CCL19-armed oncolytic adenovirus synergizes with CCR7-expressing CAR-T cells to suppress ovarian tumor growth.

Solid tumors remain refractory to chimeric antigen receptor (CAR) T cell therapy largely because they fail to infiltrate the tumor bed. We show that in vitro expanded HER2 CAR-T cells are more than 80% CCR7+ cells that migrate vigorously to CCL19. An E1B-55 kDa-deleted oncolytic adenovirus expressing CCL19 retained full oncolytic potency, secreted bioactive CCL19 and tripled CAR-T migration. In NSG mice bearing SKOV3 ovarian tumors, two intratumoral oAd-CCL19 injections followed by intravenous HER2-CAR-T cells achieved most effective inhibition of tumor growth without significant toxicity, acc

An ICOSL-armed oncolytic adenovirus activates CD4+ T cell to potentiate antitumor immunity and synergizes with anti-PD-1 or CAR-T cell therapy in colorectal cancer.

BACKGROUND: The efficacy of oncolytic adenoviruses (ADVs) in colorectal cancer (CRC) is limited by their inability to effectively expand CD4+ T cells within the tumor microenvironment (TME). This study aimed to elucidate the underlying mechanism and overcome this bottleneck through viral engineering. METHODS: We constructed an oncolytic adenovirus expressing inducible T cell co-stimulator ligand (ICOSL), designated ADV-ICOSL, based on the backbone virus ADV-NC. Its antitumor efficacy, safety, and ability to induce immunological memory were systematically evaluated in MC38 and CT26 murine colo

A CDK4/6 inhibitor-armed oncolytic adenovirus reverses T cell exhaustion through the Rb-p65-CCL5 pathway and potentiates the antitumor activity of anti-PD-1 or CAR-T therapy in colorectal cancer.

INTRODUCTION: The efficacy of oncolytic adenoviruses in colorectal cancer models is constrained by a treatment-induced limitation: the high-dose, repetitive administration required for sustained oncolysis promotes chronic antigen exposure and tumor microenvironmental stress, driving CD8+ T cells into a state of exhaustion. METHODS: To mitigate this, we constructed an oncolytic adenovirus, ADV-PTD4-D3, engineered for intratumoral expression of a peptide inhibitor of CDK4/6. This local strategy aims to retain immunomodulatory potential while minimizing systemic exposure. In syngeneic murine mod

Mucosal boosting of chimpanzee adenovirus-vectored vaccine encoding SARS-CoV-2 RBD-heterodimer provides robust protection in mice.

The major route of COVID-19 vaccination currently is via intramuscular injection. Data from clinical trials and real-world studies have demonstrated its effectiveness in preventing severe illness and death caused by SARS-CoV-2 infection. However, its protective efficacy against SARS-CoV-2 infection and transmission in situ remains relatively low. Given that SARS-CoV-2, especially the Omicron variant and its sub-variants, primarily infects and replicates in the human upper respiratory tract, mucosal immune responses are crucial for preventing viral infection. Therefore, we constructed a chimpan

Oncolytic virus-drug conjugates for increased viral replication efficiency and potentiated cancer chemoimmunotherapy.

Oncolytic viruses (OVs) therapy is limited by poor systemic delivery efficiency to tumors, low intratumoral replication efficiency and insufficient immune activation against malignant cancers. Here, we report the autophagy-activated oncolytic adenoviruses (Ads)-doxorubicin (DOX) conjugates, combining the efficient co-delivery and viral intratumoral replication enhancement for potentiated chemoimmunotherapy. Ads-DOX conjugates are composed of Ads coated with a layer of biocompatible lipid membrane, and the autophagy-responsive DOX prodrugs inserted in the lipid membrane. The intravenous adminis

Lung-resident T-cell immunity dominates protection induced by an intranasal adenoviral nucleoprotein influenza vaccine.

Frequent antigenic drift in influenza viruses necessitates broadly protective vaccines. This study evaluated an intranasal adenoviral-vector vaccine expressing influenza A nucleoprotein (NP) fused to the autophagy-inducing peptide C5. A heterologous prime-boost regimen using chimpanzee and bovine adenoviral platforms induced strong NP-specific humoral and cellular immune responses in mice. High serum and lung IgG/IgA level were detected, accompanied by enhanced antibody-dependent cellular cytotoxicity. Cellular analyses revealed potent NP-specific cytokine responses and expansion of effector m

Broad and durable protection against SARS-CoV-2 and SARS-CoV by an intranasal chimpanzee adenovirus vaccine expressing tandem RBDs and nucleocapsid.

Current intramuscular COVID-19 vaccines reduce severe disease but offer limited protection against infection, enabling viral persistence, immune escape, and transmission. We developed intranasal rare-serotype chimpanzee adenovirus 68 (AdC68)-based vaccines expressing heterologous tandem receptor-binding domains from SARS-CoV-2, SARS-CoV, and MERS-CoV, fused to the SARS-CoV-2 nucleocapsid (AdC68-4RBD(XBB.1.5)-N) to enhance both B and T cell responses. Antigen integrity was confirmed by receptor binding and recognition by multiple conformation-sensitive monoclonal antibodies. In mice, intranasal

Adenoviral E3/49K: CD45-targeted immune evasion and therapeutic potential.

Human adenoviruses (HAdVs) cause a broad spectrum of diseases, ranging from mild respiratory and gastrointestinal infections to severe, sometimes even fatal illness in immunocompromised individuals. Species D adenoviruses represent the largest group, possibly driven by strong evolutionary pressure, particularly within the highly variable early transcription unit 3 (E3), which encodes immunomodulatory proteins. Among these, E3/49K is unique to species D and utilizes a particularly intriguing immunomodulatory strategy. It is a heavily glycosylated transmembrane protein that undergoes proteolytic

Therapeutic vaccination strategy targeting the PreS1 region of L-HBsAg for HBV/HDV co-infection.

BACKGROUND & AIMS: Achieving functional cure for chronic hepatitis B (CHB) will likely require a combinatorial approach targeting multiple aspects of the complex hepatitis B virus (HBV) life cycle and host adaptive immune responses. Here, we developed a therapeutic vaccination strategy targeting the PreS1 region of L-HBsAg, required for cellular entry of both hepatitis B and D viruses. METHODS: An established potent T-cell inducing platform, recombinant adenovirus (Ad), was used as a nanoparticle scaffold for PreS1 attachment, to generate antibodies that neutralize virus entry and to establis

Enhanced Immunogenicity and Cross-Neutralizing Activity of a Trivalent Adenoviral Vector-Based SARS-CoV-2 Vaccine in Mice.

The emergence of diverse SARS-CoV-2 Omicron subvariants has reduced the efficacy of existing vaccines, highlighting the need for multivalent vaccines with broader neutralizing capacity. We developed a trivalent adenoviral vector-based vaccine (Ad5-CoV19-prototype/Delta/BF.7) encoding the spike proteins of the ancestral strain, Delta variant, and Omicron BF.7 strain, and evaluated its immunogenicity in BALB/c mice. Four- to 6-week-old female BALB/c mice were intramuscularly immunized with 10 µg of monovalent or trivalent vaccines using single-dose or two-dose (2-week interval) regimens. Neutral

Qualitative Superiority, Not Magnitude, of Lung-Resident CD8+ T Cells Determines Protective Efficacy of Influenza NP Versus PB1 Vaccines.

T cell-based universal influenza vaccines aim to generate robust lung-resident memory CD8+ T cells (TRM), yet it remains unclear whether conserved Ags elicit equally protective TRM pools. We compared recombinant adenoviral vectors expressing the influenza A nucleoprotein (rAd/NP) or polymerase basic protein 1 (PB1). After intranasal immunization, both induced comparable magnitudes of respiratory Ag-specific CD8+ T cells, yet only rAd/NP provided 100% survival after lethal influenza challenge. This disparity reflected qualitative differences in the TRM pool; nucleoprotein (NP)-specific cells we

Dual-antigen AdV5 vaccines against Crimean-Congo hemorrhagic fever virus induce superior humoral and Th1-biased cellular immunity.

Platforms based on adenoviral vectors have proven their efficiency in developing vaccines and therefore can be considered potential candidates for immunization strategies against CCHFV. In this study, four replication-deficient recombinant human adenovirus serotype 5 (AdV5) vectors were engineered to express CCHFV antigens: a dual-antigen NP-P2A-Gc construct and monocistronic constructs for nucleoprotein (NP), glycoprotein complex precursor (GPC), and mature glycoprotein (Gc). Following GMP-compatible production and purification, vaccine candidates were evaluated in BALB/c mice (n = 4 per grou

Systemic and Mucosal Immune Responses From Serotype-Switched Viral-Vectored Ebola Vaccines.

Ebola virus is a highly virulent pathogen causing severe hemorrhagic fever with 25%-90% mortality. The 2014 outbreak infected over 28,000 people, causing 11,325 deaths. Although there is one FDA-approved vaccine available for limited use, additional vaccines are needed. We developed adenoviral-vectored vaccines expressing the ebolavirus glycoprotein (GP), the sole viral surface protein and primary immunogen. Vaccine constructs spanned three species and five human adenovirus subtypes, including low-seroprevalent serotypes to minimize pre-existing immunity. We compared single-dose and prime-boos

Intranasal Adenoviral Vector Vaccination Induces Durable Antibody and T Cell Responses in the Respiratory Tract.

Background/Objectives: Respiratory pathogens such as influenza virus and SARS-CoV-2 pose major threats to global public health. Current vaccines provide limited mucosal protection against respiratory infection, while vaccine-induced immunity can wane over time. Intranasal vaccination has emerged as a promising approach to induce mucosal immunity, however relatively low immunogenicity and limited durability remain major challenges. Various strategies have therefore been explored to improve intranasal vaccine efficacy, including mucosal adjuvants and alternative vaccine platforms. In this study,

DNA Vaccine Encoding the prM-E Polyprotein of the Tick-Borne Encephalitis Virus.

We evaluated immunogenicity of the experimental DNA-vaccine construct pVAX-prM-E encoding the polyprotein prM-E of the tick-borne encephalitis virus (strain Sofjin). It was shown that pVAX-prM-E administered to mice using a needle-free jet injector induced the production of high levels of virus-neutralizing antibodies. The level of humoral response to pVAX-prM-E was comparable to that induced by administration of the commercial Tick-E-Vac vaccine. ELISpot analysis of the T-cell response showed that the obtained construct induces high levels of IFNγ in response to stimulation with a pool of spe

A replication-defective tick-borne encephalitis virus generated by NS1 trans-complementation confers robust protective efficacy.

Tick-borne encephalitis virus (TBEV) poses a severe threat to public health, causing neurological disorders with high morbidity and mortality in endemic regions. Therefore, developing safe and effective strategies for TBEV vaccines has long been a focus of attention. Herein, leveraging our well-established NS1 trans-complementation platform, we successfully constructed a high-titer replication-defective TBEV (TBEV-ΔNS1) using the BHK-21 cell line stably expressing Omsk hemorrhagic fever virus (OHFV) NS1 (designated BHKNS1). Retention of the NS1 deletion in TBEV-ΔNS1 was confirmed by continuous

Immunological Interactions Between Gammadelta T Cells and West Nile Virus in the Infected Host.

Between mosquitoes and birds, West Nile virus (WNV) is a neurotropic flavivirus, an arthropod-borne pathogen involved in an enzootic cycle. Additionally, it can infect both people and horses, leading to severe illness. Since 1999, WNV has spread across North and South America, including Mexico and the Caribbean. It is endemic in several regions of Europe, Africa, the Middle East, and Asia. WNV affects the central nervous system (CNS), causing severe disease in a small percentage of infected individuals, especially in immunocompromised or elderly hosts. This re-emerging pathogen was identified

Encephalitic alphavirus infection induces long-term humoral and cell-mediated responses in seropositive participants from Panama.

Encephalitic alphaviruses are an emerging viral threat with a history of large-scale outbreaks. Evaluating the immune response to natural infection in survivors is critical to the rational development of medical countermeasures (MCM), and yet little is known thus far. Here, we describe a comprehensive analysis of adaptive immune responses from a cohort of 37 convalescent seropositive individuals of Venezuelan equine encephalitis and/or Madariaga virus from Panama. We demonstrate persistent and polyfunctional B cell responses, as well as memory T cell cytokine production in response to viral an

Self-assembled EDIII/NS1 nanoparticle vaccines elicit protective immune responses against Japanese encephalitis virus.

Japanese encephalitis virus (JEV) is a mosquito-borne orthoflavivirus that can cause severe neurological disease in humans and reproductive disorders in swine, posing a persistent threat to public health and the swine industry. Despite being vaccine-preventable, the virus remains endemic in 24 countries and territories throughout Asia and Oceania, underscoring the urgent need for safer and more effective vaccines to counter this ongoing threat. In this study, we developed the nanoparticle vaccines by covalently conjugating 60 copies of EDIII or NS1 protein subunits to the self-assembling mi3 p

Optimized intracellular flow cytometry panel enables CD4 and CD8 T cell cytokine profiling in Syrian hamsters.

The Syrian golden hamster has emerged as a valuable animal model in challenge experiments and vaccine efficacy studies, particularly for infectious diseases that are poorly represented in mice. In recent years, research has demonstrated that hamsters can better recapitulate certain human infectious diseases compared to mice. Syrian hamsters are used in studies with Yellow fever virus, Helicobacter spp., Hantavirus pulmonary syndrome, Japanese encephalitis, Nipah virus, Adenovirus, Influenza A/B, and SARS-CoV-2, among others. However, the model has been limited by a lack of immunological tools,
