Extracellular vesicles and immune-glial remodeling in lung cancer brain metastasis: potential intersections with exercise-related myokines.

Lung cancer brain metastasis develops through reciprocal interactions between metastatic tumor cells and a brain-specific vascular, glial, and immune microenvironment. Tumor-derived extracellular vesicles (EVs) can participate in this process by altering blood-brain barrier (BBB) properties, influencing brain-directed distribution, and transferring regulatory cargo to endothelial and myeloid recipient cells. This review integrates direct evidence from non-small cell lung cancer (NSCLC) brain or central nervous system metastasis with related lung cancer EV studies, while distinguishing these fi

Differential requirements for cytokine signaling and local antigen in upper versus lower respiratory tract CD8+ TRM development.

CD8+ tissue-resident memory T cells (TRM) in the upper respiratory tract (URT) limit viral replication after transmission and prevent spread to the lower respiratory tract (LRT), whereas LRT CD8+ TRM are important for limiting immunopathology in the lung and promoting efficient viral clearance. Although the presence of local respiratory antigen is thought to be required for CD8+ TRM development in the LRT but not the URT, the relationship between local antigen encounter and cytokine signaling that influences TRM differentiation in distinct niches of the respiratory tract remains unknown. Here,

RNA-seq-based transcriptome analysis identifies TCR-expressing B1 cells as a multipotent lymphoid intermediate.

Classically, B and T lymphocytes are regarded as distinct lineages that can be readily distinguished by the receptors expressed on their surface. However, our recent study established that peritoneal B1 cells serve as precursors for extrathymic T cells through a TCR+CD19+ double-positive (DP) intermediate. To investigate the biological characteristics of these TCR-expressing B1 cells, we isolated them from the murine peritoneal cavity and compared their transcriptomes with those of B1 cells. DP cells displayed transcriptional signatures more closely associated with innate T cell immunity than

RBPJ regulates differentiation and effector programming of intestinal γδ intraepithelial lymphocytes.

Intestinal γδ intraepithelial lymphocytes (IELs) are heterogeneous tissue-resident T cells that contribute to epithelial homeostasis and host defense, but the mechanisms regulating their differentiation and functional states remain incompletely understood. Here, we investigated the role of recombination signal binding protein for immunoglobulin kappa J region (RBPJ) in intestinal γδ IELs using mice with CD8 lineage-specific deletion of Rbpj. RBPJ deficiency did not markedly affect the overall proportion of γδ IELs but altered the distribution of Thy1-defined γδCD8αα IEL subsets in the small in

Genetic and clinical evidence supports HLA-DR+ CD8+ T cells as a favorable immune biomarker in acute myeloid leukemia.

Immune dysregulation is central to acute myeloid leukemia (AML), but the contribution of specific immune subsets remains uncertain. We combined Mendelian randomization (MR), clinical flow cytometry, and transcriptomic profiling to investigate HLA-DR+ CD8+ T cells in AML. Two-sample MR using 731 immunophenotypes and AML data from UK Biobank and FinnGen identified four HLA-DR-related T-cell traits associated with lower AML susceptibility. In 85 non-APL patients, HLA-DR+ CD8+ T-cell proportions increased after complete remission, while among 46 newly diagnosed patients, a lower baseline HLA-DR+ C

Site-Specific Cutaneous Inflammation: Mechanisms and Therapeutic Implications.

Inflammatory skin diseases exhibit prominent anatomical distribution patterns that have long guided clinical diagnosis, yet the biological mechanisms underlying site specificity remain incompletely defined. These regional predilections arise from structural, immunological, and microbial heterogeneity across human skin. Epidermal barrier architecture, which varies substantially between body sites, including 10- to 100-fold differences in stratum corneum thickness, and prevalence and types of hair follicles and glands, creates distinct microenvironments that shape resident immune populations, mi

CD4+ T Cell Activation and Peripheral Immune Cell Influx into the Brain Following Wildfire Smoke Exposure is Modulated by a Saturated Fat Diet.

Over the past 40 years, wildfires across the United States have steadily increased, in terms of acreage burned. Smoke from wildfires can exacerbate several cardiovascular and respiratory diseases, with new studies highlighting potential sustained neurological outcomes. The purpose of this study was to delineate the role of peripheral immune populations in woodsmoke-induced neuroinflammation, the timeline of this response, and the impact of diet on each of these factors. Based on previous research on other pollutants and diet introduction, we included a highly saturated fatty acid (coconut oil)

The aminoadamantane derivative 3F2 modulates monocyte and NK cell activation and restores immune mediators altered by Monkeypox virus stimuli.

The monkeypox virus (MPV) is known to employ multiple immune evasion strategies that compromise antiviral defenses and disrupt immune homeostasis. Here, we characterized the immune responses induced by stimulation with inactivated MPV and evaluated the ability of aminoadamantane (AMA) and its derivative, 3F2, to restore immune function. Peripheral blood mononuclear cells that were stimulated with MPV exhibited altered activation in a subset of monocytes, along with suppressed activation of natural killer (NK) and natural killer T (NKT) cells. MPV stimulation also led to significant changes in

Liver-specific amino acid metabolism impacts on efficacy of cancer immunotherapy.

The combination immunotherapies, including those with atezolizumab and bevacizumab (Atez/Bev), have become standard therapy for hepatocellular carcinoma (HCC). However, its efficacy remains limited and reliable biomarkers are lacking. In this study, we analyzed paired tumor and background liver tissues from Atez/Bev-treated HCC patients and found that high programmed cell death-1 (PD-1) expression in CD8+ tumor-infiltrating lymphocytes (TILs) correlated with therapeutic response independent of etiology. RNA sequencing on the tumor samples revealed that the branched-chain amino acid (BCAA)-rela

EGFR-antibody-decorated cancer-cell-membrane-coated MOF nanoplatform enables targeted delivery of the STING agonist diABZI for pancreatic cancer immunotherapy.

Pancreatic ductal adenocarcinoma (PDAC) has limited immunotherapeutic effect because of poor drug penetration and tumor immunosuppression. Activation of the stimulator of interferon genes (STING) pathway may promote antitumor immunity, but systemic administration of STING agonists is constrained by inadequate tumor accumulation. Epidermal growth factor receptor (EGFR) is frequently overexpressed in PDAC and is closely associated with tumor growth, invasion, and treatment resistance. Herein, we developed an EGFR-antibody-decorated cancer-cell-membrane-coated metal-organic framework (MOF) nanopl

Safety, Pharmacokinetics, and Efficacy Study of Porcine and Rabbit Antithymocyte Globulin in the Treatment of Patients With Severe Aplastic Anemia: A Multicenter, Prospective, Randomized, Open-Label Clinical Trial.

Severe aplastic anemia (SAA) is a T lymphocyte-mediated bone marrow failure disorder. Only porcine ATG (p-ATG) and rabbit ATG (r-ATG) are available in mainland China, with the optimal first-line regimen undetermined. This study compared their pharmacokinetics, efficacy, and safety. Ninety-three SAA patients ineligible for transplantation or lacking a suitable donor (p-ATG, n = 55; r-ATG, n = 38) who were receiving immunosuppressive therapy were prospectively enrolled between 2020 and 2024. The primary endpoint was dynamic changes in lymphocyte subsets. Both ATGs peaked on treatment Day 5; r-AT

Oral Astilbin Nanogels Modulate Senescence-Associated CD4+ T Cells and Promote Treg Induction in Aged Mice with Colitis.

Inflammatory bowel disease (IBD) in older individuals is characterized by immune senescence and deteriorating intestinal homeostasis, both of which limit the efficacy of current therapies. Here, we develop fluorinated β-glucan/pectin nanogels (F2-Ast@NGs) for oral delivery of astilbin, a natural flavonoid with anti-inflammatory and immunoregulatory activities. This multifunctional platform integrates gastrointestinal protection, colon-targeted release, and local immune modulation. The β-glucan shell preserves nanogel stability during upper gastrointestinal transit while enabling β-glucanase-re

HECTD1 Directs HIF-1α to Facilitate Intestinal Inflammation and Carcinogenesis via Promoting Th17 Differentiation and Glycolysis.

Targeting glucose metabolism in Th17 cells may be beneficial for treating inflammatory bowel disease (IBD) and colitis-associated cancer (CAC), however, the molecular mechanisms orchestrating Th17 cell metabolism remain unclear. Here, we revealed that HECTD1 regulates IGF2BP3-dependent HIF-1α translation to promote Th17 cell glycolysis and differentiation and that HECTD1 expression correlates with IBD and CAC pathogenesis. T-cell-specific deletion of HECTD1 mitigated the severity of colitis and CAC in murine models. RNA-seq revealed enrichment of Th17 differentiation, glycolysis/gluconeogenesi

BeWo-Derived Extracellular Vesicles Downregulate IL-6Rα Expression via miRNAs on CD4+ T Lymphocytes.

PROBLEM: A balanced Th1/Th2/Th17/Treg status is necessary for a successful pregnancy. T cell polarization depends on the local cytokine environment and the responsiveness of T lymphocytes to cytokines. Trophoblast-derived extracellular vesicles (EVs) influence the function of maternal T lymphocytes, contributing to their polarization into the Treg phenotype. We examined whether BeWo-derived EVs affect T cell IL-6 responsiveness using the BeWo cell line as an experimental model and characterized the molecular cargo of BeWo -EVs that may contribute to this process. METHODS: Circulating lymphocy

Changes in Peripheral Immune Cell Subsets Following Endoscopic Sinus Surgery Differ Between CRSwNP and AERD.

INTRODUCTION: Changes in peripheral eosinophils follow endoscopic sinus surgery (ESS) in chronic rhinosinusitis with nasal polyps (CRSwNP), with and without aspirin sensitivity. Cytometry by time-of-flight (CyTOF) identifies more immune cell subsets than conventional assays or clinical testing. We hypothesized that ESS-associated changes in peripheral immune cell subsets differ between aspirin-tolerant (AT)-CRSwNP and aspirin-exacerbated respiratory disease (AERD) as measured by CyTOF. METHODS: Pre- and post-ESS peripheral blood from patients with CRSwNP (n = 13) or with AERD (n = 15) was ana

UcTCRp: a TCRβ-based framework for identifying and profiling MAIT- and iNKT-associated repertoire signals.

Mucosal-associated invariant T (MAIT) cells and invariant natural killer T (iNKT) cells are conventionally identified using invariant or semi-invariant TCRα chains, antigen-loaded tetramers, or transcriptomic phenotypes. These requirements limit their detection in public and clinical immune-repertoire datasets that contain only TCRβ sequences. Here we present UcTCRp, a TCRβ-only framework for detecting and profiling relative MAIT- and iNKT-associated signals in bulk immune repertoires. We first identified model-informative TCRβ features, including biased V-gene usage and reproducible CDR3β ami

ADAM9 Regulation of CXCL12-Dependent Myeloid-Derived Suppressor Cells Shifting the Immune Landscape of Lung Adenocarcinoma.

Lung cancer-mediated myeloid-derived suppressor cell (MDSC) inhibits antitumour immunity for tumour progression and is a promising target. Here, we present the ADAM9-targeting therapeutic strategy to unleash anticancer immunity. We identified a novel role for ADAM9 in promoting lung cancer immunosuppression through the upregulation of cytokines that drive the recruitment and expansion of MDSCs, with CXCL12 serving as one of the major mediators. This ADAM9-CXCL12-MDSC axis significantly impairs the function of cytotoxic CD8+ T cells. Disrupting the ADAM9-CXCL12 axis suppresses oncogenic signall

Insights into two modes of TdT action in human V(D)J recombination.

Terminal deoxynucleotidyl transferase (TdT) is a DNA polymerase expressed in pre-B and pre-T cells that functions in V(D)J recombination to add random nucleotides in a template-independent (TI) mode at junctions between antigen receptor gene segments. However, cellular, biochemical, and structural studies have also revealed a significant, short-length, template-dependent (TD) mode of TdT, in which it takes instructions from a template strand across a double-strand break. Here, we propose that this can occur through an intrachain fold-back synthesis to generate the short inverted repeats at V(D

Altered T cell immune profile and its prognostic implications in children with portal hypertension: A pilot study.

OBJECTIVES: Immune dysregulation in paediatric portal hypertension (PHT) is poorly understood. We aimed to characterise T cell phenotype and function and circulating immune mediators in children with PHT according to disease aetiology. METHODS: Forty-four children with PHT (27 chronic liver disease [CLD]-related and 17 portal vein thrombosis [PVT]-related) and 15 healthy controls were prospectively enrolled. Peripheral and portal blood samples were analysed using multiparameter flow cytometry and ELISA to assess T cell subsets, chemokine receptors, cytokines and soluble immune markers. Analys
