Disruption of the ARID1A-containing SWI/SNF complex reprograms tumor-associated macrophages and enhances immunotherapy response.

Tumor-associated macrophages (TAMs) contribute to tumor immune evasion and therapeutic resistance. However, the epigenetic and transcriptional regulators that control TAM function remain largely unidentified. Here we investigated the role of the SWI/SNF chromatin remodeling complex in TAMs and whether disruption of SWI/SNF function in TAMs could improve immunotherapy. Pharmacologic inhibition of SWI/SNF ATPase activity improved the efficacy of checkpoint blockade immunotherapy, slowing tumor growth and reprogramming transcription broadly in tumor cells, tumor-infiltrating lymphocytes, and TAMs

Single-Particle STORM Imaging Quantifies Coating Heterogeneity Among Cell Membrane-Coated Nanoparticles.

UNLABELLED: Cell membrane-coated nanoparticles (CNPs) are a potentially transformative biomimetic targeted-delivery platform, which can engage therapeutic targets through source-cell membrane protein functions or homotypic interactions. However, their therapeutic development is limited by the current inability to accurately resolve membrane coating completeness, and CNP particle-to-particle variation. Here, we introduce a STochastic Optical Reconstruction Microscopy (STORM)-based approach that resolves population composition and quantifies membrane coating coverage at the single-particle level

Engineered CTLA-4 targeting chimeras for targeted protein degradation and immune synapse reprogramming.

Despite the success of immune checkpoint blockade in cancer therapy, overcoming the immunosuppressive tumor microenvironment remains a major barrier to durable responses. Here, we introduce cytotoxic T-lymphocyte-associated antigen-4 (CTLA-4)-targeting chimeras (cTACs), a class of synthetic molecules that harness CTLA-4-mediated trans-endocytosis (TE) for targeted protein degradation across cellular boundaries. The lead candidate, cTAC4.0, is composed of the extracellular domain of CD80 fused to an anti-programmed death-ligand 1 (PD-L1) fragment via a human immunoglobulin G4 (IgG4) heavy chain

CAR T cell targeting of inflammatory myeloid progenitors in the bone marrow remodels border-associated macrophages and reverses cognitive aging.

Aging is associated with progressive neuroinflammation and cognitive decline, yet the cellular sources that sustain this process and whether they can be targeted peripherally remain unclear. Here, we identify inflammatory border-associated macrophages (BAMs) as key drivers of neuroinflammation in aging and show that their therapeutic and prophylactic elimination through intrathecal CAR T cell therapy restores cognitive performance in mouse models of aging and Alzheimer's disease. Furthermore, targeting of aged inflammatory bone marrow myeloid progenitors through either intravenous CAR T cells,

Functional Exhaustion Limits CAR-NK Therapy in Glioblastoma but Can Be Mitigated by Early Repeated Dosing.

With the commercial success of Chimeric Antigen Receptor T (CAR-T) cell therapies in the blood cancer setting, there are increasing numbers of clinical trials evaluating this treatment modality in patients with solid tumors, including brain tumors. Allogeneic CAR-natural killer (NK) cell therapy offers an attractive "off-the-shelf" option using the CAR platform. However, this approach remains comparatively untested clinically, with only one published clinical trial to date in patients with central nervous system (CNS) tumors. Building on our prior in vitro characterization of allogeneic B7H3-C

Interpretable machine learning coupled to gene regulatory networks uncovers subcircuits underlying cell fate decisions.

Gene regulatory networks (GRNs) model causal linkages that control cell fate decisions and differentiation transitions. Prioritizing regulatory subnetworks underlying cell state differences is of critical importance, but current methods including those reliant on topological metrics introduce circularity as the metrics prioritizing TFs are computed from the same networks whose assumptions they inherit. Separately, interpretable machine learning methods can identify latent factors (LFs) that discriminate cellular states with formal statistical guarantees but do not model regulatory linkages. He

The Lag3 KIEELE motif couples HPK1 to metabolic and functional regulation of Foxp3+ regulatory T cells.

Lymphocyte activation gene-3 (Lag3) is an inhibitory receptor that limits effector T cell activation and function. We previously demonstrated that Lag3 supports regulatory T (Treg) cell function through Myc-dependent metabolic reprogramming. However, the intracellular mechanisms underlying this regulation remain incompletely understood. Here, we generated mice with Treg-specific deletion of the Lag3 KIEELE motif and demonstrated that this motif is required for Treg-mediated suppression of autoimmune inflammation. Loss of the Lag3 KIEELE motif led to enhanced TCR/PI3K-Akt/MEK-ERK signaling and

Avidity-controlled biotherapeutic delivery systems for the treatment of ischemia-reperfusion acute kidney injury.

UNLABELLED: Regulatory T cells (Tregs) are key mediators of immune resolution and tissue repair following acute kidney injury (AKI), yet strategies to selectively enrich Tregs within the renal microenvironment remain limited. Interleukin-2 (IL-2) is essential for Treg survival and function, but systemic IL-2 delivery is constrained by a short half-life, narrow therapeutic window, and risk of off-target activation of cytotoxic T cells. Effective Treg-based immunomodulation may therefore require both localized cytokine signaling and targeted recruitment of precursor immune populations. Here, we

Programming primary human T cells to deliver gene editing machinery.

Clinical applications of gene editors including CRISPR-Cas9 are limited by the cell-type specificity and tissue penetrance of current in vivo delivery options. Cell-mediated gene editor delivery could overcome these obstacles. To date, systems utilizing immortalized cell lines have demonstrated gene editor delivery in vitro . Because T cells traffic throughout the organism and engage in intercellular communication with individual target cells, we wondered whether they could be engineered to deliver gene editing machinery. Here, we demonstrate that primary human T cells can produce retroviral-b

Peripheral Immune Alterations and T Cell Clonal Dynamics Across the Parkinson's Disease Spectrum.

The role of peripheral immunity in Parkinson's disease (PD) remains incompletely understood. Here, we performed paired single-cell RNA sequencing (scRNA-seq) and T-cell receptor (TCR) profiling of 630k peripheral blood mononuclear cells (PBMCs) from 168 donors spanning idiopathic and genetic PD, prodromal PD, and healthy controls. We identify broad changes in immune-cell composition across the disease spectrum, including decreased naive B cells and increased naive CD8+ T cells in idiopathic PD. Transcriptional profiling reveals disease stage-dependent remodeling of mitochondrial and oxidative

Therapeutic vaccines for NSCLC: From mechanisms to clinical translation.

Despite advances in targeted therapy and immune checkpoint inhibitors (ICIs), durable disease control remains elusive for most patients with non-small cell lung cancer (NSCLC), particularly those with advanced-stage disease. Therapeutic cancer vaccines offer the potential for sustained immune surveillance by inducing tumor-specific effector and memory T-cell responses. However, their clinical benefits have been inconsistent to date. In advanced disease, rapid tumor progression, an immunosuppressive tumor microenvironment (TME), defective antigen presentation, and immune escape collectively imp

Epithelial to mesenchymal transition and M2 macrophages drive aggressiveness in murine and human sarcomatoid urothelial carcinoma.

Patients with majority variant subtype histology including sarcomatoid urothelial carcinoma (SUC) have poor outcomes and limited treatment options, and there are few preclinical resources to investigate rare histologic subtypes. We developed three novel variant subtype histology bladder cancer (BC) cell lines by finding that basal BC tumor cells derived from mice treated with N-butyl-N-(4-hydroxybutyl)-nitrosamine (BBN) were initially not competent to grow in immune competent mice, but then acquired tumorigenicity following passaging ("recycling") in mice lacking mature B and T cells. Adoptive cell therapy in lung cancer: progress, challenges, and prospects.

Lung cancer is the leading cause of cancer-related deaths worldwide. Over the past decade, immunotherapy, represented by immune checkpoint inhibitors (ICIs) targeting programmed cell death protein-1/programmed cell death protein ligand-1 (PD-1/PD-L1) or cytotoxic T lymphocyte associated antigen-4 (CTLA-4), has revolutionized the treatment paradigm of lung cancer, reducing the risk of death by approximately 30%. However, approximately 70% of patients with advanced non-small cell lung cancer are either non-responsive to ICIs or initially respond to treatment but eventually experience disease pro

Epstein-Barr virus and hepatic failure: pathogenesis, clinical manifestations, and management.

Epstein-Barr virus (EBV) infection is ubiquitous, affecting over 90% of the global population, and while typically benign, it can rarely cause acute liver failure - a life-threatening complication that is frequently fatal. This review comprehensively reviews two overlapping clinical and pathogenetic patterns of EBV-associated hepatic failure - isolated severe EBV hepatitis and EBV-triggered hemophagocytic lymphohistiocytosis (HLH) - which are best understood as ends of a clinical spectrum rather than discrete entities. In isolated EBV hepatitis, liver injury is driven by CD8-positive T cells a

Multipathway pharmacological interventions targeting liver-derived immune resistance in colorectal cancer liver metastasis.

Immune checkpoint inhibitors provide durable benefit in mismatch repair-deficient colorectal cancer, but most microsatellite-stable tumors remain resistant, particularly in the presence of liver metastases. Immune resistance in colorectal cancer liver metastasis reflects both baseline MSS-CRC mechanisms, including low neoantigenicity, limited T-cell priming and tumor-intrinsic immune exclusion, and liver-specific or liver-amplified mechanisms, including tolerogenic antigen presentation by hepatic resident cells, macrophage-mediated loss of tumor-reactive T cells, and hepatic stromal, vascular

Shared immune perturbations in hepatocellular carcinoma-depression comorbidity and synergistic immunotherapeutic effects of fluoxetine combined with anti-PD-1.

BACKGROUND: Hepatocellular carcinoma (HCC) often coexists with depression, elevating patient mortality by 39%. The tryptophan metabolic axis bridges tumor and neuropsychiatric immune dysfunction, yet the immunometabolic crosstalk underlying HCC-depression comorbidity remains poorly defined, and reliable combinatorial immunotherapies are absent. METHODS: We integrated public multi-omics datasets including TCGA, GEO, eQTLGen and FinnGen, and performed two-sample MR as well as genetic colocalization analyses to screen hub genes associated with HCC-depression comorbidity. Tregs transduced with a Ro60-reactive TCR have increased immunosuppressive functions in Sjögren's disease.

Sjögren's disease (SjD) is a T cell-driven autoimmune condition strongly associated with the HLA-DRB1∗03:01 allele and autoantibodies against the Ro60 protein. Previous studies have identified deficiencies in regulatory T cells (Treg) in patients, indicating a breakdown in immune tolerance. Current treatments remain limited to symptomatic relief or broad immunosuppression, without addressing the underlying immune dysregulation. In this study, we present the first preclinical evidence supporting a TCR-engineered Treg (TCR-Treg) therapy specifically targeting SjD. We designed a TCR-Treg that rec

Immune checkpoint inhibitor-associated acute interstitial nephritis: Pathogenesis and heterogeneity.

Immune checkpoint inhibitor (ICI)-associated acute kidney injury is a significant complication of cancer immunotherapy, with acute interstitial nephritis (AIN) being the most common pathological type. This review systematically elaborates on the core pathogenesis, spectrum of pathological heterogeneity, and the distinct immunological underpinnings of ICI-associated AIN (ICI-AIN). The typical mechanism of ICI-AIN involves abnormal activation of the adaptive immune system, activating resident autoreactive CD8+ tissue-resident memory T cells. This triggers an interferon-gamma-driven inflammatory

A Magnetic Bead-Based Negative Selection and Flow Cytometry Protocol for Evaluating Licochalcone A-Mediated Inhibition of Murine Th17 Differentiation.

Naïve CD4⁺ T cell differentiation into Th17 cells is a key driver of autoimmune and chronic inflammatory skin diseases. Licochalcone A (Lico-A), a bioactive flavonoid from Glycyrrhiza inflata, exhibits anti-inflammatory activity in dermatological applications, yet its direct effect on Th17 commitment remains poorly characterized due to a lack of standardized evaluation protocols. To address this, this protocol establishes a complete in vitro workflow to assess Lico-A's inhibition of naïve CD4⁺ T cell differentiation into Th17 cells. The procedure comprises: isolation of highly pure naïve CD4⁺

CD8⁺ T Cell infiltration and spatial organization in cancer: mechanisms, barriers, and therapeutic strategies.

The infiltration of tumors by CD8⁺ T lymphocytes is a critical determinant of antitumor immunity and response to immunotherapy. However, the presence of tumor-specific CD8⁺ T cells does not necessarily translate into effective tumor control, as multiple biological barriers can prevent their recruitment, spatial distribution, persistence, and functional activity. This review provides a comprehensive overview of the multistep mechanisms regulating CD8⁺ T-cell infiltration, from immune priming and systemic trafficking to vascular entry, intratumoral migration, and tissue residency. We discuss how
