Influenza infection exacerbates high-fat diet-induced atherosclerosis in apolipoprotein gene-deficient mice.

Influenza is a significant public health and economic threat around the world. Although pneumonia is the most common complication associated with influenza, there are several clinical reports showing an increased risk for cardiovascular disease. Studies have shown that influenza infection correlates with increased incidence of myocardial infarction. Atherosclerosis is a common cause of coronary artery disease including myocardial infarction, stroke, and heart failure. Here, we analyzed the effect of influenza infection in high-fat diet-induced atherosclerosis in Apoe-/- mice. We found increase

Single-cell RNA sequencing reveals multiple pathways involving pulmonary immune and epithelial cells through which aryl hydrocarbon receptor activation attenuates acute respiratory distress syndrome.

Acute respiratory distress syndrome (ARDS) is a severe inflammatory lung disorder triggered by pneumonia, sepsis, trauma, and COVID-19, leading to high mortality. In this study, we investigated the effect of aryl hydrocarbon receptor (AhR) activation by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), a potent ligand, on LPS-induced ARDS in mice using single-cell RNA sequencing (scRNA-seq). scRNA-seq revealed 16 transcriptionally distinct cell clusters in the lungs. AhR activation reversed the decreased pulmonary functions caused by LPS, and significantly reduced pulmonary infiltration of monocytes

The aryl hydrocarbon receptor promotes the resolution of pulmonary neutrophilia via regulation of macrophage efferocytosis.

Cigarette smoke is the primary cause of chronic obstructive pulmonary disease (COPD), an incurable condition characterized by irreversible airflow obstruction and alveolar destruction driven by chronic inflammation of the lungs and airways. The inflammatory response caused by cigarette smoke is typified by the recruitment of innate and adaptive immune cells to the lung. Paradoxically, many of these immune cells are functionally impaired by smoke. Notable among these are lung macrophages, which have reduced ability to clear apoptotic lung epithelial cells and neutrophils by efferocytosis when e

Mitochondrial Transfer Rescues Respiration to Support De Novo Pyrimidine Biosynthesis and Tumor Progression.

UNLABELLED: Cancer cells with severe defects in mitochondrial DNA (mtDNA) can import mitochondria via horizontal mitochondrial transfer to restore respiration. Mitochondrial respiration is necessary for the activity of dihydroorotate dehydrogenase (DHODH), an enzyme of the inner mitochondrial membrane that catalyzes the fourth step of de novo pyrimidine synthesis. In this study, we investigated the role of de novo synthesis of pyrimidines in driving tumor growth in mtDNA-deficient (ρ0) cells. Although ρ0 cells grafted in mice readily acquired mtDNA, this process was delayed in cells transfecte

The Paradox of NET Involvement in the Pathogenesis of Inflammatory Bowel Disease.

Inflammatory bowel disease (IBD), namely Crohn's disease (CD) and ulcerative colitis (UC), are defined by chronic, non-resolving inflammation of the intestinal mucosa. Neutrophils are the first responders in inflammation, executing various effector functions, including chemotaxis, phagocytosis, degranulation and the release of cytokines, reactive oxygen species (ROS) and neutrophil extracellular traps (NETs). Amongst all neutrophil functions, emerging evidence increasingly suggests that NET release may be particularly relevant in underpinning the pathogenesis of IBD. NETs are extracellular str

Neutrophil Transmigration Analyses for Multiplexed Drug Screening and In Vivo Like Application Testing Using an Organ on a Chip Model.

Neutrophils are the most abundant type of innate immune cell in humans and make up for 40-60% of leukocytes in humans. As a part of the innate immunity and due to their high frequency, they form the first line of defense against harmful invaders. While neutrophils are crucial for combating infections, dysregulated neutrophil activity is a hallmark of chronic inflammation and autoimmune disorders. Here, we describe two methods to evaluate neutrophil migration, an in vitro transwell migration assay and an ex vivo organ-on-chip model, which allows quantification of the migration of neutrophils un

Neutrophil-based delivery platforms: from natural mechanisms to engineered therapeutics.

Neutrophils are one of the key components of the innate immune system, and they play essential roles in various physiological processes, including phagocytosis, chemotaxis, immune sensing, and transmigration across the vascular endothelium. The synergy of neutrophil biology with nanomaterial science has led to the development of innovative neutrophil-based drug delivery systems (NDDSs) or neutrophil-derived biomimetic delivery systems. In this review, we elucidate the mechanisms underlying neutrophil-mediated targeting strategies. By utilizing inherent properties of neutrophils, targeted deliv

Trained immunity in cardiovascular disease.

With innate immunity at the core of the pathophysiology of atherosclerosis, the discovery of new mechanisms of immune cell activation can potentially identify novel pharmacological targets to prevent or treat cardiovascular disease (CVD). One of these mechanisms is trained immunity (TRIM), defined as a recallable long-term hyperinflammatory innate immune phenotype supported by changes of metabolic and epigenetic intracellular processes. TRIM can be induced in mature innate immune cells in tissues, including monocytes/macrophages, and natural killer cells, but also in non-immune cells such as e

Innate recognition and phagocytosis of zymosan by human neutrophils require cell priming in a p38-dependent manner.

Neutrophils are the first line of defense against fungal infection and other microbial pathogens. Through alternative pathway of complement activation in innate immunity, complement fragments (opsonin) generated promote phagocytosis of the yeast forms of fungal pathogens such as Candida albicans. However, when complement levels are limited, through consumption in disease and inflammation or where microbial pathogens employ mechanisms to degrade or inhibit complement components, phagocytosis may still proceed via innate receptors, in the absence of effective opsonisation. Mechanisms of innate p

Single-cell RNA Sequencing Analysis Reveals the Molecular Mechanisms of Neutrophil Dysfunction in Chronic Bone Infection.

INTRODUCTION: Neutrophils play a key role in host immune defense. At present, neutrophils in chronic bone infections exhibit significant heterogeneity but functional alterations that remain poorly understood. MATERIALS AND METHODS: A rat model of chronic bone infection induced by Methicillin- Resistant Staphylococcus Aureus (MRSA) was established. Bone marrow cells were analyzed using scRNA-seq with Gene Ontology (GO) and pathway enrichment analysis. Differentially Expressed Genes (DEGs) were identified to assess neutrophil dysfunction, validated by immunofluorescence staining and ROS quantif

Translocator Protein 18 kDa-Targeted Total-Body PET Imaging Reveals Immune-Specific Vascular Inflammation in Large-Vessel Vasculitis: A Prospective Head-to-Head Comparison with 18F-FDG.

Accurate assessment of vascular inflammation in large-vessel vasculitis (LVV) remains challenging, especially in patients with normal inflammatory markers or inconclusive 18F-FDG PET findings. The translocator protein 18 kDa (TSPO), a marker of inflammatory process, has emerged as a promising molecular target for imaging vascular inflammation in LVV. Methods: We prospectively enrolled 11 treatment-naïve patients with LVV and 11 healthy controls who underwent 90-min dynamic total-body 18F-DPA714 PET/CT imaging. Ten patients also underwent 18F-FDG PET/CT for direct comparison. We assessed SUVs,

Neutrophil extracellular traps in colorectal cancer: a paradoxical nexus of tumor defense and disease progression.

Neutrophil extracellular traps (NETs) are extracellular networks of decondensed chromatin and granular proteins generated by neutrophils in response to activating stimuli. Growing evidence indicates that NETs participate in colorectal cancer (CRC) development through their interactions with chronic inflammation, immune regulation, coagulation, and tumor progression. However, their biological effects vary across disease settings, and the regulatory mechanisms specific to CRC have not been fully clarified. This review examines the roles of NETs at different stages of CRC, including the transitio

Biological roles and clinical translation of low-density neutrophils in rheumatic, autoimmune and inflammatory diseases: paradigm reconstruction and unresolved questions.

Neutrophils are increasingly recognized as a heterogeneous lineage rather than a uniform population of short-lived innate effector cells. Low-density neutrophils (LDNs; often termed low-density granulocytes, LDGs) are operationally defined by their co-sedimentation with peripheral blood mononuclear cells after density-gradient centrifugation, but this physical definition encompasses cells with different maturation states, origins and functions. In rheumatic and autoimmune diseases, expansion of the low-density compartment is associated with disease activity, autoantibody titers, interferon sig

Targeting innate immunity in cancer: mechanistic foundations, therapeutic vaccination, and clinical translation.

Cancer evolves within tissue ecosystems in which innate immune surveillance and tumor-promoting inflammation jointly shape disease progression and therapeutic response. Innate receptors detect cellular stress, altered-self ligands, damaged-cell products and tumor-derived nucleic acids, thereby initiating cytotoxicity, phagocytosis, antigen presentation and adaptive immune priming. However, tumors can redirect these mechanisms through suppressive cytokines, metabolic competition, hypoxia and dysregulated chemokine networks, promoting immune exclusion, metastasis and treatment resistance. This R
