Decoupling T cell cytotoxicity: a CCL21+ICAM1-based synthetic immune niche enhances tumor elimination by accelerating lytic hit delivery.

Preserving T cell cytotoxic function during ex vivo expansion remains a major challenge for adoptive cancer immunotherapy. A synthetic immune niche (SIN) composed of immobilized CCL21 and ICAM1 was shown to improve T cell expansion while preserving cytotoxicity; however, it remains unclear which specific step of the T cell killing process is enhanced by the SIN stimulation. Here, we combined advanced imaging based on time-lapse microscopy with a mean-field model to analyze the distinct stages in killing of B16 melanoma cells by CD8[Formula: see text] T cells. This framework enabled us to resol

Metabolic Reprogramming in Bladder Cancer: Molecular Drivers, Immune Regulation and Therapeutic Opportunities.

Metabolic reprogramming is increasingly recognised as a central determinant of bladder cancer heterogeneity, immune regulation and treatment response. This review integrates genetic, epigenetic, microenvironmental and therapeutic evidence to explain how bladder cancer redistributes carbon and nitrogen across glycolysis, lipid synthesis and oxidation, amino-acid metabolism and mitochondrial respiration. Alterations in FGFR3, PIK3CA, MYC, TP53/RB1 and KDM6A establish distinct, context-dependent metabolic dependencies, while hypoxia, extracellular-matrix mechanics and treatment selection reinforc

Safety and immunogenicity of a first-generation modified vaccinia virus Ankara-vectored SARS-CoV-2 vaccine candidate with mRNA-based booster vaccination: A phase 1 trial.

The rapid development of effective COVID-19 vaccines was enabled by the parallel assessment of multiple candidates early in the pandemic, including candidates based on the Modified Vaccinia virus Ankara (MVA) vector platform. This Phase 1 trial (NCT04569383) evaluated the MVA-based vaccine candidate MVA-SARS-2-S, expressing the full-length native SARS-CoV-2 spike glycoprotein (Wuhan-HU-1). Between October 2020 and May 2021, 30 healthy, SARS-CoV-2-naive adults received two doses of either 107 or 108 international units (IU) of MVA-SARS-2-S 28 days apart, with 6-month follow-up. A subgroup (n =

Epigenetic-Targeting Regimens vs. Conventional Chemotherapy in Relapsed/Refractory PTCL: A Meta-Analysis.

PURPOSE: Relapsed/refractory peripheral T-cell lymphoma (R/R PTCL) lacks a uniform salvage standard and outcomes with conventional chemotherapy (CC) remain suboptimal. This meta-analysis compared epigenetic-containing regimens (Epi-Rx) and CC in R/R PTCL. MATERIALS AND METHODS: We searched MEDLINE, Embase, and the Cochrane Library through May 2025 for prospective or retrospective studies evaluating Epi-Rx (≥1 HDAC, DNMT/hypomethylating, or EZH1/2 inhibitor, alone or in combination) or CC without epigenetic agents. Random-effects models pooled single-arm proportions, with prespecified subgroup

Apheresis in Children of Weight Under 7 kg-A Single Center Experience.

Although apheresis techniques in children are well established, published experience with very low-weight children is limited. We reviewed 68 procedures performed in 19 children weighing less than 7 kg, including hematopoietic progenitor and T-cell collections and plasmapheresis. With significant modifications of standard techniques, all but one procedure was successfully and safely completed without serious citrate toxicity. Access was a major challenge. For plasmapheresis, the mean plasma volume removed was 1.2 × total plasma volume, but the mean total blood processed was 1.74 × total blood

Real-world management of ovarian cancer in women living with HIV: treatment exposure, immune monitoring, and care barriers in a five-case series.

BACKGROUND: With improved survival among people living with HIV (PLWH), non-AIDS-defining cancers have become increasingly prominent. Women living with HIV (WLWH) remain underrepresented in ovarian cancer trials, leaving limited evidence on the tolerability of standard therapies, including chemotherapy, anti-VEGF agents, and PARP inhibitors. These patients also face unique care barriers beyond biological uncertainties. METHODS: This retrospective review included five WLWH diagnosed with ovarian cancer (June 2022-March 2024). Three received anticancer treatment; one refused chemotherapy and wa

Integrated single-cell multi-omics analysis delineates neonatal immune dysregulation associated with gestational diabetes mellitus.

INTRODUCTION: Gestational diabetes mellitus (GDM) is a common pregnancy complication which could induce immune dysfunction. Although intrauterine hyperglycemia impairs fetal immune development, its cell-type specific effects on neonatal immune cells, and cell‑cell communication remain unclear. METHODS: In this pilot study, we performed integrated 5' single‑cell RNA sequencing and TCR repertoire analysis of cord blood mononuclear cells from two GDM and one type 2 diabetes mellitus (T2DM) mother-neonate pairs, with public healthy cord blood data (GSE157007) as controls. After quality control, 5

Discovery of Avian TCRVβ2+ T Cells Recognizing the Endogenous Ganglioside via a chCD1.1-Dependent Pathway.

This study presents the discovery and first functional characterization of chicken lipid-reactive Natural Killer T (NKT)-like cells, resolving a long-standing knowledge gap in avian immunology. Using purified TCRVβ2+ lymphocytes, we demonstrate through rigorous experiments combining shRNA-mediated gene silencing, pharmacological pathway blockade, primary MACS-purified co-cultures, and intracellular cytokine flow cytometry that these cells are activated by CpG-ODN-stimulated antigen-presenting cells (APCs). Activation selectively depends on APC-driven glycosphingolipid (GSL) biosynthesis, with

Case report: Maternal engraftment of T-cells in a patient with Severe Combined Immunodeficiency due to DCLRE1C pathogenic variant - diagnostic and treatment challenges.

BACKGROUND: Severe combined immunodeficiency (SCID) is a rare but serious congenital condition caused by genetic defects that severely impair the development and function of T cells, B cells, and natural killer (NK) cells. Without early diagnosis and immune system reconstitution, SCID is usually fatal in infancy. The condition requires prompt recognition and hematopoietic stem cell transplantation (HSCT) to restore immune function. This report describes the first genetically confirmed case of SCID in Latvia - caused by a DCLRE1C gene pathogenic variant and diagnosed prior to the implementation

Cullin-5 RING E3 ligase limits CD8⁺ T-cell persistence and effector responses during chronic antigen stimulation.

T-cell exhaustion limits durable antitumor responses and remains a barrier to adoptive cell therapies (ACT) in solid tumors. To identify regulators of T-cell persistence, we performed genome-wide CRISPR knockout screens in primary human CD8⁺ T cells expressing an NY-ESO-1-specific T-cell receptor and subjected them to repeated tumor cell rechallenge. These screens revealed enrichment of single-guide RNAs targeting components of Cullin-RING E3 ligases, particularly Cullin-5 (CRL5) assemblies comprising CUL5, RNF7, and ARIH2. Disruption of CRL5 genes in T cells preserved cytotoxic activity and e

Targeting protein arginine methyltransferases to enhance cancer immunotherapy.

Arginine methylation is a post-translational modification found on many proteins, including RNA-binding proteins, signaling molecules, histones, and heat shock proteins, where it can alter protein-protein and protein-nucleic acid interactions. Arginine methylation regulates many cellular pathways, including RNA metabolism, signal transduction, gene expression, cellular stress responses, DNA repair, and, more recently, innate immune sensing. Here, we discuss recent advances in innate immune sensing and the emerging role of arginine methylation in suppressing tumor immune visibility. We describe

Dendritic-cell-targeting adeno-associated virus as potent cancer vaccine carriers.

BACKGROUND: The clinical efficacy of therapeutic cancer vaccines is critically limited by the inefficient in vivo delivery of tumor antigens to dendritic cells (DCs), the key regulators of T-cell immunity. METHODS: To address this issue, we developed a dendritic-cell-targeting adeno-associated virus (AAV) vector through rational capsid design. This was achieved by site-specifically inserting the high-affinity Clec9a-targeting peptide CBP-12, optimizing its flanking sequences, and introducing T491V/S662V gain-of-function mutations to synergistically enhance DC transduction. RESULTS: The resul

IL-23 and monocyte interactions redirect human Th17.1 cells to an IL-17AhighIFN-γhigh potentially pathogenic profile.

INTRODUCTION: IL-23-targeted therapies have demonstrated clinical efficacy in psoriatic arthritis (PsA), primarily through the reduction of IL-17A-driven inflammation. Consequently, most studies have focused on classical, high IL-17A-producing, CCR6+CXCR3-CCR4+ Th17 cells. In contrast, the impact of IL‑23 signaling on CCR6+CXCR3+CCR4- Th17.1 cells -traditionally characterized by high IFN-γ and low IL-17A production-remains largely unexplored, despite their recognized pathogenicity in autoimmune disease. The aim of this study was to characterize the effects of IL-23 on human memory Th17.1 cells

Comprehensive profiling of phenotypic features of memory T- and B-cells from birth to the onset of old age.

INTRODUCTION: Human aging is accompanied by profound and continuous remodeling of the immune system, characterized by rhythmic, stage-specific patterns. These features are characterized by waves of cellular differentiation, network reorganization, and functional recalibration, a phenomenon encompassed by the concept of immunobiography. METHODS: This study focuses on T- and B-cell memory immunophenotypes that shape adaptive immunity, including interferon-activated cells from healthy volunteers of both sexes, aged 9 months to 65 years, stratified into 12 consecutive age subgroups. Innate immunity-stroma crosstalk in pancreatic cancer: overcoming spatial and metabolic barriers to immunotherapy.

Pancreatic ductal adenocarcinoma (PDAC) exhibits profound resistance to current immunotherapies, a challenge largely attributed to its uniquely immunosuppressive and desmoplastic tumor microenvironment (TME). At the heart of this therapeutic recalcitrance lies the dynamic, bidirectional crosstalk between innate immune cells-particularly tumor-associated macrophages (TAMs) and tumor-associated neutrophils (TANs)-and stromal components, notably cancer-associated fibroblasts (CAFs). This review comprehensively delineates how the innate immunity-stroma axis establishes formidable spatial and metab

ATF6-mediated GTPase RHOQ promotes liver cancer progression via enhancing aerobic glycolysis through PAK1 phosphorylation.

BACKGROUND: Hepatocellular carcinoma (HCC) ranks among the most prevalent malignant tumours worldwide, accounting for approximately 90% of primary liver cancer cases, with the highest incidence observed in Asia. Activating Transcription Factor 6 (ATF6) is an endoplasmic reticulum stress-responsive transcription factor. Our previous studies have demonstrated that ATF6 contributes to HCC tumourigenesis and progression through signalling pathways such as ATF6-CTH-Keap1-mTORC1 and ATF6-PPM1H-RPS6KB1. METHODS: RNA sequencing identified ATF6 transcriptional targets, with Ras homolog family member Q

RNA-Binding Proteins Regulate CD8+ T Cell Biology: Mechanisms and Therapeutic Opportunities.

BACKGROUND: Immune-related diseases contribute substantially to global morbidity and mortality. CD8+ T cells play a critical role in adaptive immunity by eliminating virus-infected and malignant cells. The biology of CD8+ T cells is governed by tightly regulated gene expression programs, and disruption of these programs can lead to diverse immunopathologies and malignancies. This article evaluates our current understanding of post-transcriptional and translational regulation of gene expression in CD8+ T cells by RNA-binding proteins. METHODS: Relevant and up-to-date literature was critically

Early coconut oil exposure suppresses chronic experimental Crohn's disease via myristic acid and enhanced regulatory T cells.

Public health advisory guidelines caution against the consumption of saturated fats, especially coconut oil (CO), yet CO is widely consumed in regions with low inflammatory bowel disease prevalence (IBD) and is often perceived as beneficial by IBD patients. CO is rich in medium-chain saturated fatty acids, notably lauric acid (C12:0) and myristic acid (C14:0) which are negligible in mammalian tissues without dietary intake. Because IBD is linked to saturated fats consumption, here we tested and unexpectedly found that CO and its saturated fatty acids are beneficial suppressing intestinal infla
