Cold tumors evade APC and T-cell surveillance through low immunogenicity, defective antigen presentation, and immunosuppressive tumor microenvironments. We uncovered a non-canonical function of bacterial outer membrane vesicles (OMVs), whereby OMVs activated the TLR4-NF-κB-NLRC5 axis to induce tumor MHC I upregulation, thereby restoring tumor visibility to T cells. To further eliminate tumor invisibility to APCs, we reported a bacteria-inspired strategy that drives APCs to perceive tumor cells as bacteria. By fusing OMVs with fusogenic liposomes, we constructed fusogenic OMVs (FOs) that anchor

Persistent infection by helminths requires limiting inflammatory damage despite incomplete pathogen clearance, yet how helminths mobilize resident microbiota to establish this state remains unclear. Here we identify a cross-kingdom metabolic relay through which Trichinella spiralis promotes systemic disease tolerance. Infection selectively expanded Ligilactobacillus murinus and redirected microbial tryptophan metabolism toward indole-3-lactic acid (ILA), a direct ligand of the aryl hydrocarbon receptor (AhR). Antibiotic perturbation abrogated ILA production, AhR activation and Treg-Th17 remode

The major histocompatibility complex class I (MHC-I) immunopeptidome is dynamic and determined by numerous biochemical and cellular features. Computational tools to predict peptide-MHC-I binding focus on the affinity of a specific peptide-MHC interaction; separate tools predict complex stability or presentation. Methods used to define the immunopeptidome may preferentially recover stable complexes, and presentation predictors trained on those data may inherit that bias. However, peptides that dissociate rapidly but are generated at high rates could reach surface densities sufficient for CD8+ T

Thymic epithelial cells (TECs) provide essential signals for T-cell development, yet the mechanisms governing human TEC differentiation and the acquisition of T-cell-supporting function remain incompletely understood. Using a three-stage system that directs human embryonic stem cells (hESCs) through definitive endoderm and third pharyngeal pouch endoderm (3PPE) toward a thymic epithelial-like lineage, we investigated the role of HOXA3 during thymic epithelial-like cell (TEC-like cell) differentiation. HOXA3 knockdown during 3PPE-to-TEC-like cell transition impaired thymic epithelial differenti

Bordetella pertussis elicits both innate and adaptive cellular responses, yet their relative contributions to peripheral interferon-γ (IFN-γ) measurements and the temporal dynamics of circulating T-cell reactivity in adults remain incompletely defined. B. pertussis-reactive cellular immunity was investigated in 42 adult donors, including 26 healthy donors and 16 donors with a history of pertussis, using standard and modified IFN-γ enzyme-linked immunospot (ELISpot) assays with a chemically inactivated whole-cell B. pertussis preparation, bacterial innate-biased stimuli, and a peptide-based T-c

BACKGROUND: Acute myeloid leukemia (AML) remains the deadliest form of leukemia in children and adults despite therapeutic advances in recent decades. Chimeric antigen receptor (CAR) T-cell therapy is one such therapeutic approach that has been explored preclinically and clinically in AML; however, its efficacy is limited by antigen heterogeneity. To overcome this roadblock, we propose a dual-targeting CAR T-cell strategy, targeting the AML-associated antigens CD33 and CD70.
METHODS: To recapitulate tumor heterogeneity, we genetically edited the AML cell line Molm-13 using CRISPR-Cas9 to gene

Here we report that Dectin-1-stimulated dendritic cells (DCs) induce unique IL-10-producing T cells with strong regulatory function during fungal infection or experimental asthma. These T cells, which we call Dect-10 T cells, are induced by DC-initiated signaling that sets in motion two complementary mechanisms of IL-10-producing T cell differentiation. The first involves induction of nascent Dect-10 T cell IL-4 synthesis and attendant GATA3 production, the latter being a factor that exerts canonical and epigenetic transcriptional function at separate IL-10 promoter sites. The second involves

Acute myeloid leukemia (AML) progression involves remodeling of the bone marrow inflammatory microenvironment, yet prior studies have focused on individual cytokines rather than coordinated network-level communication. This study aimed to systematically map inflammatory network reprogramming in AML and develop a network-derived prognostic tool. Single-cell RNA sequencing was performed on bone marrow samples from three newly diagnosed AML patients and six healthy donors (47,525 cells); ligand-receptor communication was then analyzed using CellChat. Communication dominance shifted from T-cells t

CONTEXT: Atopic dermatitis is a chronic inflammatory skin disorder characterized by epidermal barrier dysfunction and dysregulated type 2 helper T cell (Th2)-mediated immune responses. Erigeron annuus (L.) Desf. is traditionally used for inflammatory conditions, but its effects on atopic dermatitis remain unclear.
OBJECTIVE: This study evaluated the protective effects of E. annuus aqueous extract (EA) and associated mechanisms.
MATERIALS AND METHODS: Atopic dermatitis-like lesions were induced in 5-week-old female BALB/c mice by repeated epicutaneous ovalbumin (OVA) sensitization, and EA was

Ageing profoundly impairs vaccine efficacy, leaving older individuals highly susceptible to infectious diseases despite vaccination. Although B cells retain the intrinsic capacity to generate germinal centre (GC) responses in older people, age-associated defects in T cells, dendritic cells and follicular dendritic cells (FDCs) limit the development of high-affinity, class-switched antibody responses. Here, we hypothesised that senescent fibroblasts disrupt B cell activation and expansion, thus contributing to the diminution of the humoral response in ageing. To test this hypothesis, we establi

PURPOSE OF REVIEW: Tissue HIV-1 (HIV) reservoirs, particularly within the central nervous system (CNS), remain understudied because of limited access to well preserved human tissues. This review aims to highlight insights from rapid research autopsy studies into HIV persistence in the CNS, with a particular focus on viable-cell approaches, BMCs/microglia as cellular reservoirs, and the mechanisms underlying HIV latency and neuroinflammation. We also discuss key knowledge gaps and how these approaches may inform strategies to target CNS HIV reservoirs.
RECENT FINDINGS: Recent rapid research au

Bispecific antibodies (BsAbs) redirect T-cells to tumor antigens in an MHC-independent manner. These universally available, off-the-shelf products have demonstrated unprecedented efficacy in heavily pretreated patients with relapsed/refractory (R/R) hematological malignancies (HMs). The indefinite treatment duration associated with most BsAb products, coupled with neutropenia, B-cell aplasia, and hypogammaglobulinemia result in significant immune-related toxicities and infections. Hence, non-relapse mortality (NRM) which represents the cumulative burden of treatment-related toxicities, defined

Most patients with solid tumors fail to sustain antitumor immunity, in part because immune cells cannot outcompete cancer cells inside the immunosuppressive tumor microenvironment. Here, we show the lipid-sensing transcription factor PPARδ is a compartment-specific determinant of cancer-immune fitness balance and immunotherapy efficacy. Activating PPARδ in cancer cells promotes their growth and impairs antitumor immunity, whereas activating it in immune cells drives tumor suppression and overrides tumor-promoting effects of cancer-intrinsic activation. Mechanistically, PPARδ enhances CD8+ T ce

T and B lymphocytes are key components of the adaptive immune system, playing a central role in maintaining host homeostasis and protecting against a wide range of pathogens. A comparable form of homeostatic regulation can be observed in microbial ecosystems, including the oral cavity. This environment is extremely unstable and can be easily disrupted by various factors or stimuli. This review article presents the different types of T and B lymphocytes and their role in the human body, and attempts to answer the question of how these two basic types of immune system cells can affect the imbala

Nature recently listed 20 major T cell milestones since the early breakthroughs of Drs. Jacques Miller and Max Cooper. However, to make the story more complete, several key discoveries may need to be mentioned. One of the major missing episodes is the discovery of the bone marrow in mice as the origin of T and B cells, revealed by Dr. Henry Claman and his colleagues. The complete absence of the critical role of B cells in the final differentiation of T cells into memory lineages could be a major oversight in modern immunology. In this short essay, I will try to summarize some of the published

Immune checkpoint inhibitors (ICIs) have reshaped cancer treatment; however, durable clinical benefit remains limited by primary and acquired resistance, frequently associated with T cell dysfunction within an immunosuppressive tumor microenvironment. Beyond their classical role in hemostasis, platelets have emerged as active immune modulators capable of shaping antitumor T cell responses. Accumulating evidence indicates that platelets engage T cells through contact-dependent receptor-ligand interactions, immunomodulatory soluble factors, and the transfer of checkpoint-associated extracellular

BACKGROUND: Gastrodin is a natural compound with anti-inflammatory, antioxidant, and antitumor activities; however, its therapeutic role and molecular mechanisms in colorectal cancer (CRC) remain unclear. In this study, we explored the anti-CRC mechanisms of gastrodin through integrated bioinformatics and experimental validation.
METHODS: The potential targets of gastrodin were obtained from public databases. Transcriptomic data of CRC were retrieved from TCGA and GEO datasets, and the differentially expressed genes were screened. The CRC-related genes were subjected to gene-set variation ana

Dendritic cells (DCs) are key initiators of antitumor immunity, yet the metabolic drivers governing their activation remain incompletely defined. Here, we show that the fungal immunomodulator β-glucan induces robust immune-metabolic reprogramming in bone marrow-derived DCs (BMDCs), characterized by coordinated increases in glycolysis and tricarboxylic acid cycle activity. This metabolic shift is essential for BMDCs activation, as inhibition of glycolysis or oxidative metabolism diminishes β-glucan-induced costimulatory molecule expression, proinflammatory cytokine production and T-cell priming

BACKGROUND: A subset of people living with HIV (PLHIV), known as immunological non-responders (INR), fails to achieve adequate CD4⁺ T cell recovery despite viral suppression with antiretroviral therapy (ART). These individuals face an increased risk of adverse clinical outcomes. This study aims to explore the intracellular mechanisms and molecular signatures underlying this incomplete immune recovery using a multi-omics approach.
METHODS: The study analysed CD4⁺ and CD8⁺ T cells from 100 ART-naïve PLHIV. Participants were classified into controls (baseline CD4⁺ > 200 cells/µL, n = 49) and cas

BACKGROUND AND AIM: The role of regulatory B cells (Bregs) cells and regulatory T cells (Tregs) has been little investigated in non-small cell lung cancer (NSCLC). Therefore, we investigated the frequency of circulating Bregs, Tregs cells, and the serum cytokine profile in NSCLC and their association with disease progression.
MATERIALS AND METHODS: In this prospective case-control study, peripheral blood from 40 patients with NSCLC and 40 healthy controls was examined, the frequency of various lymphocyte subsets including Bregs and Tregs was evaluated using flow cytometry. Serum cytokine prof
