BACKGROUND: Post-kala-azar dermal leishmaniasis (PKDL), a dermal sequel of visceral leishmaniasis (VL) is considered as a reservoir that facilitates the transmission of VL. Although PKDL lesions demonstrate an overwhelming infiltration of CD8 ⁺ T cells, characterization of lesional CD8 ⁺ T cells, their tissue-homing chemokine receptors and corresponding ligands remains poorly defined, and was the aim of this study.
METHODOLOGY: In patients with PKDL, CD8 ⁺ T cells were phenotyped in skin biopsies, in terms of senescence (CD8 ⁺ /CD57⁺), and cytotoxicity (expression of Perforin and Granzyme). T

Cystic Echinococcosis (CE), caused by the larval stage of Echinococcus granulosus sensu lato, elicits a complex host immune response characterized by the formation of a fibrotic capsule known as the adventitial layer. While the cellular composition of this layer has been previously studied, the spatial distribution of T lymphocytes remains poorly defined, particularly in relation to cyst fertility, host species, organ localization, and co-infection with Fasciola hepatica. In this study, we evaluated the distribution of CD3⁺ T cells in the adventitial layer of hepatic and pulmonary CE cysts col

Messenger RNA (mRNA) vaccines represent a transformative platform in vaccinology, with applications extending beyond SARS-CoV-2 to other infectious diseases and cancer immunotherapy. However, patients with solid tumors receiving active anticancer treatment were largely underrepresented in pivotal vaccination trials, limiting understanding of vaccine-induced immunity in this population. In this prospective exploratory study, we assessed humoral and cellular immune responses after two doses of SARS-CoV-2 mRNA vaccines in 39 patients with solid tumors undergoing active treatment. Blood samples we

V gamma 9 V delta 2 (Vγ9Vδ2) T cells, the predominant γδ T cell population in human peripheral blood, uniquely recognize nonpeptidic phosphoantigens (pAgs) independent of major histocompatibility complex molecules. This sensing mechanism relies on intracellular pAg accumulation, which triggers heteromeric cooperation between transmembrane butyrophilin 3A1 (BTN3A1) and butyrophilin 2A1 (BTN2A1) receptors to activate the γδ T cell receptor. This review synthesizes current knowledge of Vγ9Vδ2 T cell immunobiology, focusing on responses to Plasmodium falciparum and Toxoplasma gondii. We examine ho

Tegumentary leishmaniasis (TL) is a neglected tropical disease characterized by limited therapeutic options, drug-associated toxicity, and the absence of immunotherapeutic interventions. In this study, we evaluated an immunochemotherapeutic strategy combining the multi-epitope recombinant protein ChimT, monophosphoryl lipid A (MPLA), and ivermectin (IVE) for the treatment of Leishmania amazonensis infection in BALB/c mice. Infected animals were treated with ChimT, MPLA, and/or IVE; and parasitological, immunological, and biochemical evaluations were performed one and 30 days after therapy. The

Ascarids are among the most prevalent soil-transmitted helminths affecting both humans and livestock, particularly pigs. While reduced anthelmintic efficacy has been reported in humans, frequent reinfection and the lack of a vaccine highlight the need for alternative control strategies across species. In pigs, fermentable dietary fibers have been shown to enhance type 2 immune responses and mucosal barrier function and may represent a complementary strategy for parasite control. Here, we investigated the effects of a fermentable fiber diet in pigs infected with the parasite Ascaris suum (A. su

The outcome of a Plasmodium infection depends on the timely regulation of the robust pro-inflammatory response required to eliminate the parasite, but this response can cause tissue damage if not properly controlled. Interleukin-10 (IL-10) is an important regulatory cytokine that prevents immunopathology during many Plasmodium infections; however, this protection comes at the expense of less effective parasite control. This is illustrated by infection with blood-stage Plasmodium yoelii, in which mice exhibit a lower parasite load in the absence of IL-10. However, the immune components that lim

Visceral leishmaniasis (VL) is a severe disease caused by Leishmania infantum in Brazil. Relapses have increased among immunocompetent patients; however, the underlying factors remain unclear. Previous studies have linked persistently low CD4+ T cell counts to VL relapses, suggesting that impaired immune reconstitution may favor recurrence. Here, we investigated whether immune activation and thymic output influence immune recovery in VL and shape the TCRVβ repertoire, predisposing patients to relapse. Patients with VL, followed for 12 months, were stratified according to relapse (R-VL,n = 5) o

ETHNOPHARMACOLOGICAL RELEVANCE: In Nigeria and other tropical and sub-tropical regions, Azadirachta indica, or neem, is a typical medicinal plant. In traditional medicine, its stem and leaves have been used to treat malaria.
AIM OF THE STUDY: Through tryptophan catabolism and the likely mechanisms involved, we aim to examine the immunomodulatory potentials of flavonoid-rich fractions of A. indica extracts by regulating the splenic indoleamine 2, 3-dioxygenase (IDO), a crucial immune system regulator. This is to enunciate scientific proof and information about its therapeutic qualities for the

Regulatory T (Treg) cells are essential for controlling inflammation and maintaining tissue homeostasis. Transcriptional intermediary factor 1-γ (TIF1γ) has been implicated in noncanonical TGFβ-Smad signaling in hematopoietic stem cells, but its role in Treg cell function has not been explored. Here we show that TIF1γ-deficient Treg cells lose Foxp3 expression and acquire effector phenotypes in a cell-intrinsic manner upon inflammatory challenge. In vivo, TIF1γ-deficient Treg cells adopt type 1 helper T cell-like or pro-inflammatory ex-Treg cell states in autoimmune settings and display type 2

BACKGROUND: Subjects with chronic Chagas disease and no signs of heart disease exhibit decreased NKp46 expression, high CD57 expression, and IL-10 production by NK cells. This study provides a detailed characterization of the phenotype and function of NK cells according to the severity of heart disease, and evaluates how these changes following treatment with benznidazole, as well as their association with Trypanosoma cruzi-specific T-cell responses.
METHODS: The phenotype and function of NK cells in a cohort of 51 subjects infected with Trypanosoma cruzi and exhibiting varying degrees of hea

Here, we present a reproducible Seurat-based protocol to analyze PBMC CD4⁺ T-cell single-cell RNA sequencing data across malaria reinfection timepoints. This protocol demonstrates a reproducible Seurat-based workflow for analyzing PBMC CD4⁺ T-cell scRNA-seq data across malaria reinfection timepoints, using representative publicly available datasets to demonstrate its application: a Plasmodium-specific TCR-transgenic CD4⁺ T-cell dataset (GSE233703) and a polyclonal CD4⁺ T-cell dataset comparing reinfection-associated timepoints (GSE233713; D27₍₃₎ versus D30). The workflow includes standardized

Thymically-derived regulatory T (Treg) cells express the transcription factor Foxp3 and are maintained at homeostasis by the cytokine IL-2. These specialized cells mediate peripheral tolerance and determine the balance between protective and pathological T cell responses during infection. Acute infection of mice with Toxoplasma gondii results in thymic atrophy and reduced production of IL-2 that correlate with a "crash" in Treg cell populations. Here, kinetic studies of mice infected with T. gondii reveal that as acute infection resolves, there is a restoration of IL-2 and reversal of thymic h

Visceral leishmaniasis (VL) is associated with impaired Th1 responses, yet the intracellular signalling events linking immunosuppressive pathways to effector dysfunction remain poorly defined. We studied 150 adults in endemic northern Nigeria (50 active VL, 50 healed VL, 50 endemic controls) using flow cytometry, cytokine profiling, targeted RT-qPCR of 13 JAK-STAT pathway genes with dual reference gene normalisation, ex vivo checkpoint blockade, and phospho-flow cytometry for STAT1/STAT3 in both CD4+ T cells and CD14+ macrophages. Active VL was associated with reduced antigen-specific IFN-γ (2

BACKGROUND: Microgliosis and severe coagulation, including fibrinogen deposition, are features of both experimental and human cerebral malaria (CM), a lethal disease. Vascular-associated microglia migrate to coagulated cerebral vessels containing inflammatory monocytes and T cells in experimental CM. We previously showed that microglial depletion exacerbates coagulation and disease severity, including hypothermia, while anticoagulant treatment reduces microgliosis and prevents mortality. These data suggest an overall protective effect of microglia on eCM, and indicate a link between microglios

Malaria pathogenesis involves complex immunopathological mechanisms that hinder early diagnosis and effective treatment. This study integrates multi-omics data and experimental models to identify host-derived biomarkers and elucidate their functional roles. By combining human transcriptomic datasets, weighted gene co-expression network analysis (WGCNA), and machine learning (LASSO, SVM, RF), we identified C1QB as a key hub gene. In human data, C1QB was significantly upregulated in both training and validation cohorts (AUC 0.983 and 0.970). Single-gene GSEA and immune infiltration analyses link

Cancer hallmarks are characterized by wide-scale changes in gene expression programs. Pioneering studies showed how viral oncogenes target regulatory pathways, but little is known about tumorigenic mechanisms of non-viral pathogens. Theileria annulata is an intracellular parasite (related to apicomplexa parasites causing malaria) which remarkably transforms bovine leukocytes, hijacking host signaling pathways to induce cancer phenotypes, akin to human leukemias. While some host genes contribute to the proliferative or invasive hallmark phenotypes, there is still limited comprehensive understan

Chronic liver fluke infections represent a major global health burden, driving liver fibrosis and elevating cancer risk, yet the immunological mechanisms linking persistent infection to tissue damage remain poorly defined. Using Clonorchis sinensis, a carcinogenic liver fluke infecting millions worldwide, as a representative model, we map immune responses during chronic infection in mice. We show that intrahepatic CD8⁺ tissue-resident memory T cells (TRM) progressively accumulate and drive pathology by producing chemokines that promote immune recruitment and fibroblast activation. Depletion of

INTRODUCTION: Toxoplasma gondii is a significant zoonotic pathogen responsible for severe disease in immunocompromised individuals, adverse pregnancy outcomes, and substantial economic losses in the livestock industry. Given the limitations of current therapeutic strategies and vaccines, this study utilized immunoinformatics and reverse vaccinology approaches to design a multi-epitope candidate vaccine.
METHODS: A total of 49 T. gondii proteins from the SAG, GRA, MIC, and ROP families were screened. Epitopes were selected based on antigenicity, immunogenicity, cytokine-inducing potential, tox

Resistance to Leishmania (L.) major depends on the development of a L. major-specific Th1 response, while Th2 differentiation results in susceptibility. We previously showed that the early microenvironment of infected skin delivers important signals for T cell differentiation. We found increased expression levels of coagulation factor X (F10 and FX for protein, respectively) 16 h after infection in the skin of resistant mice compared to susceptible mice. Activated FX is a ligand of proteinase-activated receptor-2 (PAR2), a modulator of inflammatory responses. To assess the role of PAR2, we ana
