Review Article: Overcoming Antigen Expression Barriers in CAR-T Therapy for Solid Tumors.

Chimeric antigen receptor T cell (CAR-T) therapy has transformed the treatment landscape for hematologic malignancies, achieving deep and durable responses across multiple FDA approved products. This progress stems from foundational discoveries initiated by Zelig Eshhar, whose early demonstrations that T cells could be redirected using synthetic receptors laid the conceptual groundwork for modern CAR-T design. Despite these successes, translating CAR-T therapy into solid cancers remains challenging. Solid tumors present unique obstacles including antigen heterogeneity, immunosuppressive microe

Antibody-opsonized extracellular traps modulate macrophage responses and attenuate their T cell-associated inflammatory and costimulatory characteristics.

Macrophages and neutrophils coordinate inflammatory and reparative immunity, and dysregulated interactions between these cells contribute to the pathogenesis of immune-mediated inflammatory diseases. While extracellular traps (ETs), formed by neutrophils and other leukocytes, are enriched in citrullinated histones and exert significant pro-inflammatory effects, it remains unclear how distinct macrophage subsets interpret and resolve ET-derived signals. Here we show that macrophage subsets consistent with pro-inflammatory (GM-CSF + LPS + IFN-γ differentiated) or inflammation-resolving (M-CSF +

Case Report: CD20/BCMA bispecific CAR-T therapy for progressive multiple sclerosis.

Refractory secondary progressive multiple sclerosis (SPMS) remains an area of unmet need, particularly once disability continues to accumulate despite multiple disease-modifying therapies. Two SPMS patients are reported with C-CAR168 treatment, a bispecific CD20/BCMA CAR-T product, achieving 12- and 6-month drug-free remission. Patient 001 (EDSS 4.0→3.0) showed improvement of oculomotor dysfunction, ataxia, and cognitive impairment. Patient 002 (EDSS 6.5→6.0) demonstrated halted lower-limb progression and relieved bowel/bladder dysfunction. C-CAR168 infiltrated into the cerebrospinal fluid (CS

Mechanisms of inflammatory cell activation in periodontitis-induced bone destruction and advances in targeted interventions.

Periodontitis is a leading cause of tooth loss in adults, characterized primarily by alveolar bone destruction driven by dysregulated host immune-inflammatory responses. Central to this pathological process is the aberrant activation of inflammatory cells within the periodontal microenvironment. This review comprehensively summarizes the activation phenotypes, functions, and interaction networks of key inflammatory cells including macrophages, neutrophils, T cells, and B cells in periodontitis. It elucidates the molecular and cellular mechanisms by which these activated cells contribute to bon

Exploring tumor-derived exosomes as promising antigenic platforms: novel vaccine and immunotherapy strategies for "cold" colorectal cancer.

Colorectal cancer (CRC) remains a formidable clinical challenge, largely due to the prevalence of microsatellite-stable tumors characterized by an immunologically "cold" tumor microenvironment (TME) that is resistant to immune checkpoint inhibitors (ICIs). Tumor-derived exosomes (TEXs) play a paradoxical dual role in this landscape. Native TEXs actively orchestrate immune evasion by delivering immunosuppressive molecules, such as programmed cell death 1 ligand 1 (PD-L1) and transforming growth factor-beta (TGF-β), which impair cytotoxic T-cell function and promote the expansion of regulatory T

Immunological cross-talk between periodontitis and systemic lupus erythematosus: the role of T-B cell dysregulation in the shared pathogenic network.

Periodontitis and systemic lupus erythematosus (SLE) exhibit overlapping immunopathological features characterized by dysregulated adaptive immune responses. Emerging evidence suggests that aberrant T-B cell crosstalk represents a potential immunological link between chronic oral inflammation and systemic autoimmunity. Persistent periodontal inflammation promotes the release of pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs), leading to dendritic cell activation, neutrophil extracellular trap (NET) formation, and sustained immune stimulation. The

Mechanotransduction through LFA-1 controls upstream migration in the KG-1a stem cell line.

Immune cell recruitment into tissues requires cells to migrate on the endothelium while overcoming shear forces. Many extravasating cells, including T lymphocytes and hematopoietic stem cells, migrate upstream against the direction of shear flow on ICAM-1. Upstream migration has been shown to depend on intracellular signaling through Crk family adaptor proteins following engagement of lymphocyte function-associated antigen-1 (LFA-1). Because upstream migration is an important illustration of leukocyte mechanotransduction, we sought to identify key intracellular regulators of upstream migration

CRISPRa screen unveils noncanonical NF-κB to boost the antitumor activity of TCR-based CAR-T cells.

Engineering autologous T cells for adoptive therapies has revolutionized cancer treatment, yet limited persistence remains a major barrier to robust clinical responses and durable disease control. Although costimulation with 4-1BB markedly improves CAR-T-cell persistence, no comparably effective approach has been identified to enhance persistence in T-cell therapies that rely on native TCR signaling, including conventional TCR-T cells and synthetic T-cell receptor and antigen receptor (STAR) T cells. To identify intrinsic persistence regulators in a TCR-based context, we performed an in vivo C

Mapping the Impact of Janus Kinase Inhibitors at a Cellular Resolution in Inflammatory Bowel Disease.

The advent of biologics and targeted oral small molecules has transformed the management of patients with inflammatory bowel disease (IBD), leading to increased rates of mucosal healing, fewer hospital admissions, and reduced need for surgery. In contrast to biologics that specifically target a receptor or its substrate, Janus kinase inhibitors (JAKis) interfere with several shared signaling pathways and therefore exert broader effects. Janus kinase-signal transducer and activator of transcription signaling (JAK-STAT) occupies a central position in multiple pro-inflammatory pathways and plays

Tumor-triggered DNA fragmentation with a nucleus-targeted nanodrug for precise cGAS-STING activation and enhanced cancer immunotherapy.

Activation of the cGAS-STING pathway has emerged as a promising strategy in cancer immunotherapy due to the ability to trigger robust innate and adaptive immune response within the immunosuppressive tumor microenvironment. However, its clinical translation is restricted by multiple intractable challenges, particularly the severe inflammatory side effects caused by non-specific cGAS-STING activation. We herein developed a nucleus-targeted nanodrug with a unique characteristic of tumor-triggered DNA fragmentation and precise cGAS-STING activation. This nanodrug is engineered by encapsulating a p

Metalloimmunological hydrogel-based cancer vaccine potentiates postoperative tumor immunotherapy.

Adjuvant immune checkpoint inhibitor (AICI) therapy is limited by inadequate durable immunological memory against tumor recurrence and metastasis. Herein, we present a metalloimmunological hydrogel-based and innate immunity-focused cancer vaccine (IT-Vax) that synergizes innate-adaptive crosstalk to potentiate AICI efficacy. Through systematic screening of 9 polysaccharide hydrogels with dual crosslinking (metal coordination/Schiff base), manganese-incorporated formulations demonstrate superior dendritic cell (DC) activation capacity through orchestrating innate immune pathways. Interpretable AI model reveals spatial codes of cell recruitment in tissues.

Understanding which molecular signals recruit specific cells to precise tissue locations is fundamental to tissue biology, immunity and disease. Existing computational tools for spatially resolved transcriptomics (SRT) describe where cells reside but cannot explain why. Here we introduce SPACER, a fully interpretable multi-instance learning framework that decodes gene-level recruitment rules directly from SRT data. Applied to 37 SRT datasets spanning multiple human tumour types and mouse heart, SPACER achieved 4-21-fold improvement over existing methods in various tasks of recovering biologica

Brain-iT Immunotherapy: The Neurotransmitters Dopamine, Glutamate, GnRH-II and Neuropeptide Y Revive, Activate and Empower Healthy T Cells, Exhausted T Cells of Cancer Patients, and Genetically Engineered CAR T Cells.

Neurotransmitters are critical signaling molecules of the nervous system that activate their receptors within milliseconds, and convey accurate, coordinated, tightly regulated and important messages from one nerve cell, across a synapse, to another nerve, muscle or gland cell. It is time to realize that specific neurotransmitters also activate T cells directly and rapidly via their receptors in T cells. Subsequently, they decrease immune checkpoint inhibitors, increase expression of critical receptors, and induce a kaleidoscope of effector T cell functions. This review summarizes our discoveri

Reframing the gut-lung axis: Microbiota dysbiosis, immune-redox regulation, and the potential role of vitamin C.

The respiratory system is continuously exposed to environmental antigens, pathogens, and pollutants, requiring tightly regulated immunity to balance inflammation and host defense. Increasing evidence indicates that the microbiome-within the airways and through the gut-lung axis-plays a critical role in shaping pulmonary immune responses. Microbial metabolites such as short-chain fatty acids (SCFAs), tryptophan-derived indoles, and bile acid intermediates, influence the maturation and functional programming of alveolar macrophages, dendritic cells, and T lymphocytes. Using the emerging framewor

Association of Xeroderma Pigmentosum group F (XPF) with epigenetic regulation of T helper cell differentiation in coronary artery disease-associated prostate cancer.

BACKGROUND: Coronary artery disease (CAD) and prostate cancer share common genetic and epigenetic risk factors. Xeroderma Pigmentosum group F (XPF), a DNA repair endonuclease, has been implicated in genomic stability and epigenetic regulation. This study investigated the association between XPF expression and epigenetic changes during T helper (TH) cell differentiation in CAD-associated prostate cancer (CAD-PC). RESEARCH DESIGN AND METHODS: CD4+ T cells isolated from healthy controls and CAD-PC patients were subjected to CRISPR/Cas9-mediated XPF knockout or overexpression. TH cell differentia

Effects of polysaccharides from the rhizome of Menispermum dauricum DC on alleviating chemotherapy-induced immune damage and the immunosuppressive tumor microenvironment associated with PD-1/PD-L1 regulation.

In this study, we established a metastatic tumor model by tail vein injection of fluorescent-labeled Lewis lung cancer cells. We systematically evaluated the in vivo antitumor effects of the rhizome of Menispermum dauricum polysaccharides (MDP), a homogeneous polysaccharide obtained through sequential chromatographic purification, its efficacy in alleviating cyclophosphamide (CTX)-induced chemotherapy damage, and its ability to improve the immunosuppressive tumor microenvironment (TME). We also investigated whether its mechanism is associated with the PD-1/PD-L1 pathway. Concurrently, an in vi

TNF Receptor-2 Drives Tumoural Coagulation Factor Synthesis and Enhances Thrombosis in Clear Cell Renal Cell Carcinoma.

Clear-cell renal cell carcinoma (ccRCC), the most prevalent form of kidney cancer, is associated with an increased risk of local and systemic thromboembolism and a worse prognosis. The mechanisms that drive the coagulation program in tumours are not known. Combining RNAscope with immunofluorescence, local transcription of the key coagulation factors X and V, as well as prothrombin and fibrinogen chains (α, β, γ), across tumour cells, vascular endothelial cells and T cells is significantly enhanced in ccRCC tumours compared to adjacent normal kidney tissue. Elevated intratumoural protein expres

Exploration of Potential Molecular Mechanisms and Key Genes Related to T-Cell Exhaustion in Coronary Heart Disease from a Bioinformatics Perspective.

Coronary heart disease (CHD), a major cardiovascular disorder, is associated with T cell exhaustion (TEX) in its pathogenesis. This study aimed to identify TEX-related key genes in CHD and explore their potential mechanisms. The training set, validation set, and TEX-related genes (TEX-RGs) were downloaded from public databases. Differentially expressed genes (DEGs) were identified and intersected with TEX-RGs to obtain candidate genes. LASSO regression and Wilcoxon rank-sum test were then used to screen key genes. A nomogram for CHD prediction was constructed and validated via calibration curv
