H3K27M Mutation Modulates the Antigenic Landscape in Diffuse Midline Glioma.

BACKGROUND/OBJECTIVES: Diffuse midline gliomas (DMGs) are among the most aggressive paediatric brain tumours, with the pathognomonic H3K27M mutation present in over 80% of cases. This mutation drives epigenetic dysregulation and transcriptional reprogramming, yet its impact on the tumour antigenic landscape remains poorly understood. Given the low mutational burden of DMG, an expanded search beyond neoantigens to include epigenetically dysregulated tumour-associated antigens (TAAs) is critical for advancing antigen-specific immunotherapies. METHODS: We performed immunopeptidomic analysis of t

Flow Cytometric Lymphocyte Subset Enumeration for Lymphopenia Grading and C-Reactive Protein-Defined Inflammation Discrimination.

BACKGROUND: To determine whether flow cytometric lymphocyte subset enumeration could reproduce conventional absolute lymphocyte count (ALC)-based lymphopenia grading and add information for discriminating C-reactive protein (CRP)-defined inflammation. METHODS: We retrospectively analyzed 2896 hospitalized adults with concurrent complete blood count and flow cytometric lymphocyte subset analysis (T, CD4+ T, CD8+ T, B, and natural killer cells). CRP-defined inflammation was defined as high-sensitivity CRP ≥ 10 mg/L. The 974 patients with measured CRP constituted the prespecified primary discrim

NKG2D overexpression potentiates CAR-T cells through costimulation by tumor NKG2D ligands.

Chimeric antigen receptor (CAR)-T cell therapy has achieved remarkable success in hematologic malignancies, but durable efficacy remains limited by insufficient CAR-T cell persistence. Here, we show that activated CAR-T cells upregulate multiple NKG2D ligands (NKG2DLs), increasing their susceptibility to natural killer (NK) cell-mediated CAR-T cell cytotoxicity. To address this vulnerability, we engineer CAR-T cells to overexpress full-length NKG2D (CAR-NG). NKG2D overexpression reduces the surface accessibility of endogenous NKG2DLs without decreasing their total protein abundance. In paralle

MTHFD1L-associated branched-chain amino acid degradation transcriptional signature represents a conserved metabolic program in Epstein-Barr virus-associated malignancies.

Epstein-Barr virus (EBV)-associated malignancies encompass multiple tumor types with distinct tissue origins, yet the extent to which they share conserved metabolic programs remains poorly understood. Here, we performed an integrated cross-cancer transcriptomic analysis of nasopharyngeal carcinoma, EBV-associated gastric cancer, and EBV-associated plasmablastic lymphoma to identify conserved metabolic programs, followed by independent cohort validation and single-cell transcriptomic characterization. We identified 103 consensus differentially expressed genes and 72 conserved gene-metabolic pat

Female hormones enhance skin T cell expansion and suppress IFN-γ production.

INTRODUCTION: Estrogen receptors (ERs) regulate immune function, but their expression across immune cell subsets and hormone responsiveness in human skin are not fully understood. METHODS: We used 10-color flow cytometry to quantify ER (ERα, ERβ, and GPER1) expression in peripheral blood mononuclear cells (PBMCs) from healthy donors. We analyzed sex hormone gene expression using public single-cell RNA datasets. Skin immune responses to sodium lauryl sulfate (SLS) were assessed in pre- and post-menopausal women, with or without hormone replacement therapy (HRT). Furthermore, we measured the pr

Astragaloside IV attenuates post-ischemic neuroinflammation associated with Treg-related regulation of CD86/CD206-associated brain myeloid phenotypic responses and PI3K/AKT signaling alterations.

BACKGROUND: Ischemic stroke triggers persistent neuroinflammation, microglial activation, and peripheral immune changes that aggravate secondary brain injury. Astragaloside IV (AS-IV), a major bioactive constituent of Astragalus membranaceus, has neuroprotective and immunomodulatory effects, but its influence on Tregs and CD86/CD206-associated microglial phenotypic responses remains unclear. AIM: This study investigated whether AS-IV attenuates post-ischemic neuroinflammation in association with Treg-related immune responses, PI3K/AKT signaling alterations, and CD86/CD206-associated brain mye

Identification of a novel inflammatory mast cell subpopulation in triple-negative breast cancer.

BACKGROUND: Triple-negative breast cancer (TNBC) is associated with poor therapeutic outcomes, and increasing attention has been directed toward the tumor microenvironment (TME). Mast cells are an important component of the TME, yet their heterogeneity and functional roles in TNBC remain incompletely understood. METHODS: Single-cell RNA sequencing data of TNBC were analyzed using the Seurat pipeline. Mast cells were extracted and re-clustered, and the subpopulations were characterized by functional enrichment, transcription factor, pseudotime, drug sensitivity, and cell-cell communication ana

KLRG1+CD28⁻CD57+ Ag85A-specific CD8+ T cells mark a severity-associated terminal-differentiation imprint in pulmonary tuberculosis.

BACKGROUND: Pulmonary tuberculosis (PTB) is characterized by prolonged host-pathogen interaction, chronic granulomatous inflammation, and sustained exposure to Mycobacterium tuberculosis (Mtb) antigens. How Mtb antigen-specific CD8+ T cell differentiation states relate to clinical disease activity and severity remains incompletely defined. This study investigated the clinical relevance of a terminal differentiation-associated phenotype within Ag85A-specific CD8+ T cells across clinically defined states of Mtb infection and disease. METHODS: A total of 124 participants were enrolled, including

Broad systemic T cell immunity to Salmonella enterica serovar Typhi in U.S. healthy adults.

INTRODUCTION: T cell mediated immunity (T-CMI) is an important component of effective host immunity against intracellular pathogens such as Salmonella enterica serovar Typhi (S. Typhi; ST). Baseline T-CMI responsiveness to S. Typhi has been previously described in healthy individuals, likely reflecting cross-reactive responsiveness to other Enterobacteriaceae within the gut microbiota. Interestingly, T cell baseline responsiveness has been associated with protection or susceptibility in a human challenge model following exposure to wild-type (wt) ST. To better understand the contribution of th

Pimozide enhances antigen cross-presentation and promotes cytotoxic CD8+ T cell immunity by stimulating dendritic cells.

The activation of cytotoxic CD8+ T cells requires antigen presentation in the context of major histocompatibility complex class I (MHC-I). While endogenous antigens can directly access this pathway, the priming of CD8+ T cells against exogenous antigens-such as those derived from tumors or internalized pathogens-depends on dendritic cell (DC)-mediated antigen cross-presentation. Strategies capable of enhancing DC cross-presentation are therefore of considerable interest for the development of next-generation immunotherapies. Here, we demonstrate that pimozide, a clinically approved antipsychot

eVLP-mediated CREM editing enhances cytotoxicity of bicistronic IL-15-armored CD19/BCMA dual-target CAR-NK cells.

Chimeric antigen receptor (CAR)-engineered natural killer (NK) cells represent a promising platform for cancer immunotherapy; however, optimization of CAR architecture and additional engineering strategies remain necessary to improve their functional activity. In this study, we generated CD19/BCMA dual-target CAR constructs based on tandem and bicistronic configurations with or without membrane-bound interleukin-15 (mbIL-15), and further evaluated engineered virus-like particle (eVLP)-mediated CREM editing as an additional strategy for CAR-NK cell engineering. Four CAR configurations were eval

Identification of Akkermansia muciniphila as a candidate gut commensal involved in the pathogenesis of rheumatoid arthritis.

OBJECTIVE: The "mucosal origins hypothesis" posits that mucosal sites, such as the gut, are initial sites where immune responses first develop that may, months to years later, result in clinical rheumatoid arthritis (RA). Our goal here was to conduct a discovery-based, hypothesis-generating study using samples from a relatively small number of patients to implicate previously unrecognized highly IgA-coated commensal flora that may have a role in RA pathogenesis (primary outcome). The secondary supporting outcomes were to determine whether HLA-DR-presented microbial peptides from such flora cou

Systemic immune activation coexists with compartment-specific regulatory enrichment in matched blood, peritumor, and tumor tissue from advanced oral squamous cell carcinoma.

BACKGROUND: Oral squamous cell carcinoma (OSCC) is the most prevalent form of oral cancer and is characterized by a poor prognosis and frequent immune evasion. While the tumor microenvironment (TME) plays a central role in modulating responses to therapy, its relationship with systemic immune signatures remains poorly understood. METHODS: We performed high-dimensional immune profiling in matched peripheral blood, adjacent non-neoplastic peritumor mucosa, and tumor tissue from Human Papillomavirus (HPV)-negative, treatment-naïve patients with advanced OSCC. Soluble mediators were quantified by

Spatial immune ecosystems and therapy resistance in large B-cell lymphoma: from single-cell multi-omics to immunotherapy-matched validation.

Large B-cell lymphomas, particularly diffuse large B-cell lymphoma (DLBCL), provide a clinically important model in which tumor-intrinsic programs, immune recognition, effector-cell fitness, and tissue architecture jointly shape response and resistance to immunotherapy. Although molecular classification has refined the understanding of lymphoma biology, relapse after chemoimmunotherapy, chimeric antigen receptor (CAR) T-cell therapy, CD20×CD3 bispecific antibodies, antibody-drug conjugates (ADCs), and immune-combination strategies cannot be explained by genetics or target-antigen expression al

CAR-T cell-induced cytokine release syndrome is rapidly alleviated by Tripterygium glycosides: a case series and laboratory examination study.

BACKGROUND: Cytokine release syndrome (CRS) is a life-threatening complication of chimeric antigen receptor T cell (CAR-T) therapy. Macrophages/monocytes are mediators of CRS. Tripterygium glycosides is an immunomodulator, which could inhibit macrophages/monocytes in animal models. METHODS: Two patients with relapsed and refractory hematological malignancies developed CRS after receiving CAR-T therapy. They received short-term Tripterygium glycosides orally. RESULTS: Both patients showed rapid mitigation of fever with evidently decrease in elevated inflammatory cytokines within 72 hours. The

A positive feedback loop of kynurenine-AhR-EMT upregulates tryptophan metabolic enzymes via epigenetic modification to facilitate immune escape in triple-negative breast cancer.

BACKGROUND: Triple-negative breast cancer (TNBC) is prone to immune escape, and the synergistic mechanism of kynurenine, AhR, and EMT remains unclear. METHODS: TNBC cell lines and 4T1 orthotopic breast cancer mice were used for molecular, cellular, and in vivo intervention experiments. RESULTS: Kynurenine induced EMT in an AhR-dependent way, promoted cell migration and invasion, upregulated CCL2/CCL22, and inhibited the function of CD8⁺GrB⁺ T cells. Snail overexpression promotes H3K4me3 histone modification at IDO1 and TDO2 promoters to epigenetically amplify tryptophan metabolic enzyme expr

Distinct time-dependent S. Typhi-specific CD8+ T cell programs are associated with clinical outcome following Ty21a immunization and wild-type S. Typhi challenge in humans.

INTRODUCTION: CD8+ T cells are essential components of the adaptive immune system that play a critical role in determining the clinical severity of typhoid disease by directly killing Salmonella enterica serovar Typhi (S. Typhi)-infected cells, producing cytokines, chemokines, and establishing memory responses that have been linked to clinical outcome in a controlled human infection model (CHIM) of typhoid. Despite decades of extensive research, the role of memory CD8+ T (TM) cells and its subsets as correlates of protection against S. Typhi infection and typhoid disease remains incompletely u

Pre-transplant bone marrow lymphocyte subsets predict outcome after allogeneic stem cell transplantation.

INTRODUCTION: Recent evidence has demonstrated that both donor- and recipient-derived immune cell subpopulations play a crucial role in various immune responses following allogeneic hematopoietic stem cell transplantation (HSCT). In this retrospective single center analysis, we investigated whether different lymphocyte subpopulations in the bone marrow (BM) of the recipient prior to HSCT might have an impact on outcome parameters after HSCT. METHODS: BM aspirates of 140 patients > 18 years of age receiving a HSCT between 2008 and 2022 were analyzed. BM aspiration was performed within 3 months
