A zinc organic metal skeleton vaccine for lung cancer treatment can stimulate strong innate and adaptive anti-tumor immunity.

BACKGROUND: Tumor-associated macrophages (TAMs) function as central regulators bridging innate and adaptive immunity in the tumor microenvironment (TME). Therapeutic targeting of TAMs to promote M1 polarization represents a critical strategy for tumor suppression. METHODS: Herein, we engineered a zinc metal-organic framework functionalized with mannose (MAN) and loaded with CLDN6 antigen (Zn-MAN@CLDN6). We systematically evaluated the anti-tumor efficacy and immunomodulatory functions of Zn-MAN@CLDN6 both in vitro and in vivo. The nanoplatform exhibited the potent cytotoxicity against lung ca

Matrix-Based Specific T-Cell Activation Reporter for Nearly-Digital Mapping of Antigen-Specific T Cells via Heterotypic Cell Micro-Communities and Digital Microfluidics.

Antigen-specific T cells constitute a low percentage of peripheral blood mononuclear cells (PBMCs) and require antigen presentation by autologous antigen-presenting cells for activation and cytokine secretion. Conventional interferon-γ release assay (IGRA) averages rare activation events beyond cellular resolution, while single-cell compartmentalization disrupts the intracellular synapses for T cell immunity. Here, we developed mSTAR (matrix-based Specific T-cell Activation Reporter), an active-matrix digital microfluidic platform that partitions microliters of PBMCs into a nanoliter-scale mat

Molecular basis for developing tumor-targeted siRnas for future immunotherapy.

INTRODUCTION: Certain immune signaling pathways contribute to cancer progression by promoting tumor immune evasion through immune checkpoint activation, immunosuppressive cytokine production, metabolic depletion, and altered intracellular signaling. Small interfering RNAs (siRNAs) provide a programmable strategy for selectively silencing genes involved in immunosuppression, offering a targeted approach to enhance antitumor immunity. AREAS COVERED: This review highlights the rationale for targeting immune regulatory factors with siRNA in cancer immunotherapy. Key targets include immune checkpo

Harnessing the Bacterial Membrane-Derived Oral Vaccine to Potentiate Antitumor Immunity and Metabolic Control of T Cells.

The fluctuating gastrointestinal milieu and its restrictive epithelial barrier represent critical challenges for the design of oral cancer vaccines. Nevertheless, leveraging the gut microbiome as an interventional strategy has recently gained traction, especially in patients on immune checkpoint inhibitor (ICI) therapy. Here, microbial-derived double-layered membrane vesicles are developed as adjuvants and cell-free carriers, fused with cell-penetrating peptide-modified antigens, and encapsulated in an alginate/chitosan-microsystem to engineer oral vaccines. This design strategically harnesses

A guide to studying tumour-T-cell interactions using in vitro coculture assays.

T cells are central mediators of antitumour immunity and are the cellular basis of many successful cancer immunotherapies, including immune checkpoint blockade, T-cell receptor (TCR)-engineered therapies and chimeric antigen receptor (CAR) T cells. However, clinical responses remain variable, and many tumours evade or resist T-cell-mediated control. Understanding how T cells recognise tumour cells, deliver cytotoxic signals, produce inflammatory cytokines and shape tumour cell fate is therefore essential for improving modern immunotherapies. In vitro tumour T-cell coculture systems provide a p

CRISPR-Cas9 screen identifies eIF4G2 as a regulator of CD8+ T cell dysfunction.

CD8+ T cells directly target and kill cancer cells and are required for effective antitumor immunity. However, CD8+ T cells may become dysfunctional when exposed to chronic antigen stimulation during cancer. Dysfunction is characterized by reduced proliferation, increased expression of inhibitory receptors, and impaired effector function, yet the genetic mechanisms controlling dysfunction remain poorly understood. Therefore, we developed a murine ex vivo chronic stimulation model that recapitulates features of CD8+ T cell dysfunction observed within the tumor microenvironment. By coupling this

Recurrent thymoma-associated PRCA with persistent agranulocytosis: a case report and therapeutic considerations.

We report a case of recurrent type A thymoma accompanied by pure red cell aplasia (PRCA) and agranulocytosis. Following local recurrence after thymoma resection, the patient gradually developed anemia and leukopenia as the lesion progressed; bone marrow examination showed marked suppression of erythropoiesis and reduced granulopoiesis. Taken together with the peripheral blood findings, these results suggested thymoma-associated immune-mediated hematopoietic injury. Following multidisciplinary evaluation, the patient underwent thoracoscopic resection of the recurrent lesion, and pathological ex

Microbial cues for a stem-like lymphoid niche.

While tertiary lymphoid structures (TLSs) broadly correlate with immunotherapy efficacy, their functional heterogeneity can translate into divergent clinical outcomes. In this issue of Cancer Cell, Hu et al. uncover a microbiota-orchestrated stem-like lymphoid niche in which CD70+ ILC2s sustain progenitor exhausted T cells, and their findings redefine the functional organization underlying TLS-associated antitumor immunity.

Combating CD24 to overcome CD19 CAR-T resistance via induction of innate and adaptive immunity in B-cell malignancies.

CD24 is highly expressed in aggressive B-cell malignancies and represents a novel immunologic vulnerability to overcome resistance to CD19 chimeric antigen receptor T-cell (CAR-T) therapy. We developed a CD24 × CD3 bispecific antibody (CD24 BsAb) and CXCR3-armed CD24 CAR-T cells, both exhibiting potent antitumor activity in lymphoma and leukemia models. CD24 BsAb and CD24 CAR-T overcome CD19 CAR-T resistance driven by antigen loss and apoptosis defects in vitro and in vivo. Beyond direct cytotoxicity, CD24-targeted T cells reprogram macrophages toward an M1-like phenotype and enhance phagocyti

CRISPR integration of an FLT3 CAR into the TRAC locus generates CAR T cells for AML.

BACKGROUND: Acute myeloid leukemia (AML) remains a therapeutically challenging hematological malignancy with high relapse rates, especially in patients harboring mutations in the FMS-like tyrosine kinase 3 (FLT3) gene. FLT3, a receptor tyrosine kinase involved in hematopoietic cell proliferation and survival, is frequently mutated or overexpressed in AML, contributing to leukemic proliferation and poor prognosis. Although FLT3 inhibitors have demonstrated clinical benefit, resistance often develops, underscoring the need for alternative treatment strategies. METHODS: In this study, we develop

Vitamin D3 Inhibits Expression of Interleukin-6 and Calcitonin Gene-Related Peptide by Transformed Dorsal Root Ganglion Neurons.

Interleukin-6 (IL-6) and the neuropeptide calcitonin gene-related peptide (CGRP) promote the generation of T helper 17 (Th17) cells. Th17-type immunity is involved in several autoimmune disorders, including psoriasis. Also, there is evidence that sensory nerves play a role in Th17-type immunity, including in psoriasis. Vitamin D (VD) blood levels vary inversely with the severity of psoriasis, and topical VD analogs are effective treatments for this disease. We have now asked whether 25-dihydroxyvitamin D3 [25(OH)D3] and the more potent, active analog 1α,25-dihydroxyvitamin D3 [1α,25(OH)2D3] in

Pbx1 as a regulator of T-cell fate in autoimmunity.

Pbx1 is a highly conserved transcription factor that regulates cell fate, proliferation, and apoptosis in multiple cell types, including stem cells and lineage precursors, and mature immune cells. The expression of a novel dominant-negative splice isoform of Pbx1, as well as a non-coding polymorphism near its 3' end in T cells and B cells, respectively, have been associated with susceptibility to systemic lupus erythematosus. This review, focused on T cells, presents the evidence that Pbx1 regulates the balance between regulatory and effector CD4+ T cells, with the lupus-associated dominant-ne

Fueling the sentinels: organelle remodeling and metabolic adaptation in tissue-resident memory T cell persistence.

Tissue-resident memory T (TRM) cells persist at barrier and parenchymal sites where oxygen, nutrients, and inflammatory signals fluctuate. Their durability therefore cannot be explained by tissue retention alone. Evidence from skin, liver, intestine, lung, tumors, and inflamed tissues supports a coupled program in which local nutrient capture, mitochondrial fuel selection, organelle dynamics, autophagy, mitophagy, and proteostatic responses sustain survival while preserving rapid effector competence. We propose "organelle-niche matching" as a testable four-step working model: tissue inputs eng

From gut to lung: the microbiome-directed causal mechanisms and therapeutic horizons in asthma.

The gut-lung axis represents a predominantly gut-to-lung communication network through which gut microbiota and their metabolites influence pulmonary immunity. In asthma, gut dysbiosis-characterized by reduced microbial diversity, depletion of short-chain fatty acid (SCFA)-producing taxa, and expansion of pro-inflammatory pathobionts-has been consistently associated with disease severity and type 2 inflammation. While the axis is conceptually bidirectional, the vast majority of mechanistic evidence to date describes unidirectional gut to lung regulation. This review critically synthesizes evid

Macrophage states and immunometabolic adaptation in peritoneal metastases from colorectal cancer.

Immunotherapy responses in colorectal cancer peritoneal metastases (CRCPM) are not determined by mismatch repair (MMR) status alone. Most MMR-proficient/microsatellite-stable (pMMR/MSS) tumors have low baseline sensitivity, while MMR-deficient/microsatellite instability-high (dMMR/MSI-H) tumors do not respond uniformly, particularly in patients with ascites. This Mini Review compares evidence across compartments and disease stages: human associations, macrophage-resolved functional studies and interventions altering immunotherapy efficacy in mice. Human ascites and tissue studies identify resi

T-cell dysfunction and death in sepsis: a mechanistic review.

Sepsis is a life-threatening syndrome of multi-organ dysfunction caused by a dysregulated host response to infection. Despite advances in supportive care, the underlying immunopathological mechanisms remain incompletely understood. A hallmark of sepsis is immunoparalysis, characterized by an early hyperinflammatory phase followed by sustained immunosuppression, predisposing patients to secondary infections and adverse outcomes. A central feature of this process is profound lymphopenia accompanied by impaired T-cell effector function. This review synthesizes current evidence on T-cell perturbat

Heart-specific T cell responses: from antigens to disease mechanisms.

The field of immunology has transformed our understanding of myocardial cell biology and paved the way for new pathophysiological concepts in cardiology. Researchers have sought to elucidate the contributions of nonspecific inflammatory mechanisms to cardiac diseases, and an increasing number of clinical trials have explored anti-inflammatory approaches in cardiovascular disease. Moreover, adaptive immune mechanisms directed to cardiac proteins released by dying cardiomyocytes have been reported across a broad spectrum of conditions, including acute myocarditis, myocardial infarction and heart

The Gut Microbiota-inflammation-immune Axis in Cancer: Anti-inflammatory Mechanisms and Therapeutic Implications.

Through its mediation between inflammatory and immune pathways, the gut microbiota has proved to be a key regulator of cancer development, progression, and treatment response. Changes in microbial composition known as dysbiosis might facilitate chronic inflammation, immune dysregulation, and genomic instability, thereby facilitating carcinogenesis. Conversely, positive commensal microbes can generate short-chain fatty acids, bile acids, and tryptophan derivatives, which help maintain the epithelial barrier, cytokine signaling, and innate and adaptive immune responses. These microbial metabolit

Yiguanjian alleviates ocular surface inflammation and lacrimal gland immune dysregulation in experimental dry eye disease: evidence from in vivo and in vitro models.

BACKGROUND: Persistent ocular surface inflammation and lacrimal gland immune dysregulation are central to the progression of dry eye disease (DED). Th17/Treg imbalance contributes to sustained inflammatory activation in the ocular surface and lacrimal gland. Yiguanjian (YGJ) has anti-inflammatory and immunomodulatory potential, but whether it can alleviate ocular surface inflammation and restore lacrimal gland immune homeostasis in DED remains unclear. METHODS: A benzalkonium chloride (BAC)-induced rat DED model was used to assess the effects of YGJ on tear secretion, tear film break-up time,
