Vaccination coverage against COVID-19 among people living with HIV infection in the Czech Republic.

OBJECTIVES: The pandemic of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has affected hundreds of millions of individuals globally. Vaccination has become a promptly effective preventive procedure. People living with HIV (PLHIV) are in increased risk of many infections including COVID-19. We aimed to analyse the first phase of vaccination (January 2021-June 2022) in the Czech Republic in PLHIV focusing on vaccination coverage, vaccine hesitancy and breakthrough infections. METHODS: In this single-centre, open-label, prospective study we have enrolled 1,758 PLHIV with median ag

HDAC6 Promotes Host Defense Against Chlamydial Lung Infections by Regulating M2-Th2 Responses.

Histone deacetylase 6 (HDAC6), a member of the class IIb HDAC family, plays a crucial role in epigenetic regulation and cytoskeletal dynamics, while participating in host anti-infective immune responses. However, its precise functions and mechanisms during Chlamydia muridarum (C. muridarum) infection remain incompletely defined. Our study demonstrated that C. muridarum respiratory infection upregulates HDAC6 expression at the infection site and in immune organs. Comparative analysis of wild-type (WT) and HDAC6-deficient (HDAC6-/-) mice in this infection model revealed that HDAC6 deficiency exa

Molecular Regulation of HIV-1 Expression and Persistence Across Diverse Cellular Reservoirs.

Despite the remarkable success of antiretroviral therapy (ART) in suppressing human immunodeficiency virus type 1 (HIV-1) replication, viral persistence remains a major barrier to cure. This persistence is sustained by heterogeneous cellular reservoirs in which viral expression is tightly regulated by host-dependent molecular mechanisms. Beyond the canonical cluster of differentiation 4 (CD4+) T-cell reservoirs, HIV-1 establishes long-lived infection in myeloid cells, glial populations within the central nervous system (CNS), and additional non-canonical cellular niches, each characterized by

Transcriptomic Profiling Identifies TALAM1 and LINC00702 as HIV-1-Responsive lncRNAs in Microglia.

Microglia, the resident macrophages of the central nervous system (CNS), serve as the primary reservoir of HIV-1 in the brain and play a crucial role in the development of HIV-1-associated neurocognitive disorders (HAND). While long non-coding RNAs (lncRNAs) have emerged as essential regulators of HIV-1 replication in T cells and macrophages, their role in microglia remains poorly understood. Here, we performed RNA sequencing of polyadenylated transcripts from a human microglial cell line exposed to HIV-1 infection or TNF-α stimulation to investigate transcriptional responses and identify lncR

PIK3CA mutant cervical cancer is selectively suppressed by PI3Kα inhibition (Alpelisib/BYL-719 and Inavolisib/GDC-0077) and cooperates with HPV directed T cell therapy.

Cervical cancer is largely driven by human papillomavirus (HPV) infection, yet clinically actionable molecular subtypes and effective targeted therapies remain limited. Here, we define biologically and clinically relevant subtypes and evaluate targeted therapeutic strategies. Analysis of public datasets, coupled with functional studies in cervical cancer cell lines and immune assays, identified three subtypes: (I) PIK3CA wild-type without YAP1 amplification, (II) PIK3CA-mutant without YAP1 amplification, and (III) PIK3CA wild-type with YAP1 amplification. Notably, YAP1 amplification is associa

Antiretroviral therapy interferes with pseudovirus neutralization assays while Gag-specific T-cells influence mRNA vaccine outcomes in HIV patients.

People living with HIV infection (PLWH) often have attenuated responses to infections and vaccination. This study aimed to better understand how HIV-associated inflammation and chronic T-cell activation influenced the immune responses to mRNA vaccination or neutralization assay analyses. PLWH on ART or healthy donor controls were analyzed using systems serology and viral T-cell phenotyping to Spike or HIV-1 Gag peptide stimulation after primary mRNA COVID-19 vaccination. Neutralization assays using a lentiviral pseudovirus construct were compromised by the presence of integrase strand transfer

Impact of SARS-CoV-2 vaccination on CD4+ and CD8+ T lymphocyte profiles in an HIV-positive and HIV-negative female cohort.

INTRODUCTION: The COVID-19 pandemic led to rapid global vaccine deployment, especially among high-risk groups, such as individuals living with HIV. Data are limited, however, on the immunologic effects of SARS-CoV-2 vaccination-specifically, on CD4+ and CD8+ T lymphocyte levels-in HIV-positive women in South Africa, a population with high HIV prevalence. METHODS: This prospective cross-sectional study included 40 women (aged 14-42 years) admitted to a South African tertiary-care hospital, stratified by HIV and SARS-CoV-2 vaccination status. Flow cytometry (BD Multitest [BD Biosciences]) was u

Acute HSV-2 infection with a low-passage clinical strain: Analysis of viral tropism, immunity, and disease manifestations in a dual animal model system.

Herpes simplex virus type 2 (HSV-2) is the primary causative agent of genital herpes and represents a substantial global health concern. Although laboratory-adapted HSV-2 strains have been widely studied, the natural virological and pathogenic features of clinical isolates remain less well understood. In this study, we characterized HSV-2/KM-1, a low-passage clinical isolate (passage 6) from China, using both guinea pig and mouse models. Intravaginal inoculation resulted in acute genital lesions, viral shedding (which peaked between 2 and 5 days post-infection (DPI), and systemic symptoms. Vir

Systematic discovery of pro- and anti-HIV host factors in primary human CD4+ T cells.

Host factors that promote or restrict human immunodeficiency virus (HIV) infection in human CD4+ T cells have not been comprehensively identified. We employed orthogonal genome-wide CRISPR activation (CRISPRa) and CRISPR knockout screens in primary CD4+ T cells to discover pro- and anti-HIV host factors systematically. Secondary pooled screens and individual perturbations validated high-confidence hits and revealed diverse mechanisms of action. CRISPRa uncovered multiple potent antiviral factors, including PI16, PPID, SHISA3, and ITM2A. PI16 interacts with host factors involved in HIV fusion a

Immunological evaluation of vaccines to prevent Chlamydia trachomatis infection using the human in vitro MIMIC system.

The Modular Immune In vitro Construct (MIMIC) system is an established laboratory-based methodology that reproduces in vitro the human in vivo T-cell immune response. In this study, we utilized the MIMIC system to establish a dendritic cell and CD4+ T-cell co-culture model that recapitulates T helper cell responses to Chlamydia trachomatis infection in humans. We demonstrate that C. trachomatis infection induces C. trachomatis-specific IFN-γ-producing CD4+ T-cell responses, with IFN-γ as the dominant cytokine, accompanied by significant production of TNF-α, IL-13, IL-8, and IL-6. Expansion of stemlike HIV-specific CD8+ T cells and limited viral epitope diversity characterize durable posttreatment control of HIV.

Individuals who durably control HIV after cessation of antiretroviral therapy (ART) are of great interest for HIV cure efforts, but the correlates of posttreatment control remain incompletely understood. Here, we show across a diverse cohort of posttreatment controllers (PTCs), posttreatment noncontrollers (NCs), and spontaneous elite controllers (ECs) that robust expansion of HIV-specific CD8+ T cells and reduced viral reservoir diversity were strongly associated with durable (>1 year) viral suppression after ART cessation. In addition, HIV-specific CD8+ T cells in PTCs exhibited a stemlike m

A New Type of Nonsuppressible Viremia Produced by HIV-Infected Macrophage.

BACKGROUND: HIV-1 RNA typically declines rapidly after initiation of antiretroviral therapy (ART), often reaching undetectable levels within a few weeks and remaining undetectable by standard assays. However, some patients receiving ART have persistent nonsuppressible viremia (NSV) that does not respond to treatment optimization or intensification. NSV can emerge at the time of ART initiation (primary NSV) or after being ART suppressed (secondary NSV). Here, we examine mechanisms producing primary NSV in 4 people undergoing ART. METHODS: Blood samples were collected from 4 participants with a

SARS-CoV-2 infection disrupts the immune control status in a male HIV-1 elite controller.

OBJECTIVE: To report a case of an HIV elite controller (EC) who developed disease progression following SARS-CoV-2 infection, characterized by the breakdown of prior immune control. CASE PRESENTATION: We report a case of a Chinese male EC with HIV-1 CRF01_AE and protective HLA-B*57:01, who had sustained undetectable viral load and stable CD4+ T-cell counts for over 10 years. Following SARS-CoV-2 infection, he developed persistent HIV viral rebound (peaking at 4430 copies/ml) for eight months, accompanied by a profound CD4+ T-cell decline to 174 cells/µl, and subsequently initiated antiretrovi

Wnt7a mediates the Wnt/β-catenin signaling pathway to influence ferroptosis in T lymphocytes of HIV-infected individuals.

Human Immunodeficiency Virus (HIV) infection is characterized by the depletion of human CD4 + T cells, in which ferroptosis has been identified as a contributing mechanism. Dysregulation of the canonical Wnt/β-catenin signaling pathway is strongly associated with HIV infection. Our findings demonstrate that Wnt7a activates this pathway. Subsequently, β-catenin translocates to the nucleus, where it interacts with the transcription factor 4 (TCF4) to regulate ferroptosis in T lymphocytes from HIV-infected individuals. Analysis of peripheral blood mononuclear cells (PBMCs) from HIV-infected indiv

Targeting PD-1+ T cells with chimeric antigen receptors to reduce the HIV reservoir.

The unique ability of chimeric antigen receptor (CAR) T cells to infiltrate tissues is revolutionizing our perspectives for tackling severe, refractory and otherwise untreatable diseases. In HIV, CAR-T cells have been designed to target viral biomarkers, with limited success so far. Here, we investigated the possibility of redirecting CAR-T cells against a cellular biomarker of the HIV reservoir, the programmed cell death protein 1 (PD-1). We designed two second-generation 4-1BB-CARs using the scFv of either a blocking (bPD1-CAR) or a nonblocking (nbPD1-CAR) anti-PD-1 monoclonal antibody. The

Pre-Human Immunodeficiency Virus (HIV) infection Th17 CD4+ T cells as predictors of early HIV disease progression.

Interleukin-17-producing T helper (Th17) CD4+ T cells are highly susceptible to HIV infection and are depleted early in people living with HIV. Here, we investigated whether systemic Th17 cell levels prior to HIV infection are associated with subsequent HIV disease progression. We analyzed archived cryopreserved peripheral blood mononuclear cells (PBMCs) collected within one year prior to HIV acquisition from participants enrolled in a South African cohort (HIV Vaccine Trials Network [HVTN] 503; n = 35) and an East African cohort (Partners Pre-exposure Prophylaxis/Couples' Observational Study

Cannabidiol prevents mucosal HIV-1 transmission by targeting Langerhans cells, dendritic cells, macrophages and T-cells.

HIV-1 transmission depends on the structure and immune cell composition of mucosal epithelia. Transmission mechanisms involve direct infection of CD4+ T-cells or macrophages, and indirect viral transfer to CD4+ T-cells from Langerhans cells (LCs) or dendritic cells (DCs). LCs-mediated HIV-1 transfer is inhibited by the neuropeptide calcitonin gene-related peptide (CGRP), due to upstream activation in LCs of the transient receptor potential vanilloid 1 (TRPV1) ion channel. Herein, we investigated the potential anti-HIV-1roles of cannabidiol (CBD), the non-psychoactive compound in marijuana, whi

Therapeutic HIV-1 Tat vaccination promotes durable immune reconstitution and reservoir reduction in ART-treated adults with clade C infection: a 12-year follow-up study.

INTRODUCTION: Antiretroviral therapy (ART) suppresses HIV replication but fails to eradicate viral reservoirs or fully restore immune competence. Therapeutic vaccination targeting HIV-1 Tat, a key viral protein persistently expressed also during ART, may enhance immune reconstitution and limit reservoir maintenance. Long-term data in individuals infected with HIV-1 clade C, particularly in sub-Saharan Africa, are lacking. METHODS: We conducted a 12-year extended follow-up of 161 ART-treated adults previously enrolled in a randomized, placebo-controlled phase 2 trial of therapeutic HIV-1 Tat v
