AA in Obesity and Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) Nonalcoholic Fatty Liver Disease (NAFLD).

Obesity and MASLD (NAFLD) are chronic, low-grade systemic inflammatory diseases resulting from complex interactions among diet, adipose tissue, gut microbiota, liver, muscle, pancreas, and the brain. Central to this integrative framework are AA and other polyunsaturated fatty acids (PUFAs), which regulate inflammation, insulin sensitivity, neuroendocrine signaling, and energy homeostasis.Excess consumption of energy-dense foods rich in saturated and trans fats leads to adipocyte hypertrophy, altered membrane fluidity, increased antigenicity, and enhanced recruitment of macrophages and T cells

AA-Induced Lipid Peroxidation at the Center of Tumor Cell Apoptosis and Ferroptosis.

Polyunsaturated fatty acids (PUFAs), especially gamma-linolenic acid (GLA), arachidonic acid (AA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), can block HMG-CoA reductase activity and thus decrease cholesterol synthesis/formation. Cholesterol has antioxidant actions and thus reduces the formation of ROS (reactive oxygen species). In contrast, PUFAs augment the generation of ROS. PUFAs undergo peroxidation to form lipid peroxides that are toxic to tumor cells but not normal cells. GPX4 (glutathione peroxidase) reduces the formation of lipid peroxides and thus enhances tumor cel

Multiparametric Flow Cytometry Analysis of Naïve, Memory, and Effector T-Cells.

Polychromatic flow cytometry enables the detection and characterization of markers that are helpful in defining the phenotype of various cell subsets. Here, we described a flow cytometry-based method to characterize the phenotype of naïve, memory, and effector T cells. Being able to differentiate these cells is crucial in understanding immune response and immune profiling. Naïve T cells enable the body to fight off new, unrecognized infections and diseases, and Memory T cells are enriched for response to recall antigens. Furthermore, the antigen-experienced T-cell populations can be broadly di

Enumeration of Benign and Malignant Plasmacytoid Dendritic Cells in Peripheral Blood and Bone Marrow by Flow-Cytometric Analysis.

Dendritic cells (DCs) are powerful antigen-presenting cells involved in regulating immune response. Plasmacytoid dendritic cells (pDCs) are a subtype of DCs present in small quantities in the bone marrow, peripheral blood, and lymph nodes. They are an important component of our immune system's regulation and response to diseases. They activate T cells and play a critical role in immune tolerance. In this chapter, we review the immunophenotypic features of pDCs and provide a practical protocol for pDCs enumeration in the peripheral blood and in bone marrow samples.

Immunophenotyping of Human Regulatory T-Cells from Peripheral Blood Mononuclear Cells.

Regulatory T cells, or Tregs, are a unique type of CD4⁺ T cell that act as the immune system's brakes-they stop it from turning against the body's own tissues. For about twenty years now, researchers have been digging into what sets Tregs apart from other T cells, both in their composition and function (1). Markers like CD25 (IL-2Rα), Foxp3, and CTLA-4 usually show up when Tregs are active, but these markers aren't always reliable-they can shift depending on where the cells are and what they are doing. Foxp3 is a transcription factor that Tregs need to develop and function. Mutations on Foxp3

Immunophenotyping of Cellular Therapy Products Against Oncogenic and Viral Targets.

Flow cytometric analysis can provide valuable information on the viability, phenotype, and therapeutic potential of cell therapy products using a minimal amount of cell numbers. In this chapter, we describe methods for the analysis of clinical products that incorporate extra considerations within the protocols to help maintain consistency and confidence of staining. This can include methods for evaluating the transduction efficiency for T cells expressing chimeric antigen receptors (CAR-T), characterizing the frequency of diverse lymphocyte populations within polyclonal cell cultures and posti

Nucleofection-Based CRISPR/Cas Delivery in Human T Cells for Immunotherapy Applications.

Electroporation-based delivery of CRISPR/Cas systems has emerged as a powerful and versatile approach for gene editing in primary human T cells, enabling efficient, transient, and nonviral modification while minimizing genomic integration risks. This chapter focuses on the principles and practical implementation of electroporation (nucleofection) for the delivery of Cas9 ribonucleoprotein (RNP) complexes into human T cells, highlighting critical parameters that influence editing efficiency, cell viability, and scalability for research and clinical applications. We provide a comprehensive proto

Mechanisms underlying rheumatic heart disease immune-mediated pathogenesis: T-cell responses, cytokine networks, and immune dysregulation as drivers of valvular injury.

Acute rheumatic fever is an immune-mediated sequela of Streptococcus pyogenes (group A Streptococcus) infection, presenting with fever, arthritis, chorea, rash, and, crucially, carditis. Without effective secondary prophylaxis and care, acute rheumatic fever can progress to chronic rheumatic heart disease, a leading yet under-recognised cause of cardiovascular morbidity and death that disproportionately affects children and young adults aged 5-24 years in low-resource settings. Robust evidence implicates the role of antigenic mimicry-particularly between streptococcal M protein and cardiac or

Quantification of Human T Cell Responses to Chlamydia trachomatis: Intracellular Cytokine Staining (ICS) and Activation-Induced Marker (AIM) Assays.

High-resolution characterization of antigen-specific T cell responses is essential for understanding cellular immunity to Chlamydia trachomatis infection in humans. This chapter describes two complementary flow cytometry-based approaches for measuring human antigen-specific T cell responses: intracellular cytokine staining (ICS) and activation-induced marker (AIM) staining. Both assays enable single-cell-level analysis following antigen stimulation but differ in their readouts. ICS quantifies functional cytokine production in response to short- or long-term stimulation, whereas the AIM assay i

Human MAITregs possess diverse TCR repertoires and are functionally supported by glycolysis.

Mucosal-associated invariant T (MAIT) regulatory cells (MAITregs) represent a specialized subset with immunosuppressive functions, yet their properties and molecular basis are largely unknown. We demonstrate that MAITregs, while sharing T cell receptor (TCR) repertoires with conventional MAIT cells, undergo selective clonal expansion during in vitro generation, leading to biased V(D)J profiles and restricted CDR3 diversity. Moreover, integrated transcriptomics revealed that MAITregs preferred glycolysis, which was supported by chromatin remodeling at glycolytic gene loci. Functionally, glycoly

INTRODUCTION: Eosinophilic esophagitis (EoE) is a chronic, antigen-driven inflammatory disease characterized by esophageal dysfunction and tissue remodeling. Despite increasing prevalence, its pathogenesis remains incompletely understood. This review aims to systematically synthesize current evidence on the immunological, genetic, epithelial, and extracellular matrix mechanisms driving EoE. The goal is to identify upstream molecular targets and biomarkers to support precision medicine approaches. METHODS: A PRISMA-based systematic literature search was conducted in PubMed, Web of Science, and

Insufficient ferroptosis in radiation-induced tumor cells promote Treg cell generation to induce radiotherapy resistance.

Recently, cellular mechanisms underlying radioresistance in the tumor microenvironment (TME) attract increasing attention, but how the resistant tumor cells reshape the TME is still unclear. In this study, we find that the ferroptosis-resistant genes are upregulated in radioresistant tumor cells, resulting in the impairment of radiation-induced ferroptosis in radioresistant cells. Additionally, the release of transforming growth factor beta 1 (TGF-β1) is increased in radioresistant tumor cells, contributing to Treg cell infiltration in radioresistant tumor tissues. Hence, the ferroptosis induc

CXCR2 blockade potentiates HDAC3 deletion mediated antitumor immunity in colorectal cancer.

HDAC3 is highly expressed in colorectal cancer (CRC) and associated with poor prognosis. This study reveals that HDAC3 deletion dually regulates chemokine expression through epigenetic mechanisms, promoting CD8⁺ T cell infiltration (via CXCL10) on one hand, while enhancing the recruitment of myeloid-derived suppressor cells (MDSCs) (via CXCL1/2/3/5) on the other. The latter, mediated through CXCR2 signaling, counteracts the intrinsic anti-tumor effect of HDAC3 deletion. We further demonstrate that combining a CXCR2 inhibitor effectively blocks MDSC infiltration and significantly enhances the i

Enhancing tumor T cell antigen prediction by integrating deep protein representations.

BACKGROUND: Cancer is a major threat to human health, and cancer immunotherapy is considered the most promising treatment option. It offers high effectiveness and precision, with fewer side effects than traditional treatments. Tumor T cell antigens (TTCA) are the proteins or protein fragments that stand on the surface of cancer cells and that are recognized by the immune system. However, physical experimental approaches to infer them might be costly and time-consuming, even though they are important for cancer immunotherapy. RESULTS: We propose DEEPTHYBRID to predict TTCAs by combining chemic

VSIG3 preserves mitochondrial homeostasis and restrains CD4⁺ T-Cell infiltration in myocardial ischemia-reperfusion injury through regulation of mitochondrial-derived vesicles.

Myocardial ischemia-reperfusion (MI/R) injury remains an inevitable and severe clinical challenge during cardiac surgery, primarily characterized by mitochondrial dysfunction and robust CD4+ T cell infiltration. In this study, we investigated the therapeutic potential of the immune checkpoint molecule VSIG3 (V-set and transmembrane domain-containing protein 3) in mitigating MI/R injury. In clinical samples, ELISA-detectable VSIG3-related plasma immunoreactivity of an undefined molecular form was associated with myocardial injury and inflammation. Recombinant VSIG3 administration improved cardi

Bioinformatics identification of candidate biomarkers associated with T cell proliferation in atherosclerosis via WGCNA and machine learning.

BACKGROUND: Atherosclerosis (AS) is a complex chronic disease caused by the development of atherosclerotic plaques. T cell proliferation exerts a vital influence on development of the AS. This research aimed to conduct a comprehensive analysis to computationally screen candidate T cell proliferation-related biomarkers in AS and to explore their potential molecular mechanisms. METHODS: The transcriptional datasets of AS patients were obtained from the Gene Expression Omnibus (GEO) repository. To identify differentially expressed-T cell proliferation-related genes (DE-TPRGs), we integrated diff

An updated patent review of STAT6 inhibitors and modulators (2020 - present).

INTRODUCTION: Signal Transducer and Activator of Transcription 6 (STAT6) is a critical transcription factor in the interleukin-4 (IL-4) and IL-13 signaling pathways and serves as a key pathogenic driver of type 2 (TH2) immune responses. Dysregulated STAT6 activity is implicated in asthma, atopic dermatitis, autoimmune diseases, eosinophilic disorders, and certain cancers. As such, targeting STAT6 presents a promising therapeutic strategy for these diseases. AREAS COVERED: This review provides a comprehensive analysis of patent strategies for targeting STAT6 from 2020 to the present. We catego
