Ischemic Stroke in Hypereosinophilic Syndrome: A Clinicopathologic Study of Two Cases.

Ischemic stroke is a rare complication of hypereosinophilic syndrome (HES). Manifestations of stroke in HES have been described in the radiologic literature; however the pathologic characterization of central nervous system (CNS) involvement in HES is limited. In this small case series, we present two post-mortem neuropathologic examinations of patients exhibiting CNS infarcts associated with HES and review the relevant literature.

Clinical characteristics, management, and hematopoietic cell transplantation of patients with TLR8 gain-of-function.

Toll-like receptor 8 (TLR8) gain-of-function (GOF) somatic variants were recently identified as causing severe neutropenia, lymphoproliferation, and immune dysregulation. We report the expanded clinical and laboratory phenotype and management of 10 patients, including the original cohort of 6 male patients. We identify the first female patient with TLR8 GOF who presented during infancy with pure red cell aplasia due to a germ line TLR8 variant, and 2 new disease-causing somatic variants in male patients. Eight patients had somatic mosaicism with peripheral blood variant allele fractions of 7%

Pro-resolving lipid mediators in diseases: exploring the molecular basis and clinical implication.

Inflammation resolution is now understood as an active and highly coordinated biological process rather than a passive decline in inflammatory signals. When this program fails, inflammation may become persistent and gradually shift to a chronic pathological state. Such unresolved inflammation is increasingly recognized as a core driver of cardiovascular disease, metabolic disorders, autoimmune pathologies, neurodegeneration, and tumor progression. Although conventional anti-inflammatory drugs can suppress inflammatory mediators, they do not restore immune balance or actively promote resolution

Alkaloid fraction of Duguetia furfuracea demonstrated anti-inflammatory and antinociceptive effects through decrease of tumour necrosis factor alpha in mice models.

Duguetia furfuracea, popularly known as "araticum-seco," is traditionally used in folk medicine for the treatment of pain and inflammatory disorders. This study aimed to analyze the chemical composition and investigate the anti-inflammatory and antinociceptive activities of the alkaloid fraction obtained from the leaves of D. furfuracea (DfFAlk). The chemical composition of DfFAlk was characterized using ultra-fast liquid chromatography (UFLC) coupled with mass spectrometry (UFLC/ESI). In vivo anti-inflammatory activity was assessed using the carrageenan-induced paw oedema model. The effects o

Mutations in VPS18 lead to a neutrophil maturation defect associated with disturbed vesicle homeostasis.

Neutrophils, the first cells to arrive at the site of inflammation, are rather short-lived cells and thus have to be constantly replenished. During neutrophil development, vesicle dynamics need to be fine-tuned and impaired vesicle trafficking has been linked to failure in neutrophil maturation. Here, we characterized the role of VPS18 as a central core component of CORVET & HOPS tethering complexes for neutrophil development. Using CRISPR/Cas9-engineered Hoxb8 cells with heterozygous mutations in Vps18, we found that VPS18 deficiency interfered with neutrophil development due to tethering com

Genetic Determinants of Hematopoietic Toxicity Risk in Thai Pediatric Patients Undergoing 6-Mercaptopurine Treatment.

The nucleoside diphosphate-linked moiety X-type motif 15 (NUDT15) has been identified as a key genetic determinant of 6-mercaptopurine (6-MP)-induced hematopoietic toxicity in populations with a high frequency of NUDT15 variants but a low frequency of thiopurine S-methyltransferase (TPMT) variants. However, evidence remains limited in populations where both genetic variants are prevalent. This study aimed to characterize the impact of NUDT15 and TPMT polymorphisms on 6-MP-induced hematopoietic toxicity in Thai pediatric patients, a population with a high prevalence of both variants. A total of

Multimodal evaluation of mannose engineered poly lactic glycolic acid nanoparticles with granulocyte colony stimulating factor focused delivery to bone marrow for neutropenia.

Neutropenia, characterized by a critical reduction in neutrophils, demands targeted therapeutic strategies to enhance the delivery efficiency of granulocyte colony-stimulating factor (G-CSF) specifically to bone marrow macrophages. This study focused on engineering mannose-modified poly(D, L-lactide-co-glycolide) (PLGA) nanoparticles (NPs) to achieve ligand-directed delivery of G-CSF. Mannose anchoring was achieved via Ethylenediamine (EDA)-mediated chemical ligation using N-hydroxysulfosuccinimide (NHS) and dicyclocarbodiimide as coupling agents, resulting in Mn-EDA-PLGA NPs. G-CSF-loaded and
