Hypogammaglobulinemia associated with accelerated catabolism of IgG secondary to its inteaction with an IgG-reactive monoclonal IgM.

Hypogammaglobulinemia due to a new pathophysiological mechanism was studied in a patient with Sjögren's syndrome, a monoclonal IgM and a mixed (IgM-IgG) cryoglobulinemia. The IgM (IgMdk) component of the cryogel possessed light chains of lambda-type with highly restricted electrophoretic mobility analagous to those of a Waldenström's macroglobulin. IgMdk reacted specifically with native IgG, with IgG subclasses 1, 2, and 4, and with the Fc piece of IgG to form a cryogel. Serum concentrations of IgG 1, 2, and 4 were 10% of normal, whereas the IgG3 level was slightly increased and the IgM level

IgA deficiency in children. A clinical study with special reference to intestinal findings.

Clinical, immunological, and intestinal studies on 26 children with IgA deficiency in the age range 2 to 16 years are reported. 9 of these children were suffering from autoimmune disease, namely thyroiditis (5), thyrotoxicosis (1), rheumatoid arthritis (2), and probable Sjögren's syndrome (1). The last-mentioned patient had defective cellular immunity. Altogether 11 patients were subject to recurrent respiratory tract infections. The symptomatology of the remaining patients was variable. In a boy with growth retardation, a chromosome anomaly was found, and endocrinological studies indicated to

Effects of therapeutically useful thiols (DL-penicillamine and alpha-mercaptopropionylglycine) on immunoglobulins.

The action of 0·05 M DL-penicillamine (DL-P) and α-mercaptopropionylglycine upon immunoglobulins, both in their native serum and after isolation, has been studied. IgM was always affected, apparently with breakdown of 19S polymers and release of 7S sub-units. This depolymerization leads both to a change in electrophoretic mobility and to increased diffusibility in gels, as well as to decrease in serum viscosity. IgG, as a rule, is not affected when studied in whole serum. The exception to the rule is provided by monoclonal proteins of the IgG3 subgroup. The observed effects are variable, and m

The effect of hydrocortisone on immunoglobulin metabolism.

The serum concentration of all subclasses of IgG (gamma(2)A, gamma(2)B, and gamma(1)) as well as IgA and IgM were reduced in normal and low pathogen mice receiving hydrocortisone acetate. Turnover studies using (131)I-labeled gamma(2)A subclass of IgG demonstrated that high dose corticosteroids cause a significantly shortened survival (increased catabolic rate) which contributes to the observed hypogammaglobulinemia. This increase in fractional catabolism is not due to excess loss in the urine or stool but reflects an increase in endogenous catabolism.

The transformation of human lymphocytes by monkey antisera to human immunoglobulins.

Cultures of human peripheral leucocytes were stimulated to incorporate tritiated thymidine when incubated with monkey antisera to human immunoglobulins. Twenty-five of forty-four monkey antisera were active and stimulated 90 per cent of leucocyte (WBC) cultures to incorporate a small but significantly greater amount of tritiated thymidine (TdR3H) than that incorporated by controls. This stimulation of TdR3H uptake correlated with an increase from 2 to 8 per cent lymphoblasts in the cultures. Leucocytes washed free of serum immunoglobulins responded to a greater degree to the anti-immunoglobuli
