INTRODUCTION: While there is evidence that Ebola virus (EBOV) antagonizes the antiviral type I interferon (IFN-I) response, the role of IFN-I for EBOV disease remains controversial, as initially protective responses may contribute to disease pathogenesis later.
METHODS: We analyzed patient data from the 2014-2016 West Africa epidemic with a combination of machine learning and mathematical modeling to identify predictive immune mediators and reconstruct their temporal dynamics in survivors and non-survivors.
RESULTS: Our results suggest that IFN-I response in survivors occurs before symptom o

BACKGROUND: Ebola virus (EBOV) poses a significant public health threat due to its high case fatality rates and proved human-to-human transmission. While licensed vaccines exist for EBOV, their intramuscular administration presents some logistical challenges in outbreak settings. Oral vaccines offer advantages in accessibility, compliance, and scalability but have not been explored for EBOV.
METHODS: We developed an orally-administered EBOV vaccine using virus-like particles (VLPs) incorporating the EBOV glycoprotein (GP), nucleoprotein (NP), and viral protein 40 (VP40). VLPs were produced vi

The immune response plays a crucial role in eliminating dengue virus infection from the human body. Both humoral and cellular immune responses are naturally involved in combating the virus. This study presents a mathematical model that captures the dynamics of humoral and cellular immune responses to dengue virus infection, incorporating key interactions between B cells, T cells, and viral particles. Additionally, the model considers hematocrit level as an indicator of disease severity, which is influenced by the interaction between the virus and immune responses. The basic reproduction number

Dengue is an arboviral disease of global significance caused by Orthoflavivirus denguei (DENV), which has four antigenically distinct serotypes. The envelope (E) protein plays a critical role in viral entry and eliciting immune responses. This study aimed to compare the biochemical and immunological properties of the E protein across the four DENV serotypes using in silico approaches. E protein reference sequences were retrieved from RefSeq and analyzed with various bioinformatics tools. Sequence alignment revealed identities ranging from 63.08% to 77.69%. Biochemical analysis showed minimal v

Despite deployment of vector control and prevention strategies, dengue remains a substantial threat to public health. Development of an effective vaccine against dengue has been challenging due to the need for simultaneous immunity against all four dengue virus serotypes, durable protection regardless of prior dengue exposure, and the complex epidemiological patterns of serotype co-circulation. As part of the clinical development of TAK-003, a live attenuated tetravalent dengue vaccine based on a DENV-2 backbone, extensive profiling of post-vaccination immune responses has been performed in pa

Hemorrhagic fever with renal syndrome (HFRS) is a severe and often fatal zoonotic disease primarily caused by the Hantaan virus (HTNV), a member of the Orthohantavirus genus. Although immune cells are known to play critical roles in HTNV-induced HFRS, the detailed dynamic changes, phenotypic alterations, and functional profiles of distinct immune subsets over the course of the disease remain poorly understood. In this review, we summarize current understanding of key immune populations during HTNV infection, including monocytes, macrophages, dendritic cells (DCs), natural killer (NK) cells, ne

Currently, the role of dengue virus-specific T-cell responses in disease pathogenesis and protection is not well understood, including potential differences in individuals with obesity. We sought to investigate the functionality and phenotype of T cells in patients with acute dengue fever (DF) or dengue haemorrhagic fever (DHF). T-cell function was assessed in paired samples (first sample on ≤4 days and second 5-6 days since onset of illness) in patients with DF (n = 50) and DHF (n = 12), using 109 peptides representing CD8+ epitopes and 90 peptides targeting CD4+ T-cell epitopes. Phenotypic a

Dengue causes over 390 million annual infections globally. Qdenga (TAK-003), the only widely available dengue vaccine, has an incompletely defined immunological profile. We performed longitudinal immunological profiling of 110 adults from a dengue-endemic region, including older adults (≥65 years), to assess the effects of immune imprinting and vaccine design on humoral and cellular immunity. Qdenga elicited B and T cell activation across groups but generated serostatus-dependent, serotype-skewed antibody responses. Only 8% of DENV-naïve individuals developed tetravalent neutralizing responses

The recent outbreak of the Marburg virus (MARV) in Rwanda was effectively controlled, but concerns remain regarding the virus's persistence and its long-term effects. MARV can remain in the testes of nonhuman primates, where it harms Sertoli cells and compromises the blood-testis barrier, which may have implications for male infertility. This persistence is aided by immunosuppressive regulatory T cells (Tregs), which allow the virus to evade the immune system. Drawing comparisons with Ebola and Zika viruses, we propose that MARV may contribute to male infertility and recommend further studies

Dengue virus (DENV), as a widely circulating arbovirus, is prone to causing dengue fever and poses a serious threat to human health. Nevertheless, there is currently no ideal safe and effective vaccine for DENV. In particular, vaccination with approved DENV vaccines may increase the chance of infection with heterotypic serotypes of DENV and the risk of severe dengue upon infection. Antibody-dependent enhancement (ADE) is considered a major mechanism contributing to severe disease in secondary infections, which seriously restricts the safety and efficacy of the vaccine. Notably, many studies ha

Dengue virus (DENV) is a major global health threat, with secondary heterotypic infections potentially inducing detrimental memory immune responses. Antigen-specific CD8 + T cells contribute to both protection and pathogenicity, yet how their phenotypic heterogeneity relates to disease severity remains unclear. Here, we performed plate-based single-cell RNA sequencing of circulating DENV-specific CD8 + T cells identified by HLA tetramers loaded with DENV NS3-derived epitopes. Using tetramer binding to peptides corresponding to the currently and serologically inferred dominant previously infect

Dengue is classified into mild dengue, which is treated at outpatient clinics, dengue with warning signs (WS+), which requires hospitalization, and severe dengue, which can be fatal. It is assumed that severe manifestations result from uncontrolled immune activation. Regulatory T cells (Tregs) are crucial for controlling inflammation in various diseases, but are poorly studied in dengue. This study aimed to investigate the Tregs phenotype in different clinical forms of dengue infection. Plasma and peripheral blood mononuclear cells (PBMCs) were collected from healthy donors and dengue patients

Yellow fever (YF) remains a significant public health concern despite the success of the live-attenuated YF-17D vaccine. While YF-17D serves as a benchmark for durable protection, its inherent drawbacks, including cold-chain dependence, contraindications for vulnerable populations, and rare severe adverse events, necessitate the development of safer, more deployable alternatives. The core of next-generation vaccine design lies in the systematic identification and functional validation of highly effective YFV epitopes. The robust immunogenicity of YF-17D is characterized by a polyfunctional, mu

Recombinant vesicular stomatitis virus (rVSV) is a powerful vaccine platform that elicits durable cellular and humoral immune responses. rVSV vaccines have successfully been used for various hemorrhagic fever-causing viruses. Here, we create and evaluate rVSV vaccines for Machupo virus (MACV), a highly pathogenic New World (NW) mammarenavirus that causes Bolivian hemorrhagic fever. VSV genome rearrangements impact attenuation of the viral vector and the immune response to the vaccine. Therefore, in this study, we directly compare the effect of altering the MACV glycoprotein complex (GPC) posit

BACKGROUND: Dengue fever is a mosquito - borne infectious disease caused by the dengue virus (DENV). Understanding its immune response mechanisms is crucial for developing effective treatments and vaccines, especially regarding the dynamic changes of peripheral blood lymphocyte subsets. Traditional immunological methods have limitations in accurately analyzing lymphocyte subsets and their functional dynamics. However, the combination of single - cell RNA sequencing and flow cytometry can explore the immune response of dengue patients more deeply.
METHODS: We integrated four public datasets fr

O'nyong-nyong virus (ONNV) is a mosquito-borne alphavirus first isolated in Uganda in 1959. Since its discovery, ONNV has caused several outbreaks in Africa, manifesting clinically as fever, rash, and joint/muscle pain lasting months. Currently, we have a limited understanding of ONNV infection and disease in relevant animal models, which restricts the evaluation of vaccines and therapeutics. In 1967, Binn et al. reported that infection of rhesus macaques (RMs) with ONNV failed to induce viremia in two animals. This may be attributed to the potential attenuation of the virus through extensive

BACKGROUND: Rift Valley fever virus (RVFV) is a zoonotic arbovirus endemic to Africa and the Arabian Peninsula that can cause disease in humans and livestock. There are currently no licensed vaccines for human use, but several candidates are at the clinical trial stage including DDVaxTM, a preparation of live-attenuated RVFV lacking the nonstructural proteins NSs and NSm (ΔNSsΔNSm). Multiple pre-clinical studies have shown that ΔNSsΔNSm is safe and immunogenic.
METHODS: The longitudinal study described in this manuscript examined the additional features of duration, sex and dose in immunogeni

Viral encephalitis (VE) is a universal menace accounting for severe morbidity and mortality among the affected individuals. VE is the cerebral inflammation triggered by viral infections. In the central nervous system, microglia are the frontline responders that provide defense against invading pathogens, such as neurotropic viruses. Upon viral recognition, microglia become activated and mount antiviral immune responses by producing type I interferons, presenting viral antigens to T cells, and phagocytosing viral components and virus-infected cells during the acute phase of infection. However,

BACKGROUND: Childhood neuropsychiatric systemic lupus erythematosus (cNPSLE) represents one of the most severe complications of systemic lupus erythematosus. Aberrant CD8+ T cell activation and hyperactivated IFN-I signaling contribute to cNPSLE immunopathogenesis. IFI27, a key IFN-I-stimulated gene, has not been functionally characterized in cNPSLE.
METHODS: The scRNA-seq data from cNPSLE patients were analyzed to characterize immune cell heterogeneity and interactions using cell clustering, differential gene screening, GO enrichment, and CellChat analysis. NPSLE mouse models were establishe

Human bornavirus encephalitis (BVE) is a rare, emerging and fatal zoonotic disease mainly caused by the Borna disease virus 1 (BoDV-1), a non-cytolytic RNA virus. Despite increasing recognition, the immunopathogenesis of human BoDV-1 infection remains insufficiently characterised. Complete coronal and sagittal brain sections from four fatal BoDV-1 cases were analysed using digitised immunohistochemistry to quantify viral distribution and tissue responses. Transcriptome-based analyses characterised local immune cell profiles in relation to viral loads measured by RT-qPCR. BoDV-1 viral loads var
