Recombinant PsoSP4 confers superior 80% protection against psoroptic mange in rabbits in association with a Th1-biased immune response.

Psoroptic mange, caused by Psoroptes ovis, is a severe global ectoparasitic disease for which no commercial vaccine is available. Here, we evaluated four recombinant P. ovis proteins (rPsoSP4, rPsoPRF, rPsoMIF, and rPsoSP5) as subunit vaccine candidates in a rabbit model. Three antigens conferred significant and graded protection, with final protection rates of 80% (rPsoSP4), 60% (rPsoPRF) and 20% (rPsoMIF), compared to 0% in the control and rPsoSP5-vaccinated groups. The lead candidate, rPsoSP4, reduced ear lesion scores, crust weights, and mite burden by up to 88.57%, 88.30%, and 81.80%, res

A prime-and-trap vaccination strategy to evaluate minimal requirements for malaria liver stage protection in mice.

BACKGROUND: The Plasmodium liver stage is an attractive prophylactic vaccine target since blocking parasites in the liver has the potential to prevent both clinical disease and transmission. However, immunity to Plasmodium liver stages is difficult to study, and highly efficacious liver stage vaccines have been difficult to develop. Prime-and-trap vaccination induces liver CD8+ T cells that can provide complete liver stage protection in mice. However, the minimal vaccine requirements for complete liver stage protection remain unclear. METHODS: In this study, a prime-and-trap vaccination strat

The LEF1-LAG3 axis regulates CD4+ T cell function during Plasmodium yoelii NSM infection.

BACKGROUND: CD4⁺ T cells are pivotal in coordinating anti-malarial immunity, while co-inhibitory receptors such as LAG3 critically regulate their function. However, the phenotype of LAG3⁺CD4⁺ T cells during Plasmodium infection and the upstream molecular mechanisms regulating LAG3 expression remain incompletely elucidated. METHODS: We established a murine model using Plasmodium yoelii NSM (P. yoelii NSM). A multifaceted approach, incorporating single-cell RNA sequencing (scRNA-seq), flow cytometry, magnetic bead-based cell sorting, real-time quantitative polymerase chain reaction (RT-qPCR), d

Exploring Autologous Dendritic Cells for T Cell Modulation: A Step Towards Personalized Medicine in Leishmaniasis.

Leishmaniasis caused by Leishmania infantum is a zoonotic disease endemic in many regions worldwide. The antigen-presenting dendritic cells (DCs) bridge the innate and adaptive immune response by activating T lymphocytes. Therefore, the present study examines whether T lymphocyte activation can be directed by autologous DCs primed by extracellular vesicles (EVs) derived from L. infantum. For this, lymphocytes were co-cultured with monocyte-derived DCs (moDCs) that were primed by EVs. moDC signaling and activation were examined by gene expression of toll-like receptors and cytokines. The antige

Comprehensive Immunophenotyping of Monocytes and Dendritic Cells Suggests Distinct Pathophysiology in Chronic Fatigue Syndrome and Long COVID.

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) and long Coronavirus Disease 2019 (long COVID) are complex chronic conditions that often follow infectious triggers with overlapping clinical features but poorly defined pathophysiological relationships. This study aimed to identify disease-specific immune signatures through multiparameter immunophenotyping of monocytes, dendritic cells, and T cell subsets. A total of 207 participants were included (ME/CFS: n = 103; long COVID: n = 63; healthy controls: n = 41). Peripheral blood mononuclear cells were analyzed using multiparameter flo

Early Regulatory and Th2-Associated Responses Shape Resistance to Leishmania panamensis Infection in C57BL/6 Mice.

Characterizing the specific interactions of Leishmania species with different host systems is essential for the development and validation of experimental infection models and for identifying potential therapeutic targets. Leishmania parasites elicit diverse host immune responses that result in different levels of disease severity. Here, we developed a murine model of L. panamensis infection and compared the responses of BALB/c and C57BL/6 mice following intradermal ear inoculation. BALB/c mice developed progressive ulcerative lesions associated with high parasite burden, whereas C57BL/6 mice

CD73 activity controls cytotoxic CD4 T-cell response driving myocardial pathology in chronic Chagas disease.

Chagas disease, caused by Trypanosoma cruzi, is the major cause of infectious cardiopathology worldwide. Although cytotoxic CD4 T-cells (CD4 CTLs) have recently been recognized as crucial effectors in infections and inflammation, the mechanisms that control their differentiation and impact on pathological outcomes remain largely undefined. Here, we demonstrate that the ectonucleotidase CD73, which generates adenosine from extracellular AMP, acts as a key immunoregulator of CD4 CTL response during T. cruzi infection. Using murine models, we found that infection induced a robust expansion of CD4

Integrating network pharmacology and experimental validation to uncover the therapeutic mechanisms of Chaigui decoction in Schistosoma japonicum-induced liver fibrosis.

This study investigated the therapeutic effects and underlying mechanisms of Chaigui decoction (CGD) against hepatic fibrosis induced by Schistosoma japonicum. Utilizing a network pharmacology approach, 113 common targets were identified by intersecting CGD-associated genes with fibrosis-related databases, with subsequent enrichment analyses revealing 435 Gene Ontology terms and 197 Kyoto Encyclopedia of Genes and Genomes pathways. In vitro, serum containing CGD significantly attenuated the pro-fibrotic phenotype induced by transforming growth factor-beta 1 (TGF-β1) in hepatic stellate cells.

Schistosomal extracellular vesicles: Mechanisms of immune modulation and emerging clinical applications.

Schistosomiasis remains a persistent global health challenge and a leading cause of parasitic morbidity worldwide. The complex, enduring relationship between Schistosoma parasites and their human hosts is underpinned by a sophisticated molecular dialogue, much of which is mediated by extracellular vesicles (EVs). These parasite-derived vesicles, which carry proteins, lipids, and nucleic acids, act as a double-edged sword in host-parasite interactions. On one hand, they facilitate immune evasion by promoting Th2 polarization, expanding regulatory T cells, and driving alternative macrophage acti

Pan-pathogen vaccine approaches: Toward broad-spectrum immunity in a One Health era.

Emerging and re-emerging infectious diseases (EIDs) represent an escalating threat to global public health, as exemplified by outbreaks of COVID-19, Ebola, Zika, and other zoonotic viruses. Traditional pathogen-specific vaccines, although effective for individual diseases, face significant limitations in addressing EIDs due to long development timelines, resource intensity, and restricted adaptability to novel pathogens or variants. Pan-pathogen vaccines-designed to provide broad-spectrum immunity across multiple related or unrelated pathogens-offer a transformative approach to pandemic prepar

Prophylactic Administration of Gypsophila oldhamiana Extract Restricts Acute Toxoplasma gondii Infection via the DC-IL-12-CD8⁺ T Cell Axis in a Murine Model.

INTRODUCTION: Toxoplasma gondii (T. gondii) is a globally prevalent parasite that poses significant medical and veterinary challenges. Although current therapies such as pyrimethamine and sulfadiazine are effective, they often cause severe adverse effects, including myelosuppression, highlighting the need for safer and more effective alternatives. In this study, we evaluated the immunostimulatory and antiparasitic activities of three traditional herbal extracts, Stellaria dichotoma L. var. lanceolata Bge. (LDS), Stellaria aquaatic (SA) and Gypsophila oldhamiana (GO), in a murine model of T. go

Identification of a persistent Ascaris-derived Kalirin epitope associated with chronic T cell activation in the lung.

Ascariasis remains a dominant global health burden due to its vast prevalence and associated morbidity. The obligatory migration of Ascaris larvae through pulmonary tissue triggers intense type-2 inflammation which typically presents as acute allergic airway disease. Even after the parasite is eliminated, a single episode of larval migration can result in chronic lung damage and dysfunction, which may be driven by the long-term retention of helminth antigens in macrophages. However, the molecular identity of these retained antigens, and the mechanisms by which they sustain chronic T cell respo

β-catenin-driven innate and metabolic reprograming in macrophages fuel T-cell-dependent inflammation in Toxoplasma gondii infection: implications for therapeutic intervention.

Toxoplasma gondii activates innate immunity via TLR11/12 in mice, but the lack of functional human counterparts leaves a gap in understanding parasite sensing in humans. Here, we bridge this gap by uncovering a host-intrinsic sensing mechanism, wherein β-catenin signaling mediates immune recognition of T. gondii. Notably, this parasite hijacks the PI3K-AKT-β-catenin pathway in macrophages to promote its replication. While β-catenin ablation, either genetically or pharmacologically (XAV939), disavows this process, thereby inhibiting replication. Phospho-β-catenin-TCF4 drives IRF4 transcription,

In vivo reduction of Treg expansion in rodent helminth-malaria coinfection.

BACKGROUND: Parasitic helminths modulate host immunity to establish chronic infections, often expanding regulatory T cells (Tregs) to suppress inflammation. However, how helminths adjust immunomodulation during coinfection with fast-replicating microparasites remains unclear. Here, we investigated the immune modulation dynamics in a murine model coinfected with the nematode Heligmosomoides polygyrus (Hp) and the protozoan Plasmodium yoelii (Py). METHODS: We compared Treg expansion in single Hp infected and coinfected mice, assessed Hp excretory-secretory products (HES) for Treg induction in v

ST2 blockade modulates IL-33-driven immune responses and is associated with reduced hepatic fibrosis in alveolar echinococcosis.

BACKGROUND: Infection with Echinococcus multilocularis causes alveolar echinococcosis (AE), a severe parasitic disease characterised by progressive hepatic infiltration and fibrosis. Current treatment options remain limited, and the immune-mediated mechanisms underlying fibrosis are not fully understood. This study investigated the role of the IL-33/ST2 axis in AE-associated immunopathology and evaluated the effects of ST2 blockade in an experimental model. METHODS: A murine model of AE was established in C57BL/6 mice, which were allocated to untreated infected, anti-ST2-treated infected and

Host immune responses and immune evasion mechanisms in Trypanosoma cruzi: implications for the immunopathogenesis of Chagas disease.

Chagas disease remains a major neglected tropical disease in Latin America and represents a significant public health challenge in Brazil. The progression of Trypanosoma cruzi infection is highly heterogeneous and reflects a complex interaction between parasite persistence and host immune responses. This narrative review summarizes current knowledge on host immune responses and immune evasion mechanisms during T. cruzi infection and discusses their implications for the immunopathogenesis and clinical progression of Chagas disease. The review included publications indexed in major biomedical da

Acriflavine attenuates inflammatory pathology in cutaneous leishmaniasis independently of parasite control.

Cutaneous leishmaniasis is a parasitic skin disease for which current treatments often fail, highlighting the need for new therapeutic approaches. While high parasite burdens are associated with delayed healing, excessive protective immune responses, including elevated IFN-γ production, have also been linked to worse clinical outcomes, indicating that immunopathology contributes significantly to disease progression. Acriflavine is an antimicrobial compound with reported anti-leishmanial activity and the ability to inhibit hypoxia-driven responses. Because Leishmania -infected skin is hypoxic i

Restoring balance in atopic disorders: insights into type 2 immunity and chronic inflammation.

Atopic diseases arise from an immunological imbalance where regulatory mechanisms are unable to preserve or restore homeostasis, leading to chronic inflammatory conditions affecting epithelial organs. This may involve homeostasis, deficient or insufficient regulatory T cells (Tregs) or other aberrant regulatory mechanisms. Type 2 (T2) immunity is a conserved response that evolved to combat large helminth parasites (worms), venoms, and toxins involving both innate and adaptive immune pathways. Many T2 cytokines and alarmins act to recruit and activate innate and adaptive immune cells, and they

Memory regulatory T cells reprogram into protective TFH cell-like effectors in recurrent malaria.

Although people living in malaria-endemic areas experience repeated infections with Plasmodium, the role of regulatory T (Treg) cells in recurrent malaria remains poorly understood. During a primary infection with Plasmodium, Treg cells suppress protective immunity by inhibiting germinal center (GC) reactions, thereby impeding the control of parasitemia. In contrast, memory Treg (mTreg) cells remaining after the clearance of initial Plasmodium infection acquire protective functions after recall. Longitudinal studies in humans and mice show that mTreg cells undergo antigen-driven expansion and

T-Bet Modulates Plasmodium-Specific CD4+ T Cell Differentiation and Anti-Malarial Immunity During Blood-Stage Infection.

CD4+ T helper (Th)1 cells are theorized to control Plasmodium parasite burden during blood-stage malaria. However, their exact role in controlling parasitemia in vivo remains inadequately characterized. Here, we used a CD4+ T cell-restricted Cre-Lox T-bet (the master regulator of Th1 cells) excision mouse model to examine the role of Plasmodium-specific Th1 cell responses in controlling blood-stage infection. Our results found that T-bet deficiency in CD4+ T cells markedly enhanced the growth of blood-stage Plasmodium yoelii (P. yoelii) 17XNL and Plasmodium chabaudi (P. chabaudi) AS, but not P
