Single-cell characterization of the CD8+ T cell landscape and repertoire in HIV-1 Infection following antiretroviral therapy.

Although antiretroviral therapy (ART) effectively suppresses HIV-1 viremia, immune dysfunction often persists. Here, we integrated single-cell RNA and T cell receptor (TCR) sequencing data to delineate the alterations of CD8⁺ T cells. Based on cross-sectional samples from different individuals (healthy donors, treatment-naïve individuals, and ART-treated individuals), we found ART promoted the transition of CD8⁺ T cells from exhausted toward naïve and memory-like subsets. Weighted gene co-expression network analysis identified distinct modules, including three (SM2-SM4) highly expressed in tre

Synergistic Interaction Between HPV-16 E7 Oncoprotein and Severe Vitamin A Deficiency in Regulating Adaptive Immunity in a Preclinical Cervical Cancer Model.

Persistent infection with High-Risk Human Papillomavirus (HR-HPV), particularly HPV-16, is the main driver of cervical intraepithelial neoplasia (CIN) and cervical cancer (CC). However, HR-HPV infection alone is insufficient for malignant progression and nutritional cofactors such as Vitamin A deficiency, may influence cervical neoplasia progression through immune dysregulation. This study aimed to explore the interaction between HPV-16 E7 oncoprotein expression and severe Vitamin A Deficiency (VAD), as well as the effects of a Vitamin A-Normal Diet Reintegration (NDR) on adaptive immune regul

Pharmacological and genetic modulation of IL-32 expression in intestinal epithelial cells does not impact HIV-1 outgrowth in co-cultured CD4+ T-cells.

BACKGROUND: The crosstalk between intestinal epithelial cells (IEC) and CD4+ T-cells is essential for the maintenance of mucosal homeostasis, but its role in governing HIV-1 latency versus reactivation in gut-homing/resident T-cells remains poorly documented. We previously demonstrated that the Th17-lineage cytokine IL-17A transcriptionally reprograms IEC for promoting viral outgrowth in CD4+ T-cells of antiretroviral therapy (ART)-treated people with HIV-1 (PWH). These effects coincided with the downregulation of IL-32, a cytokine with documented antiviral properties and identified as a marke

Monocyte Dll4 Contributes to Systemic Inflammation in People Living With HIV.

BACKGROUND: In people living with HIV (PLWH), persistent systemic inflammation drives comorbidities, including cardiovascular and cerebrovascular diseases. Monocyte-mediated inflammation, rather than T-cell activation, is a major contributor to chronic inflammation. However, the underlying mechanisms remain elusive. Here, we investigated the role of Dll4 (delta-like 4)-Notch signaling in monocyte-mediated inflammation. METHODS: In clinical specimens, we quantified monocyte mDll4 (membrane-bound Dll4) expression and plasma levels of exDll4 (extracellular Dll4). We also analyzed the association

Spatial cellular order underlies locally-confined mechanisms of immune resistance in oropharyngeal cancer.

Oropharyngeal squamous cell carcinomas (OPSCCs) frequently result from oncogenic human papilloma virus (HPV) infections (HPV-OPSCC). The mechanisms underlying effective immune escape, despite abundant viral antigens, are incompletely understood. Here, we performed single-cell spatial gene expression profiling of HPV-OPSCC to characterize cellular organization and mechanisms of immune resistance. We describe distinct tumor-parenchymal immune foci that differ in cytokine expression, spatial location, immune cell infiltration and cancer cell states. Furthermore, immune foci display profound diffe

Metabolic reprogramming of CD4⁺ T cells by Zaprinast induces HIV-1 latency reversal ex vivo.

BACKGROUND: HIV-1 latency and persistence of viral reservoirs within memory CD4+ T cells remain a fundamental obstacle to achieving a cure despite suppressive antiviral treatments. HIV-1 persistence is sustained by the dynamic interactions between viral regulatory mechanisms and the host cellular environment. At the intersection between immunometabolism and virology, the quiescent metabolic profile of resting CD4+ T cells, defined as the balance between oxidative phosphorylation (OXPHOS) and aerobic glycolysis, supports the long-term maintenance of latent viral reservoirs. Existing "Shock and

Dynamics of HIV reservoirs following repeated anti-SARS-CoV-2 vaccination.

Due to persistent immune dysfunction, People with HIV (PWH) are at increased risk of more severe infections with SARS-CoV-2 and hospitalizations. In several countries, they were listed as a priority group for receiving anti-SARS-CoV-2 vaccines when these vaccines were in scarce supply. These prophylactic vaccines do not completely prevent infections in vaccinated individuals, especially with a heterologous vaccine strain. However, they persistently reduce the severity of breakthrough infections, hospitalization rates and deaths. Although antiretroviral therapy (ART) suppresses HIV replication

Central Nervous System T-cell immune architecture, and not HIV burden, tracks with cognition under long-term viral suppression.

Despite effective antiretroviral therapy, HIV persists in the central nervous system (CNS) and may contribute to neuroinflammation and cognitive impairment. How viral persistence, immune responses, and regional CNS T-cell architecture relate to cognitive functioning remains unclear. We performed a cross-sectional, multi-compartmental immune-genomic study in 12 people with HIV on long-term viral suppression enrolled in the Last Gift rapid autopsy program. Quantitative HIV reservoir measures (total-episomal DNA, unspliced-multiply spliced RNA) and paired αβ T-cell receptor repertoire (TCRR) sequ

Long-Acting Cabotegravir/Rilpivirine Reduces Immune-Activation and -Senescence in People With HIV With CMV Co-Infection.

Despite the effectiveness of combined antiretroviral therapy (ART), HIV infection remains a chronic condition characterised by persistent inflammation and immune activation, likely associated with viral persistence and other factors such as cytomegalovirus (CMV) co-infection. Long-acting (LA) injectable formulations, a newer class of ART with improved and sustained bioavailability, may help modulate the HIV-associated immunoinflammatory state. Therefore, we analysed dynamic changes in lymphoid immune-activation and -senescence markers in people with HIV (PWH) switching to LA injectable caboteg

Preventive and therapeutic efficacy of mRNA/LNP vaccines encoding HPV16 E6 and E7 in an early-intervention HPV tumor mouse models.

Persistent infection with high-risk human papillomavirus (HPV) drives the development of cervical and other anogenital cancers, with limited therapeutic options for early-stage disease and precancerous lesions. There is a critical unmet need for effective therapeutic vaccines capable of inducing durable immune responses to eliminate early tumor cells and prevent disease progression. In this study, we developed lipid nanoparticle (LNP)-formulated mRNA vaccines encoding HPV16 E6 and E7 oncoproteins and evaluated their efficacy in a murine model using a low tumor inoculum (5,000-10,000 TC-1 cells

CD4/CD8 ratio is associated with structural reorganization of vaccine-induced immune responses in people living with HIV.

BACKGROUND: Despite effective antiretroviral therapy and sustained viral suppression, people living with HIV (PLWH) continue to exhibit persistent immune activation and low-grade inflammation. Although the CD4/CD8 ratio is routinely monitored as a marker of immune health, the mechanisms underlying residual immune dysregulation remain incompletely understood. METHODS: Peripheral blood mononuclear cells from PLWH and HC were collected before and one month after two doses of mRNA vaccination and analyzed by single-cell RNA sequencing with paired TCR profiling. Differential gene expression (DGE)

Research progress on HIV-1 structural proteins and antiviral therapies.

Human Immunodeficiency Virus (HIV), including HIV-1 and HIV-2, is a retrovirus that causes Acquired Immunodeficiency Syndrome (AIDS). HIV primarily targets the human immune system, especially CD4+ T lymphocytes, leading to progressive immune system dysfunction and rendering patients susceptible to severe opportunistic infections and malignancies. HIV-1 accounts for the majority of global HIV infections, and most structural, mechanistic, and therapeutic studies have been conducted in HIV-1 systems. The structural proteins Gag, Pol, and Env of HIV-1 play a pivotal role in the viral life cycle, m

Bioactive enhanced adjuvant chemokine oligonucleotide nanoparticles (BEACONs) for mucosal vaccination against genital herpes.

Genital herpes, caused by herpes simplex virus 2 (HSV-2), remains a prevalent sexually transmitted infection with no available vaccine. Effective local immunity, mediated in part by tissue-resident memory T cells (TRM cells) and luminal antibodies, provides immediate viral control. Here, we developed bioactive enhanced adjuvant chemokine oligonucleotide nanoparticles (BEACONs) formed via electrostatic interactions between CpG oligodeoxynucleotides (CpG ODNs) and the chemokine CXCL9. This adjuvant enhances antigen-presenting cell engagement and innate immune signaling, promotes CD8 T cell recru

Generation and evaluation of an EcoHIV mouse model to study HIV-1 integration and expression.

Antiretroviral therapy suppresses HIV replication but does not purge the latent reservoirs, which remains a major barrier to cure. There is an urgent need to establish a reservoir in a mouse model for investigation of viral replication and clearance. We established an EcoHIV infection model in conventional mice by replacing HIV-1 gp120 protein with an ecotropic murine leukemia virus envelope to enable entry via the murine mCAT-1 receptor. EcoHIV can infect immunocompetent mice and multiple mutant mouse strains and replicate in CD4+ T cells, macrophages, and immune cells in the brain. We create

Targeting HIV-1 integrase: A review of options, challenges and future directions.

HIV-1 infects CD4+ T-cells, causing immunodeficiency and, if untreated, progression to AIDS. Global antiretroviral therapy scale-up has reduced deaths and new infections by nearly half over the past decade, yet lifelong treatment and emerging drug resistance remain critical challenges. This review provides an overview of the HIV-1 lifecycle, with emphasis on the structure, function, and multiple roles of IN in viral replication. We discuss the development, mechanisms of action, clinical utility, and resistance profiles of current integrase inhibitors, underscoring the need for innovative appro

Type-1 inflammatory imprinting and programmed responsiveness to CD40L enhance Siglec-1-dependent HIV-1 trans-infection by dendritic cells.

Dendritic cells (DC) play a central role in host immunity as they carry environmental cues from sites of infection to the lymphatics, where they subsequently direct appropriate adaptive immune responses. Importantly, this process is exploited by pathogens such as HIV-1, which utilize DC to efficiently facilitate HIV-1 trans-infection of CD4+ T cells. In this study, we show that monocyte-derived DC matured under type-1 proinflammatory conditions, either through exposure to soluble mediators of type-1 immunity or through bystander activation of cytotoxic T cells, display enhanced trans-infection

Case report: A case of HIV seroconversion in a high-risk individual after 35 years of resistance with a CCR5Δ32 heterozygous genotype.

Numerous studies have examined the factors contributing to the variation in disease progression among people living with HIV-1, identifying a range of genetic, immunologic and virologic factors that interact in a complex, multifactorial dynamic. However, investigating individual susceptibility to HIV-1 infection adds an additional layer of complexity. In this observational report, we focus on the case of a man (Case-2021) who previously remained HIV-1 negative despite 35 years of self-reported high-risk sexual behavior with other men while enrolled in the MACS/WIHS Combined Cohort Study (MWCCS

Three immunoregulatory signatures define non-productive HIV infection in stem cell memory CD4+ T cells.

The persistent HIV reservoir constitutes the main obstacle to curing HIV/AIDS disease. Our understanding of how non-productive HIV infections are established in primary human CD4+ T cells during the first round of infection is still incomplete. In this study, we leverage the HIV reporter virus pMorpheus-V5 to delineate cellular expression patterns upregulated in non-productively infected stem cell memory (TSCM) CD4+ T cells. We find that CD4+ TSCM harboring non-productive proviruses display a distinct transcriptomic signature comprising 118 upregulated genes, distinct from that of productively

Predictive value of peripheral lymphocyte subsets in relation to opportunistic infections in people living with HIV/AIDS.

BACKGROUND: Opportunistic infections (OIs) remain a major clinical challenge in people living with HIV/AIDS (PLWHA), and CD4⁺ T-cell counts alone may not fully reflect immune dysfunction. This study aimed to establish predictive cut-off values for lymphocyte subsets to improve the assessment of OIs risk in PLWHA. METHODS: Peripheral blood samples were collected to assess lymphocyte subsets using flow cytometry between August 17, 2021, and April 19, 2023. Correlation analysis was performed to identify lymphocyte subset indicators strongly associated with CD4+ T-cell counts. Receiver operating

Pulmonary Immune Cell Landscape Altered by Exposure to HIV, Schistosoma and Their Combination.

Local immune cell activation and vascular remodelling are characteristic pathogenic features of pulmonary arterial hypertension (PAH). HIV and schistosome infections have been individually associated with PAH. However, whether co-infection with these pathogens has a distinct impact on the development of pulmonary vascular disease remains poorly understood, partly due to the lack of experimental animal models. In a novel non-infectious model of HIV and Schistosoma pulmonary co-exposure based on lung embolisation of S. mansoni eggs in HIV-transgenic (HIV) mice, we previously reported exacerbated
