetic Analysis Prioritizes IRF1 as a Candidate Regulatory Gene Associated with Ulcerative Colitis.

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory bowel disease with a complex and incompletely understood pathogenesis. This study aimed to prioritize candidate genes associated with UC and characterize their potential genetic and immune-related molecular context using an integrative transcriptomic and genetic framework. METHODS: Bulk transcriptomic analysis was first performed using the discovery cohort GSE87466. After duplicated gene symbols were collapsed by mean expression, differential expression analysis and weighted gene co-expression network analysis (WGCNA) were conducte

Targeting hypoxia-driven histone lactylation in myeloid cells synergizes with CD137 agonism to expand effector-like exhausted CD8+ T cells in glioblastoma.

BACKGROUND: Overwhelming presence of immunoregulatory myeloid cells and dysfunctional T cells is a major barrier to effective immunotherapies in glioblastoma (GBM). Therapeutic strategies that simultaneously address myeloid-driven immunosuppression and impaired T cell function are therefore needed to restore antitumor immunity in GBM. METHODS: We employed single-cell RNA sequencing, immunophenotyping, and functional analyses in an orthotopic murine GBM model (SB28) to investigate how hypoxia shapes the tumor immune microenvironment and limits CD8+ T cell responses. Pharmacologic hypoxia modul

Association of early dynamic changes in peripheral blood immune-inflammatory indices with subsequent acquired resistance to immune checkpoint inhibitors in advanced non-small cell lung cancer: a landmark analysis.

BACKGROUND: Acquired resistance to immune checkpoint inhibitors (ICIs) remains a major challenge in advanced non-small cell lung cancer (NSCLC). This study evaluated whether early dynamic changes in peripheral inflammatory and nutritional indices predict subsequent acquired resistance. METHODS: This retrospective landmark cohort included 138 patients with advanced NSCLC who achieved disease control after ICI-containing therapy. Systemic immune-inflammation index (SII), derived neutrophil-to-lymphocyte ratio (dNLR), platelet-to-lymphocyte ratio, prognostic nutritional index (PNI), and lactate

Diagnostic Performance of the Neutrophil-to-Lymphocyte Ratio for the Identification of Insulin Resistance in Patients with Fibrotic Chronic Hepatitis C: a Case-Control Study.

BACKGROUND/AIMS: Chronic hepatitis C virus (HCV) infection is accompanied by persistent hepatic inflammation, progressive fibrogenesis and metabolic disturbance, of which insulin resistance (IR) is the most consistently reported. The neutrophil-to-lymphocyte ratio (NLR), derived from a routine complete blood count, has been proposed as an inexpensive index of systemic inflammatory activity, but its diagnostic accuracy for IR in fibrotic chronic hepatitis C has not been quantified in Iraqi patients. To quantify the cross-sectional diagnostic performance of NLR for the identification of insulin

Mast Cells Regulate Epithelial Barrier Integrity and Myeloid Immune Responses in Experimental Colitis.

Mast cells (MCs) are increasingly recognized as modulators of intestinal homeostasis and inflammation; however, their precise contribution during colitis remains incompletely defined. Here, we investigated the role of MCs in dextran sulfate sodium (DSS)-induced colitis using a Kit-independent MC-deficient model (Cpa3-Cre; Mcl1fl/fl), in which selective deletion of the anti-apoptotic gene Mcl1 ablates MCs. Despite efficient and tissue-wide MC depletion, MC-deficient mice exhibited macroscopic and histological disease severity comparable to control mice. However, MC deficiency significantly alte

Indole-3-Propanoic Acid Alleviates Pulmonary Inflammation in a Mouse Model of Bronchial Asthma by Regulating PD-1/PD-L1.

BACKGROUND: Bronchial asthma is a prevalent chronic inflammatory disorder of the airways, with children representing a particularly vulnerable population. Early intervention is critical for the management of pediatric asthma. The pathogenesis of asthma is primarily characterized by a Th2-immune response that drives excessive secretion of inflammatory cytokines, leading to allergic airway inflammation. Emerging evidence indicates that gut microbiota-derived metabolites modulate immune cells and influence asthma development and progression. Among these metabolites, indole derivatives with immuno

The dipeptidyl peptidase 1 inhibition landscape in bronchiectasis and its future direction.

Bronchiectasis (BE) is a heterogeneous lung disease characterised by chronic neutrophilic inflammation. A "vicious vortex" develops through the interactions between neutrophilic inflammation, infection and impaired mucociliary clearance, leading to progressive structural lung damage and worsening lung function. As part of this neutrophilic inflammation, neutrophil serine proteases (NSPs) overwhelm their natural inhibitors, with high NSP levels being associated with pulmonary exacerbations in people with BE. NSPs, activated by dipeptidyl peptidase 1 (DPP1) during neutrophil maturation in the bo

Identification and validation of genes associated with mitochondria in cerebral infarction and benign prostatic hyperplasia.

BACKGROUND: Cerebral infarction (CI) and benign prostatic hyperplasia (BPH) are prevalent conditions in middle-aged and elderly individuals. Previous studies have demonstrated that mitochondrial dysfunction plays a critical role in both CI and BPH. This study aims to identify the mitochondria-related genes (MRGs) in CI and BPH, providing theoretical clues and preliminary candidate gene resources for further research on mitochondria targeted therapeutic strategies across different diseases. METHODS: The transcriptomic data of BPH and CI were obtained from public databases, while MRGs were deri

RAGE in neutrophils triggers cerebral vasospasm in the hyperacute phase after subarachnoid hemorrhage.

Subarachnoid hemorrhage (SAH) due to the rupture of an intracranial aneurysm is a highly fatal type of stroke. Cerebral vasospasm (CVS) is a major post-SAH complication leading to delayed ischemic neurological deficits, thereby worsening patient outcomes. Previously, we found that lower plasma levels of the soluble receptor for advanced glycation end products (RAGE) predict symptomatic CVS in patients with SAH. However, the molecular mechanisms underlying CVS remain unclear. Here, using an SAH mouse model with endovascular perforation, we found that neurological deficits, CVS in the circle of

The impact of neutrophil-to-lymphocyte ratio on staging and mortality risk stratification of cardiovascular-kidney-metabolic syndrome.

BACKGROUND: Cardiovascular-kidney-metabolic (CKM) syndrome is a systemic disorder involving pathophysiological interactions among metabolic risk factors, renal system, and cardiovascular system, with consequent systemic inflammatory and oxidative stress responses. Neutrophil-to-lymphocyte ratio (NLR) has been reported to be a prognostic indicator of systemic inflammation and immunity, but its mortality predictive value in CKM syndrome has not been investigated. METHODS: Totally 19,525 participants were included from NHANES database (1999-2018), and were divided into 4 groups by their NLR leve

Neutrophil Extracellular Traps Mediate in vitro and in vivo Degradation of α-Synuclein Amyloid Fibrils.

PURPOSE: Neutrophil extracellular traps (NETs) are chromatin-based structures decorated with neutrophil enzymes such as elastase and myeloperoxidase (MPO). We previously demonstrated that amyloid fibrils (AFs), regardless of their protein composition, induce NET release in human neutrophils in vitro through a mechanism dependent on reactive oxygen species generated by NADPH oxidase 2 (NOX2). In this study, we investigated whether α-synuclein amyloid fibrils (αSF) induce NET formation in vivo and whether NETs modulate amyloid-associated pathology. METHODS: αSF were instilled intratracheally in
