Quantifying the economic burden of zoonotic Salmonella in poultry using a stochastic modelling approach for the Netherlands, Belgium, and Denmark.

Zoonotic Salmonella remains a major target of control programmes in European poultry production, yet its economic burden at the animal production level is not well quantified across countries. This study aimed to develop an animal cost-of-illness (a-COI) framework and apply it to estimate the economic burden of zoonotic Salmonella infections in poultry populations under surveillance in the Netherlands, Belgium, and Denmark. The framework combined publicly available epidemiological data, economic inputs, and production-impact parameters to estimate direct tangible costs associated with surveill

Consumption of Unpasteurized (Raw) Milk and Milk Products by Pregnant People and Children: Policy Statement.

Sales of unpasteurized (raw) milk and milk products remain legal in at least 27 US states. Unpasteurized milk and milk products from cows, goats, and sheep continue to be a source of infections attributable to several virulent pathogens, including Listeria monocytogenes, Campylobacter jejuni, Salmonella species, Brucella species, and Escherichia coli O157. These infections can occur in both healthy and immunocompromised individuals, including older adults, infants, young children, and pregnant people and their fetuses, potentially resulting in life-threatening illness and fetal miscarriage. Prevalence, Species Distribution, and Parasitic Burden of Cryptosporidium Infection in Cancer Patients: A Case-Control Study in Egypt.

Cryptosporidiosis causes a significant impact on immunocompromised patients, but its epidemiology among Egyptian cancer patients is poorly studied. This study was conducted to evaluate the prevalence, species distribution, and Cryptosporidium DNA copy number among Egyptian cancer patients versus immunocompetent controls. This case-control study included 600 cancer patients and 400 immunocompetent controls. In addition to modified acid-fast staining, molecular analysis was performed using eight new, in silico and in vitro validated species-specific primer sets targeting Cryptosporidium parvum,

A Single Amino Acid in the A2 Linker of Cholera Toxin Affects the Efficiency of Toxin Disassembly and Resulting Toxicity.

Cholera toxin (CT) from Vibrio cholerae and heat-labile enterotoxin (LT) from enterotoxigenic Escherichia coli are highly related AB5-type protein toxins that contain a catalytic A1 subunit, an A2 linker, and a cell-binding homopentamer of B subunits. The two proteins bind the same GM1 receptor and have similar levels of enzymatic activity toward their shared Gsα target, yet CT is more potent than LT. This difference has been mapped to a sequence of 11 amino acids (residues 226-236) near the C-terminus of the A2 linker. We now identify a single amino acid, aspartic acid at position 229, as a k

Genomic Epidemiology of Salmonella enterica Serovar Hadar Strain Linked to Poultry-Associated Salmonellosis Outbreaks, United States.

Nontyphoidal Salmonella enterica serovar Hadar causes poultry-associated salmonellosis outbreaks in the United States. One persisting strain of Salmonella Hadar caused 5 multistate outbreaks resulting in ≈2,000 human cases during 2020-2023. The Centers for Disease Control and Prevention designated the strain as reoccurring, emerging, or persisting (REP), related within 26 core-genome allele differences. That REP strain has caused human illnesses by consumption of commercial poultry food products or contact with backyard poultry. To investigate the REP strain's evolution and identify possible m

Antibiotic Resistance Trends and Prescribing Patterns in Pediatric Non-Typhoidal Salmonella Enteritis: A 10-Year Single-Centre Retrospective Study.

BACKGROUND: Non-typhoidal Salmonella (NTS) enteritis is a major cause of infectious diarrhoea in children and increasing antibiotic resistance is limiting treatment effectiveness. There is limited longitudinal data on resistance development and on how well prescribing practices align with resistance patterns in hospitalised children. This study aimed to assess trends in antibiotic resistance, the prevalence of multidrug resistance (MDR), and treatment concordance over a decade from a single Chinese Centre. METHODS: The study included children with microbiologically confirmed NTS enteritis wit

Rare Fatal Event of Multidrug-resistant Shigella Septicemia and Cryptococcal Meningitis in an Immunocompromised Host.

Bacteriemia due to Shigella spp. is a rare event. However, an individual with impaired immunity remains at risk of such invasion where the infective dose is as low as 10 viable bacilli. This case report describes one such fatal event where a 40-year-old immunocompromised male presented to a tertiary-level healthcare facility with a history of diarrhea for 10 days and seizures. Prompt microbiological workup established multidrug-resistant Shigella flexneri septicemia and cryptococcal meningitis.

Salmonella infection and biofilms drive anti-C1q and anti-dsDNA autoantibodies in systemic autoimmunity.

Systemic lupus erythematosus (SLE) is characterized by autoantibody production alongside inflammatory manifestations, including nephritis. In SLE, infections are frequent and associated with disease flares, increasing morbidity and mortality. Additionally, dysbiosis, often marked by overgrowth of Pseudomonadota, is a prominent feature of SLE and lupus nephritis. We previously demonstrated that complexes of DNA and curli, an amyloidogenic protein from biofilms of Pseudomonadota, including Salmonella Typhimurium and E. coli, can induce anti-dsDNA and anti-chromatin autoantibody responses. Here,

Epigastric typhoid ileal perforation unmasking an incidental adult midgut malrotation: a rare clinical co-existence.

Peritonitis due to typhoid ileal perforation is a life-threatening surgical emergency that remains common in endemic regions but is rarely encountered in developed countries. Anatomically, its presentation is mostly localized to pain in right iliac fossa. A 33-year-old man presented with six days of fever, severe epigastric pain, vomiting, and septic shock. Because of the marked epigastric tenderness and pneumoperitoneum on X-rays, perforated peptic ulcer was the leading preoperative diagnosis. Emergency laparotomy instead revealed severe peritoneal contamination, incidental incomplete midgut

Expanded prevalence of group 4 capsule expression and its genetic requirements in a broad array of Shigella strains.

UNLABELLED: The expression of a group 4 capsule (G4C) has been identified in Shigella sonnei and Shigella flexneri serotype 6 and demonstrated to play a significant role in pathogenic processes in S. sonnei. However, it has not been examined phenotypically in S. flexneri 6. Despite the impact of G4C, no systematic analysis of its expression or the presence of the genomic determinants has been performed across a large collection of S. sonnei and S. flexneri serotypes 2a and 6 or any of the other >46 Shigella strains and serotypes that cause disease. Development of novel human monoclonal antibodies for botulinum neurotoxin type F subtype 1.

Different types of Botulinum neurotoxins (BoNTs) cause human botulism. Among them, type F botulism, caused by genetically diverse BoNT/F subtypes, is rare and may be overlooked despite its clinical importance. Toxin-neutralizing human antibodies, which carry the minimum risks of inducing serum sickness or anaphylaxis, are ideal for the treatment of human botulism. In this study, we generated human monoclonal antibodies (mAb) against BoNT/F using trans-chromosomic monoclonal antibody (TC-mAb) mice. A recombinant toxin fragment consisting of the light chain and the heavy chain N-terminal domain

Bacteriophage Sf14 survival under simulated gastrointestinal conditions.

Shigella flexneri is an enteric pathogen responsible for causing shigellosis, a type of bacillary dysentery of significant global health concern. The increasing prevalence of antibiotic resistance has driven the search for alternative treatments, including bacteriophage (phage) therapy. However, the effectiveness of orally administered phages for enteric pathogens depends on their ability to survive passage through the gastrointestinal tract. In this study, the ability of bacteriophage Sf14 to maintain infectivity was evaluated under simulated gastrointestinal conditions using a modified INFOG

Effect of oral gavage or intracloacal administration of Escherichia coli or Salmonella at hatch on enteric colonization and early performance in broiler chicks.

Minimal research has evaluated the efficacy of challenging chicks with Enterobacteriaceae via intracloacal gavage. The objectives of these studies were to: (1) evaluate day-of-hatch (DOH) colonization by Escherichia coli or Salmonella Enteritidis (SE), (2) determine the effects of bacterial challenge route on broiler performance, and (3) assess oral and intracloacal inoculation methods in broiler challenge models. In Exp 1, chicks were randomized, weighed, and challenged at DOH via oral or intracloacal gavage with saline (negative control), E. coli, or SE at doses of 103, 104, or 105 CFU/0.25

Antimicrobial resistance and genomic characterization of Salmonella Minnesota, ESBL-producing Escherichia coli, and ESKAPE pathogens isolated from Brazilian broiler chicken carcasses.

INTRODUCTION: Brazil is the world's largest exporter of chicken meat, and poultry products are recognized as important reservoirs of foodborne pathogens. Salmonella enterica serovar Minnesota has emerged as the predominant serovar in the poultry industry and is frequently associated with multidrug-resistant strains carrying multiple antimicrobial resistance genes. In addition, Escherichia coli and ESKAPE pathogens have been increasingly isolated from poultry products worldwide, representing a growing public health concern due to their antimicrobial resistance and dissemination potential. Toward an Aptamer-Based Biosensor for Cross-Serotype Detection of Botulinum Neurotoxin: Binding Mechanism from a Comprehensive Computational Investigation.

Botulism is a potentially fatal illness in humans and many animals caused by the botulinum neurotoxins (BoNTs) produced mainly by Clostridium botulinum, which can be found in a wide variety of foods. The treatment of this illness requires antitoxin drugs given to the patients within a 48 h window to be effective. Therefore, there is a strong need for the development of rapid and sensitive biosensors for the early detection of the toxins in foods. In this work, by using comprehensive atomistic computer modeling and simulation, we investigate aptamers that bind BoNT serotypes A, B, and C to serv
