Annexin A5-engineered nanovaccines synchronize spatiotemporal antigen release with phagocytosis checkpoint blockade for potent in situ cancer immunotherapy.

In situ generation of autologous tumor vaccines from dying tumor cells offers a promising route for personalized cancer immunotherapy, yet is often limited by phosphatidylserine (PS)-mediated immunosuppressive clearance that can restrict downstream immune activation and favor M2-like macrophage polarization. Here, we report a sequentially orchestrated nanovaccine (NT@ANX5-P) that couples photodynamic therapy (PDT)-induced immunogenic tumor-cell damage with on-demand PS masking to enhance in situ tumor vaccination. The nanovaccine consists of a photosensitizer core cloaked with genetically engi

TNFα promotes brain-predominant EAE independent of IFNγ and IL-17A in HLA-DR3 transgenic mice.

Multiple sclerosis (MS) is thought to be mediated by human leukocyte antigen (HLA) class II-restricted autoreactive CD4 T cells producing interleukin (IL)-17A and/or interferon γ (IFNγ), but the necessity of these cytokines for disease pathogenesis is unclear. Utilizing HLA-DR3 (DRβ1*0301) transgenic mice lacking IFNγ or IL-17A in experimental autoimmune encephalomyelitis (EAE), a model of MS, we found that neither cytokine is essential for disease induction, although both have distinct roles. The absence of IFNγ caused atypical, brain-predominant EAE characterized by high levels of IL-17A, wh

Adipokine regulation of immune cell polarization in oral cancer and its impact on chemotherapy sensitivity.

Oral squamous cell carcinoma (OSCC) is the most common malignant tumor in the head and neck region. In locally advanced patients, neoadjuvant chemotherapy centered on cisplatin and 5-fluorouracil often leads to primary/acquired resistance, with a 5-year survival rate of less than 50%. Tumor microenvironment (TME) immunosuppressive remodeling is the core cause of chemotherapy failure. Adipokines secreted by cancer-associated adipocytes (CAAs) are the key mediators that link metabolic disorders, immune polarization and chemotherapy resistance. The adipokines leptin, adiponectin, chemerin, resist

Peripheral effector-memory CD4+ T cells reflect clinical symptom burden in pediatric allergic rhinitis.

BACKGROUND: Pediatric allergic rhinitis (AR) can substantially affect quality of life and school performance, yet conventional sensitization markers such as allergen-specific immunoglobulin E (sIgE) do not consistently reflect symptom severity. We examined whether activation-induced marker (AIM)-defined allergen-specific peripheral effector-memory CD4+ T-cell (peff-mCD4) frequencies were associated with symptom burden. METHODS: In this pilot, single-center observational study, house dust mite (HDM)-specific responses were analyzed in 20 children and Japanese cedar pollen (JCP)-specific respon

Immune Profiles Elicited by Different HBV Vaccine Modalities and Their Associations with Serological and Intrahepatic Viral Outcomes.

Therapeutic vaccination for chronic hepatitis B aims to restore HBV-specific T-cell immunity and contribute to a functional cure. We systematically compared the immune profiles and antiviral efficacy of four vaccine modalities: recombinant HBsAg (rHBsAg), antigen-antibody immune complex (IC), a DNA vaccine encoding HBsAg (DNA-HBsAg), and an HBsAg-Fc fusion DNA vaccine (DNA-HBsAg-Fc). In naïve mice, the rHBsAg vaccine predominantly induced anti-HBs IgG responses, whereas the IC vaccine elicited strong anti-HBs IgG and detectable CD4+ T-cell responses. In contrast, DNA vaccines preferentially in

The cerebrospinal fluid immune microenvironment in lung cancer leptomeningeal metastasis: cellular ecosystems and therapeutic possibilities.

Leptomeningeal metastasis (LM) is a devastating form of central nervous system (CNS) dissemination in lung cancer, particularly non-small cell lung cancer (NSCLC). Current treatment frameworks emphasize CNS drug penetration, oncogenic driver evolution, and intrathecal therapy. Emerging evidence supports viewing the cerebrospinal fluid (CSF) compartment as an active tumor-immune space, but the field remains early, with limited lung cancer-specific mechanistic and single-cell datasets. Lung cancer cells in CSF appear to adapt to low protein content, limited nutrients, fluid shear, weak matrix su

Antiviral and inflammatory immune signatures in anti-CD20-treated multiple sclerosis: a cross-sectional study.

CD20-targeting monoclonal antibodies are highly effective therapies for relapsing multiple sclerosis (MS). We examined how anti-CD20 therapy reshapes antiviral T-cell immunity, innate inflammatory tone, and blood markers of neuroaxonal injury. In this cross-sectional study, adults with relapsing MS were enrolled at two neurological centers in Rome. An immunophenotyping cohort (45 untreated, 25 anti-CD20-treated, all receiving ocrelizumab) was profiled by high-parameter flow cytometry across more than 40 immune populations. A separate antiviral cohort (24 untreated, 13 treated, 19 healthy donor

An Ex Vivo 3D Co-Culture Model of the Bone Marrow Niche Better Predicts Clinically Relevant Epigenetic Target Modulation to Improve Immunotherapy in Multiple Myeloma.

Background: In the last decade, treatment options for multiple myeloma have increased remarkably, including antibodies or CAR-T cells targeting myeloma surface markers with unprecedented efficacy. To enhance immunomodulation and clinical efficacy even more, the combination of these agents with other drugs is highly relevant. A significant association between CD38 expression on myeloma cells and the cytotoxicity of the anti-CD38 antibody daratumumab has been shown. All-trans retinoic acid, the pan-histone deacetylase inhibitor panobinostat, and the enhancer of the zeste homolog 2 inhibitor taze

Targeting SERPINB5 and MSLN for Pancreatic Cancer Therapy.

Background/Objectives: Pancreatic cancer (PC) is a leading cause of cancer-related death in the United States and lacks efficient therapeutic options. Serine peptidase inhibitor, clade B, member 5 (SERPINB5) is non-inhibitory toward serine proteases, yet it has diverse biological functions, including regulating cell migration, cell death, gene expression, and stress responses. Mesothelin (MSLN) is a tumor-associated antigen that is broadly overexpressed on various malignant tumor cells and plays a crucial role in promoting proliferation, invasion, and metastasis. Methods: We integrated public

Research Progress on Targeted Therapies for Pemphigus Vulgaris and Bullous Pemphigoid: Mechanisms, Opportunities and Challenges.

Autoimmune bullous diseases (AIBDs) are a group of severe disorders characterized by autoantibodies that attack structural proteins within the skin/mucosa, leading to blister and bullae formation. Pemphigus vulgaris (PV) and bullous pemphigoid (BP) are among the most prevalent representative AIBDs. Traditional treatments involving corticosteroids and immunosuppressive agents are often associated with substantial adverse effects and a high rate of relapse. In recent years, the therapeutic landscape has evolved toward precision targeted therapies, encompassing biological agents, small-molecule d

Towards the development of an efficient nonlinear controller for ART and cART against HIV infection.

The Human Immuno-deficiency Virus (HIV) is responsible for attacking the CD4+ T cells present in human blood and causes the human immune system to become vulnerable for numerous contagious and opportunistic diseases. It is hence pertinent to curb the spread of HIV infection using anti-HIV drugs. This study integrates sigmoid function with conventional sliding mode control and incorporate it within the framework of multi-objective optimal model predictive controller to reduce the impact of HIV infection through anti-retro-viral therapy. The proposed algorithm is designed using a deterministic H

Benzo[a]pyrene and bisphenol A interact to induce combined pulmonary toxicity through an integrated transcriptomic-metabolomic network.

Widespread atmospheric pollutants such as benzo[a]pyrene (BaP) and bisphenol A (BPA) persistently coexist, yet their combined risks to the respiratory system remain largely overlooked. In this study, C57BL/6J mouse and A549 cell models were employed to investigate the combined pulmonary toxicity induced by BaP and BPA, with an integrated transcriptomic-metabolomic framework established to characterize their response patterns. Combined exposure to BaP and BPA caused pathological damage in mouse lung tissue and specifically induced a broader spectrum of adverse effects. In vitro experiments demo

Reprogramming immunosuppressive TMEs with Salmonella-delivered sialidase and COX-2 blockade synergistically enhances cancer immunotherapy.

Sialic acid is a component of the extracellular matrix that is upregulated in multiple tumors and plays critical roles in tumorigenesis and progression. Sialic acid engages sialic acid-binding immunoglobulin-like lectins (Siglecs) on immune cells to suppress immune activation. Despite the potential of sialylation-targeted therapies to reactivate antitumor immunity, their clinical translation remains limited by suboptimal drug delivery systems and insufficient tumor specificity. To overcome these obstacles, we engineered a sialidase-expressing S. typhimurium and termed it SLpnanH. The engineere

Bench-to-Bedside Perspectives on the Development of Soluble TCR-Based Protein Biologics: Current Landscape, Control Strategy and Future Direction.

Soluble T-cell receptor (TCR)-based biologics (protein-based) are an emerging class of immunotherapeutic agents that recognize antigenic peptides presented by the human leukocyte antigen (HLA) on the surface of target cells. Furthermore, they simultaneously bind to cluster of differentiation 3 (CD3) subunits on T cells, recruit T cells to target cells, and promote T-cell activation and degranulation, ultimately eliminating tumor cells. Soluble TCR-based protein biologics target intracellular antigens that are inaccessible to conventional antibody-based drugs. Consequently, the development of s

CTH Promotes Liver Cancer Progression and Exploratory Evaluation Supports Enhanced Antitumor Activity of PPG Plus GPC3 CAR-T Cells.

Although hydrogen sulfide (H2S) is a key regulator in cancer biology, the mechanism by which the H2S-producing enzyme cystathionine-γ-lyase (CTH) functions in hepatocellular carcinoma (HCC) remains elusive. Using the slow-releasing H2S donor GYY4137, we observed enhanced migration and invasion in HuH7 and HepG2 cells. Subsequently, we observed consistent protumorigenic effects of CTH not only in vitro but also in subcutaneous and orthotopic mouse models. Here, our findings suggest that CTH-H2S sulfhydrates Rab7a, which may be associated with HCC progression. We further showed that CTH modulate

Immune Landscape and Intercellular Crosstalk in the Cornea of an Autoimmune-Mediated Dry Eye Disease.

PURPOSE: The purpose of this study was to comprehensively characterize the identity and signaling profiles of resident and infiltrating immune cells, and their interactions with non-immune cells, in a spontaneous mouse model of autoimmune-mediated dry eye disease (DED). METHODS: Corneal tissues from wild-type (WT) and autoimmune regulator-deficient (Aire-/-) mice were analyzed by whole-mount immunofluorescence to define immune cell composition and spatial distribution within the limbus and central cornea. Immune cell identities and cell-cell communication were further characterized using sing

Latent effector capacity governs reversible T cell exhaustion: A theoretical mathematical model of PD-1 blockade response.

T cell exhaustion is often viewed as a terminal state marked by irreversible loss of function during chronic infection and cancer. However, the rapid recovery of antitumor activity in clinical responders after PD-1 blockade suggests that some exhausted T cells retain hidden functional potential. To explain this paradox, we present a theoretical framework that separates latent effector capacity from active effector function. Latent effector capacity represents a history-dependent state reflecting preserved epigenetic accessibility and regulatory readiness, distinct from ongoing gene expression.

Effects of pyridaben on the spleen and T lymphocytes and intervention strategy in grass carp (Ctenopharyngodon idella).

The widespread use of pyridaben (PDB) poses serious threat to aquatic organism health. 6-Gingerol, a bioactive component of ginger, has been demonstrated to possess both antioxidant and immunomodulatory properties. However, the effects of spleen injury of PDB in aquatic species and the potential mitigating effects of 6-Gingerol remain insufficiently characterized. The present study aimed to characterize the toxic effects of PDB on the spleen of grass carp and to evaluate whether 6-Gingerol could effectively counteract PDB-induced toxicity. The results showed that PDB exposure could induce sple

Akkermansia muciniphila attenuates macrophage efferocytosis to enhance anti-PD-1 efficacy in microsatellite-stable colorectal cancer.

Approximately 85% of colorectal cancers (CRCs) are microsatellite stable (MSS) and thus exhibit intrinsic resistance to anti-programmed death-1 (PD-1) therapy. In this study, we demonstrate that oral administration of Akkermansia muciniphila (Akk) suppresses tumorigenesis and reprograms the tumor microenvironment in MSS CRC mouse models. Akk attenuates macrophage efferocytosis through the TLR2/NF-κB pathway, an effect that leads to the accumulation of apoptotic cancer cells, which activates the cGAS-STING pathway and potentiates type I interferon production. This immunostimulatory cascade acti
