The central nervous system (CNS) is a dynamic immunological environment, where specialized interfaces, immune reservoirs, and metabolic clearance pathways, such as the glymphatic system, orchestrate immune surveillance and homeostasis. Within this intricate network, purinergic signaling, mediated by nucleotides like ATP and adenosine, emerges as a critical regulator of immune responses, inflammation resolution, and tissue repair. Adenosine, drives immunosuppression and immune exhaustion, shaping the balance between neuroprotection and neurodegeneration. Purinergic signaling influences a divers

PURPOSE: Radiation-induced intestinal injury (RIII) is a serious complication of radiation therapy without effective therapies. The alarmin, S100A8/A9, a biomarker of gastrointestinal inflammation, is highly elevated following abdominal irradiation (ABI). However, it remains unclear whether it functions as an active driver of RIII pathogenesis. The study aims to assess the effects of S100A8/A9 on the pathogenesis of RIII. Our goal is to identify its dynamic alteration during RIII and determine whether it can be used as a therapeutic target in the injury and repair phases of acute RIII.

Sleep disturbance disrupts immune homeostasis and impairs oral mucosal healing, yet effective local immunomodulatory strategies remain limited. Here, we identify excessive neutrophil extracellular traps (NETs) as a key mediator linking sleep deprivation to delayed oral wound closure. NETs-associated proteases compromise epithelial barrier function and suppress repair cell migration. To address this, we developed a melatonin-loaded niosome-incorporated silk fibroin methacrylate (SilMA)/gelatin methacryloyl (GelMA) composite hydrogel microneedle (Nio-MT@S-G MN) for localized melatonin delivery a

BACKGROUND: Methotrexate (MTX) is commonly used in patients with autoimmune disorders with the therapeutic objective of inducing immunosuppression predominantly through the modulation of T-lymphocyte activity. Additional studies suggest that the regulation of polymorphonuclear neutrophils (PMNs) may also play a significant role in disease modification. However, the molecular mechanism underlying this effect remains incompletely understood. Moreover, most studies investigating the interaction between MTX and PMNs have relied on centrifugation for cell isolation, a method meanwhile known to alte

BACKGROUND: Sepsis is a life-threatening syndrome of immune dysregulation; preexisting lymphopenia and an elevated neutrophil-to-lymphocyte ratio (NLR) heighten host susceptibility. Chronic low-dose exposure to lead (Pb), cadmium (Cd), and mercury (Hg) is immunotoxic, but its association with these pre-sepsis markers in the general population remains unclear.
METHODS: We analyzed 12,580 adults from NHANES 2011-2018. Outcomes were lymphopenia (absolute lymphocyte count <1.0 × 109/L) and high NLR (top quartile). Survey-weighted logistic and Weighted Quantile Sum (WQS) regressions evaluated indi

Neutrophils are postulated to contribute to the severity of acute lung injury induced by SARS-CoV-2 infection. Neutrophils play both protective and damaging roles in bacterial and viral pneumonia. The present study examined the hypotheses that neutrophils play multiple roles in the pathophysiology of SARS-CoV-2 infection and that granule exocytosis is a contributor to immune cell recruitment and lung injury. Neutrophils were depleted beginning one day prior to, or one day after, infection of 8-10 month old C57BL/6 mice with a mouse-adapted SARS-CoV-2. In separate experiments neutrophil exocyto

BACKGROUND: Obesity drives chronic inflammation and a prothrombotic state, contributing to metabolic dysfunction and cardiovascular disease (CVD). Neutrophil extracellular traps (NETs), web-like DNA-protein structures released by neutrophils, trap pathogens through a specialized cellular process called NETosis, which involves chromatin decondensation and cell death. While NETs protect against infection, excessive NET formation promotes tissue damage, insulin resistance, endothelial dysfunction, and thrombosis in obesity. Elevated NETosis fuels systemic inflammation, disrupts insulin signaling,

PURPOSE: Idiopathic pulmonary fibrosis (IPF) is a devastating condition that is characterized by dysregulated repair responses to alveolar epithelial damage and progressive remodeling of the lung interstitium. Neutrophils are critical responders to tissue injury, but their role in organ fibrosis pathogenesis remains unclear. Neutrophil numbers are increased in bronchoalveolar spaces of IPF patients, and neutrophil activation markers are elevated in fibrotic lung tissue. However, the key mediators driving neutrophil movement in fibrotic lung diseases are unknown. Transforming growth factor-beta

BACKGROUND: Neuroendocrine neoplasms (NEN) are mainly located in the gastrointestinal tract or the lungs. In up to 14%, the primary tumor remains unidentified, classified as cancer of unknown primary (CUP). Due to their rarity and heterogeneity, CUPs are poorly studied.
METHODS: Retrospective data analyses with data provided by the SwissNET registry from 2008-2022 including patients diagnosed with CUP or metastasised NEN with known primary (mNEN). Main objective was to assess the demographics, tumor characteristics and the diagnosis and treatment of CUPs in comparison to mNEN. Secondary objec

BACKGROUND: Citrullination represents a post-translational modification primarily mediated by peptidylarginine deiminase (PADI) 2 and 4 and resulting in the conversion of positively charged peptidylarginine to neutrally charged peptidylcitrulline. Molecular consequences of citrullination include the generation of neoepitopes which provoke the production of autoantibodies implicated in the development of autoimmune diseases. As citrullination initiates, promotes, and is enhanced by aseptic inflammation which plays a pivotal role in atherosclerosis, we proposed that citrullination might accompan

BACKGROUND: Faecal calprotectin (FC) has limitations including inconvenient sampling and limited accessibility, while several blood markers derived from complete blood counts demonstrate suboptimal diagnostic performance in monitoring ulcerative colitis (UC) disease activity. There is an urgent need for superior noninvasive biomarkers. The pan-immune inflammation value (PIV), a novel composite index derived from complete blood counts, has shown prognostic value in malignancies but remains unexplored in UC activity assessment.
METHODS: A retrospective analysis was conducted on 196 patients wit

BACKGROUND: Acute pancreatitis (AP) is a disease with high morbidity and mortality. Neutrophils are highly correlated with the occurrence and severity of the inflammatory response in AP. However, the mechanism by which neutrophils act on AP is currently unclear. We aimed to target genes coexpressed with neutrophils to identify new possibilities for diagnosing and treating AP.
METHODS: We analyzed the differences in immune infiltration in AP and the differential expression of neutrophil-coexpressed genes. Then, models were built through various machine learning methods, and the hub neutrophil

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory bowel disease that can lead to malignancies over time. Sphingosine-1-phosphate (S1P) receptor signaling affects lymphocyte trafficking and vascular integrity, influencing intestinal inflammation. This study aimed to identify S1P-related key genes in UC.
METHODS: Differentially expressed genes (DEGs) between the UC and control groups were analyzed in the GSE87473 (training) dataset. Genes overlapping between the DEGs and S1P-related genes were considered candidate genes. These genes were incorporated into machine learning algorithms

INTRODUCTION: Disturbed immunity and inflammation have been associated with the pathogenesis of schizophrenia (SZ) and suicidal behaviors. We hypothesized that the neutrophil-tolymphocyte ratio (NLR) was associated with the increased risk of suicide ideation (SI) in patients with SZ.
METHODS: A total of 97 drug-naïve first-episode patients with SZ (15 with SI and 82 without SI) were recruited in the present study. The symptom severity was assessed using the Positive and Negative Syndrome Scale (PANSS), and differential white cell counts and SI were assessed for all patients.
RESULTS: Patient

BACKGROUND: With the viral variants and waning of vaccine-induced immunity over time, SARS-CoV-2 breakthrough infections have become the next challenge following the COVID-19 pandemic, highlighting the need for broader and sustained immune responses. As the first defense against viral invasion, mucosal immunity is inducible by SARS-CoV-2 infection. Previous studies focused on serum antibodies and cellular immunity in the acquired immune, but the specific role of mucosal immunity in breakthrough infection remains to be elucidated.
METHODS: We performed single-cell RNA sequencing (scRNA-seq) on

BACKGROUND & AIMS: Fibroblast growth factor receptor 2 gene (FGFR2) rearrangements are among the most common oncogenic drivers in intrahepatic cholangiocarcinoma (iCCA). Although FGFR inhibitors are clinically approved, primary and secondary resistance remain major limitations. Preclinical investigation of resistance mechanisms, including cancer cell-extrinsic crosstalk, is hampered by current models that rely on human FGFR2 fusion transgenes in immunodeficient hosts. We therefore aimed at generating an entirely murine FGFR2 fusion-driven iCCA (Ff-iCCA) model to study immunomodulatory mechanis

Neutrophils are central to immune responses in health and disease, and their ability to deform correlates with their functional responses. In health, reorganization of the neutrophil cytoskeleton allows neutrophils to carry out their effector mechanisms. However, in the disease state, dysregulation of neutrophil cytoskeletal properties results in disruption of neutrophil structural integrity, in turn altering their immune response capabilities. We outline how actin and actin polymerization are essential for cell processes, the role of actin in neutrophil function, and how disruption of actin c

BACKGROUND AND AIMS: Venous thromboembolism (VTE) is a disease related to high mortality and complications. Neutrophil extracellular trap (NET) formation promotes thrombo-inflammatory responses, exacerbating VTE. P2Y14 receptor (P2Y14R), which is highly expressed on neutrophils mediates NET formation, but its role and mechanism in VTE are unclear. This study aims to explore the role and mechanism of P2Y14R in VTE and to investigate the feasibility of P2Y14R-targeting therapy for VTE.
METHODS: Venous blood of VTE patients was collected to detect the expression of P2Y14R. Deep vein thrombosis a
