The antibody mediated prevention (AMP) trials confirmed that broadly neutralizing antibodies (bNAbs) can prevent HIV-1 acquisition, and provided paradigm-shifting insights into HIV-1 biology and protective thresholds. HIV-associated pulmonary arterial hypertension (HIV-PAH) is rare and understudied. We pooled 12 right heart catheterization-confirmed cohorts (n = 537, 1986-2021) and found that HIV-PAH presents with severe hemodynamics and advanced functional limitation, with only 64% receiving PAH-targeted therapy at diagnosis. Survival improved progressively over three decades alongside increasing therapy use. These findings highlight persistent treatment gaps and support consideration of contemporary combination PAH therapy in people with HIV-PAH.

Severe combined immunodeficiency (SCID) is a disease of adaptative immunity and the most severe form of primary immunodeficiency. Without early diagnosis and proper management, infants are at increased risk of severe recurrent infections and death. The addition of a T-cell receptor excision circle (TREC) assay to newborn screening (NBS) has made an enormous impact on early disease diagnosis and management, resulting in improved survival. In this review, we will discuss the diagnosis and management of SCID and the impact of the TREC assay addition to NBS.
