Time activity curves were recorded over the spleen, chest and liver, following intravenous bolus injection of autologous radio-labelled platelets. Circulating erythroid progenitors from 10 patients with polycythaemia vera, nine with secondary polycythaemia and 10 normal subjects were studied in culture to determine their frequency, requirements for erythropoietin (ESF) and physical characteristics. A radioimmunoassay (RIA) method for erythropoietin (Epo) was developed and validated against the polycythaemic mouse assay. The correlation was good, with a r = 0.94. Several other criteria of specificity were also filled by the RIA, which had a lower detection limit of 5 microU/ml. Erythrocyte 2,3-diphosphoglycerate (2,3-DPG) and adenosine triphosphate (ATP) were measured in 16 patients with polycythaemia vera and found to be normal. The determination of erythrocyte organic phosphate concentration can be helpful in the differential diagnosis of different types of polycythaemia due to the lack of 2,3-DPG rise in patients with polycythaemia vera, as compared to the 2,3-DPG increase usually observed in patients with secondary polycythaemia.

Blood viscosity, cerebral blood flow (CBF) and cerebral oxygen carriage (CBF X arterial oxygen content) were measured in 12 patients with polycythaemia secondary to hypoxic lung disease. Certain platelet functions were evaluated in 24 patients with secondary polycythaemia (SP) and in a large number of patients suffering from myeloproliferative disorders (MD'S): 89 patients with chronic myeloid leukaemia (CML) at different stages of development, 58 with polycythaemia vera (PV), 23 with essential thrombocythaemia (ET), and 25 with agnogenic myeloid metaplasia (AMM). Bleeding time, epinephrine-induced platelet aggregation and adhesiveness agreed with those generally reported in the literature; they are independent of thrombocytosis, the haemoglobin level and the leucocyte count.

Arterial oxygenation, breathing pattern, and electroencephalogram were studied during sleep in patients with chronic bronchitis and emphysema and in healthy subjects. Erythrocytosis in a young Sinhalese man is described. The patient was known to have had a raised Hb and PCV for at least 10 years. The clinical and radiological features of 23 patients with cor pulmonale due to chronic obstructive airways disease were reviewed. Total red cell volumes and splenic red cell pools were measured in 31 patients with polycythaemia. 22 had polycythaemia vera (PV), 12 of whom had clinically detectable splenomegaly, and nine patients had secondary polycythaemia (PS). The unsaturated B12 binding capacity (UBBC) of the serum and the binding capacity of each of the 3 vitamin B12 binders--the transcobalamins (TC) I, II and III were determined in 21 patients with polycythaemia vera (PV) during the course of the disease and following treatment, using the recently described charged cellulose filter technique. Serum iron concentration, serum iron binding capacity and saturation coefficient were assayed in polycythaemia rubra vera and secondary polyglobulia. In the exacerbation stage of polycythaemia rubra, significantly lower Se Fe and SC values were found while in the remission stage and in secondary polyglobulias these values did not differ from those of the normal control.
