Hepatic sarcoidosis presenting with portal hypertension without histologically established cirrhosis and reticular hepatic imaging features.

We report a 50-year-old man who was referred for evaluation of suspected hepatic sarcoidosis after non-caseating granulomas were incidentally identified on the liver surface during surgery. Laboratory tests showed a cholestatic pattern with elevated alkaline phosphatase and gamma-glutamyl transpeptidase, mild thrombocytopenia, and increased angiotensin-converting enzyme and soluble interleukin-2 receptor levels. Contrast-enhanced computed tomography demonstrated marked surface irregularity and reticular hypoattenuated linear structures within the liver. Gadoxetic acid-enhanced magnetic resonan

Involvement of the spleen in the anti-prion activity of hydroxypropyl methylcellulose in mice.

Prion diseases are a unique group of fatal infectious neurodegenerative disorders. Previously, we reported that hydroxypropyl methylcellulose (HPMC) exerts long-term anti-prion activity in prion-infected rodents after a single peripheral administration, but the mechanism of action is not well understood. Interestingly, the distribution of HPMC in mice partially resembles that of peripheral pathogenic prions, since peripherally administered HPMC remains in the spleen and lymphoid tissues for a long period. Here, we characterized the anti-prion activity of HPMC in relation to the spleen in mice.

Effects of dietary phytobiotic mixtures on cellular and humoral immune parameters and production performance in broiler chickens.

The intensification of poultry farming and the numerous accompanying stress factors lead to increased bird mortality and decreased production rates. Therefore, the main goal of this study was to verify the hypothesis that adding various phytobiotic compounds to poultry feed exerts immunomodulatory effects by altering selected cellular and humoral immune parameters, which may have implications for the health and productivity of the birds. The effect of using mixtures of polyphenolic compounds with tannins and essential oils (carvacrol, cinnamaldehyde, capsaicin) in the diet of Ross 308 broiler

Identification, molecular typing, antimicrobial susceptibility, and experimental pathogenicity of Streptococcus agalactiae isolated from Tilapia.

Streptococcosis caused by Streptococcus agalactiae is widespread in the world, especially in south Asia and southeast Asia. In this study, an S. agalactiae isolate collected from diseased fish in India was molecularly confirmed through GAPDH-targeted PCR, and further validated by sequencing. Genotyping revealed distinct genomic relationships among regional isolates; pulsed-field gel electrophoresis identified three pulsotypes with the Indian isolate clustering closely with two Taiwanese isolates under both ApaI and SmaI digestion, while ERIC-PCR grouped the Indian strain with a Taiwanese strai

Preoperative transcatheter arterial embolization enables safe resection of a giant hypervascular pancreatic acinar cell carcinoma: A case report.

Pancreatic acinar cell carcinoma (ACC) is a rare malignant neoplasm that accounts for 0.4%-0.7% of all pancreatic tumors. It often presents as a large, bulky mass owing to its expansive growth pattern. We report a case of a large pancreatic ACC that achieved remarkable long-term recurrence-free survival after successful surgical resection supported by preoperative interventional radiology (IR). A 64-year-old male presented to our hospital with weight loss and abdominal distension. A firm mass was palpable in the left upper abdomen. CT revealed a giant, heterogeneously enhancing tumor measuring

DNA vaccination against viral hemorrhagic septicemia virus (VHSV) affects the capacity of kidney leukocytes to respond to a posterior viral encounter.

Given the lack of antiviral prophylactic measures authorized in aquaculture, vaccination is the most effective method to prevent and control diseases provoked by viral agents. Viral hemorrhagic septicemia virus (VHSV) is an important fish pathogen, known to induce high mortality rates and large economic losses worldwide in many aquaculture fish species, including rainbow trout (Oncorhynchus mykiss). DNA vaccination against VHSV has been proven a very effective method to induce a long-term protection against the virus, yet the mechanisms through which this vaccine induces protection are still u

CD80+ Macrophage-Induced IL-17+CD8+ T cells accumulate in hyperlipidemic patients and murine vascular lesions to promote atherosclerotic progression.

BACKGROUND: Tc17 cells (IL-17 + CD8+T) are found in many inflammatory diseases. However, the distribution, regulation, and function of Tc17 cells in ApoE deficient-induced atherosclerosis are currently largely unexplored. METHODS: The percentage of Tc17 cells, monocytes and IL-1β+ monocytes in blood of hyperlipidemic patients or healthy donors were analyzed by flow cytometry. The content of IL-1β in plasma was detected by ELISA. Correlations between parameters of clinical samples were assessed using Spearman r correlation analysis or linear regression analysis as appropriate. Aorta, blood, spl

Risk factors for drug-related gastrointestinal ulcer:a retrospective pharmacovigilance study.

OBJECTIVE: Despite the fact that many medications have been linked to gastrointestinal ulcers, the extent to which most of these drugs contribute to such ulcers is not well understood. This study investigates the risk factors linked to gastrointestinal ulcers caused by drugs by analyzing large datasets obtained from the FDA Adverse Event Reporting System (FAERS). METHODS: The data from the first quarter of 2015 to the third quarter of 2025 were analyzed using the report odds ratio (ROR), combined with single-factor, LASSO, and multivariate regression analysis methods, to thoroughly investigat

One-component ionizable amphiphilic Janus dendrimers as a delivery platform for efficient mRNA vaccine development.

There have been rapid advances in nucleoside-modified messenger RNA-based vaccines, particularly during the SARS-CoV-2 pandemic, demonstrating a rapid and cost-effective approach for addressing infectious diseases. A major challenge remains in developing a delivery system that protects mRNA from degradation and facilitates its passage through tissue and cellular barriers. Current four-component lipid nanoparticles enable efficient endosomal escape and are a pivotal technology for the delivery of mRNA vaccines. However, challenges such as stability, availability, durability, and adverse effects

Integrated pan-cancer profiling and experimental validation identify CCDC59 as a key driver and therapeutic biomarker in liver hepatocellular carcinoma.

BACKGROUND: CCDC59, a coiled-coil domain-containing (CCDC) family member, is involved in multiple diseases, yet its pan-cancer role remains largely unclear. METHODS: We analyzed CCDC59 across cancers using The Cancer Genome Atlas (TCGA), Gene Expression Omnibus (GEO), Human Protein Atlas (HPA), along with R and tools like STRING, TISIDB, and TISCH2. CCDC59 expression was validated in clinical samples via quantitative real-time PCR (RT-qPCR) and western blot. Functional assays (proliferation, colony formation, and transwell) and cell cycle analysis were performed in liver hepatocellular carcin

DNA-directed assembly of multivalent lipid nanoparticles for targeted T cell gene delivery.

Lipid nanoparticles (LNPs) are a powerful emerging tool for in vivo T cell engineering to treat diseases ranging from B cell lymphomas and other cancers to autoimmune diseases. Key challenges in designing these therapeutics include achieving both precise cell targeting and efficient mRNA translation. While single-targeted LNPs have been extensively studied, bispecific LNPs have only been briefly explored. Multiple receptor engagement offers the opportunity for enhanced mRNA delivery, expression, and T cell targeting. Here, a DNA-tethering method was developed to enable rapid modification of li

CXCR4-antagonistic peptide-decorated lipid nanoparticles for co- delivery of AML1-ETO siRNA to enhance chemotherapy in refractory AML.

Fusion protein AML1-ETO and chemokine receptor CXCR4 are crucial therapeutic targets for AML, the former plays a key role in differentiation blockade and leukemogenesis, and the latter is closely associated with drug resistance and relapse. This work developed a unified platform (referred as to E5-LNP@siAE) to integrate AML1-ETO-targeted RNA interference (RNAi) and CXCR4 antagonism by employing lipid nanoparticle (LNP) technology and antagonistic peptide E5 modification. The E5-LNP@siNC was slightly negative charged with 121.8 nm in diameter, showing homogenous spherical morphology and stabili

Acyl-CoA-binding protein (ACBP): a poor-prognosis biomarker in sepsis and a target for disease mitigation.

Sepsis remains a major clinical challenge, with high mortality and long-term disability despite current interventions. Here, we identify the tissue hormone acyl-CoA-binding protein (ACBP), also known as diazepam-binding inhibitor (DBI), as a biomarker and driver of poor outcome in sepsis. ACBP/DBI was elevated in the plasma of septic patients and associated with organ dysfunction and increased mortality. In murine models of endotoxemia, Escherichia coli infection, and polymicrobial sepsis, genetic deletion or antibody-mediated neutralization of ACBP/DBI conferred robust protection by dampening

Delivering cytokine mRNA to secondary lymphoid organs for robust cancer immunotherapy.

Recombinant cytokine therapeutics, among the earliest clinical immunotherapies, have yet to reach their full potential despite bioengineering advances. Here, we propose a novel approach focusing on in vivo cytokine production by immune cells in lymphoid organs to mimic these proteins' physiological function. We developed apolipoprotein nanoparticles (aNPs) to deliver mRNA encoding cytokines, such as interleukin-2 (IL-2), to monocytes in secondary lymphoid tissues. To this end, we engineered a scalable manufacturing process that yields stable aNP-mRNA formulations suitable for long-term storage

Discovery of frailty-linked proteins through multi-omics integration of brain and plasma proteomes with GWAS datasets.

BACKGROUND: Frailty, a clinical state of increased vulnerability to stressors with aging, imposes significant strain on healthcare systems. Its genetic underpinnings remain incompletely explored, highlighting the need to identify novel therapeutic targets for aging. METHODS: We integrated brain (N = 376) and plasma (N = 35,559) proteomes with a large-scale GWAS of Fried Frailty Score in 386,565 UK Biobank participants. We employed a combined analysis using Mendelian randomization (MR), Steiger filtering, Bayesian colocalization, proteome-wide (PWAS), and transcriptome-wide (TWAS) association

Systemic Inflammation Modulates Clearance and Drives Extra-Hepatic Distribution of Extracellular Vesicles.

Extracellular vesicles (EVs) are promising vehicles for targeted therapeutic delivery, capable of encapsulating and transporting biomolecules to specific cells and tissues. Given that inflammation is central to many acute and chronic diseases, understanding EV biodistribution under inflammatory conditions is essential for therapeutic optimisation. This study examines how acute systemic inflammation influences EV biodistribution, clearance and plasma half-life, with a focus on the role of macrophages and their polarisation states. Using a lipopolysaccharide (LPS)-induced inflammation model in w

Natural SARS-CoV-2 infection in farmed minks (Neovison vison) causes lung pathology, systemic viral spread, and transmission risk, even in asymptomatic animals.

INTRODUCTION: In 2020, the first disease outbreak of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in farmed minks (Neovison vison) was reported in the Netherlands, followed by outbreaks in other countries. The disease in minks is characterized by interstitial pneumonia and viral replication in both the upper and lower respiratory tracts, resulting in respiratory disease and, in some cases, death. A major concern, besides animal health problems, is that minks are a potential reservoir for SARS-CoV-2, with a zoonotic impact for human Coronavirus Disease-19 (COVID-19), underscorin

Systemic delivery of drug-free polymeric nanoparticles reprograms innate immunity in a sex-dependent manner after spinal cord injury.

Sex differences influence distinct inflammatory responses after spinal cord injury (SCI), yet their impact on immune-modulating nanotherapeutics remains unclear. Here, we investigated the sex-dependent effects of drug-free poly(lactic-co-glycolic acid) (PLGA)-based nanoparticles (NPs) following SCI. Systemic NP administration enhanced locomotor recovery in both sexes and eliminated the functional gap observed in controls. Mechanistically, NPs engaged distinct immune pathways between sexes. Females accumulated more NPs in the spleen, leading to reduced monocyte-derived macrophage infiltration,
