Ligustilide enhances TNTs-mediated mitochondrial transfer to accelerate the reactivation of TECs in sleep deprivation induced postpartum mouse.

ETHNOPHARMACOLOGICAL RELEVANCE: Angelica sinensis (Oliv.) Diels (Danggui) has been widely used by puerperae in China for postpartum recovery for more than a thousand years. Its key active ingredient, ligustilide (LIG), has been documented to exert thymus-repairing effects; however, its role and underlying mechanisms in postpartum thymic regeneration impairment remain unclear. AIM OF THE STUDY: During the puerperium, the acutely atrophied maternal thymus gradually returns to its non-pregnant state. However, frequent sleep deprivation (SD) resulting from neonatal breastfeeding often slows down

Clinical significance of circulating regulatory T Cells, soluble CD25, and CXCL9 in assessing disease activity of vitiligo.

Vitiligo is an autoimmune skin disorder characterized by T-cell-mediated melanocyte destruction and loss of immune tolerance. Dysregulation of regulatory T cells and altered chemokine signaling have been implicated; however, their relationship with disease activity remains unclear. Therefore, this study aimed to evaluate peripheral immune cell subsets, particularly Tregs, sCD25, and CXCL9 biomarkers in patients with vitiligo and their relationship with disease activity. This study included 44 patients with non-segmental vitiligo and 44 matched healthy controls. Peripheral blood lymphocyte subs

PET pharmacokinetics of [¹⁸F]F-AraG as a marker of T cell activation during immunotherapy with a STING agonist with and without anti-PD-1.

BACKGROUND: There is an urgent need for clinical biomarkers to monitor the mechanistic efficacy of immunotherapy in oncology. The [18F]F-AraG tracer, developed to image activated T cells, may provide the means to establish such a biomarker. This study extends previously reported pharmacokinetic findings in untreated NSCLC across additional tumor types undergoing immunotherapy, and examines [18F]F-AraG kinetics in various organs. Four patients with advanced solid tumors were included in this phase 1 trial investigating a stimulator of interferon genes agonist with anti-programmed cell death pro

Cross-omics mapping of the human HIV restrictome across clinical states, immune-cell compartments, and protein responses.

Host restriction factors can block HIV at multiple stages, but they are often studied as individual genes or within a single molecular layer. I performed an evidence-structured computational reanalysis of three independent public datasets to determine how the human HIV restrictome varies across clinical states, immune-cell compartments, and protein responses. The analysis used bulk monocyte RNA sequencing (GSE157198), PBMC single-cell/CITE-seq data (GSE256089), and primary CD4 T-cell proteomics (PXD012263) within a prespecified restrictome-immunometabolism framework. In GSE157198, ART-naive mo

Dock2 deficiency compromises erythroid differentiation of hematopoietic stem cells.

Anemia is a prevalent pathological characteristic observed in a variety of hematologic disorders, predominantly resulting from impaired red blood cell production or increased erythrocyte degradation. DOCK2, a pivotal guanine nucleotide exchange factor within the DOCK protein family, is uniquely expressed within the hematopoietic system. Previous research has established that the absence of Dock2 significantly disrupts the development and function of several immune cell subsets, including T cells, B cells, NK cells, and granulocytes. However, the role of DOCK2 in erythroid development and assoc

β-Hydroxybutyrate alleviates intestinal aGVHD via remodeling gut microbiota-bile acid axis and inhibiting TLR4-NF-κB-driven Th1/Th17 differentiation.

The intestine is a major target organ in acute graft-versus-host disease (aGVHD), and its microbial homeostasis critically influences disease pathogenesis and progression. Emerging evidence indicates that beta-hydroxybutyrate (BHB) can remodel the gut microbiota and dampen pro-inflammatory immune activity. To investigate the therapeutic potential of BHB in aGVHD, we first reanalyzed publicly available transcriptomic and single-cell RNA sequencing data from patients with aGVHD, which revealed that genes involved in ketone body metabolism and transport were significantly decreased in intestinal

High Frequency of Circulating IL-4R+SELL+ Naïve B Cells Is Associated with Functional Cure in Chronic Hepatitis B Patients Receiving Antiviral Treatment.

BACKGROUND AND AIMS: Although hepatitis B surface antigen (HBsAg)-specific B cells have been partially characterized in chronic hepatitis B (CHB), the role of naïve B cells and their potential relevance to functional cure remain insufficiently explored. Here, we aimed to characterize the naïve B-cell subsets in functionally cured and uncured patients with CHB. METHODS: In this retrospective study, we applied single-cell RNA sequencing to compare peripheral B-cell profiles in functionally cured and uncured CHB patients. Key findings were validated by flow cytometry in an independent cohort and

Rapid CAR-T manufacturing with low-intensity priming enhances CAR-T cell fitness and limits inflammatory programming in a phase 1 study.

Conventional chimeric antigen receptor (CAR)-T manufacturing requires prolonged ex vivo activation and expansion, which may compromise T cell fitness and promote inflammatory differentiation. Here, we developed PrimeCAR, a 2-day manufacturing strategy combining low-intensity anti-CD3 priming with IL-7 and IL-21. Compared with conventionally manufactured CAR-T cells, PrimeCAR products exhibited a distinct transcriptional state characterized by preserved stemness-associated programs and a PrimeCAR-enriched primed naive T cell (Tn)-like state. Following antigen stimulation, PrimeCAR cells prefere

Targeting CDK2 ameliorates allergic asthma through suppression of epithelial chemokine production and T cell inflammatory responses.

Allergic asthma is a chronic inflammatory airway disease driven by dysregulated type 2 immunity and aberrant epithelial-immune crosstalk. Despite advances in biologic therapies, many patients remain refractory to current treatments, underscoring the need for novel strategies that simultaneously target multiple pathogenic axes. Here, we show that cyclin-dependent kinase 2 (CDK2) is upregulated in bronchoalveolar lavage cells from asthmatic patients and its expression correlates with enhanced cell cycle activity, inflammatory signatures, and T cell activation in bronchial epithelium across six i

Delayed Drug Hypersensitivity: Mechanistic Advances and Emerging Therapies.

PURPOSE OF REVIEW: Delayed drug hypersensitivity reactions span mild cutaneous eruptions, severe cutaneous adverse reactions (SCARs), and internal-organ injury. We review how pharmacogenomic, single-cell, and spatial methods have shown that these syndromes are driven by shared effector mechanisms, and how identifying which one predominates opens the way to targeted treatment. RECENT FINDINGS: HLA associations are necessary but not sufficient to explain who reacts. Susceptibility is increasingly attributed to a pre-existing, often virus-shaped T-cell repertoire and to where tissue-resident mem

Chronic Hepatitis B: Current Understanding and Clinical Implications of Immune Exhaustion.

Chronic hepatitis B (CHB) infection leads to progressive dysfunction and exhaustion of antiviral immune cells, which is a primary obstacle to clinical cure. Although the phenotypic hallmarks of exhausted T cells are well described, the hierarchical trajectory, subset-specific heterogeneity, and underlying mechanisms of exhaustion across the full spectrum of immune cells in the unique hepatic microenvironment remain poorly defined. This review provides a comprehensive and critical synthesis of recent advances in immune exhaustion during CHB. We delineate the common and distinct exhaustion progr

Allogeneic Dendritic Cells: Transforming the Impact of Cancer Immunotherapy.

Aggressive tumors are treated with surgery, radiation, chemotherapy, targeted therapy, and various immunotherapies. Cancer immunotherapies under investigation encompass monoclonal antibodies, including checkpoint inhibitors, bispecific antibodies, and T cell redirecting antibodies, as well as gene-modified cell therapies such as CAR-T cells, donor lymphocyte infusions (DLI), antigen-specific DLI, and TCR-engineered T cells. While dendritic cell (DC)-based vaccines derived from peripheral blood have shown promise in preclinical studies, clinical trials have been less effective. Recently, a nove

Engineered Dendritic Cells Imprint Dual Gut-Homing Specificity on Human Tregs for Pan-Intestinal Immunotolerance.

Current therapeutic strategies for inflammatory bowel disease often rely on systemic immunosuppression or single-target biologics, which can lead to significant side effects or incomplete remission. To overcome this therapeutic hurdle, we recently proposed engineering dendritic cells (DCs) to de novo synthesize high concentrations of the active vitamin D (1,25[OH]2D) and A (retinoic acid or RA) because 1,25(OH)2D directly stimulates the expression of Foxp3 (the master regulator of CD4+ Tregs) and RA induces gut-homing receptor expression. In our previous studies, we engineered mouse DCs using

The narrow path to CD8+ T cell recognition of mutant p53.

Shared cancer driver mutations offer attractive targets for cancer immunotherapy, but their expression does not necessarily put them within the reach of a T cell response. In this issue of Immunity, Haratani et al. identify distinct barriers to the processing, presentation, and recognition of neoantigens generated from mutant p53 that help explain this gap.

Astrocytes go into presenter mode.

Astrocytes are glial cells important for neuronal health, but their functions in CNS immunopathology are less understood. In a recent issue of Nature, Lee et al. report that CD40 engagement and proinflammatory cytokines promote lipid droplet accumulation in astrocytes and subsequent antigen presentation, boosting pathogenic CD4+ T cell responses during experimental autoimmune encephalomyelitis.

Immunogenicity of recombinant porcine parainfluenza virus 5 expressing the spike protein of porcine epidemic diarrhea virus.

The porcine epidemic diarrhea virus (PEDV) induces severe diarrhea in neonatal piglets, often resulting in high mortality rates. Consequently, the development of effective and safe vaccines is a critical priority for controlling PEDV infection. Vaccines vectored by the parainfluenza virus 5 (PIV5) exhibit distinct advantages in terms of safety, genomic stability, and immunogenicity. In this study, two novel porcine-origin PIV5 strains (PIV5-HuB and PIV5-DP1-1) were isolated from pigs in China, and their whole-genome sequences and phylogenetic relationships were analyzed. Subsequently, the PIV5
