Kcnn4 (KCa3.1) is a calcium-activated potassium channel involved in cell volume regulation and immune cell function, but its role in salivary gland homeostasis remains poorly understood. In this study, we generated constitutive Kcnn4 knockout (Kcnn4-/-) mice to investigate the impact of Kcnn4 loss on salivary glands and systemic inflammation. Histological examination revealed that Kcnn4-/- mice had enlarged salivary glands with increased collagen deposition (fibrosis) but no major disorganization on H&E staining. mRNA-seq identified 473 differentially expressed genes (DEGs), with significant e

BACKGROUND: Pulmonary endarterectomy (PEA) remains one of potentially curative options for chronic thromboembolic pulmonary hypertension (CTEPH). The operation can remove proximal thromboembolic materials and their secondary branches. However, the cellular landscape and pathological features across different anatomical regions remain limited understood.
METHODS: Ten patients with CTEPH undergoing PEA surgery and six reference distal pulmonary arteries were conducted single-cell transcriptomics to profile the cellular composition of the main and its bifurcate portion.
RESULTS: Pronounced musc

BACKGROUND: Atherosclerosis (AS) is a chronic inflammatory disease driven by complex immune responses. Among the involved immune cells, CD4⁺ T cells are closely associated with plaque progression. An imbalance between inflammatory and regulatory T-cell responses can sustain vascular inflammation. However, the relationship between routinely measured blood biomarkers and these T-cell states remains unclear.
MAIN BODY: This narrative review examines how LDL-C, HDL-C, HbA1c, high-sensitivity C-reactive protein (hsCRP) and creatinine relate to biological processes involved in CD4⁺ T-cell regulatio

Triple-negative breast cancer (TNBC) treatment is often hindered by tumor hypoxia, cellular survival compensation, and the immuno-suppressive micro-environment. Through the "molecular-electron pumping" strategy, a cyanine photosensitizer BrCy5 with excellent Type I ROS generation ability (two kinds of Type I ROS production of BrCy5 are 308% and 206% higher) was developed to realize photodynamic therapy-mediated immunotherapy under tumor hypoxia. In the work, a multifunctional nanoenzyme HMB loaded with Type I photosensitizer was further designed, which precisely delivers BrCy5 and Fe3+ to tumo

Patients with acute-on-chronic liver failure (ACLF) have severe CD8+ T cell dysfunction. This study examined G protein-coupled receptor 82 (GPR82) in CD8+ T cell exhaustion in hepatitis B virus-related acute-on-chronic liver failure (HBV-ACLF). Using GEO analysis, clinical samples, GPR82 manipulation, transforming growth factor beta (TGF-β)/SMAD intervention, dual-luciferase assays, and a mouse hepatitis virus strain 3 (MHV-3) mouse model, we found GPR82 overexpression in ACLF liver and HBV-ACLF CD8+ T cells, correlating with T cell exhaustion. GPR82 knockdown enhanced CD8+ T cell function, wh

We previously demonstrated that anemoside B4 (AB4) exerts a beneficial effect in a dextran sulfate sodium (DSS)-induced murine model of ulcerative colitis (UC). In this study, we further explored the mechanism of AB4 against UC from the perspective of gut microbiota-immune interplay. The gut microbiota was depleted in two DSS-induced colitis models to investigate the modulatory effects of the gut microbiome on immune responses and the activity of AB4. In parallel, adoptive transfer of CD4[Formula: see text]CD62L[Formula: see text] T cells into Rag1-deficient (Rag1-/-) mice was used to elucidat

Matrix metalloproteinase-8 (MMP-8) regulates inflammatory responses, but its contributions to the pathogenesis of influenza A viral (IAV) disease are not known. Plasma MMP-8 levels were increased 30-fold in subjects infected with H1N1 or seasonal IAV and were inversely associated with the PaO2/FiO2 ratio, suggesting potential value as a candidate marker of lung injury severity that requires validation in larger cohorts. MMP-8 levels were elevated in the lungs of H1N1-infected wild-type (WT) mice and localized to airway epithelial cells, macrophages, and polymorphonuclear neutrophils (PMNs). H1

INTRODUCTION: IKZF2 (Helios) maintains the suppressive function of regulatory T cells (Tregs) and is an emerging immuno-oncology target. Its degradation destabilizes Tregs, strengthens antitumor immunity, and may remodel the tumor microenvironment (TME) with reduced off-target toxicity. Recent patents focus on selective cereblon (CRBN)-recruiting molecular glues, combination therapies, and next-generation degraders.
AREAS COVERED: SciFinder and Google Patents were searched for IKZF2 molecular glue degraders reported in patents and peer-reviewed journals from January 2020 to September 2026.

Marek's disease virus (MDV) is a highly contagious, oncogenic alphaherpesvirus that induces profound immunosuppression and T-cell lymphomas in poultry, serving as a prominent paradigm for investigating virus-induced tumorigenesis and pathogen evolution under vaccine-driven selection pressure. Despite half a century of widespread vaccination, the continuous circulation and progressive enhancement of MDV virulence within flocks highlight a critical gap in our understanding of host-virus immunological interplay and co-evolution. This review systematically delineates the dynamic molecular mechanis

Despite advances in adoptive T cell therapy, therapeutic efficacy in solid tumors remains inconsistent, and the mechanistic determinants of treatment failure are incompletely understood. Increasing evidence suggests that effective antitumor responses depend not only on direct recognition of malignant cells but also on interactions with the tumor microenvironment, including non-malignant host cells that cross-present tumor antigens. Using an HLA-A2-transgenic syngeneic mouse cancer model that enables near-physiological presentation of human tumor antigens by both malignant and host cells, we in

B-cell-based immune cell therapies represent a potential strategy for restoring immune tolerance by expanding regulatory T cells (Tregs). CD4+Foxp3+ Tregs are essential for maintaining immune homeostasis and represent an attractive therapeutic target. However, clinically established strategies to safely and durably expand endogenous Tregs remain lacking. We previously identified IgDlow/- B cells as a splenic B-cell subset that promotes Treg expansion through a glucocorticoid-induced TNF receptor ligand (GITRL)-dependent mechanism, whereas other B cell populations do not. In B-cell-specific GIT

BACKGROUND: Cholangiocarcinoma is a heterogeneous biliary malignancy characterized by late diagnosis, stromal desmoplasia, immune exclusion, and frequent resistance to chemotherapy, immunotherapy, and molecularly targeted therapy. Although genomic stratification has enabled biomarker-directed treatment for selected patients, cancer cell-intrinsic alterations alone do not fully explain disease progression or therapeutic failure.
MAIN BODY: Recent advances in single-cell RNA sequencing, single-cell multi-omics, spatial transcriptomics, spatial proteomics, multiplex imaging, and pathomics have r

Cytotoxic CD8+ T cells have been the focus of research efforts to improve the efficacy of anticancer adoptive cell therapies. However, emerging evidence is highlighting the critical role of CD4+ T cells in these treatments, including chimeric antigen receptor-T cells. Although the presence of CD4+ T cells is known to be important for efficacy of cell therapies, the mechanisms by which CD4+ chimeric antigen receptor-T cells exert their beneficial effects remain incompletely understood. In this article, we review the roles of CD4+ T cells in adoptive cell therapy, highlighting understudied areas

γδ T cells are promising cancer-immunotherapy effectors because they recognize tumors without major histocompatibility complex (MHC) restriction and carry low graft-versus-host-disease risk in allogeneic settings, yet few bibliometric studies have integrated immunotherapy-specific research trends with clinical-trial mapping and translational-gap analysis. We characterized the global research output, intellectual foundations, emerging frontiers, and clinical translation of γδ T cells in cancer immunotherapy (2000-2025) by combining bibliometric analysis of 3,273 Web of Science publications (bib

Acute myeloid leukemia (AML) is an aggressive hematologic malignancy associated with a poor prognosis and an unmet need for more effective treatment options, particularly after relapse. One potential approach to address this need, which has been used successfully in solid tumors, is adoptive transfer of T cells engineered to express specific T cell receptors (TCRs). Particularly attractive are TCRs that can target epitopes derived from oncogenes on which cancer cells depend while sparing normal cells that do not harbor the mutation. One promising oncogenic target in AML is mutated nucleophosmi

UNLABELLED: Bispecific T cell engagers (BTCEs) have transformed the treatment of B cell malignancies, but how they remodel non-conventional T cell circuits in patients, and how they should be ideally combined with existing treatment backbones, remains unclear. Here we show that CD3-engaging BTCEs convert human FOXP3⁺ regulatory T cells (Tregs) into an AP-1-imprinted hybrid cytotoxic state that contributes to tumor clearance. Leveraging longitudinal bone marrow and peripheral blood sampling from patients with newly diagnosed multiple myeloma (NDMM) treated with BTCE-based induction in the phase

UNLABELLED: Partially preserved function of recombination-activating genes (RAG) 1 and 2 can be linked to divergent and hard-to-predict clinical and immunological phenotypes, even among close relatives. It is unclear how intrinsic RAG hypomorphism interacts with extrinsic environmental pressures to shape clinical trajectory. We dissect this phenomenon in a unique cohort of 18 Old Order Mennonite individuals with a novel missense founder RAG1 variant (p.C176F) with 25.8% recombinase activity of wild-type protein. After studying a myriad of markers, we demonstrate varying severity and progressio

Background: Intrahepatic cholangiocarcinoma (ICC) is the second most common primary liver cancer. Macrophages are crucial components of the tumor microenvironment (TME). Triggering receptor expressed on myeloid cells 2-positive (TREM2+) macrophages have recently emerged as potent pro-tumor progress players in several cancers. However, the heterogeneity and tumor-promoting mechanisms of macrophages especially TREM2+ macrophages in ICC remain systematically unexplored. Therefore, this study aimed to systematically elucidate the heterogeneity of macrophages in ICC, with a specific focus on the pr

Helicobacter pylori is a highly adaptable bacterial pathogen that affects nearly half of the global population and plays a significant role in chronic gastritis, peptic ulcer disease, and gastric carcinoma. The immunokinetic profile of H. pylori infection is characterized by a paradox of cellular response that does not eliminate the bacterium, alongside inconsistent mucosal antibody protection. Reported studies generally demonstrate an early increase in mucosal interleukin-8 (IL-8), followed by increased T helper 1 cells (Th1)- and T helper 17 cells (Th17)-associated cytokine responses, includ

Alternative splicing and alternative promoters generate splicing variants to expand proteomic and transcriptional diversity to orchestrate physiological activities and dictate tumorigenesis. Splicing variants are attractive tumor-associated antigens (TAAs) for the clinical development of cancer therapies. Here, recent progress in claudin-18 isoform 2 (CLDN18.2)- and fibroblast growth factor receptor 2 isoform b (FGFR2b)-targeted therapeutics is summarized, and perspectives in this field are discussed. Zolbetuximab and bemarituzumab are first-generation monoclonal antibodies (mAbs) targeting CL
