AA in Obesity and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) Nonalcoholic Fatty Liver Disease (NAFLD).

Obesity and MASLD (NAFLD) are chronic, low-grade systemic inflammatory diseases resulting from complex interactions among diet, adipose tissue, gut microbiota, liver, muscle, pancreas, and the brain. Central to this integrative framework are AA and other polyunsaturated fatty acids (PUFAs), which regulate inflammation, insulin sensitivity, neuroendocrine signaling, and energy homeostasis.Excess consumption of energy-dense foods rich in saturated and trans fats leads to adipocyte hypertrophy, altered membrane fluidity, increased antigenicity, and enhanced recruitment of macrophages and T cells

AA-Induced Lipid Peroxidation at the Center of Tumor Cell Apoptosis and Ferroptosis.

Polyunsaturated fatty acids (PUFAs), especially gamma-linolenic acid (GLA), arachidonic acid (AA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), can block HMG-CoA reductase activity and thus decrease cholesterol synthesis/formation. Cholesterol has antioxidant actions and thus reduces the formation of ROS (reactive oxygen species). In contrast, PUFAs augment the generation of ROS. PUFAs undergo peroxidation to form lipid peroxides that are toxic to tumor cells but not normal cells. GPX4 (glutathione peroxidase) reduces the formation of lipid peroxides and thus enhances tumor cel

Multiparametric Flow Cytometry Analysis of Naïve, Memory, and Effector T-Cells.

Polychromatic flow cytometry enables the detection and characterization of markers that are helpful in defining the phenotype of various cell subsets. Here, we described a flow cytometry-based method to characterize the phenotype of naïve, memory, and effector T cells. Being able to differentiate these cells is crucial in understanding immune response and immune profiling. Naïve T cells enable the body to fight off new, unrecognized infections and diseases, and Memory T cells are enriched for response to recall antigens. Furthermore, the antigen-experienced T-cell populations can be broadly di

Enumeration of Benign and Malignant Plasmacytoid Dendritic Cells in Peripheral Blood and Bone Marrow by Flow-Cytometric Analysis.

Dendritic cells (DCs) are powerful antigen-presenting cells involved in regulating immune response. Plasmacytoid dendritic cells (pDCs) are a subtype of DCs present in small quantities in the bone marrow, peripheral blood, and lymph nodes. They are an important component of our immune system's regulation and response to diseases. They activate T cells and play a critical role in immune tolerance. In this chapter, we review the immunophenotypic features of pDCs and provide a practical protocol for pDCs enumeration in the peripheral blood and in bone marrow samples.

Immunophenotyping of Human Regulatory T-Cells from Peripheral Blood Mononuclear Cells.

Regulatory T cells, or Tregs, are a unique type of CD4⁺ T cell that act as the immune system's brakes-they stop it from turning against the body's own tissues. For about twenty years now, researchers have been digging into what sets Tregs apart from other T cells, both in their composition and function (1). Markers like CD25 (IL-2Rα), Foxp3, and CTLA-4 usually show up when Tregs are active, but these markers aren't always reliable-they can shift depending on where the cells are and what they are doing. Foxp3 is a transcription factor that Tregs need to develop and function. Mutations on Foxp3

Immunophenotyping of Cellular Therapy Products Against Oncogenic and Viral Targets.

Flow cytometric analysis can provide valuable information on the viability, phenotype, and therapeutic potential of cell therapy products using a minimal amount of cell numbers. In this chapter, we describe methods for the analysis of clinical products that incorporate extra considerations within the protocols to help maintain consistency and confidence of staining. This can include methods for evaluating the transduction efficiency for T cells expressing chimeric antigen receptors (CAR-T), characterizing the frequency of diverse lymphocyte populations within polyclonal cell cultures and posti

Nucleofection-Based CRISPR/Cas Delivery in Human T Cells for Immunotherapy Applications.

Electroporation-based delivery of CRISPR/Cas systems has emerged as a powerful and versatile approach for gene editing in primary human T cells, enabling efficient, transient, and nonviral modification while minimizing genomic integration risks. This chapter focuses on the principles and practical implementation of electroporation (nucleofection) for the delivery of Cas9 ribonucleoprotein (RNP) complexes into human T cells, highlighting critical parameters that influence editing efficiency, cell viability, and scalability for research and clinical applications. We provide a comprehensive proto

Mechanisms underlying rheumatic heart disease immune-mediated pathogenesis: T-cell responses, cytokine networks, and immune dysregulation as drivers of valvular injury.

Acute rheumatic fever is an immune-mediated sequela of Streptococcus pyogenes (group A Streptococcus) infection, presenting with fever, arthritis, chorea, rash, and, crucially, carditis. Without effective secondary prophylaxis and care, acute rheumatic fever can progress to chronic rheumatic heart disease, a leading yet under-recognised cause of cardiovascular morbidity and death that disproportionately affects children and young adults aged 5-24 years in low-resource settings. Robust evidence implicates the role of antigenic mimicry-particularly between streptococcal M protein and cardiac or

Quantification of Human T Cell Responses to Chlamydia trachomatis: Intracellular Cytokine Staining (ICS) and Activation-Induced Marker (AIM) Assays.

High-resolution characterization of antigen-specific T cell responses is essential for understanding cellular immunity to Chlamydia trachomatis infection in humans. This chapter describes two complementary flow cytometry-based approaches for measuring human antigen-specific T cell responses: intracellular cytokine staining (ICS) and activation-induced marker (AIM) staining. Both assays enable single-cell-level analysis following antigen stimulation but differ in their readouts. ICS quantifies functional cytokine production in response to short- or long-term stimulation, whereas the AIM assay i

Primary adrenal small cell neuroendocrine carcinoma in a patient with sarcoidosis and synchronous colon adenocarcinoma.

Primary adrenal neuroendocrine carcinoma (NEC), particularly the small cell subtype, is exceptionally rare. In patients with sarcoidosis, inflammatory lymphadenopathy may mimic a metastatic disease, making disease detection difficult. A 76-year-old man with sarcoidosis was referred for an enlarging left adrenal incidentaloma that had increased in size from 7 mm to 19 × 14 mm over 3 years. Hormonal evaluation revealed unremarkable findings; laparoscopic adrenalectomy was performed because malignancy could not be excluded. Histopathological examination revealed small cell NEC with positive synap

Complex hybrid repair of multifocal visceral and aortoiliac pseudoaneurysms associated with neurosarcoidosis.

We report the case of a 36-year-old man with neurosarcoidosis who developed multiple pseudoaneurysms of the superior mesenteric artery (SMA), infrarenal aorta, and bilateral iliac arteries. Fluorodeoxyglucose-positron emission tomography demonstrated no significant uptake within the aneurysmal segments. Given the complex vascular anatomy, a staged hybrid approach was undertaken. Initial treatment consisted of a hepatic artery-to-distal SMA bypass followed by plug embolization of the SMA aneurysms. Subsequently, aortoiliac reconstruction was performed with a left aortouni-iliac endograft, femor

Diagnostic challenges of masked arginine vasopressin deficiency in neurosarcoidosis complicated by sodium-glucose cotransporter 2 inhibitor use.

A wide variety of endocrine disorders caused by sarcoidosis have been reported to date. We report the case of an 82-year-old male patient who presented with hypercalcemia and anterior pituitary hormone deficiency at the time of his sarcoidosis diagnosis and was subsequently diagnosed with arginine vasopressin deficiency (AVP-D) 6 months after the initiation of glucocorticoid therapy. Magnetic resonance imaging revealed an enlarged pituitary gland, thickened pituitary stalk, and loss of the posterior pituitary bright spot, raising the suspicion of hypothalamic-pituitary sarcoidosis. Despite imp

Heerfordt-Waldenström Syndrome With Cardiac Sarcoidosis.

Heerfordt-Waldenström syndrome is a rare presentation of sarcoidosis with only a handful of cases reported worldwide. It consists of a tetrad of uveitis, parotitis, and VII nerve palsy and chronic fever. Symptomatic cardiac involvement in sarcoidosis is an uncommon but potentially lethal manifestation. A timely diagnosis and treatment can result in the prevention of morbidity and mortality. We report a case of a young woman with multisystem sarcoidosis who presented with constitutional features, uveitis, multiple cranial nerve palsies, leptomeningitis, and ankle oligoarthritis. Intermittent fasting exacerbates microplastic-induced gut inflammation via Rikenella-mediated regulation of purine metabolism and Th17/Treg balance.

The pervasive accumulation of microplastics (MPs) in ecosystems and human bodies has prompted growing concern regarding their biological effects. However, whether intermittent fasting (IF) modulates MP-induced gut injury remains unclear. We established a murine model of chronic oral exposure to 1 μm polystyrene MPs combined with a daily 8-h feeding window. Multi-omics analyses, including 16S rRNA sequencing, untargeted metabolomics, and immune profiling, were performed. In a murine model of chronic oral exposure to 1 μm polystyrene microplastics, IF aggravated colonic inflammation and oxidativ

TCRζ-driven pre-signaling organization of Lck in Rab11⁺ endosomes shapes TCR activation.

BACKGROUND: T cell activation relies on the precise spatiotemporal regulation of T cell receptor (TCR) signaling at the immunological synapse, where vesicular trafficking coordinates the delivery of key signaling molecules. Endosomal pools of lymphocyte-specific protein tyrosine kinase (Lck) and its immediate substrate, the TCRζ chain, have been linked to TCR signaling competence, yet the mechanisms that regulate and coordinate their trafficking routes remain unresolved. METHODS: We simultaneously tracked the endosomal localization dynamics of endogenous Lck, ζ chain, and the TCR in unperturb

Efferocytosis activates a DNMT3A-mediated oxidized DNA repair pathway to enable tissue resolution.

Efferocytosis, the clearance of apoptotic cells by macrophages, promotes tissue resolution. Efficient resolution requires efferocytosis-induced macrophage proliferation (EIMP) to expand pro-resolving macrophages. Here, we show that efferocytosis activates base excision repair (BER) to remove 8-OHdG from DNA, enabling EIMP. Mechanistically, efferocytosis promotes poly(ADP-ribose) polymerase 1 (PARP1) chromatin binding and PARylation to facilitate DNA repair complex assembly and increases nuclear MutT homolog 1 (MTH1) (gene NUDT1), which hydrolyzes 8-OHdG. Both processes require DNA-methyltransf

Locoregionally delivered PD-1 blockade augments B7-H3 targeting CAR T cell efficacy in models of glioblastoma.

Glioblastoma (GBM) is a lethal brain tumor with a 95% five-year mortality rate, and remains a major unmet clinical need. An innovative approach to target GBM is administration of chimeric antigen receptor (CAR) T cells. In this study we identified B7-H3 as a favorable target antigen for CAR T cell therapy of GBM and developed a B7-H3 targeting CAR T cell. This CAR demonstrates robust, antigen-specific cytolysis and cytokine production, low tonic signaling, and preservation of a stem/naïve cell phenotype. Using orthotopic xenograft models of GBM we show that a single intravenous administration
