Systemic immune activation coexists with compartment-specific regulatory enrichment in matched blood, peritumor, and tumor tissue from advanced oral squamous cell carcinoma.

BACKGROUND: Oral squamous cell carcinoma (OSCC) is the most prevalent form of oral cancer and is characterized by a poor prognosis and frequent immune evasion. While the tumor microenvironment (TME) plays a central role in modulating responses to therapy, its relationship with systemic immune signatures remains poorly understood. METHODS: We performed high-dimensional immune profiling in matched peripheral blood, adjacent non-neoplastic peritumor mucosa, and tumor tissue from Human Papillomavirus (HPV)-negative, treatment-naïve patients with advanced OSCC. Soluble mediators were quantified by

Spatial immune ecosystems and therapy resistance in large B-cell lymphoma: from single-cell multi-omics to immunotherapy-matched validation.

Large B-cell lymphomas, particularly diffuse large B-cell lymphoma (DLBCL), provide a clinically important model in which tumor-intrinsic programs, immune recognition, effector-cell fitness, and tissue architecture jointly shape response and resistance to immunotherapy. Although molecular classification has refined the understanding of lymphoma biology, relapse after chemoimmunotherapy, chimeric antigen receptor (CAR) T-cell therapy, CD20×CD3 bispecific antibodies, antibody-drug conjugates (ADCs), and immune-combination strategies cannot be explained by genetics or target-antigen expression al

CAR-T cell-induced cytokine release syndrome is rapidly alleviated by Tripterygium glycosides: a case series and laboratory examination study.

BACKGROUND: Cytokine release syndrome (CRS) is a life-threatening complication of chimeric antigen receptor T cell (CAR-T) therapy. Macrophages/monocytes are mediators of CRS. Tripterygium glycosides is an immunomodulator, which could inhibit macrophages/monocytes in animal models. METHODS: Two patients with relapsed and refractory hematological malignancies developed CRS after receiving CAR-T therapy. They received short-term Tripterygium glycosides orally. RESULTS: Both patients showed rapid mitigation of fever with evidently decrease in elevated inflammatory cytokines within 72 hours. The

A positive feedback loop of kynurenine-AhR-EMT upregulates tryptophan metabolic enzymes via epigenetic modification to facilitate immune escape in triple-negative breast cancer.

BACKGROUND: Triple-negative breast cancer (TNBC) is prone to immune escape, and the synergistic mechanism of kynurenine, AhR, and EMT remains unclear. METHODS: TNBC cell lines and 4T1 orthotopic breast cancer mice were used for molecular, cellular, and in vivo intervention experiments. RESULTS: Kynurenine induced EMT in an AhR-dependent way, promoted cell migration and invasion, upregulated CCL2/CCL22, and inhibited the function of CD8⁺GrB⁺ T cells. Snail overexpression promotes H3K4me3 histone modification at IDO1 and TDO2 promoters to epigenetically amplify tryptophan metabolic enzyme expr

Distinct time-dependent S. Typhi-specific CD8+ T cell programs are associated with clinical outcome following Ty21a immunization and wild-type S. Typhi challenge in humans.

INTRODUCTION: CD8+ T cells are essential components of the adaptive immune system that play a critical role in determining the clinical severity of typhoid disease by directly killing Salmonella enterica serovar Typhi (S. Typhi)-infected cells, producing cytokines, chemokines, and establishing memory responses that have been linked to clinical outcome in a controlled human infection model (CHIM) of typhoid. Despite decades of extensive research, the role of memory CD8+ T (TM) cells and its subsets as correlates of protection against S. Typhi infection and typhoid disease remains incompletely u

Pre-transplant bone marrow lymphocyte subsets predict outcome after allogeneic stem cell transplantation.

INTRODUCTION: Recent evidence has demonstrated that both donor- and recipient-derived immune cell subpopulations play a crucial role in various immune responses following allogeneic hematopoietic stem cell transplantation (HSCT). In this retrospective single center analysis, we investigated whether different lymphocyte subpopulations in the bone marrow (BM) of the recipient prior to HSCT might have an impact on outcome parameters after HSCT. METHODS: BM aspirates of 140 patients > 18 years of age receiving a HSCT between 2008 and 2022 were analyzed. BM aspiration was performed within 3 months

replicateFest: an R package and shiny app for analysis of T cell receptor repertoire data from the functional expansion.

MOTIVATION: The Functional Expansion of Specific T cell (FEST)-based assays combine short-term peptide stimulation with TCR sequencing to identify clonotypes that expand in response to specific antigens. These approaches have proven invaluable for detecting antigen-specific T cell responses, guiding vaccine development, and assessing checkpoint blockade efficacy. However, variability introduced by biological and technical replicates poses challenges for reproducibility and interpretation, and existing computational tools do not address replicate-level analysis in these assays. RESULTS: We dev

Anti-PD-L1 scFv-secreting fifth-generation GD2-CAR T cells enhance antitumor function against GD2-low retinoblastoma.

Retinoblastoma (RB) is an aggressive pediatric intraocular malignancy in which chemoresistant or relapsed disease often requires enucleation, leading to vision loss. GD2-targeted chimeric antigen receptor (CAR) T cells are promising for GD2-positive tumors, but PD-L1-mediated immunosuppression and reduced surface antigen density can limit their activity. We developed a fifth-generation anti-GD2 CAR (GD2-CAR5), defined here as a fourth-generation backbone that additionally drives secretion of an anti-PD-L1 single-chain variable fragment (scFv) from the same transgene cassette, incorporating a h

Chronic XBP1s-mediated activation of the unfolded protein response impairs CD8+ T cell anti-tumor immunity.

CD8+ T cells play a crucial role in anti-tumor immunity; however, their functionality is compromised by exhaustion driven by chronic antigen exposure, poor tumor microenvironment, and metabolic dysfunction. Activated CD8+ T cells undergo extensive proteomic remodeling, which includes the induction of the IRE1-XBP1 signaling axis of the unfolded protein response. However, the functional consequences of sustained IRE1-XBP1 activation in exhausted CD8+ T cells remain elusive. Using a model of constitutive XBP1 activation in CD8+ T cells, we demonstrate that sustained XBP1s signaling leads to redu

Split intein-mediated reconstitution of regulatable CAR T-cells.

INTRODUCTION: Chimeric antigen receptor (CAR) T-cell therapy has transformed the treatment of B-cell malignancies. However, strategies that improve the modularity and controllability of CAR signaling could broaden its applicability and safety across additional clinical contexts. Here, we describe a regulable CAR T-cell platform based on split intein-mediated protein trans-splicing (PTS) to conditionally reconstitute functional receptors in primary T-cells. METHODS: As a proof of concept, we engineered a split CAR targeting CD1a (NCT05679895), a surface antigen expressed in T-cell acute lympho

Resveratrol as an immune checkpoint inhibitor to enhance the efficacy of breast cancer immunotherapy.

A major goal in cancer treatment is to convert "cold" tumors into "hot" ones by altering the tumor microenvironment (TME). Breast cancer is often a cold tumor and is resistant to immunotherapy. Resveratrol is a polyphenolic compound found in grapes, with antioxidant, anti-inflammatory, and potential cardioprotective and anti-aging properties. Resveratrol promotes the conversion of immunologically cold breast tumors into hot tumors, thereby increasing the efficacy of immunotherapy. This article reviews the potential of resveratrol to convert immunologically "cold" breast tumors into "hot" tumor

Nidogen 1-enriched extracellular vesicles drive HCC progression by inducing mregDC differentiation and Treg infiltration.

Aim: Nidogen 1 (NID1) is a secreted pro-tumorigenic protein. While extracellular vesicle (EV)-NID1 is known to accelerate hepatocellular carcinoma (HCC) progression, its role in reshaping the HCC immune microenvironment remains undefined. This study investigated how EV-NID1 modulates dendritic cells (DCs) to establish an immunosuppressive niche that drives liver cancer growth. Methods: The TIMER3.0 database was used to assess the correlations of NID1 with DCs and regulatory T cells (Tregs). A Transwell co-culture system using bone marrow-derived DCs was employed to evaluate the paracrine effec

Question-Driven Selection of Psoriasis Animal Models: A Structured Narrative Review of 48 Model Records.

Animal models connect psoriasis mechanisms to preclinical testing, but no model is informative for every question. This structured narrative review used PubMed, supplemented by reference-list screening and manual checks, to identify 29 original studies contributing 48 model records. One reviewer conducted screening and extraction, with targeted verification by a second reviewer. Within the selected records, IMQ- and IL-23-induced models primarily addressed acute inflammatory pathways, genetically engineered and composite models tested defined perturbations, humanized models examined human immu

Association of phenotypic and functional characteristics of regulatory T cells from minor salivary glands with expression of the delta 7 FoxP3 splice variant in patients with Sjögren's disease.

BACKGROUND: Sjögren's disease (SjD) is a systemic autoimmune disorder characterized by lymphoplasmacytic infiltration of exocrine glands, including minor salivary glands (MSG), with potential formation of ectopic germinal centers. Regulatory T cells (Tregs) are critical suppressors of autoimmune responses, and their dysfunction has been implicated in SjD pathogenesis. FoxP3, the master transcription factor of Tregs, undergoes alternative splicing to produce four variants: full-length (FL), Δ2, Δ7, and Δ2Δ7. While FoxP3 splice variants are known to influence Treg proliferation and suppressive a

Lipid-based mRNA chimeric antigen receptor T cells for enhanced in vitro antitumor activity.

Chimeric antigen receptor (CAR) T cell therapy provides substantial clinical benefits in hematologic malignancies. However, cell engineering using conventional viral vectors is costly, labor intensive, and prone to safety risks, while electroporation-induced cellular damage limits manufacturing scalability. This study reports a safe, high-efficiency, nonviral platform for generating mRNA-engineered CAR T cells. Herein, eight ionizable lipids are synthesized and formulated into lipid nanoparticles. Systematic screening and structural optimization identified lipid 6 as the lead candidate. Transf

From high-salt diet to cancer immunity: gut microbiota and immune system interactions and their mechanisms.

High-salt diet (HSD) has traditionally been studied in the context of cardiovascular disease, hypertension, and kidney disease. Increasing evidence now suggests that salt intake may also influence cancer immunity through direct effects on immune cells and indirect effects mediated by the gut microbiota. HSD can regulate inflammatory activation in T cells, macrophages, NK cells, and other immune populations. It can also reshape gut microbial composition and alter microbial metabolites, including short-chain fatty acids (SCFAs), bile acids (BAs), and tryptophan (Trp) metabolites. These changes m

Gene expression signatures from single-cell transcriptomics predict Sjögren's syndrome.

Sjögren's syndrome (SjS) is a systemic autoimmune disorder characterized by lymphocytic infiltration of exocrine glands, leading to dry eyes and mouth. While previous genome-wide association studies (GWAS) and transcriptomic analyses have identified genes associated with SjS, predictive models based on single-cell resolution are limited. In this study, single-cell RNA sequencing (scRNA-seq) data from peripheral blood mononuclear cells (PBMCs) of SjS patients were analyzed to map immune cell alterations linked to the disease. Compared with healthy controls, SjS patients displayed decreased prop

Microbial influences on immune modulation and colorectal cancer progression through combined transcriptomic and microbiomic analysis.

Schematic representation of the distinct tumor microenvironment characteristics in CA1 and CA2 subtypes. CA1 illustrates an immune-active tumor microenvironment characterized by elevated infiltration of immune cells, particularly CD8 + T cells and macrophages, and enhanced IFN-γ signaling. This subtype shows enrichment of specific bacterial species (including P. gingivalis and B. fragilis) that contribute to immunomodulation through metabolic pathways such as γ-aminobutyric acid (GABA) and butanoate metabolism. Despite high immune activity, CA1 exhibits a poor prognosis, potentially linked to

Microbiome-pancreatic cancer crosstalk: From tumor biology to therapeutic intervention.

Pancreatic cancer (PC) remains one of the most lethal malignancies worldwide, characterized by dense desmoplastic stroma, profound immunosuppression, and poor responsiveness to conventional therapies. Accumulating evidence positions the intratumoral microbiome, encompassing bacteria as well as understudied fungal and viral components, as a critical regulator of PC pathogenesis, tumor microenvironment (TME) remodeling, and therapeutic efficacy. In this review, we synthesize the current understanding of the pancreatic intratumoral microbiome, including its distinct composition relative to normal
