INTRODUCTION: miR-155 regulates diverse inflammatory responses; however, its role in neurotropic virus-induced CNS inflammation, particularly in specific immune cell subsets, remains unclear. This study aimed to identify the immune cell population in which miR-155 is required for protection against Japanese encephalitis virus (JEV) and to elucidate the underlying mechanisms.
METHODS: Conditional miR-155 knockout mice with cell type-specific deletion in myeloid cells, dendritic cells (DCs), T cells, or B cells were generated using Cre-LoxP technology. Following JEV infection, we assessed survi

BACKGROUND: Chimeric antigen receptor T cell therapy has limited efficacy in the treatment of glioma due to the blood-brain barrier and T cell exhaustion. Enhancement with rabies virus glycoprotein (RVG29) has shown improved brain-related efficacy.
METHODS: In this study, we developed CD70R CAR-T cells by modifying anti-CD70 CAR-T cells with RVG29 and tested their tumor-killing ability in vitro.
RESULTS: Transcriptomic profiling via RNA-seq revealed the activated signaling pathways in modified CD70R CAR-T cells. These cells displayed effective in vitro cytotoxicity against CD70+; glioma cell

Rabies is one of the oldest diseases recognized by humanity, caused by the rabies virus (RABV). This zoonosis has a worldwide distribution and still causes the death of approximately 60,000 people a year. Despite advances in research, there are still gaps in knowledge about how RABV is maintained in the wild, especially aspects related to its biology and its mechanisms for adapting to its hosts. In recent years, reports of the isolation of RABV in different bat species, with varying feeding and behavioral habits, have increased public health concerns. The possibility of these animals developin

Rabies virus (RABV) is a zoonotic pathogen with an almost 100% case-fatality rate and limited post-exposure prophylaxis options. mRNA-based antigen delivery represents a promising approach but requires further optimization. We prepared a cationic lipid nanoparticle (LNP) delivery system using the thin-film hydration method and characterized its physicochemical properties and delivery efficiency. An mRNA encoding a fusion antigen comprising rabies virus glycoprotein (G) and herpes simplex virus type 2 infected cell polypeptide 35 (ICP35), lacking canonical 5' and 3' untranslated region sequence

Progressive multifocal leukoencephalopathy (PML) is a rare brain infection caused by JC polyomavirus (JCPyV) in immunocompromised individuals, for which no effective antiviral therapy exists. Diverse cellular therapy approaches using polyomavirus-specific T cells (PyVST) has emerged as a therapeutic option. We aimed to evaluate the real-world effectiveness of PyVST in patients with PML. We retrospectively analyzed 64 patients from 13 centers who received PyVST alongside standard care. Conditions predisposing to PML included mainly hematologic malignancies (33/64, 51.6%). One-year survival was

BACKGROUND: Progressive multifocal leukoencephalopathy (PML) is a fatal demyelinating disease caused by JC polyomavirus (JCV) that affects immunosuppressed individuals and has no approved therapy. Here we evaluate the safety and efficacy of third-party, off-the-shelf, partially HLA-matched BK virus-specific T cells (BK-VST) in patients with PML.
METHODS: We conducted a single-center phase 2 study where 37 patients received most closely HLA-matched BK-VST at 2.0 x 105 T cells/kg, with additional infusions permitted for subjects who did not achieve a complete response. Twenty-three patients had

BACKGROUND AND OBJECTIVES: Bispecific antibodies (BisAbs) have transformed the management of relapsed and refractory multiple myeloma (MM), achieving high response rates in heavily pretreated patients. However, these therapies induce profound immune perturbation, including plasma cell aplasia, hypogammaglobulinemia, and T-cell exhaustion, predisposing patients to serious infections. Progressive multifocal leukoencephalopathy (PML) has rarely been reported in this setting.
METHODS: We report on 5 patients with MM who developed PML following BisAbs therapy. Clinical presentation, prior treatmen

Programmed cell death protein 1 (PD-1) is expressed by T cells during progressive multifocal leukoencephalopathy (PML), a life-threatening brain disease caused by the human-only JC polyomavirus (JCPyV). PD-1 checkpoint immunotherapy has benefited some PML patients, but reasons for its variable outcomes are unclear. Using mouse polyomavirus (MuPyV), we show that PD-1 loss acts in a brain-autonomous manner to increase the magnitude of brain-infiltrating CD4+ and CD8+ T cells and the function of virus-specific CD8+ T cells; in concert, brain virus levels decline and neuroinflammation increases. INTRODUCTION: Available work from North America indicates that same-sex attracted (SSA) individuals enjoy aspects of rural life but nonetheless report encountering homophobia and experiencing isolation from SSA networks. The experience of prejudice and social isolation are often associated with psycho-social distress among the general population of same-sex attracted individuals. Little is known of how SSA women experience life in rural areas of Australia and how this influences their psycho-social wellbeing.
METHODS: This was a small-scale qualitative study using guided interviews to explore

BACKGROUND: Substantial outbreaks of scrub typhus, coupled with the discovery of this vector-borne disease in new areas, suggest that the disease remains remarkably neglected. The objectives of this study were to map the contemporary and potential transmission risk zones of the disease and to provide novel insights into the health burden imposed by scrub typhus in southern China.
METHODS: Based on the assembled data sets of annual scrub typhus cases and maps of environmental and socioeconomic correlates, a boosted regression tree modeling procedure was used to identify the environmental niche

Integrated single-cell RNA sequencing, spectral flow cytometry, immune-receptor repertoire profiling, and plasma proteomics revealed coordinated peripheral immune remodeling during acute scrub typhus. Patients exhibited expansion of cytotoxic, clonally amplified CD8+ T cells with exhaustion-associated features, depletion and apoptosis of CD4+ T cells, B-cell contraction with enhanced plasmablast/plasma-cell differentiation, and inflammatory myeloid remodeling. CD14+CXCR6+ and CD14+GNLY+ myeloid-cell frequencies positively correlated with bacterial burden, defining prominent immune features of

BACKGROUND: Adolescent girls and young women (AGYW) in sub-Saharan Africa are disproportionately affected by Chlamydia trachomatis (CT) infections, a leading cause of pelvic inflammatory disease, infertility and increased HIV susceptibility. CT-induced genital inflammation disrupts mucosal barriers and activates HIV target cells, compounding immunological risks. We investigated CT infection in AGYW from two South African regions, focusing on genital inflammation and immune activation.
METHODS: An observational cohort of 298 sexually active AGYW (aged 16-22 years) from Cape Town and Johannesbu

CD4 T cells are essential for protective immunity against Chlamydia in the female reproductive tract (FRT), yet the characteristics of protective mucosal effector CD4 T cells remain poorly defined. We previously identified the transcription factor BHLHE40 as a key regulator of polyfunctional effector CD4 T cell differentiation during Chlamydia infection. Here, we identify the chemokine receptor CXCR6 as a marker of these protective T cells. Following intravaginal Chlamydia muridarum infection, Bhlhe40-deficient mice exhibited reduced frequencies of CXCR6+ CD4 T cells that correlated with impai

Immune responses to tick-transmitted Borrelia burgdorferi (Bb) have not been characterized in vivo. We analyzed the reservoir host's local and systemic immune responses to Bb during tick feeding. C3H/HeN mice were challenged via infected or uninfected nymphal Ixodes scapularis ticks or by subcutaneous injection of cultured multi-strain Bb. Skin and spleen tissues were evaluated by flow cytometry, serum was evaluated by cytokine proteome array, and all data were analyzed by comparison between each of the three challenged groups against the subcutaneously inoculated PBS control. Lyme disease (LD), a multifaceted condition caused by Borrelia burgdorferi (Bb), remains poorly understood, particularly regarding metabolic pathways. This study aimed to evaluate the role of glycolysis-related genes (GRGs) in LD pathogenesis and identify key genes and mechanisms relevant to diagnosis and therapy. Differentially expressed GRGs were identified and further analyzed by correlation analysis and Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses. Key genes were screened using least absolute shrinkage and selection operator (LASSO) and support vector machi

The role of epigenetic regulation in HIV latency remains incompletely understood. We show that histone deacetylase 3 (HDAC3) inhibits trans-activator of transcription (Tat)-mediated HIV transcription through histone decrotonylation (HDCR), independent of deacetylase activity. Chemical biology approaches identified selective HDCR inhibitors (HDCRis) that reverse HIV latency with minimal impact on other histone acylations. Although HDAC2, HDAC3, and HDAC8 exhibit HDCR activity, genetic and chemical studies reveal that the HDCRi citarinostat is selective for HDAC3 and HDAC8. Molecular docking sug

Tumor spatial architecture significantly influences immune responses, but the impact of regional variations on T cell exhaustion and clonality in human papillomavirus (HPV)-related cervical squamous cell carcinoma (CESC) remains poorly understood. Using single-cell RNA sequencing (scRNA-seq, n = 13) and TCR sequencing (scTCR-seq, n = 9), this study profiled T cells from paired tumor core and edge samples of patients with CESC. This was supplemented by bulk RNA-seq data (n = 41), TCGA datasets from three cancer types (CESC, head and neck squamous cell carcinoma, and lung squamous cell carcinoma

BACKGROUND: People with human immunodeficiency virus (HIV) receiving antiretroviral therapy (ART) have increased risks of non-AIDS comorbidities. Growth differentiation factor 15 (GDF-15) is a mitokine released upon mitochondrial stress and is a validated aging biomarker. Herein, we assessed associations between plasma GDF-15 levels, inflammation, and HIV reservoir markers in ART-treated people with HIV (PWH).
METHODS: Blood samples were collected from 78 ART-naive, 140 ART-treated PWH (median ART duration, 15.6 years) and 83 uninfected control participants. GDF-15 and markers of inflammation

Oral tenofovir-based HIV-pre-exposure prophylaxis (PrEP), approved for use in preventing human immunodeficiency virus (HIV) infection, has been implicated in altering the immune landscape. We leveraged longitudinal samples from heterosexual couples with the male partner living with HIV and the female partner without HIV initiating PrEP after enrollment. Applying high parameter flow cytometry to cells from ectocervical and vaginal biopsies and peripheral blood samples, we found a decrease in activated ectocervical CD8+ and CD4+ T cell subsets after oral PrEP exposure. Our data expand the unders
