STAT3 is a central mediator of pro-survival signaling within the STAT family, functioning through both canonical transcriptional activity and noncanonical mechanisms. As a pleiotropic regulator, STAT3 integrates cytokine, growth factor, and stress signals to coordinate diverse cellular processes, including epithelial architecture, T cell development, macrophage polarization, apoptosis, metabolism, and inflammation-associated wound healing. This integrative capacity positions STAT3 as a critical signaling hub at the interface of tissue homeostasis and immune regulation. Dysregulated STAT3 activ

Salmonella is a leading cause of foodborne illness in the United States, yet rural-urban differences in incidence and their underlying drivers remain poorly characterized. We conducted an ecological analysis of deidentified probable and confirmed Salmonella cases from the National Notifiable Diseases Surveillance System (NNDSS), 2011-2023, to evaluate temporal, regional, and population-level determinants of Salmonella incidence. Multivariable negative binomial models assessed rural-urban differences nationally and by U.S. Census region, and Blinder-Oaxaca decomposition quantified contributions

SUMMARYInvasive non-typhoidal Salmonella (iNTS) disease is a severe bloodstream and focal invasive infection caused by non-typhoidal serovars of Salmonella enterica, especially S. Typhimurium and S. Enteritidis. Its clinical importance is disproportionate to its routine visibility because disease often presents as undifferentiated fever, sepsis, meningitis, severe anaemia-associated deterioration, recurrent bacteraemia, or focal infection, while definitive recognition depends on blood or other sterile-site microbiology. Global estimates indicate substantial mortality, but the true incidence re

Vibrio cholerae is the causative agent of the human diarrheal disease cholera. Previous studies identified "hyperswarming" (HS) mutant V. cholerae with enhanced motility and decreased virulence factor expression, but the genetic basis of HS was unknown. Through whole-genome sequencing, we have identified point mutations within toxT, which encodes the master virulence regulator, as being responsible for the HS phenotype. These mutations are loss of function, and they eliminate transcription of cholera toxin and toxin-coregulated pilus genes. Using single-cell video tracking, these HS strains we

BACKGROUND: Information on the burden of disease caused by foodborne pathogens in Northwest China is limited. This study aims to estimate the burden of acute gastroenteritis (AGE) and foodborne salmonellosis, shigellosis and norovirus gastroenteritis in Gansu Province.
METHODS: Using data from population surveys conducted during 2011-2015, the burden of AGE was estimated in Gansu Province. By determining the number of pathogen-specific cases from sentinel hospital surveillance conducted during 2014-2019, with adjustments applied for healthcare-seeking behavior and stool specimen submission ba

Balamuthia mandrillaris is a rare and lethal amoeba that causes granulomatous amoebic encephalitis (GAE), a severe brain infection with a high mortality rate. This manuscript presents a case study of a 5-year-old female patient with B. mandrillaris amoebic encephalitis. She presented from an outside hospital (OSH) with daily fevers, fatigue, and worsening mental status. She was without comorbidities, and sick contacts were denied. Diagnosis was challenging due to nonspecific encephalitis symptoms and nonspecific neurological imaging. Neuroimaging revealed symmetric diffuse restriction in the m

New therapeutic approaches are needed to treat infections due to multidrug resistant (MDR) Gram-negative bacteria. The use of antibody-recruiting molecules (ARMs) is a promising approach to address the MDR problem given their potential to engage both the adaptive and innate immune systems to treat bacterial infections. Here we present two new strategies to create ARMs. First, we show that a 9-mer antimicrobial peptide (K6) can be fused to a 11-mer mimotope peptide recognized by antibodies targeting the Vibrio cholerae O1 O-antigen to produce an ARM peptide (FZ5) that can render E. coli more se

Tetanus neurotoxin (TeNT) invades the central nervous system (CNS) via retrograde transport through motoneurons. Whether a host factor is needed to mediate TeNT trafficking has remained unknown. Here, using pull-down and mass spectrometry analysis, and by developing a luciferase-based in vivo transport assay, we identify receptor protein tyrosine phosphatase sigma (PTPRS; PTPσ) as the retrograde transport receptor for TeNT. Global or motoneuron-specific PTPRS knockout (KO) abolishes TeNT entry into the CNS in mouse models. PTPRS-TeNT interactions occur in endosomes, prevent toxin translocation

The characterization of pathogens affecting Antarctic wildlife has been historically limited and sporadic. With the introduction of H5N1 high pathogenicity avian influenza virus (HPAIV) in Antarctica in the austral summer 2023/2024, concerns about the health of Antarctic seabirds and mammals prompted immediate surveillance efforts. In March 2024, we visited the Danger Islands archipelago, that hosts two mega-colonies of Adélie penguins, and observed an unusual mortality estimated in thousands of Adélie penguins and other species. Swabs and tissues were collected for molecular detection of infe

BACKGROUND: Blastocystis sp. is a highly prevalent anaerobic intestinal protozoan increasingly associated with gastrointestinal disorders. Its survival within the gut microenvironment depends on metabolic pathways such as glycolysis, where glyceraldehyde-3-phosphate dehydrogenase (GAPDH) plays a central role, alongside interactions with nitric oxide (NO) derived from host defenses and surrounding microbiota. The emergence of metronidazole resistance necessitates exploration of alternative therapeutic strategies.
OBJECTIVE: To evaluate the in vitro effects of metronidazole, metformin, and Alli

Between July and October 2024, 216 cases with genetically related Salmonella Blockley sequence type 52 were identified in France (n = 56), Germany (n = 30), the United Kingdom (UK) (n = 129) and the United States (US) (n = 1). Case interviews in the UK identified tomatoes as a possible vehicle of infection. A rapid case-control study with 29 cases and 87 controls was conducted to investigate this hypothesis, using pre-existing control data. Multivariable, Firth penalised logistic regression was used to explore associations between illness, food exposures and retailers. Consumption of tomatoes

UNLABELLED: Vibrio cholerae , the facultative pathogen underlying cholera, naturally inhabits warm aquatic estuaries. Environmental persistence is enhanced by the ability of V. cholerae to colonize host reservoirs and form multicellular biofilms, causing seasonally endemic outbreaks in many tropical regions. Most toxigenic strains utilize the type IVa mannose-sensitive hemagglutinin (MSHA) pilus for host reservoir colonization and biofilm formation. Temperature and salinity can alter V. cholerae biofilm formation, yet their impact on MSHA production specifically remains largely unknown. Here,

Non-typhoidal Salmonella (NTS) is a leading cause of bloodstream infections associated with substantial mortality mostly in sub-Saharan Africa (sSA) in children under 5 years. Salmonella enterica serovars Typhimurium and Enteritidis account for more than 90% of invasive NTS (iNTS) infections. Typhoid fever is caused by S. enterica serovar Typhi and is relatively common in South and Southeast Asia and sSA, especially among children. While there are licensed vaccines to prevent S . Typhi infections, the only current intervention for iNTS disease is treatment with antibiotics. This work presents

Botulinum neurotoxin (BoNT) injection into the trapezius muscle is increasingly performed for aesthetic contouring. However, iatrogenic pneumothorax has not been previously documented for this cosmetic indication, despite the inherent anatomical risks. We report a 26-year-old woman who developed immediate severe radiating pain during bilateral trapezius BoNT type A (BoNT-A) injections using a 4.8-cm, 25-gauge needle. During the injection, the patient experienced immediate severe radiating pain, followed by pleuritic chest pain and dyspnea within 30 minutes. Computed tomography confirmed a 30%

Salmonella remains one of the most important foodborne pathogens associated with poultry products, despite substantial progress in processing hygiene and antimicrobial interventions. This review critically examines the sources, transmission routes, and control strategies for Salmonella in vertically integrated broiler production systems. Attention is given to contamination originating from breeder flocks, hatcheries, feed, water, litter, grow-out farms, transport, processing environments, and finished products. Preharvest interventions, including biosecurity, breeder and hatchery control, vacc

Adherent-invasive Escherichia coli (AIEC) is implicated in Crohn's disease (CD) pathogenesis, but whether fecal microbiota transplantation (FMT) clears mucosal AIEC and alters fecal and mucosal microbiomes has not been studied. In this open-label pilot study, thirty adults with CD and seropositivity for serum anti-E.coli antibody (AEcAb) received a single colonoscopic FMT from one of two donors (ClinicalTrials.gov identifier NCT05611866). The primary outcome was adverse events (AEs) within 12 weeks after FMT; secondary outcomes included changes in mucosal AIEC, clinical measures, and fecal and

BACKGROUND: Typhoid fever is a severe infectious disease prevalent in developing countries. This enteric fever can cause ulceration with perforation, typically in the terminal ileum. There is insufficient evidence regarding the prevalence and morbidity of perforated typhoid in endemic areas such as Togo, West Africa. Our aim was to evaluate and improve mortality rates in patients with typhoid perforation at Hospital of Hope (HOH).
MATERIALS AND METHODS: A retrospective review of 180 patients with perforated typhoid at HOH in Togo, West Africa, examined surgical outcomes from January 1, 2020,

Cholera remains a persistent public health threat, complicating efforts to track transmission across borders and undermining control measures. Surveillance systems often remain fragmented and reactive, and many aspects of Vibrio cholerae evolution continue to be challenging, restricting the development of effective, data-driven interventions. Although genomics is increasingly transforming our understanding of cholera by revealing new insights into pathogen biology, transmission dynamics, and evolution, its routine integration into public health practice remains low. In this Personal View, we p
