CLAD: translating basic science to clinical practice.

Chronic lung allograft dysfunction (CLAD), a fibrotic manifestation of chronic rejection, remains the major barrier to long-term survival after lung transplantation. CLAD is increasingly recognised as the final common pathway of diverse alloimmune injuries, augmented by non-alloimmune insults, that culminate in irreversible airway and parenchymal remodelling. Recent advances in molecular profiling, single-cell technologies, and biomarker discovery, with corroboration by novel animal and in vitro models, are redefining CLAD pathogenesis. Emerging data implicate distinct pathways involving cytot

Association of donor single-cell cytokine secretion & circulating cytokine levels with the development & trajectory of severe primary graft dysfunction in lung transplantation.

Whether donor immune biology at procurement is associated with primary graft dysfunction (PGD) trajectory is unknown. Pre-transplant donor T-cell function and circulating plasma proteins were profiled to address this gap. Twenty-one recipients were classified as no PGD (NOGD, n=7), recovery after initial PGD3 (RECV, n=5), or sustained PGD3 (SGD, n=9). The polyfunctionality of T-cells isolated from donor peripheral blood mononuclear cell (PBMC) after stimulation and plasma levels of 92 proteins were assessed. CD8+ polyfunctionality strength index (PSI) was lowest in RECV (p=0.019), driven by ef

CXCR4-enriched B cells foster immunosuppression and gastric cancer progression in male mice via the adenosine-IL-10 axis.

BACKGROUND & AIMS: B cells are one major component of immune cells infiltrated in human solid tumors. However, the role of tumor-infiltrating B cells in mediating disease progression of human gastric cancer (GC) remains elusive. METHODS: Flow cytometry, immunohistochemistry and immunofluorescence were performed to analyze the prevalence, phenotypic and functional features of B cells in different samples of GC patients. B cells and T cells were purified and cultured ex vivo for regulation and functional assays. Tumor responses to B cell deficiency or adoptively transferred IL10 knockout B cell

Circular RNAs in Breast Cancer Immunometabolism: Mechanisms of Immune Regulation and Metabolic Reprogramming.

Metabolic reprogramming and immune dysfunction are increasingly recognized as interconnected hallmarks of breast cancer progression, collectively shaping tumor evolution, immune evasion, and therapeutic response. Emerging evidence identifies circular RNAs (circRNAs), a highly stable class of endogenous non-coding RNAs generated through back-splicing, as pivotal regulators of the immunometabolic crosstalk within the breast cancer tumor microenvironment (TME). Through diverse mechanisms, including microRNA sequestration, interactions with RNA-binding proteins, and modulation of transcriptional a

Leucine-derived isovaleric acid remodels the tumor microenvironment by degrading EIF3I.

Metabolic reprogramming and immune evasion are hallmarks of cancer. Although leucine is well known for regulating protein synthesis, its role in immunometabolic interactions within the tumor microenvironment (TME) remains unclear. In this study, we found that leucine supplementation significantly inhibited tumor growth. Metabolomic analysis revealed that isovaleric acid (IVA) was the main differential metabolite produced during leucine degradation by the gut microbiota. Mechanistically, IVA directly binds to EIF3I at lysine 17 (K17), increasing its ubiquitination and promoting its proteasomal

Effects of Saccharomyces cerevisiae boulardii CNCM I-1079 on immune phenotype during the periparturient period.

This study aims to characterize systemic and colonic intestinal mucosal immune phenotype in dairy cows across the periparturient period, and to determine whether Saccharomyces cerevisiae boulardii CNCM I-1079 (SCB) supplementation affects those components. Holstein cows (n = 83) were blocked by parity and randomly assigned to either control (CON; n = 43) or SCB (n = 40; targeted supplementation: 1.0 × 1010 cfu/cow/d of SCB top dressed) dietary treatments from d -28 prepartum to d 70 postpartum. Blood samples were collected on d -30, -7, 7, 21 and 70 relative to calving and colon biopsies at d

The conformation-selective PI3Kδ inhibitor roginolisib in patients with advanced or metastatic cancer, including metastatic uveal melanoma: first-in-human clinical trial.

Phosphatidylinositol 3-kinase delta (PI3Kδ) promotes tumour cell growth directly or indirectly by activating immune-suppressive cells. In contrast to previous PI3Kδ inhibitors, roginolisib is a selective, non-ATP competitive inhibitor, which locks PI3Kδ into an inactive state and causes an allosteric modulation. In a First-in-Human dose study, continuous daily dosing of roginolisib was investigated in patients with solid and haematologic malignancies. In Part A, 24 patients received escalating doses of roginolisib. In Part B, 20 mostly pre-treated metastatic uveal melanoma (mUM) patients recei

Microbial cues and T cell immunity: deciphering their interactions in chronic liver disease.

Chronic liver disease (CLD) poses a major global health challenge, as its progression to cirrhosis and hepatocellular carcinoma significantly contributes to worldwide morbidity and mortality. Mounting evidence has identified the gut microbiota as a critical modulator of CLD progression, primarily via the gut-liver axis. The liver is continuously exposed to gut-derived signals through the portal circulation, while reciprocally secreting bioactive mediators that shape intestinal microbial composition and function. This bidirectional network enables intricate crosstalk between the gut microbiota

Mitochondria transfer via tunneling nanotubes drives macrophage immunosuppression and metabolic reprogramming in pediatric AML.

Although immunotherapy has revolutionized cancer treatment, its benefits in pediatric acute myeloid leukemia (AML) remain limited, partly due to an incomplete understanding of the AML microenvironment. While most studies focus on T cells, innate immune populations such as macrophages are less well explored. In pediatric AML patients, we demonstrate that immunosuppressive macrophages dominate the AML microenvironment and support growth of RUNX1::RUNX1T1 and KMT2A-rearranged leukemic blasts. In co-culture experiments, AML cells reprogrammed naïve and pro-inflammatory macrophages toward an anti-i

LAG3 checkpoint function and its mechanism of action in therapeutic targeting.

Lymphocyte activation gene 3 (LAG3) protein is a key immune inhibitory checkpoint receptor that suppresses T cell activation, proliferation and effector functions to maintain immune homeostasis and tolerance. In persistent pathological settings, chronic T cell receptor (TCR) signalling, together with sustained LAG3 signalling and other inhibitory receptor signalling, drives T cell dysfunction, highlighting LAG3 as a promising therapeutic target. However, more than 30 years after its discovery, the fundamental mechanisms governing LAG3 signalling and function remain incompletely defined. Elucid

Lymph node contraction links sex-biased naive CD8+ T cell decline to compromised antigen recognition during middle age.

The abundance of diverse naive CD8+ T cell clones is essential for broad protection against infection and cancer, but how sex and aging jointly shape this compartment remains poorly understood. Here, using mouse models with supporting human data, we uncover a sex-biased mechanism of immune aging in which early male-biased depletion of naive CD8+ T cells driven by accelerated, antigen-agnostic differentiation into virtual memory cells combines with thymic involution, limiting naive CD8+ T cell replenishment. These mechanisms led to more rapid lymph node contraction and reduced local naive T cel

Understanding the Immune Response Changes to Clinical Interventions for Epstein-Barr Virus Infection Prior to Lymphoma Development in Children After Organ Transplants (UNEARTH).

BACKGROUND: Post-transplant lymphoproliferative disorders (PTLD) remain a major complication of solid organ transplants (SOT), occurring in 1%-10% of patients, but in greater proportions in children. About 50%-80% of cases are Epstein-Barr Virus (EBV) associated, through partially known and still unknown mechanisms. Even in EBV seronaive SOT recipients, transplant of an SOT from an EBV seropositive donor does not always lead to peripheral blood EBV replication. Longitudinal peripheral blood EBV DNA nucleic acid testing (NAT) after SOT has not improved the individual prediction of PTLD occurren

Intramuscular Non-replicating Tiantan Vaccinia Drives Superior Antibody Maturation and Sterilizing Protection Against Lethal Orthopoxvirus Challenge in Mice.

The global resurgence of orthopoxviruses, particularly MPXV, necessitates safe yet highly protective vaccines. Here, we systematically evaluated a non-replicating Tiantan vaccinia virus (NTV)-derived from the historically efficacious vaccinia Tiantan strain-across intramuscular (IM), subcutaneous (SC), and dose-sparing intradermal (ID) routes in lethal murine models. While all regimens elicited robust polyfunctional T cells, cross-neutralizing antibodies against MPXV, VTT, and ECTV, and protective immunity against standard lethal VACV-WR and ECTV challenges, under extreme viral exposure (70 LD

Hypoxia during GPC2-CAR T-cell manufacturing compromises cell expansion and cytotoxic capacity.

BACKGROUND AND AIM: Hypoxia is a hallmark of the solid tumor microenvironment and is known to impair CAR T-cell function. In order to condition CAR T-cells to this microenvironment during manufacturing, we evaluated whether varying oxygen tension during the manufacturing of GPC2-CAR T-cells improves cellular characteristics and function against neuroblastoma. METHODS: Cell products were produced over 8-9 days under normoxic (21% oxygen), transitional (21%, then transitioned to 1% on Day 2), or maintained hypoxic conditions (1% from Day 0 onward). RNA-sequencing, metabolic assays, phenotyping,

Immune responses induced by a subunit vaccine against Mycoplasma gallisepticum.

Mycoplasma gallisepticum (MG) is an avian respiratory pathogen responsible for global economic loss to the poultry industry. We previously developed an efficacious subunit vaccine to address the drawbacks of current bacterin and live-attenuated vaccine designs. To improve our understanding of how the subunit vaccine works, we sought to understand if the vaccine influences the biology of the pathogen, whether the subunit vaccine has correlates of protection when analyzing antibody responses or tracheal-associated cells, and whether each component of the subunit vaccine contributes to protection

Lung Resident T Cells Across the Spectrum of TB Infection and Disease.

Tuberculosis (TB) remains the leading infectious cause of death worldwide, yet immunological protection against Mycobacterium tuberculosis (Mtb) remain poorly understood. We performed integrated single-cell transcriptomics and functional T-cell cloning on paired bronchoalveolar lavage (BAL) and peripheral blood samples from those exposed to TB, using IGRA and PET-CT to study potentially protective responses across the spectrum of TB infection/disease. Recent Mtb exposure alone was sufficient to remodel the pulmonary T-cell compartment. Longitudinal follow-up demonstrated that all participants

A trivalent gastric cancer antigen conjugated to a diphtheria toxin epitope synergizes with an NKT cell agonist to boost antitumor immunity.

The gastric cancer-associated antigen MG7-Ag is aberrantly expressed in multiple gastrointestinal malignancies and represents a promising broad-spectrum target for pan-gastrointestinal tumor peptide vaccines; however, its clinical application is hampered by low immunogenicity. To overcome this limitation, we covalently conjugated a diphtheria toxin-derived T-helper epitope to the trivalent antigen MG3, which was constructed by tandem repetition of the monovalent MG1 peptide from the MG7-Ag. A liposomal vaccine was subsequently developed by incorporating DT-MG3 together with the natural killer
