Insights into two modes of TdT action in human V(D)J recombination.

Terminal deoxynucleotidyl transferase (TdT) is a DNA polymerase expressed in pre-B and pre-T cells that functions in V(D)J recombination to add random nucleotides in a template-independent (TI) mode at junctions between antigen receptor gene segments. However, cellular, biochemical, and structural studies have also revealed a significant, short-length, template-dependent (TD) mode of TdT, in which it takes instructions from a template strand across a double-strand break. Here, we propose that this can occur through an intrachain fold-back synthesis to generate the short inverted repeats at V(D

Altered T cell immune profile and its prognostic implications in children with portal hypertension: A pilot study.

OBJECTIVES: Immune dysregulation in paediatric portal hypertension (PHT) is poorly understood. We aimed to characterise T cell phenotype and function and circulating immune mediators in children with PHT according to disease aetiology. METHODS: Forty-four children with PHT (27 chronic liver disease [CLD]-related and 17 portal vein thrombosis [PVT]-related) and 15 healthy controls were prospectively enrolled. Peripheral and portal blood samples were analysed using multiparameter flow cytometry and ELISA to assess T cell subsets, chemokine receptors, cytokines and soluble immune markers. Analys

A modular synthetic promoter toolkit for programmable gene control in activated and chronically stimulated T cells.

Synthetic circuits that couple transgene expression to endogenous transcription factor (TF) activity enable cellular response to changing functional states. In T cells, antigen stimulation results in dynamic changes of transcription factor expression programs. Harnessing these endogenous TF dynamics for programmable gene control remains an important challenge in engineered T cells. Here, we develop a library of synthetic promoters (SPs) responsive to seven TFs identified through transcriptomic analysis of T-cell activation programs. These promoters couple endogenous TF activity to transcriptio

Topically Delivered Treg-Attracting Microparticle-Hydrogel Composites Recruit Regulatory T Cells and Limit Cardiac Injury After Severe Ischemia.

Ischemic heart disease is a highly prevalent and debilitating condition throughout the world. Following an ischemic injury, the immune response can become overly active and spread damage to nearby healthy tissue. Because the heart lacks regenerative capacity, preventing excessive damage is critical. Existing immunosuppressive therapeutics provide indiscriminate suppression of inflammatory and repair functions of infiltrating immune cells. Regulatory T cells (Treg) are the body's highly sophisticated system for regulating immunologic activity. These cells can provide powerful immune suppression

Antigen-specific CD8 T cells accumulate and functionally reactivate in the bone marrow following brain infection.

BACKGROUND: Bone marrow is a primary immune organ responsible for hematopoiesis, yet its role in shaping adaptive immune responses remains incompletely understood. In particular, whether antigen-specific CD8 T cell responses can arise and persist within the bone marrow following brain infection is unclear. METHODS: We used intracranial infection with Theiler's Murine Encephalomyelitis Virus (TMEV) in C57BL/6 mice, along with complementary antigen delivery (OVA + Poly I:C) and systemic infection (LCMV) models. Antigen-specific CD8 T cells were quantified in the brain, secondary lymphoid organs

Glioblastoma immune landscape remodeling following treatment with CD70-targeted CAR-T cells.

Chimeric antigen receptor (CAR)-T cell therapy has shown limited efficacy in glioblastoma (GB), largely because of the immunosuppressive tumor microenvironment (TME). We have previously demonstrated that the ligand-based mCD70-directed construct mCAR27 specifically eliminates CD70⁺ GB cells in vitro, and induces durable tumor remission with prolonged survival in a syngeneic model of orthotopic GB. Despite the therapeutic promise of CD70 as a GB CAR-T cell target, the impact of CD70-directed CAR-T cells on the brain tumor immune landscape after target interception remains unexplored. Here, we i

Poloxamer-Based Th1-Biased NKT Cell Nano-Agonist for Tumor Immunotherapy.

BACKGROUND: Natural killer T (NKT) cells bridge innate and adaptive immunity and are promising targets for tumor immunotherapy. However, its agonist α-GalCer induces NKT cells to secrete both Th1-type and Th2-type cytokines. Coupled with the challenges of in vivo delivery, these drawbacks collectively limit its clinical application. METHODS: In this study, we constructed Th1-biased NKT cell nano-agonists (LGCP Ms) by using Th1-preferred glycolipids (Lys-α-GalCer) and Poloxamer through thin-film hydration combined with probe sonication to enhance NKT cell-mediated anti-tumor immunity. RESULTS

CD8+ T cell transcriptional reprogramming toward TCF7-low cytotoxicity drives immune aging in viral ARDS.

Viral acute respiratory distress syndrome (ARDS) exhibits distinct clinical outcomes across age groups, but the immune mechanisms underlying this disparity remain unclear. Here, we integrated single-cell transcriptomic profiling, T cell receptor repertoire analysis, serum proteomics, and functional validation to characterize age-dependent T cell remodeling in influenza-associated ARDS. Adult ARDS was characterized by reduced T cell receptor diversity, expansion of CD8+ effector memory T cells, and enhanced cytotoxic programs marked by increased granzyme B (GZMB) expression compared with pediat

[Tissue resident memory cells in psoriasis].

Tissue resident memory cells (Trm-cells) function as both benign and pathogenic memory cells. Benign Trm-cells play an important role in adaptive immunity by protecting against external pathogens, whereas pathogenic Trm-cells can induce relapses in several chronic skin diseases, including psoriasis. Early and intensive treatment with biologic therapy can lead to effective, long-term disease control by limiting local memory formation in the skin. However, this review finds it important not to compromise the beneficial role of Trm-cells in adaptive immunity when pursuing long-term treatment stra

Syntaxin11 deficiency inhibits CRAC channel priming to suppress cytotoxicity and gene expression in T lymphocytes independent of membrane trafficking.

Mutations in Syntaxin11 (STX11), a Q-SNARE, result in a fatal immune disorder known as familial hemophagocytic lymphohistiocytosis 4 (FHLH4) in human patients. A key diagnostic feature of FHLH4 is defective T and natural killer cell cytotoxicity. Here, we show that STX11 directly binds and regulates Orai1, the pore-forming subunit of calcium release-activated calcium (CRAC) channels. CRAC channels enable store-operated calcium entry (SOCE) from the extracellular space and are crucial for granule exocytosis and nuclear factor of activated T cells (NFAT)-dependent gene expression in activated ly

From immune heterogeneity to targeted therapies in IgG4-related disease.

PURPOSE OF REVIEW: Recent advances in single-cell, spatial, and multiomic technologies have provided new insights into the immunopathogenesis and clinical heterogeneity of IgG4-related disease (IgG4-RD). At the same time, several mechanism-based therapies have entered clinical development. RECENT FINDINGS: Recent studies have identified distinct B-cell, T-cell, myeloid, and stromal populations and their interactions within affected tissues. Type 2 peripheral helper T-cell-associated, granzyme K+ cytotoxic T-cell-associated, and age-associated B-cell-related programs may contribute to tissue i

Oncolytic Vaccinia Virus Armed With CLEC2A Enhances Antitumor Immunity via ROS-Autophagy Feedback Loop and Cytokine Activation.

Tumors often exhibit immunosuppressive microenvironments (TMEs) and present as immunologically "cold." Oncolytic viruses (OVs), particularly engineered vaccinia viruses (oncoVVs), are an encouraging therapy that directly lyses cancer cells and reprograms the TME. In this study, we evaluated the therapeutic potential of an oncolytic vaccinia virus expressing human C-type lectin domain family 2 member A (oncoVV-CLEC2A) in melanoma and ovarian cancer cells. OncoVV-CLEC2A elevated reactive oxygen species (ROS) levels, suppressed Nrf2-mediated antioxidant responses, and triggered autophagy. These c

An impaired T-cell-mediated mitogen response as a potential biomarker to aid in clinical diagnosis of pulmonary tuberculosis.

PURPOSE: A considerable proportion of suspected pulmonary tuberculosis (pTB) cases are identified and treated on the basis of clinical diagnoses, as it is often not possible to confirm Mycobacterium tuberculosis (Mtb) infection. In the absence of pathogen detection, supplementary indicators, such as immunological biomarkers, would facilitate TB diagnostic decision-making. METHODS: In this prospective case-control study conducted in Ghana, patients with suspected pTB (n = 83) were divided into three subgroups: confirmed cases with high (pTBhigh) or low (pTBlow) Mtb sputum load, and clinically

Immune Checkpoint Dysfunction Remodels Adaptive Immune Receptor Repertoires in CTLA-4 Insufficiency and LRBA Deficiency.

Cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) is a critical negative regulator of T-cell activation, and its function is impaired either directly in CTLA-4 insufficiency or indirectly through LRBA deficiency. Although the immunological consequences of these closely related inborn errors of immunity (IEI) have been extensively characterized, their impact on adaptive immune receptor repertoires (AIRRs) remains poorly understood. Here, we performed a comprehensive analysis of αβ T-cell receptor (TCR) and B-cell receptor (BCR) repertoires in patients with CTLA-4 insufficiency and LRBA defic

New insights into β-cell vulnerability in type 1 diabetes-Lessons from nature.

Type 1 diabetes results from an autoimmune-mediated destruction of pancreatic β-cells, yet considerable heterogeneity exists regarding disease onset, progression and severity. Emerging evidence suggests that this variability reflects not only differences in immune responses, but also differences in the architecture and immune landscape of the endocrine pancreas. Our recent Steve Morgan Foundation Grand Challenge studies have provided important 'lessons from nature', revealing that the endocrine tissue in the human pancreas exists as a continuum of endocrine objects (EOs), ranging from isolated

Boiling Histotripsy with 1.1/3.3 MHz Dual-Frequency Inducing Long-Duration Anti-Tumor Immunity in Mice Breast Cancer.

Triple-negative breast cancer (TNBC) has a poor prognosis and high risk of recurrence and metastasis. Histotripsy is an ultrasound-guided, non-invasive, non-thermal ablation method that mechanically lyses target tissues. A boiling histotripsy (DF-MS BH) technique combining 1.1/3.3 MHz superposition with a multi-stage ultrasound pulse scheme was proposed, aiming to enhance anti-tumor immune response by optimizing pulse dose and number of treatments. A custom-designed 1.1-/3.3-MHz two-element confocal-annular array and multi-stage ultrasound pulses were used for treatment. For 6-mm-diameter tumo

Structural and mechanistic insights into IL2RG-associated immunodeficiency through computational approaches.

Interleukin-2 receptor γ chain (IL2RG, CD132) act as a signaling subunits for various cytokine receptors, required for lymphocyte growth and maturation. Genetic variants within the IL2RG gene are associated with X-linked Severe Combined Immunodeficiency (X-SCID) disease. This study uses an in-silico method to investigate the structural and functional impacts of two IL2RG variants, Trp240Arg (W240R) and Arg226Cys (R226C), located in the extracellular domain, a region important for receptor stability. Homology modeling was generated for Wild-Type (WT) and Variant IL2RG structures and subsequentl

Germline Genomic and DNA Methylation Differences Between MGUS and Multiple Myeloma Among Patients of African Ancestry.

Individuals of African ancestry (AA) have approximately twofold higher risk of multiple myeloma (MM) than individuals of European ancestry, yet the molecular determinants between monoclonal gammopathy of undetermined significance (MGUS) and MM remain poorly defined. We performed whole-exome sequencing (WES) and genome-wide DNA methylation profiling in 28 AA patients with MM and 28 frequency-matched AA patients with MGUS without documented progression. After quality control, the WES analysis included 28 patients with MGUS and 27 with MM, while the DNA methylation analysis included 27 patients w

The Converging Roles of DNA Damage Resolution and Redox Dysregulation in Plasma Cell Transformation During the Evolution of Multiple Myeloma.

Increasing evidence indicates that dysregulated DNA damage response (DDR) pathways and persistent oxidative stress contribute to genomic instability, increase mutational burden, and promote disease progression. This study investigated the interplay between DDR-related parameters and redox status throughout Multiple Myeloma (MM) evolution. Bone marrow plasma cells (BMPCs) from 16 patients with monoclonal gammopathy of undetermined significance (MGUS), 18 with smoldering multiple myeloma (SMM) and 15 with MM, were analyzed. Measurements included redox status, assessed by the reduced-to-oxidized
