NKG2A inhibition promotes NK cell-CD8+ T cell interactions to improve anticancer immunity in ovarian carcinoma.

Natural killer (NK) cells contribute to tumor immunosurveillance, yet their heterogeneity across cancer types remains incompletely understood. Transcriptomic, spatial, and functional assays reveal that non-small cell lung carcinoma (NSCLC) is enriched in NK cells that mediate clinically relevant effector functions, whereas high-grade serous ovarian carcinoma (HGSOC) contains dysfunctional NK cells that express co-inhibitory receptors including NKG2A. Analysis of HGSOC patient samples and syngeneic mouse models indicates a crosstalk between NK cells and CD8⁺ T cells critical for effective antit

Homotypic interactions between SLAMF1 receptors on innate T cells and neutrophils regulate killing of fungi.

Innate lymphocytes and myeloid cells communicate and play an essential part in activating neutrophils and other effector cells to kill fungi. Here, we identified that Signaling Lymphocytic Activation Molecule 1 (SLAMF1) orchestrates a cellular and molecular signaling network that activates phagocytes. We uncovered innate lymphocytes including innate CD4+ or TCRγδ+ T cells augment neutrophil killing of Blastomyces dermatitidis (Bd) in a SLAMF1 dependent manner. SLAMF1 expression on neutrophils enabled homotypic SLAMF1:SLAMF1 interactions with innate CD4+ T cells, which released soluble factors

Combination adjuvants drive long-lived plastic Th17 cells that convert to multi-functional Th1 cells and protect mice against fungal infection.

Th1 cells are viewed as a cornerstone of immunity to fungi and other intracellular pathogens. Despite the widely accepted role of Th1 cells in antifungal resistance, the development of protective strategies harnessing them is stunted by a limited understanding of how best to promote their development. We and others have reported a requisite role for Th17 cells in resistance to fungi. We have long been puzzled about how to reconcile seminal roles for both Th1 and Th17 subsets. Here, we report that Th17 cells convert into polyfunctional Th1 cells producing multiple cytokines, including IFN-γ, TN

Clinical and immunological features of a patient exhibiting delayed puberty, microcephaly, scoliosis and epilepsy caused by a novel mutation in IGSF10.

Members of the immunoglobulin superfamily (IgSF) are cell-surface proteins essential for a wide range of biological processes, including molecular transport, morphoregulation, and cell differentiation. IGSF10, a member of this family, has been identified as a disease-associated gene in delayed puberty and cleidocranial dysplasia. In this study, a Chinese patient with delayed puberty, microcephaly, scoliosis, and epilepsy was investigated. The genetic defect and immune alterations were assessed using whole-exome sequencing, Sanger sequencing, and single-cell RNA sequencing. After variant filter

Prognostic Value of Aspergillus-Specific T Cells in Invasive Pulmonary Aspergillosis Among Patients with Hematologic Malignancies.

In the OPTIFIL study that included 39 patients with localized invasive pulmonary aspergillosis complicating hematological malignancies, Aspergillus-specific IFNγ-producing T cells were detectable in 41% of the patients and predicted better 6-weeks outcomes, while their absence correlated with uncontrolled disease. Lymphopenia was the main limitation to Aspergillus ELISpot assay interpretation.

Analysis of CD4+ and CD8+ T Lymphocytes and Expression of Immune Checkpoint Proteins (PD-1 and PD-L1) in Primary and Persistent Periapical Lesions.

INTRODUCTION: Persistent periapical lesions (PePL) represent a clinical challenge because of their inadequate response to conventional endodontic therapies. Studies indicate differences in immunoinflammatory mechanisms between primary periapical lesions (PrPL) and PePL but the findings remain conflicting. This study aimed to evaluate CD4+ and CD8+ T lymphocytes and the expression of immune checkpoint proteins (programmed cell death protein 1 [PD-1] and its ligand [PD-L1]) in PrPL and PePL, correlating the findings with the intensity of the inflammatory infiltrate and pattern of the cystic epit

Checkpoint blockade-related CNS adverse events are characterized by neurotoxic T helper cells with sustained neuroantigen reactivity.

The immune-related adverse events of central nervous system (irAEs-CNS) are rare yet life-threatening complications arising from immune checkpoint inhibitor therapies, with no effective treatments. The intracranial immune landscape in irAE-CNS remains poorly characterized. Through longitudinal multi-omics profiling of cerebrospinal fluid from three independent clinical cohorts-combined with serial cerebral positron emission tomography/computed tomography (PET/CT) imaging, neurofunctional assessments, and 15-month clinical follow-up, we identify a defined proinflammatory memory T helper cell po

Immunoinflammatory cells and Beta-defensin-3 in radicular cysts of primary and permanent teeth.

The expression of hBD-3 and quantification of immune and inflammatory response cells (non-degranulated and degranulated mast cells, mature and immature plasmacytoid dendritic cells, mature and immature Tregs, T lymphocytes, cytotoxic T lymphocytes, and B lymphocytes) were evaluated in the radicular cysts (epithelium/capsule) of primary and permanent teeth. The relationship between the size of the radiographic lesion and expression of hBD-3 was also evaluated. Radicular cysts were subjected to immunohistochemical analysis to quantify the immune and inflammatory response cells and to evaluate hB

Tempol Protects Against Radiation Injury in the Submandibular Gland Through the β-Catenin Signaling Pathway.

BACKGROUND: Radiation damage severely impacts salivary gland function and cell survival, especially in head and neck radiotherapy. Tempol (TPL), a free radical scavenger, has shown protective effects against radiation damage. This research aimed to investigate the protective effects of TPL on radiation-induced damage in Hs917.T cells and the submandibular gland (SMG) of C57BL/6 mice, along with the mechanisms involved. METHODS: Human parotid fibroblasts (Hs917.T) were pre-treated with TPL and exposed to ionizing radiation (IR). Protective effects were evaluated using MTT, clonogenic survival

Exploring dysregulation of cuproptosis-related genes molecular clusters and candidate biomarkers in pterygium.

Pterygium is a common ocular surface disorder, with its prevalence strongly correlated to ultraviolet (UV) exposure in geographic regions. Epidemiological investigations reveal significant demographic variations, with higher incidences observed in areas with intense UV radiation and within specific populations, notably rural individuals. Despite surgical interventions being the standard treatment, recurrent cases underline the necessity for understanding the underlying biological mechanisms contributing to pterygium pathogenesis. Recent advancements in cellular death mechanisms point to cuprop

LOX-1 mediated lipid peroxidation leads to dysfunction of tumor infiltrating CD8+ T cells in ovarian cancer.

Abnormal lipid accumulation in the tumor microenvironment (TME) promotes lipid peroxidation in tumor-infiltrating CD8+T cells (CD8+TILs), driving their dysfunction and impairing antitumor immunity in ovarian cancer (OC). To elucidate the underlying immunosuppressive mechanisms, we integrated clinical specimens, mouse models, and in-vitro validation. Notably, CD8+TILs from OC patients exhibited pronounced lipid peroxidation signatures. Elevated oxidized low-density lipoprotein (oxLDL) levels were detected in ovarian tumor interstitial fluid (TIF), tumor mass-derived supernatants, and tissue sec

Elevated RIPK2 Expression in Atopic Dermatitis Skin Is Associated with Mixed Th1/Th2 Cytokine Responses in CD4+ T Cells.

Receptor-interacting protein kinase 2 (RIPK2) is a critical mediator of the immune system leading to production of pro-inflammatory cytokines. Its role in the pathogenesis of atopic dermatitis (AD) has not been defined. This work aims to assess RIPK2 expression in the skin in AD and to evaluate the association with Th1-, Th2-, and Th17-related cytokine expression in lesional skin. AD patients' skin biopsies and healthy controls were collected. An OPAL multiplex immunohistochemistry method was used to simultaneously detect TNF-α, IL-4, IL-13 and IL-17 in CD4+ RIPK2+ and CD4+ RIPK2- T cells. Qua

Immunological Interplay at the Ocular Surface of Stevens-Johnson Syndrome, Ocular Cicatricial Pemphigoid, and Ocular Graft Versus Host Disease.

Rare ocular surface inflammatory disorders (OSIDs), such as ocular cicatricial pemphigoid (OCP; 1.3-2/million), Stevens-Johnson Syndrome (SJS; 1-5/million), and ocular graft versus host disease (oGVHD; occurring in >50% of chronic GVHD cases) share overlapping clinical phenotypes. Their defining features include conjunctival cicatrization and chronic inflammation. An incomplete understanding of the underlying immunopathogenesis of these conditions has hindered the development of targeted therapies, leaving patients at risk of permanent vision loss and highlighting the need to define distinct o

The neurotoxic legacy of CAR-T cells: where do we stand?

Chimeric antigen receptor T (CAR-T) cell therapy has transformed outcomes for relapsed/refractory B-cell malignancies and is increasingly reshaping the therapeutic landscape of autoimmune disorders and solid tumors, offering curative potential where options were previously limited. Its broader deployment is, however, constrained by immune-mediated toxicities, chiefly cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS). ICANS spans a heterogeneous spectrum from mild aphasia and tremor to seizures, cerebral edema, coma, and death, and remains diffic

Exploratory comparison of immune cell compositions across organs in IgG4-related disease.

OBJECTIVES: IgG4-related disease (IgG4-RD) is a systemic fibroinflammatory disorder affecting multiple organs, but the extent to which immune microenvironments differ across tissues remains unclear. We compared immune cell compositions between submandibular gland and pancreatic lesions using transcriptome-based deconvolution analysis. METHODS: Bulk RNA sequencing data were obtained from patients with IgG4-related submandibular sialadenitis (n = 40) and autoimmune pancreatitis (n = 8). Immune cell fractions were estimated using CIBERSORTx with the LM22 immune signature matrix. Relative immune
