Toxoplasma gondii is a widespread intracellular parasite that establishes chronic infection through the persistence of tissue cysts, for which no effective treatment or licensed vaccines for humans are currently available. Preventive vaccination of animal hosts represents a promising strategy to reduce parasite transmission and disease burden. In this study, we evaluated the immunogenicity and protective efficacy of recombinant subunit vaccines based on T. gondii GRA7, ROP2, or profilin (TgPF), formulated with the Immune-Stimulating Particle Adjuvant (ISPA), a saponin-based particulate adjuvan

Hookworms migrate through the lung as part of their lifecycle, causing significant tissue damage that activates tissue-resident populations such as ILC2s. These drive type 2 immune responses critical for wound healing and the development of protective immunity. Monocyte-derived macrophages that seed the lung after infection are central effectors of these responses, but the cellular and molecular factors guiding their development and function remain poorly understood. We find that ILC2s and IL-4/IL-13-producing CD4+ T cells regulate alveolar macrophage populations during helminth infection. In

BACKGROUND: Toxoplasmosis is a prevalent zoonotic disease worldwide, affecting approximately one-third of the global human population. Primary infection with Toxoplasma gondii during pregnancy can induce miscarriage or congenital infection, leading to irreversible damage to the foetus. Moreover, reactivation of T. gondii infection in immunosuppressed individuals can result in fatal outcomes. No vaccine exists to prevent human disease caused by this parasite. Thus, a vaccine that could induce complete and lasting protection against human toxoplasmosis is an unmet need.
METHOD: In this work, BA

A major limitation on the development of a malaria vaccine is the lack of validated T cell epitope targets. Plasmodium falciparum is the most prevalent malaria parasite affecting humans in Africa, whereas Plasmodium vivax is more widespread and is the main species that causes malaria in the Americas and Asia1. P. vivax exclusively infects peripheral-blood reticulocytes, which retain RNA and the capacity for host protein synthesis2. We previously reported that reticulocytes infected with P. vivax express human leukocyte antigen class I (HLA-I), which enables recognition and killing of the paras

γδT cells are a subset of innate lymphocytes that play an essential role in anti-infection immunity. However, the functional role of CD8αβ+ γδT cells, a distinct subset of γδT cells, remains poorly characterized during malaria infection. Female C57BL/6 mice were intraperitoneally injected with 1 × 106 Plasmodium yoelii-infected red blood cells (iRBCs). At 12 days post-infection (12 dpi, the peak of parasitemia), spleens were harvested and lymphocytes were isolated. Fluorescence-activated cell sorting (FACS) was performed to determine the frequency, phenotypic features and functional properties

BACKGROUND: Experimental infection with Plasmodium falciparum in malaria-naive European adults induces CD25+FoxP3+ regulatory CD4+ T cell (Treg) expansion. Increased Treg activity is associated with reduced inflammatory cytokine responses and increased parasite growth. Whether similar associations occur in semi-immune adults from malaria-endemic regions remains unclear. In this study, we aimed to examine how inflammatory cytokines, regulatory cytokines, and Tregs influence parasitic and clinical outcomes during controlled human malaria infection in semi-immune adults from three regions in Keny

HLA class I presentation of pathogen-derived peptides is essential for CD8+ T-cell recognition of Toxoplasma gondii. While in vitro MHC ligands have been documented, the in vivo ligandome during infection progression remains poorly characterized. Here, we employed an MS-based immunopeptidomics approach to directly profile the T. gondii immunopeptidome presented by HLA-A *02:01 in transgenic mice. By employing a hierarchical discovery funnel, our analysis identified a comprehensive repertoire of 3,744 unique T. gondii-derived peptides. Subsequent filtering for canonical 8-12mers, matching the t

Infection with Plasmodium falciparum or Plasmodium vivax during pregnancy can result in adverse outcomes for both mother and child. Submicroscopic infections are common in malaria-endemic areas but often remain chronic, asymptomatic, and untreated. Chronic infections persistently stimulate the immune system, altering expression of immune mediators and promoting expansion of regulatory T cells. These immune changes could compromise responses to routine vaccinations. This study evaluated the immunomodulatory effects of submicroscopic infection by P. falciparum and P. vivax during pregnancy and t

UNLABELLED: Chronic infections require mechanisms that limit tissue damage while preserving pathogen control, yet the contribution of regulatory T (Treg) cells to this balance remains unclear. In this study, we characterized Treg cell responses during a chronic parasitic infection using experimental Trypanosoma cruzi infection as a model of persistent low-level parasitism and chronic tissue inflammation. We found that, although Treg cell numbers decline in the spleen, they accumulate in parasite-affected tissues such as skeletal muscle, where they adopt a combined Th1-associated and tissue-rep

Schistosomiasis, caused by parasitic worms, affects over 250 million people globally, with limited treatment options due to praziquantel's inability to prevent reinfection or reduce immunopathology. In Schistosoma mansoni (S. mansoni) infection, egg-induced granulomatous inflammation in the liver and intestines is driven by CD4 T helper (Th) cell responses, with severe pathology in some individuals mediated by Th17 cell activation. We previously demonstrated that the stimulator of interferon genes (STING) mitigates schistosome egg-induced immunopathology by promoting type I Interferon (IFN-I)

Type 2 immunity is a defining feature of helminth infection and is traditionally viewed as protective, promoting parasite expulsion, tissue repair, and/or suppression of bystander antigens. However, work in allergic disease has demonstrated that Th2 cells are not functionally uniform but instead comprise specialized subsets, including highly differentiated pathogenic Th2 populations characterized by sustained IL-5/13 production, epithelial alarmin responsiveness, and stable tissue residency. The presence of these cells is associated with immunopathology and worse clinical outcomes. Interesting

Leishmanization, the deliberate inoculation of live parasites into the skin, was historically the only effective immunization strategy against leishmaniasis. Advances in molecular biology have since transformed this concept, particularly through gene-editing technologies targeted attenuation of virulence-associated genes, leading development of a new generation of live attenuated parasites. These next-generation leishmanization approaches preserved broad antigenic diversity while offering improved safety profiles inducing durable protective immunity. Deletion of the kharon1 gene in Leishmania

Diffuse cutaneous leishmaniasis (DCL), caused by Leishmania mexicana, is a chronic, treatment-refractory form of leishmaniasis characterized by multiple nodular lesions and high parasite load. During chronic infection, T cells acquire an exhausted immune state marked by elevated expression of PD-1 and comprise progenitor-like (CXCR5+TIM-3-), intermediate (CXCR5+TIM-3+), and terminally-differentiated (CXCR5-TIM-3+) exhausted T-cell (Tex) subsets. It remains unknown whether anti-PD-1 immune checkpoint therapy is effective during chronic L. mexicana infection and how it affects the Tex landscape.

Type 1 regulatory (Tr1) T cells are a major source of IL-10-mediated immune regulation, yet their phenotypic definition and role in human disease remain incompletely understood. In this issue of the Journal of Clinical Investigation, Nideffer et al. provide insight into human Tr1 cells during pediatric Plasmodium falciparum (Pf) infection. The authors identified Tr1 cells as a major component of the malaria-specific CD4+ T cell response, producing both IL-10 and IFN-γ. They proposed that, in this context, Tr1 cells may be better identified by CD127 downregulation combined with CXCR6 expression

BACKGROUND: Both T helper 2 (Th2) and B cell responses are crucial for controlling gastrointestinal (GI) nematode infections. However, adaptive immune determinants of host susceptibility to GI nematode infection are still not fully understood.
METHODS: Adaptive immune responses against the small intestinal nematode Heligmosomoides polygyrus bakeri were compared between susceptible C57BL/6 and resistant BALB/c mice. Parasite-specific Th2 responses were quantified via combined detection of CD40-L and type 2 cytokine expression in CD4+GATA-3high cells stimulated with parasite products. Follicula

Crusted scabies is a rare parasitic hyperinfestation, commonly associated with impaired host immunity. Dysregulation of T cell homeostasis has been reported to drive disease progression. We characterized a profoundly imbalanced T cell phenotype in an immunocompetent male with crusted scabies, potentially induced by prior tralokinumab and corticosteroid treatment for presumed atopic dermatitis.

The aim of this study was to assess the impact of combining albendazole with gold and silver nanoparticles on the Th1/Th2 cytokine balance, specifically the IFN-γ/IL-4 ratio, in BALB/c mice with hydatidosis. Sixty healthy female BALB/c mice (6-8 weeks old, 25-35 g) were infected with 1500 live Echinococcus granulosus protoscoleces and sorted into six groups: albendazole (200 mg/kg), nanosilver (25 mg/kg), ABZ-loaded silver nanoparticles (100 mg/kg), nanogold (25 mg/kg), ABZ-loaded gold nanoparticles (100 mg/kg), and a control group (not treated). After a five-month incubation and 12-week treat

The immunopathology of cutaneous leishmaniasis (CL) caused by Leishmania aethopica is not well understood. This study aimed to characterize the host's localized immune response in formalin-fixed paraffin-embedded (FFPE) skin biopsies taken from skin lesions of patients who had developed localized CL due to infection with L. aethiopica. A retrospective cross-sectional study was conducted on 40 FFPE skin biopsy samples collected from 2018 to 2022 in the ALERT hospital, Addis Ababa, Ethiopia. Immunohistochemical techniques were used to examine localized immune T helper 1 (Th1) and T helper 2 (Th2

Toxoplasma gondii remains a significant parasitic pathogen with limited effective vaccines against its acute virulent form. This study aimed to evaluate the protective and immunological efficacy of a total glycoprotein-based vaccine isolated from T. gondii tachyzoites against infection with the virulent RH strain in Swiss albino mice. A glycoprotein-enriched fraction was isolated from T. gondii tachyzoites using Concanavalin A affinity column chromatography. Mice were divided into four groups (n = 12/group): group I received three subcutaneous doses of 25 μg total isolated glycoproteins emulsi

Eosinophils are evolutionarily conserved granular innate cells that can trace back to 100 million years to the emergence of gnathostomes, which now is involved in regulation of diverse homeostatic conditions and diseases in mammals. They have traditionally been associated with Th2 responses, primarily in eosinophilic allergic asthma and helminth parasitic infections. These Th2 responses are correlated with basic cationic proteins and specific cytokines release from preformed eosinophilic vesicles, associated with tissue damage and remodeling. Consequently, eosinophils have historically been ch
