Expanded proinflammatory CD4 T cells expressing toll like receptor 4 in trauma associated acute lung injury.

Acute lung injury (ALI) is characterised by hypoxia and inflammation that derives from diverse aetiologies. Delineating the molecular pathogeneses may identify treatable subgroups and inform clinical trials. Using a multi-omics approach, we analysed trauma-associated ALI (n = 9), COVID-19 ALI (n = 11), ICU controls (n = 13) using scRNA-seq, serum cytokine profiling, and bronchoalveolar lavage (BAL) microbiome analysis. We identified a population of activated CD4+ T cells expressing Toll-like receptor 4 (TLR4) and downstream NOD-, LRR- and pyrin domain-containing protein 3 (NLRP3) inflammasome

NIR-II-Responsive Carbon Dot/Bi2S3 Enables Mild Hyperthermia-Amplified Sonocatalytic Therapy Through Heterojunctions Engineering.

Sonodynamic therapy (SDT) demonstrates significant promise for noninvasive therapy of deep solid tumors such as cervical cancer, but its therapeutic efficacy is greatly restricted by low-efficiency sonosensitizers and the immunosuppressive tumor microenvironment (TME). Herein, we report the development of a novel Type-II carbon dot (CD)/Bi2S3 heterojunction for the mild NIR-II photothermal therapy (mPTT) amplified SDT/nanocatalytic therapy (NCT). The formation of heterojunctions drives the spatial separation of electron-hole pairs under ultrasound excitation, amplifying the generation of react

[The roles of TNFR2 signaling in tumors and advances in TNFR2 immunotherapy].

Tumor necrosis factor receptor 2 (TNFR2) is a type I transmembrane glycoprotein receptor expressed in various tumor cells and certain immune cells, particularly regulatory T cells. TNFR2 coordinates multiple signaling pathways to promote tumor initiation and growth, activate immunosuppressive cells, modulate the tumor microenvironment, and facilitate immune escape and tumor progression. In recent years, immunotherapy has been incorporated into the treatment of malignant tumors, yet monotherapy with immune checkpoint inhibitors often suffers from low response rates and frequent immune related a

Affinity and costimulatory architecture-associated toxicity in mesothelin-targeted TCR-mimetic CAR-T cells.

INTRODUCTION: TCR-mimetic CAR-T combines antibody-based targeting with physiological TCR signaling, yet suboptimal co-stimulation and safety risks remain key barriers for solid tumor therapy. The prevailing assumption that stronger co-stimulation yields superior efficacy has not been systematically tested in TCR-mimetic CAR-T architectures. METHODS: We developed a Mesothelin-targeted scFv-CD3ϵ TCR-mimetic CAR-T platform and systematically compared co-stimulatory modules-4-1BB, BRM-truncated 4-1BBΔBRM, and the 80BB chimera-alongside CD3ζ overexpression. To define the requirements for toxicity,

Mammalian peripheral nerve specific autoimmunity: proposed mechanisms and redefinition of molecular mimicry.

Molecular mimicry is considered essential for peripheral nerve-specific autoimmunity: a process by which antibodies, systemically generated primarily as a consequence of infection with bacteria or viruses possessing epitopes that are similarly expressed by peripheral axon complex gangliosides or peripheral nerve myelin proteins, are hypothesized to induce peripheral nerve injury in genetically susceptible individuals. Human in vitro studies have proposed that recently activated antigen-specific effector memory CD4+ T-cells initiate tissue specific autoimmunity by T-cell receptor-driven transmi

Tph and Tfh cells as biomarkers and targets in rheumatic diseases.

The role of B cell-helper T cells, including T follicular helper cells (Tfh), T peripheral helper cells (Tph), and others, in autoimmune rheumatic diseases is reviewed, focusing on clinical applications. A case is presented where the presence of B helper T cells could help decide the next medication to use in a patient with rheumatoid arthritis refractory to TNF inhibitors. The review covers topics including the biology of B cell and T cell interactions, the origin and function of B cell-helper T cells, disease associations, B cell-helper T cells as a marker of disease phenotype and treatment

INSR Armoring Protects CAR-NK and CAR-T Cells Against Tumor Stroma-Induced CAR Shedding by Maintaining Cholesterol Homeostasis.

While chimeric antigen receptor (CAR)-engineered T and NK cell therapies have revolutionized hematological malignancy treatment, translating this success to solid tumors remains a persistent challenge. Here, we demonstrated a stromal-associated metabolic mechanism in which cancer-associated fibroblasts (CAFs) and macrophage-associated populations in gastrointestinal cancers contribute to CAR loss through CYP27A1-derived 27-hydroxycholesterol (27HC). 27HC triggered membrane cholesterol depletion, destabilizing lipid rafts and promoting the release of full-length CAR in ARF6+CD81+ microvesicle-e

Adjuvant pegylated interferon-alpha after nucleos(t)ide analog withdrawal drives a temporally coordinated expansion of regulatory B cells and contraction of HBV-specific T cells.

BACKGROUND AND AIMS: The NUC-B study showed that a 4-month course of pegylated interferon-alpha (PEG-IFNα) enhanced the outcome and reduced hepatic flares following nucleos(t)ide analog (NUC) withdrawal in HBeAg-negative chronic hepatitis B. Here, we investigated differential T- and regulatory B-cell (BREG) changes before and during these interventions in a substudy of 41 representative patients. APPROACH AND RESULTS: Antigen-specific T cells were primarily evaluated using quality-controlled IFN-γ ELISpot with 15 pools of genotype-matched HBV core/small-surface peptides, confirmed by multipar

Influenza A virus infection induces immune dysregulation in the placenta and fetus without vertical transmission in nonhuman primates.

Influenza A virus (IAV) infection during pregnancy is associated with stillbirth and preterm birth. We hypothesized that maternal IAV infection disrupts placental and fetal immune networks in ways that correlate with disease severity. Pregnant pigtail macaques were inoculated with IAV [A/California/07/2009 (H1N1)] and underwent delivery and fetal necropsy 5 days postinfection (N = 11), with comparison to uninfected controls (N = 16). Stillbirth occurred in 18% of infected pregnancies and in no controls. Although vertical transmission was not observed, low levels of viral RNA were detected in t

SUMO2 deletion changes chromatin accessibility and enhances cytotoxic T cell activation and tumor infiltration.

Cytotoxic T cells [cytotoxic lymphocytes (CTLs)] are crucial for adaptive immunity leading to prolonged survival and potential cures for cancer. Recent clinical data have shown that pharmacological inhibition modifies the tumor microenvironment (TME) and activates CTLs, although the mechanism is not well described. In this study, we found that T cell-specific knockout (KO) of the most prevalent SUMO paralog, Sumo2/SUMO2, in both mouse and human CD8+ T cells significantly enhanced CD8+ T cell activation without enhanced IFN-I (type I interferon)-responsive genes in vitro but increased chromatin

Extracellular matrix-targeted TNC-scFv armoring enhances CAR T cell persistence and antitumor activity in solid tumors.

Chimeric antigen receptor T cell (CAR T cell) therapy has shown limited efficacy in solid tumors, largely due to physical and immunosuppressive barriers imposed by the tumor microenvironment (TME). Tenascin-C (TNC), an extracellular matrix protein highly expressed in multiple solid malignancies, contributes to immune exclusion and T cell dysfunction. Here, we engineer an armored CAR T cell platform that locally targets the extracellular matrix by secreting a TNC-specific single-chain variable fragment (TNC-scFv) linked to conventional CAR architecture. TNC-scFv-armored CAR T cells exhibit enha

Decoding the immune regulation of ovarian aging: mechanisms and therapeutic strategies.

BACKGROUND: Ovarian aging is among the earliest functional declines in humans and acts as a pacemaker of female aging. It encompasses both physiological age-related ovarian decline and pathological accelerated aging, exemplified by premature ovarian insufficiency (POI). Increasing evidence positions the immune system as both a regulator of ovarian homeostasis and a driver of ovarian dysfunction. Age-related immunosenescence and chronic low-grade inflammation (inflammaging) coordinately drive ovarian aging through bidirectional crosstalk between systemic immunity and the ovarian immune microenv

Gut microbial dysbiosis is associated with altered splenic immune homeostasis in chickens.

Gut microbiota plays a crucial role in animal health and immunity, and the broiler gut community is particularly sensitive during the early growing and finisher phases, when it is still maturing and immune development is active. In this study, we investigated the association between changes in the intestinal flora and splenic immune parameters in Qingyuan partridge chickens. A total of 40 twenty-week-old roosters were used. The results showed that antibiotic treatment (ABX) reduced the proportion (percentage) of splenic CD3+ T cells and increased the levels of inflammatory cytokines (IL-6, IL-

Multi-omics integrated analysis reveals the therapeutic mechanism of 919 formula in metabolic dysfunction-associated steatotic liver disease.

BACKGROUND: Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most prevalent chronic liver disease worldwide and is driven by metabolic stress-induced inflammation. Emerging evidence implicates cytotoxic CD8⁺ T cells as key contributors to the progression of MASLD. However, therapeutic strategies specifically targeting pathogenic CD8⁺ T cell responses in MASLD are still scarce. The clinical efficacy of the 919 formula in MASLD has been demonstrated, yet its cellular targets and molecular mechanisms remain unclear. METHODS: Single-cell transcriptomics profiled peripheral

First-line toripalimab in combination with platinum-doublet chemotherapy for advanced B3 thymoma and thymic carcinoma: an open-label prospective phase Ⅱ trial.

BACKGROUND: Several studies have demonstrated the efficacy of programmed cell death protein 1/programmed death-ligand 1 (PD-L1) inhibitors in patients with thymic epithelial tumors (TETs). Previous trials suggest that the efficacy of immunotherapy in TETs is associated with specific features of the tumor immune microenvironment. PATIENTS AND METHODS: Patients received a combination of paclitaxel, carboplatin, and toripalimab every 3 weeks for up to six cycles, followed by toripalimab maintenance every 4 weeks until disease progression or other discontinuation criteria. MAIT cells show promise as an immunotherapy tool with both adaptive and innate characteristics.

May unconventional MAIT cells be an effective innovative immunotherapy tool against liver cancer? This question is addressed by Yang and colleagues in this issue of Med.1 Their findings with adoptive transfer of expanded human MAIT cells to cancer xenograft mouse models suggest that these cells may hold greater promise in the cancer field than previously thought.

Lymphatic constraint of germinal centers optimizes protective antibody responses.

Immunization strategies are central to pathogen control, where efficacy relies on antigen distribution and inflammatory context. Lymphatic vessels are necessary conduits for antigen transport that actively remodel in peripheral tissues and draining lymph nodes (LNs) in response to their inflammatory context. In this study, we found that inhibiting viral-induced lymphatic remodeling in skin and LNs prompted germinal center (GC) expansion but, paradoxically, decreased the protective antibody response. While GCs scaled proportionately, larger GCs failed to support proliferative bursts consistent

Single-cell analyses of global cohorts outline determinants of immune aging and link the ratio of GZMK+ to GZMB+ Tem cells to health trajectories.

Human immune aging is heterogeneous, with immune responses becoming increasingly variable over the lifespan. Here, we integrated seven large-scale public single-cell peripheral blood mononuclear cell (PBMC) datasets and a newly generated cohort, encompassing 2,609 ostensibly healthy individuals across diverse ages, ancestries, and biological sexes. We identified both conserved and cohort-associated age-associated remodeling across 59 immune populations, together with sex- and ancestry-dependent immune differences. CD8+ T cells emerged as major drivers of aging heterogeneity, with the GZMK+/GZM
