TCRαβ repertoire plasticity defines universal CD8+ T cell responses directed at influenza-specific HLA-A*02:01-PB1413-421 epitope.

HLA-A*02:01-restricted PB1413+CD8+ T cells provide the broadest reported cross-reactive immunity against influenza A, B, and C viruses. Here, we investigated T cell receptor (TCR) clonotypic signatures within these universal A2/PB1413+CD8+ T cell pools, compared with immunodominant influenza A-specific M158+CD8+ T cells and codominant influenza B-specific A2/HA543+CD8+ memory T cells. Using single-cell reverse transcriptase polymerase chain reaction and RNA sequencing methodologies, we defined TCRαβ repertoire composition and diversity. Our study shows that CD8+ T cells responding to these 3 e

Annexin A5-engineered nanovaccines synchronize spatiotemporal antigen release with phagocytosis checkpoint blockade for potent in situ cancer immunotherapy.

In situ generation of autologous tumor vaccines from dying tumor cells offers a promising route for personalized cancer immunotherapy, yet is often limited by phosphatidylserine (PS)-mediated immunosuppressive clearance that can restrict downstream immune activation and favor M2-like macrophage polarization. Here, we report a sequentially orchestrated nanovaccine (NT@ANX5-P) that couples photodynamic therapy (PDT)-induced immunogenic tumor-cell damage with on-demand PS masking to enhance in situ tumor vaccination. The nanovaccine consists of a photosensitizer core cloaked with genetically engi

TNFα promotes brain-predominant EAE independent of IFNγ and IL-17A in HLA-DR3 transgenic mice.

Multiple sclerosis (MS) is thought to be mediated by human leukocyte antigen (HLA) class II-restricted autoreactive CD4 T cells producing interleukin (IL)-17A and/or interferon γ (IFNγ), but the necessity of these cytokines for disease pathogenesis is unclear. Utilizing HLA-DR3 (DRβ1*0301) transgenic mice lacking IFNγ or IL-17A in experimental autoimmune encephalomyelitis (EAE), a model of MS, we found that neither cytokine is essential for disease induction, although both have distinct roles. The absence of IFNγ caused atypical, brain-predominant EAE characterized by high levels of IL-17A, wh

Adipokine regulation of immune cell polarization in oral cancer and its impact on chemotherapy sensitivity.

Oral squamous cell carcinoma (OSCC) is the most common malignant tumor in the head and neck region. In locally advanced patients, neoadjuvant chemotherapy centered on cisplatin and 5-fluorouracil often leads to primary/acquired resistance, with a 5-year survival rate of less than 50%. Tumor microenvironment (TME) immunosuppressive remodeling is the core cause of chemotherapy failure. Adipokines secreted by cancer-associated adipocytes (CAAs) are the key mediators that link metabolic disorders, immune polarization and chemotherapy resistance. The adipokines leptin, adiponectin, chemerin, resist

Peripheral effector-memory CD4+ T cells reflect clinical symptom burden in pediatric allergic rhinitis.

BACKGROUND: Pediatric allergic rhinitis (AR) can substantially affect quality of life and school performance, yet conventional sensitization markers such as allergen-specific immunoglobulin E (sIgE) do not consistently reflect symptom severity. We examined whether activation-induced marker (AIM)-defined allergen-specific peripheral effector-memory CD4+ T-cell (peff-mCD4) frequencies were associated with symptom burden. METHODS: In this pilot, single-center observational study, house dust mite (HDM)-specific responses were analyzed in 20 children and Japanese cedar pollen (JCP)-specific respon

18F-Fluorodeoxyglucose PET in Rheumatologic and Autoimmune Disorders.

FDG PET detects the silent inflammation of autoimmune and rheumatologic diseases even before tissue damage, as activated leukocytes overexpress glucose transporters. It has an important role in large vessel vasculitis which includes early diagnosis, disease monitoring and response assessment to treatment. In inflammatory arthritides, it quantifies synovitis and enthesitis and detects early treatment response. In connective tissue and immunoglobulin G4 related diseases, whole body imaging maps silent disease, helps exclude malignancy, and guides biopsy. It is central to cardiac sarcoidosis and

Immune Profiles Elicited by Different HBV Vaccine Modalities and Their Associations with Serological and Intrahepatic Viral Outcomes.

Therapeutic vaccination for chronic hepatitis B aims to restore HBV-specific T-cell immunity and contribute to a functional cure. We systematically compared the immune profiles and antiviral efficacy of four vaccine modalities: recombinant HBsAg (rHBsAg), antigen-antibody immune complex (IC), a DNA vaccine encoding HBsAg (DNA-HBsAg), and an HBsAg-Fc fusion DNA vaccine (DNA-HBsAg-Fc). In naïve mice, the rHBsAg vaccine predominantly induced anti-HBs IgG responses, whereas the IC vaccine elicited strong anti-HBs IgG and detectable CD4+ T-cell responses. In contrast, DNA vaccines preferentially in

The cerebrospinal fluid immune microenvironment in lung cancer leptomeningeal metastasis: cellular ecosystems and therapeutic possibilities.

Leptomeningeal metastasis (LM) is a devastating form of central nervous system (CNS) dissemination in lung cancer, particularly non-small cell lung cancer (NSCLC). Current treatment frameworks emphasize CNS drug penetration, oncogenic driver evolution, and intrathecal therapy. Emerging evidence supports viewing the cerebrospinal fluid (CSF) compartment as an active tumor-immune space, but the field remains early, with limited lung cancer-specific mechanistic and single-cell datasets. Lung cancer cells in CSF appear to adapt to low protein content, limited nutrients, fluid shear, weak matrix su

Antiviral and inflammatory immune signatures in anti-CD20-treated multiple sclerosis: a cross-sectional study.

CD20-targeting monoclonal antibodies are highly effective therapies for relapsing multiple sclerosis (MS). We examined how anti-CD20 therapy reshapes antiviral T-cell immunity, innate inflammatory tone, and blood markers of neuroaxonal injury. In this cross-sectional study, adults with relapsing MS were enrolled at two neurological centers in Rome. An immunophenotyping cohort (45 untreated, 25 anti-CD20-treated, all receiving ocrelizumab) was profiled by high-parameter flow cytometry across more than 40 immune populations. A separate antiviral cohort (24 untreated, 13 treated, 19 healthy donor

An Ex Vivo 3D Co-Culture Model of the Bone Marrow Niche Better Predicts Clinically Relevant Epigenetic Target Modulation to Improve Immunotherapy in Multiple Myeloma.

Background: In the last decade, treatment options for multiple myeloma have increased remarkably, including antibodies or CAR-T cells targeting myeloma surface markers with unprecedented efficacy. To enhance immunomodulation and clinical efficacy even more, the combination of these agents with other drugs is highly relevant. A significant association between CD38 expression on myeloma cells and the cytotoxicity of the anti-CD38 antibody daratumumab has been shown. Targeting SERPINB5 and MSLN for Pancreatic Cancer Therapy.

Background/Objectives: Pancreatic cancer (PC) is a leading cause of cancer-related death in the United States and lacks efficient therapeutic options. Serine peptidase inhibitor, clade B, member 5 (SERPINB5) is non-inhibitory toward serine proteases, yet it has diverse biological functions, including regulating cell migration, cell death, gene expression, and stress responses. Mesothelin (MSLN) is a tumor-associated antigen that is broadly overexpressed on various malignant tumor cells and plays a crucial role in promoting proliferation, invasion, and metastasis. Methods: We integrated public

Baseline Liver Metabolic Health in Newly Diagnosed People Living with HIV Compared with HIV-Negative Controls: Cross-Sectional Findings from the Prospective MASHIV-TR Cohort.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is an important comorbidity in people living with HIV (PLWH), yet most data derive from individuals receiving antiretroviral therapy (ART). In the baseline cross-sectional analysis of the prospective multicentre MASHIV-TR cohort (nine centres, Türkiye), we compared 250 newly diagnosed, ART-naïve PLWH and 161 HIV-negative controls. Hepatic steatosis and liver stiffness were assessed by vibration-controlled transient elastography; MASLD was defined as hepatic steatosis, indicated by CAP ≥ 248 dB/m together with at least one cardiom

Research Progress on Targeted Therapies for Pemphigus Vulgaris and Bullous Pemphigoid: Mechanisms, Opportunities and Challenges.

Autoimmune bullous diseases (AIBDs) are a group of severe disorders characterized by autoantibodies that attack structural proteins within the skin/mucosa, leading to blister and bullae formation. Pemphigus vulgaris (PV) and bullous pemphigoid (BP) are among the most prevalent representative AIBDs. Traditional treatments involving corticosteroids and immunosuppressive agents are often associated with substantial adverse effects and a high rate of relapse. In recent years, the therapeutic landscape has evolved toward precision targeted therapies, encompassing biological agents, small-molecule d

Towards the development of an efficient nonlinear controller for ART and cART against HIV infection.

The Human Immuno-deficiency Virus (HIV) is responsible for attacking the CD4+ T cells present in human blood and causes the human immune system to become vulnerable for numerous contagious and opportunistic diseases. It is hence pertinent to curb the spread of HIV infection using anti-HIV drugs. This study integrates sigmoid function with conventional sliding mode control and incorporate it within the framework of multi-objective optimal model predictive controller to reduce the impact of HIV infection through anti-retro-viral therapy. The proposed algorithm is designed using a deterministic H

HIV-1 compartmentalization in the central nervous system during cryptococcal meningitis: Analysis of viral, fungal, and host factors.

Opportunistic infections in the central nervous system (CNS), such as cryptococcal meningitis (CM) have profound consequences for people living with HIV-1 (PWH), including blood-brain barrier impairment and neuroinflammation. To investigate how CM may shape the HIV-1 populations in the CNS, we characterized viral variants in the blood and cerebrospinal fluid (CSF) compartments in PWH with active CM. Thirty-nine participants with CM had paired plasma and CSF viral load measurements prior to the start of anti-retroviral therapy (pre-ART), with overall significantly higher HIV-1 viral loads in pl

Expert intuition vs. Algorithmic clustering: Mapping radiological subtypes in pulmonary sarcoidosis.

BACKGROUND: Pulmonary sarcoidosis is a heterogeneous granulomatous disease with a broad spectrum of clinical and radiological manifestations. While the traditional Scadding classification based on chest radiography remains a cornerstone of disease staging, it provides limited information regarding parenchymal involvement and poorly correlates with functional impairment. High-resolution computed tomography (HRCT) offers superior sensitivity in detecting subtle and early interstitial changes, yet standardized HRCT-based classification systems for sarcoidosis remain underdeveloped. Client, health care provider, and policymaker perspectives on point-of-care/near-point-of-care testing for sexually transmitted infections in community pharmacies in Uganda.

INTRODUCTION: Point-of-care or near-point-of-care (POC/near-POC) tests for sexually transmitted infections (STIs) are often unavailable and unaffordable in low-resource settings, despite the World Health Organization's (WHO) recommendation that such testing be offered, leading to empiric treatment. Private community pharmacies (CPs) are often the first point of contact for people with symptoms of STIs, where pharmacists dispense treatment without diagnostic testing or prescriptions. This study evaluated clients,' providers,' and policymakers' perspectives on POC/near-POC STI testing in communi

Benzo[a]pyrene and bisphenol A interact to induce combined pulmonary toxicity through an integrated transcriptomic-metabolomic network.

Widespread atmospheric pollutants such as benzo[a]pyrene (BaP) and bisphenol A (BPA) persistently coexist, yet their combined risks to the respiratory system remain largely overlooked. In this study, C57BL/6J mouse and A549 cell models were employed to investigate the combined pulmonary toxicity induced by BaP and BPA, with an integrated transcriptomic-metabolomic framework established to characterize their response patterns. Combined exposure to BaP and BPA caused pathological damage in mouse lung tissue and specifically induced a broader spectrum of adverse effects. In vitro experiments demo

Reprogramming immunosuppressive TMEs with Salmonella-delivered sialidase and COX-2 blockade synergistically enhances cancer immunotherapy.

Sialic acid is a component of the extracellular matrix that is upregulated in multiple tumors and plays critical roles in tumorigenesis and progression. Sialic acid engages sialic acid-binding immunoglobulin-like lectins (Siglecs) on immune cells to suppress immune activation. Despite the potential of sialylation-targeted therapies to reactivate antitumor immunity, their clinical translation remains limited by suboptimal drug delivery systems and insufficient tumor specificity. To overcome these obstacles, we engineered a sialidase-expressing S. typhimurium and termed it SLpnanH. The engineere
